Physical inactivity and aging cause significant impairment of skeletal muscle functionality and mechanical properties, as well as remodeling of the extracellular matrix (ECM). The purpose of this work was to study the effect of chronic decrease in physical activity and age on the biogenesis of ECM in skeletal muscle. Biopsy samples from m. vastus lateralis were taken for quantitative mass spectrometry-based proteomic analysis and RNA sequencing in 15 young healthy volunteers and 8 young and 37 elderly patients with long-term primary osteoarthritis of the knee/hip joint as a model to study the effects of chronic muscle motor decline. In total, 1022 mRNAs and 101 ECM proteins and ECM-associated proteins (matrisome) were detected. An increase in the expression of two dozen highly abundant matrisome proteins specific to elderly and young patients (relative to young healthy subjects) was identified; however, changes in the expression of mRNAs encoding matrisome regulators (enzymatic regulators and secreted proteins) were similar. Comparison with previous proteomic and transcriptomic data showed that the described matrisome changes were markedly different from those induced by aerobic physical training in young healthy individuals, in particular in the expression of dominant ECM proteins and, especially, in the expression of mRNAs of ECM enzymatic regulators and secreted proteins. Matching the expression profiles of these regulatory genes may be useful for finding pharmacological targets for preventing adverse changes/activation of ECM biogenesis in various pathological conditions/physical training.
This study investigated the effect of knockout of six Hsp70 genes (orthologues of the mammalian genes Hspa1a, Hspa1b, Hspa2, and Hspa8) on age-related changes in gene expression in the legs of Drosophila melanogaster, which contain predominantly skeletal muscle bundles. For this, the leg transcriptomic profile was examined in males of the w(1118) control strain and the Hsp70(-) strain on the 7th, 23rd and 47th days of life. In w(1118) flies, an age-related decrease in the locomotion (climbing) speed (a marker of functional state and endurance) was accompanied by a pronounced change in the transcriptomic profile of the leg skeletal muscles, which is conservative in nature. In Hsp70(-) flies, the median lifespan was shorter and the locomotion speed was significantly lower compared to the control; at the same time, complex changes in the age-related dynamics of the skeletal muscle transcriptome were observed. Mass spectrometry-based quantitative proteomics showed that 47-day-old Hsp70(-) flies, compared with w(1118) flies, demonstrated multidirectional changes in the contents of key enzymes of glucose metabolism and fat oxidation (glycolysis, pentose phosphate pathway, Krebs cycle, beta-oxidation, and oxidative phosphorylation). Such dysregulation may be associated with a compensatory increase in the expression of other genes encoding chaperones (small Hsp, Hsp40, 60, and 70), which regulate specific sets of target proteins. Taken together, our data show that knockout of six Hsp70 genes slightly reduced the median lifespan of flies, but significantly reduced the locomotion speed, which may be associated with complex changes in the transcriptome of the leg skeletal muscles and with multidirectional changes in the contents of key enzymes of energy metabolism.
