Equipment on the head of the horse has the potential to affect horse behavior, learning ability, and wellbeing. The therapeutic riding (TR) horse is usually ridden by a participant while being led by a horse leader, which presents a unique situation for equipment used on the head of the horse. The purpose of this study was to describe the types of equipment used on the heads of TR horses in the United States. An anonymous online survey was distributed to certified TR instructors through social media and email listservs, ultimately resulting in 403 complete responses. Summary statistics were calculated, and open-responses were category-coded. The types of equipment used most often were bitted bridles (75.9%), traditional halters (75.2%), and sidepull bitless bridles (54.6%). Respondents used halters alone (65.9%), placed halters under bridles (43.6%), and placed halters over bridles (11.1%). Most respondents (75.9%) attached the lead rope to the bottom ring of the halter, though 23.8% of respondents used a tie ring on the underside of the bitted/bitless bridle noseband. Respondents attached reins to the halter square side buckles (60.1%), to the bit (56.7%), to bitless bridle rings (53.8%), or to halter jowl rings (31.6%). Overall, many prevalent equipment use practices were atypical of designed uses, or of the rest of the horse industry. Further research should examine the effect of such unique equipment practices.
OBJECTIVE:To determine whether dexmedetomidine prolongs corneal anesthesia following subconjunctival ropivacaine injection in healthy equine eyes. METHODS:Ten horses with normal ophthalmic exams were enrolled in a randomized, blinded, crossover experiment with a minimum 4-week washout between treatments. Baseline corneal touch thresholds (CTT, cm) were measured via esthesiometry and a subconjunctival injection was performed in the treatment eye with ropivacaine (R, 0.3 mL 0.5%) or dexmedetomidine-ropivacaine (DR, 0.1 mL 0.5 mg/mL + 0.2 mL 0.5%) and saline (control, 0.3 mL) in the contralateral eye. CTT was measured every 10 min up to 120 min, every 30 min up to 360 min, and every 60 min thereafter until return to baseline. Ambient humidity was concurrently recorded. Subconjunctival hemorrhage was scored 0, 2, and 7 days after injection. CTT values were converted to corneal pressure tolerance (CPT, g/mm2) using humidity-adjusted exponential regression. Data were compared between treatments using mixed model analysis (significance p < 0.05). RESULTS:Mean ± standard deviation total anesthesia time was significantly longer with DR (196 ± 121 vs. 103 ± 47 min R, p = 0.02). Evidence of significant differences between treatments was not seen for minimum CTT, duration of minimum CTT, maximal CPT, or hemorrhage scores. However, time to minimum CTT was longer for DR (32 ± 12 vs. 16 ± 10 min R, p < 0.001). Subconjunctival hemorrhage resolved within 7 days of injection. CONCLUSIONS:Dexmedetomidine prolongs corneal anesthesia from subconjunctival ropivacaine in horses but delays the onset. Further studies are needed to assess efficacy in diseased eyes.
Purpose: High cancer-related fatigue (CRF) and low energy are commonly reported by survivors of cancer both during and following treatment(s) and can be alleviated by regular exercise. However, the immediate or "acute" effects of exercise for improving CRF and energy among survivors of cancer is not well understood. This study examined changes in CRF and energy from immediately before to immediately after exercise sessions during an 8-week videoconference exercise program. Methods: Videoconference exercise sessions were held 1x per week and consisted of light to moderate intensity aerobic and resistance exercise. Energy and CRF were self-reported immediately before and after each exercise session using a visual analog scale (0 = low, 10 = high). Separate linear mixed models were used for CRF and energy, with time (pre/post), treatment status (ie, receiving chemotherapy at the time of the program) and time x treatment interaction included as fixed effects. Results: Participants (N = 55) were M = 59 +/- 9.94 years old, 96% female, with ovarian (61.8%), breast (30%), or other (7.2%) cancer, and 27% were receiving chemotherapy. There was a main effect of time for CRF [F(1, 380) = 18.96, P < .01; M Delta = - 0.63 +/- -0.04], and for energy [F(1,380) = 93.91, P < .01; M Delta = 1.16 +/- .001], but no effect of treatment status. Conclusion: Regardless of treatment status, CRF and energy improved from immediately before to after exercise sessions during an 8-week videoconference exercise program. Future research is needed to determine how to leverage the positive, acute effects of exercise to enhance exercise motivation and adherence, and to understand how exercise intensity may impact CRF and energy immediately following exercise.
