The phenomenon of antibody is dependent on the cell in which occurs the main mechanism of action of all targeted antitumor preparations containing monoclonal or polyclonal antibodies. Successful manifestation of this phenomenon is possible only with the correct ratio between the antigens of cancer cells, antibodies specific to them and the required amount of complement. The following paper shows the need to monitor the quantitative content of complement in the patient's serum and correct its content by introducing the required volume of fresh frozen plasma as a source of exogenous complement. The required amount of specific antibodies (dosage of the drug) must be determined after proper correction of the amount of complement in the patient's blood. If this condition is met, the maximum efficacy of the drug and the increased success of the treatment of tumor diseases will be achieved.
The electrophoretic mobility (EPM) of cells from rat ovary carcinoma lines differring in ploidity and metastatic capacity has been studied. It was shown that the reduction of EPM of metastatic cells after treatment with hyaluronidase and alcian blue (AB) sorption was 2-3 times as much as the nonmetastatic cells. The data obtained by scanning electron microscopy have shown marked differences in cell surface microstructure of the metastatic and nonmetastatic cells after treatment with hyaluronidase and AB.
Changes in concentrations of protein- and lipid-associated sialic acids (PASA, LASA) in the blood serum, ascitic fluid and cells of clonal lines in ovary tumor of rats have been studied. A considerable increase of PASA and LASA concentrations in serum as against the standard has been found. The concentrations correlate with those in the ascitic fluid, but not in the tumor cells. Maximum frequency of the circulating PASA and LASA concentrations was determined in rats with polyploid unmetastatic tumors, and minimum - in animals with metastatic ovary tumors irrespective of the tumor ploidity level. Possible relation between concentration of circulating sialoconjugates and tumor cell composition is under discussion.
The relation was studied between the density of the electrical charge of the surface (DECS) and the karyological structure of the rat ovary tumour (OT) cells. For this purpose the sedimentation rate and electrical mobility of cells from two OT clones were measured. The OT1 and OT2 clones differed in their modal chromosome numbers (44 and 80-90, respectively). An increase in DECS by 4-5% on the average occurs in cells with the chromosome set close to the tetraploid one. The further increase in the chromosome number to the octaploid one does not change DECS. The results obtained show stability of DECS with significant changes in the structure of the OT cells karyotype.