ЦЕЛЬ: скелетные мышцы играют ключевую роль в регуляции углеводно-жирового обмена, а также в развитии метаболических нарушений в организме. При ожирении и особенно при сахарном диабете 2 типа (СД2Т) происходят выраженные изменение транскриптома скелетных мышц человека. Однако транс- криптомный ответ в скелетных мышцах на инсулин охарактеризован плохо, как и механизмы регуляции экспрессии инсулинозависимых генов. МАТЕРИАЛЫ И МЕТОДЫ: мы исследовали ранний транскриптомный ответ скелетных мышц людей на инсулин, индуцированный приемом смешанной пищи. В исследовании участвовали 8 здоровых людей и пациенты с ожирением с (n=8) и без СД2Т (n=8); биопсии из m. vastus lateralis были взяты до и через 1 ч после приема еды, нормированной на массу тела (Nestle Resource 2.0; 3 мл/кг массы тела). Пациенты были разделены на две группы: с сильным и слабым ответом С-пептида (и инсулина) на пищу. Как и ожидалось, вторая группа в основном состояла из людей с ожирением и СД2Т. Изменение транскриптомного профиля в мышце оценивали с помощью РНК секвенирования (NextSeq 550, Illumina). РЕЗУЛЬТАТЫ: анализ транскриптома показал, что у здоровых людей, несмотря на слабое повышение уровня С-пептида после приема пищи, несколько сотен генов изменили экспрессию. Используя метод позиционно-весовых матриц и наши данные по положению открытого хроматина вокруг стартов транс- крипции в скелетных мышцах человека (Makhnovskii et al. 2022), мы предсказали транскрипционные факторы, ассоциированные с генами, изменившими экспрессию. Примечательно, что пациенты со слабой реакцией С-пептида на прием пищи фактически не показали изменений в транскриптомном профиле, что указывает на наличие резистентности к инсулину с точки зрения экспрессии генов. Удивительно, но у паци- ентов с сильной реакцией С-пептида на прием пищи также было обнаружено лишь небольшое изменение транскриптомного профиля. Это означает, что резистентность к инсулину в регуляции экспрессии генов появляется на начальной стадии развития метаболических нарушений. ВЫВОДЫ: таким образом, наше исследование впервые показало, что развитие метаболических на- рушений у человека может быть связано с дефектом регуляции инсулин-зависимой генной экспрессии в скелетной мышце.
Гипертрофический эффект силовых физических упражнений связывают с влиянием как механических, так и метаболических стимулов. При использовании упражнений в условиях ограниченного кровоснабжения работающих мышц, то есть в условиях, когда метаболические сдвиги усиливаются, тренировочный эффект может достигаться при использовании гораздо меньших внешних отягощений (20% максимальной произвольной силы (МПС)). Были сопоставлены эффекты 8-недельной обычной высокоинтенсивной (8085% МПС) силовой тренировки и тренировки с низкой интенсивностью (50% МПС) без расслабления работающих мышц. Оказалось, что обычная тренировка приводит к несколько большему приросту силовых возможностей и размеров тренируемых мышц, чем тренировка без расслабления. При обычной тренировке происходит преимущественное увеличение площади, занимаемой на срезе мышечными волокнами II типа, а при тренировке без расслабления волокнами I типа. Упражнения без расслабления вызывают более выраженное увеличение секреции в кровь гормона роста, инсулино-подобного фактора роста-1 и кортизола. После обычных силовых упражнений и упражнения без расслабления происходит различное изменение экспрессии гена-регулятора миогенеза миостатина. Силовая тренировка (50% МПС) без расслабления эффективное средство для роста силы и гипертрофии мышц, которое может найти применение в восстановительной медицине, спорте и физической культуре, как тренировочный режим с относительно низкими нагрузками.
Hypertrophic effect of strength training is known to originate from mechanical and metabolic stimuli. During exercise with restricted blood supply ofworking muscles, that is under conditions of intensified metabolic shifts, training effect may be achieved with much lower external loads (20% of one repetition maximum (1 RM)). The aim of the study was to compare the effects of 8 wks high-intensity (80-85% MVC) strength training and low-intensity (50% 1 RM) training without relaxation. The high-intensity strength training leads to somewhat higher increments in strength and size of trained muscles than training without relaxation. During high-intensity training an increase of area occupied by type II fibers at muscle cross section prevails while during training without relaxation - an increase of area occupied by type I fibers takes place. An exercise session without relaxation leads to a more pronounced increase in secretion of growth hormone, IGF-1 and cortisol. Expression of gene regulating myogenesis (Myostatin) is changed in different ways after high-intensity strength exercise session and after exercise session without relaxation. Low-intensity strength training (50% 1 RM) without relaxation is an effective way for inducing increases of strength and size of trained muscles. This low intensive type of training may be used in restorative medicine, sports and physical culture.