The objectives of this study were 1) to understand stakeholder perceptions regarding the bison industry, 2) to benchmark live animal characteristics and production parameters of the bison industry, and 3) to identify live animal factors related to animal welfare and their effect on specific quality characteristics of bison meat. A survey was conducted both online and in-person at the National Bison Association Winter Conference (2022). Descriptive statistics were performed on a total of 110 surveys. Most stakeholders (94%, n = 104) agreed that the industry should continue to grow, and the majority (99%, 108) agreed that animal welfare impacts meat quality. Facility design (80%, n = 88), animal handling (78%, 86), employee training (56%, 62), and transportation duration (56%, 62) were selected as the factors that affect animal welfare. More than half of the stakeholders selected flavor (67%, n = 74) as the most important quality attribute of bison meat. For the in-plant antemortem and postmortem parameters data was collected from three plants in the United States over the course of a year. A total of 2,284 bison (bulls: n = 1,101; cows: n = 199; heifers: n = 984) were included in the study. Antemortem measurements such as distance traveled, vocalization, prod use, mobility, and head bumps were measured, followed by postmortem measurements that included bruise score, live weight, dressing percentage, ribeye area, and instrumental color. Approximately 97% of bison (n = 2,213) had at least one bruise. The average distance traveled from producer to slaughter plant was (mean +/- SD; 823 +/- 583 km) and the average dressing percentage was (mean +/- SD; 60.5 +/- 3.3%). Average (mean +/- SD) fat thickness and ribeye area were 1.4 +/- 1.1 cm and 62.6 +/- 9.8 cm2, respectively. Approximately 30% (n = 676) of the bison in this study head bumped between 1 and 5 times in the restraining chute or the single-file gate before being stunned. Linear regression indicated that differences in lean a* were associated with plant, number of head bumps in the chute, ribeye area, fat thickness, live weight, and sex class (P < 0.05). Logistic regression indicated that season, sex class, live weight, plant, and season were associated with differences (P < 0.05) in bruising. These results can be used as a baseline for current production parameters and serve as the foundation for future research to monitor improvement.
During the preslaughter phase, cattle are transported from their place of origin to a slaughter facility, experiencing transportation, lairage, environmental factors, and novel environments. Although research exists that has focused how the preslaughter phase impacts cattle welfare and meat quality, some significant preslaughter management factors and subsequent welfare and meat quality outcomes have not been thoroughly explored. The objective of this study was to assess the effects of preslaughter management factors on welfare and meat quality outcomes in fed beef cattle in the United States. Transportation factors, environmental characteristics, lairage factors, cattle characteristics, and several meat quality variables were collected from 5 federally inspected commercial processing facilities in the United States. After excluding slaughter lots that included <75% complete datasets, a total of 619 slaughter lots representing 84,508 head of cattle were used for further analysis. Predictor variables of interest included processing plant, cattle breed, sex class, operation shift at the plant, distance traveled to the plant, truck waiting time to unload at the plant, lairage duration and space allowance, temperature humidity index, and windspeed. Outcome variables of interest included cattle mobility, carcass bruising, dark cutting (DC), quality grades, and hot carcass weights. Logistic and linear regressions were used to analyze the associations between the predictor and outcome variables of interest. Increased distance traveled and truck waiting time were associated with higher odds of mobility impairment (P = 0.0009 and P = 0.007, respectively), with each 10 km increase in distance traveled having an odds ratio (OR) of 1.001 (95% confidence interval [CI]: 1.000 to 1.001) and each 1-min increase in waiting time having an OR of 1.003 (CI: 1.001 to 1.004). Conversely, a 10-km increase in distance traveled decreased the odds of carcass bruising (OR: 0.997, CI: 0.996 to 0.998; P < 0.0001). Longer lairage was associated with increased odds of DC (P = 0.0415), with each 60-min increase in duration having an OR of 1.034 (CI: 1.001 to 1.068). The results demonstrate the importance of truck arrival management (i.e., scheduling, prioritizing unloading) on mobility. Focusing on lairage management (i.e., density and time) may provide some opportunities to improve meat quality.
Animal welfare is an integral component of beef cattle slaughter, with regulations, industry guidelines, and company-specific programs ensuring standards are met to minimize stress and discomfort during the preslaughter marketing process. This marketing process involves many inherent stressors, including various environmental and human factors, which may have negative impacts on cattle welfare and meat quality. This research aimed to investigate the relationship between several preslaughter factors (i.e., distance traveled to the plant, truck waiting time at the plant, space allowance and time in holding pens, and season) on physiological stress parameters, mobility, bruising, and dark cutting in a population of finished beef cattle at one commercial slaughter facility in the Southwestern United States. Results of the study indicate that truck waiting time, space allowance, and time spent in holding pens are associated with physiological stress parameters, cattle mobility, and carcass bruising. Additionally, the summer season is associated with negative animal welfare outcomes. This data underscores the importance of the preslaughter period on cattle welfare and meat quality and identifies areas for continuous improvements such as reducing truck waiting times, shortening the time cattle spend in pens, and providing more space for cattle at the plant.
OBJECTIVE:The objective of this study was to use shotgun label-free tandem mass spectrometry (LF-MS/MS) to evaluate aqueous humor (AH) from horses with uveitis (UH) compared to ophthalmologically healthy horses (HH).ANIMALS STUDIED:Twelve horses diagnosed with uveitis based on ophthalmic examination and six ophthalmologically healthy horses (postmortem) purchased for teaching purposes.PROCEDURES:All horses received a complete ophthalmic examination and physical exam. Aqueous paracentesis was performed on all horses and AH total protein concentrations were measured with nanodrop (TPn) and refractometry (TPr). AH samples were analyzed with shotgun LF-MS/MS and proteomic data were compared between groups using Wilcoxon rank-sum test.RESULTS:A total of 147 proteins were detected, 11 proteins had higher abundance in UH, and 38 proteins had lower abundance in UH. Proteins with higher abundance included apolipoprotein E, alpha-2-macroglobulin (A2M), alpha-2-HS-glycoprotein, prothrombin, fibrinogen, complement component 4 (C4), joining chain for IgA and IgM, afamin, and amine oxidase. There were positive correlations between TPn (p = .003) and TPr (p = .0001) compared to flare scores.CONCLUSION:Differential abundance of A2M, prothrombin, fibrinogen, and C4 indicate upregulation of the complement and coagulation cascade in equine uveitis. Proinflammatory cytokines and the complement cascade have potential as therapeutic targets for equine uveitis.
Preslaughter management factors and their impacts on cattle welfare and meat quality are well documented in current literature. However, certain management factors related to transportation and lairage are underrepresented. Benchmarking preslaughter management factors that can impact welfare and meat quality outcomes will allow the industry to identify areas for improvement. The objective of the current study was to benchmark preslaughter management factors for a nationwide sample of commercial fed cattle processing facilities. Five processing facilities in the West, Midwest, and Southwest regions of the United States were sampled from March 2021 to July 2022. Data were collected on a total of n = 637 slaughter lots representing n = 87,220 head of cattle. Variables of interest included general cattle characteristics, distance traveled to the plant, truck wait times to unload, environmental conditions, lairage density, and lairage duration. Additionally, mobility was scored using a four-point locomotion scale (one being normal, not lame, and four being extremely reluctant to move). Carcasses were also observed for bruising using the following scale: no bruises, bruises smaller or larger than a deck of cards, and having multiple bruises. Descriptive statistics were performed on the data at the lot and individual animal level. On average, cattle traveled 155.8 +/- 209.6 km (mean +/- SD) from the feedlot to the processing facility and waited 30.3 +/- 39.7 min to unload. Once in lairage pens, cattle were held for 200.7 +/- 195.0 min. The mean lairage stocking density was 3.1 +/- 2.0 m2 per head. A majority of the cattle scored a mobility score of 1 (n = 77,645, 91.8%), 7.8% (n = 6,125) were scored as a two and the remaining less than one percent of cattle were scored as either a 3 or 4 (n = 265). Carcasses with bruises less than or equal to the size of a deck of cards (n = 22,672, 27.1%) were less frequent than bruises measuring greater than the size of a deck of cards (n = 34,427, 42.6%). Of carcasses that were bruised, 65.2% (n = 39,856) had multiple bruises of varying size. This baseline data on preslaughter management factors identifies opportunities for improvement in wait times, lairage densities, and factors that cause bruising. Future studies should explore the relationships between these factors and their impacts on welfare and meat quality, report the economic value of these outcomes, and explore industry acceptability and adoptability of optimal preslaughter management practices.
OBJECTIVE To determine if tissue oxygen saturation (StO 2 ) correlates with oxygen delivery (DO 2 ) and/or cardiac output (CO) in a canine hemorrhagic shock model. ANIMALS 8 healthy purpose-bred dogs. METHODS Dogs were anesthetized, and hemorrhagic shock was induced by withdrawing up to 60% of total blood volume, targeting a mean arterial pressure (MAP) of 40 mm Hg. The withdrawn blood was returned to the patient in 2 equal aliquots. Data was collected at 4 time points: 10 minutes after MAP was stabilized under anesthesia (time point [TP]-1), 10 minutes after up to 60% of blood volume was removed to target a MAP of 40 mm Hg (TP2), 10 minutes after the return of 50% of shed blood (TP3), and 10 minutes after the return of the remaining 50% of shed blood (TP4). Total blood volume withdrawn, StO 2 , CO, heart rate, and MAP were recorded, and DO 2 was calculated at each TP. RESULTS Mean StO 2 significantly decreased between TP1 (77.8% [± 9.54]) and TP2 (44.8% [± 19.5]; P < .001 vs TP1). Mean StO 2 increased to 63.1% (± 9.85) at TP3, but remained significantly lower compared to TP1 ( P = .002). There was no difference between mean StO 2 at TP4 (82.5% [± 12.6]) versus TP1 ( P = .466). StO 2 has a strong, positive correlation to both CO ( r = 0.80; P < .001) and DO 2 ( r = 0.75; P < .001). CLINICAL RELEVANCE A decrease in StO 2 may be used in conjunction with physical examination findings and diagnostic parameters to support a diagnosis of shock. The return of shed blood was correlated with increases in StO 2 , DO 2 , and CO, suggesting that StO 2 may be used as a marker of adequate resuscitation.
OBJECTIVE:To determine whether shock index (SI) positively correlates with percentage blood loss and negatively correlates with cardiac output (CO) in a canine hemorrhagic shock model and whether SI and metabolic markers may be used as end point targets for resuscitation.ANIMALS:8 healthy Beagles.PROCEDURES:Between September and December 2021, dogs underwent general anesthesia for experimental induction of hypotensive shock, with the total volume of blood removed, CO, heart rate, systolic blood pressure, base excess, blood pH, and concentrations of hemoglobin, lactate, ionized calcium recorded, and SI calculated at 4 time points (TPs): after anesthetic induction when the dog had been stable for 10 minutes (TP1), 10 minutes after the mean arterial pressure stabilized to a target of 40 mm Hg following jugular removal of up to 60% blood volume to induce hemorrhagic shock (TP2), 10 minutes after autotransfusion of 50% of the removed blood (TP3), and 10 minutes after autotransfusion of the remaining 50% of the removed blood (TP4).RESULTS:Mean SI increased between TP1 (1.08 ± 0.35) and TP2 (1.90 ± 0.73) and did not return to the prehemorrhage values for TP3 or TP4. SI correlated positively with percentage blood loss (r = 0.583) and negatively with CO (r = -0.543).CLINICAL RELEVANCE:An increase in SI may support diagnosis of hemorrhagic shock; however, SI cannot be used as the sole end point of resuscitation. Significant differences in blood pH, base excess, and lactate concentration suggested they may be useful markers of hemorrhagic shock and need for blood transfusion.
Objective: Develop reliable, valid questions to assess changes in food resource management (FRM) behav-iors in adults with limited incomes. Design: Questionnaire development using a mixed-methods approach: content validity (subject matter and curricula), face validity, temporal reliability (test-retest), sensitivity to change, and exploratory factor analy-sis (EFA). Setting: Community settings in 12 states. Participants: Convenience samples of English-speaking Expanded Food and Nutrition Education Program (EFNEP) or EFNEP-eligible adults: 105 (cognitive interviews), 181 (test-retest), 185 (sensitivity), and 389 (EFA) adults. Variables Measured: Behaviors related to FRM skills: planning, shopping, and budgeting. Analysis: Consistency and agreement in cognitive interviews and temporal reliability; sensitivity at posttest (paired t tests, Wilcoxon signed-rank tests); internal consistency of scales identified in EFA (Cronbach a). P < 0.5. Results: All questions had acceptable temporal reliability ranges for the intraclass correlation coefficient (0.48-0.74) and Spearman rank-order correlation (0.48-0.73). All questions were sensitive to change at posttest (P < 0.001). Planning and saving scales, revealed by EFA, demonstrated internal consistency (> 0.80 Cronbach a). Conclusions and Implications: The 9 FRM behavior questions have acceptable temporal reliability and content and face validity and can be used nationally by EFNEP to assess participants' self-reported behavior changes. Other nutrition programs with similar audiences and content could use these questions to measure changes in FRM behaviors.
OBJECTIVE To evaluate the sedative and cardiopulmonary effects of various combinations of acepromazine, dexmedetomidine, hydromorphone, and glycopyrrolate, followed by anesthetic induction with propofol and maintenance with isoflurane in healthy dogs. ANIMALS 6 healthy adult female Beagles. PROCEDURES Dogs were instrumented for hemodynamic measurements while anesthetized with isoflurane. Two hours after recovery, dogs received 1 of 4 IM combinations in a crossover design with 1 week between treatments: hydromorphone (0.1 mg/kg) and acepromazine (0.005 mg/kg; HA); hydromorphone and dexmedetomidine (0.0025 mg/kg; HD); hydromorphone, acepromazine, and dexmedetomidine (HAD); and hydromorphone, acepromazine, dexmedetomidine, and glycopyrrolate (0.02 mg/kg; HADG). Sedation was scored after 30 minutes. Physiologic variables and cardiac index were measured after sedation, after anesthetic induction with propofol, and every 15 minutes during maintenance of anesthesia with isoflurane for 60 minutes (target expired concentration at 760 mm Hg, 1.3%). RESULTS Sedation scores were not significantly different among treatments. Mean ± SD cardiac index was significantly higher for the HA (202 ± 45 mL/min/kg) and HADG (185 ± 59 mL/min/kg) treatments than for the HD (88 ± 31 mL/min/kg) and HAD (103 ± 25 mL/min/kg) treatments after sedation and through the first 15 minutes of isoflurane anesthesia. No ventricular arrhythmias were noted with any treatment. CLINICAL RELEVANCE In healthy dogs, IM administration of HADG before propofol and isoflurane anesthesia provided acceptable cardiopulmonary function with no adverse effects. This combination should be considered for routine anesthetic premedication in healthy dogs.
ABSTRACT Background Malnutrition and diarrhea are leading causes of death in children aged <5 y. Rice bran is a nutrient-dense prebiotic available globally. Objectives The objective of this secondary analysis was to evaluate the effects of daily rice bran supplementation on environmental enteric dysfunction (EED) markers, total fecal secretory IgA (sIgA), and microbiota in infants at high risk of malnutrition. Methods Six-month-old Malian and Nicaraguan infants were randomly assigned to control or daily rice bran supplementation cohorts (1 to 5 g/d). Feces were collected monthly for 6 mo to evaluate fecal sIgA, markers of EED, and microbiota diversity. Statistical methods included linear mixed models, generalized mixed models, Spearman correlation, and Wilcoxon rank-sum tests. Results Six-month-old Malian infants had significantly elevated sIgA (4.0× higher, P < 0.001), fecal myeloperoxidase (31.6× higher, P < 0.001), fecal α1-antitrypsin (1.8× higher, P = 0.006), and lower fecal neopterin (0.13× higher, P < 0.001) than the age-matched Nicaraguan infants. In the Nicaraguan rice bran cohort from 6 to 12 mo of age, there was a significant decrease in sIgA concentrations (0.4×, P < 0.05) and a correlation between sIgA and the EED marker α1-antitrypsin (0.523, P < 0.0001) at 12 mo of age. In Malian infants, daily rice bran ingestion resulted in decreased EED scores (0.71×, P = 0.02) and a stable sIgA concentration over time. The rice bran group of Malian infants also had correlation between sIgA and the EED marker neopterin (0.544, P < 0.001) at 12 mo of age and a significant (P < 0.05) increase in microbiota α-diversity at a younger age (9 mo with rice bran compared with 10 mo in control group), which supports earlier microbiota maturation. Conclusions These results support rice bran as a functional food ingredient targeting gut mucosa in children at high-risk of malnutrition.
This double-blind, randomized, prospective clinical trial was conducted to obtain exploratory data comparing the efficacy of intra-articular allogeneic mesenchymal stem/stromal cells (MSC) to high molecular weight hyaluronic acid (HA) for the treatment of pain associated with canine osteoarthritis (OA). Objective gait analysis (%Body Weight Distribution, %BWD), accelerometry, clinical metrology instruments and veterinary exams were used as outcome measures during various time points throughout the 48-week study period. Fourteen dogs with elbow or coxofemoral OA were enrolled and assigned in a 2:1 ratio to the treatment groups. Each patient received a set of two injections 4 weeks apart. Self-limiting joint flare was observed in seven patients, with six of these in the MSC group. Ten patients completed all follow-up appointments. Both treatment groups showed evidence of mild improvement following the treatment, but the results were inconsistent among the various outcome measures assessed. Overall, dogs enrolled in the HA group showed greater improvement compared to the MSC group. The primary outcome measure, %BWD, showed evidence of improvement, when compared to baseline values, at 36 weeks after injection for the HA group only (p = 0.048, estimated difference: 4.7). Similarly, when treatment groups were compared, evidence of a difference between treatment groups (with the HA-group showing greater improvement) were identified for weeks 24 and 36 (p = 0.02 and 0.01, respectively). The small sample size of this exploratory study does not allow firm conclusions. However, until studies with larger sample sizes are available, the current literature combined with our data do not support the clinical use of intra-articular MSC therapy over high molecular weight HA for the treatment of canine OA at this time.
Variability in carcass size can influence carcass chilling rates which could result in issues associated with beef tenderness and color. Moreover, the usage of electrical stimulation can affect postmortem metabolism and meat quality. However, few studies have looked at the combined impact of chilling and electrical stimulation on temperature decline, postmortem biochemistry, and color among the current consist of US beef. Beef carcasses (N =162, < 30 months) were randomly selected from two beef processing plants. One side of each carcass was electrically stimulated (ES), while the opposing side was not electrically stimulated (NES). Matched sides were subjected to either conventional spray chilling (CONV) or delayed-spray chilling (DELAY). Deep tissue (10.5-cm under the pelvic bone) and surface temperature (1.5-cm under the loin fat) were continuously monitored during chilling in addition to temperature and pH measurements from the semimembranosus (SM), longissimus lumborum (LL), and psoas major (PM) muscles at an initial time (45 to 60 min), 6 h, 12 h, and after chilling (28 to 36 h) postmortem. Further, the instrumental (L*, a*, and b*) and visual color of the PM were evaluated. For data analysis, carcasses with hot carcass weights above or below the average were considered heavy or light, respectively. A nonlinear regression model was fitted to the continuous deep and surface temperatures, whereas other parameters were evaluated using a mixed model. Electrical stimulation improved (P < 0.05) the color of PM in light weight carcasses, but not (P > 0.05) in heavy weight carcasses. Temperature decline was faster (P < 0.05), and pH decline slower (P < 0.05) in the SM and LL of light weight carcasses under CONV. Exponential decay models for deep and surface temperatures indicated the rate of cooling differed (P < 0.05) due to the combination of treatment factors. Heavy weight carcasses in DELAY had slower rates of temperature decline (P < 0.05). Overall, the variability in carcass size affected temperature decline and postmortem metabolism. Therefore, postmortem management practices should consider carcass weights to ensure homogeneous meat quality.
BACKGROUND:The aim of this study was to evaluate the levels of pro-inflammatory cytokines in aqueous humor (AH) from dogs with anterior uveitis and post-operative ocular hypertension (POH) following phacoemulsification, in AH from dogs with primary glaucoma, and in normal healthy eyes with no signs of anterior uveitis or other ocular diseases.METHODS:An exploratory study including 21 samples of AH collected from 15 dogs; post-phacoemulsification with anterior uveitis and POH ('POH group', n = 10 samples), primary glaucoma ('glaucoma group', n = 6 samples), and normal ('normal group', n = 5 samples). Target mass spectrometry via multiple reaction monitoring (MRM-MS) with the Canine Cytokine SpikeMix™ as internal standard was used to measure the pro-inflammatory cytokine levels.RESULTS:The MRM-MS method measured 15 pro-inflammatory cytokines. Tumor-necrosis-factor-alpha (TNFα) and interleukin-18 (IL-18) levels in AH were different between all three groups (glaucoma>POH>normal) (p = .05, p = .02, respectively). Additionally, IL-6 was higher in the 'POH group' compared to the 'glaucoma group' (p = .04) and IL-4 was higher in the 'POH group' compared to the 'normal group' (p = .04). Intraocular pressure (IOP) was positively associated with increased AH levels of IL-18 (Spearman correlation = .64, p = .03).CONCLUSIONS:MRM-MS using the Canine Cytokine SpikeMix™ as an internal standard was established as a method to detect pro-inflammatory cytokine levels in canine AH. The study demonstrated increased levels of IL-4, IL-6, IL-18, and TNFα in AH from canines with POH following phacoemulsification. Primary glaucomatous eyes had the highest levels of IL-18 and TNFα which may indicate that inflammation plays a role in the pathogenesis of primary glaucoma in dogs.
Kidney tissues from cats with naturally occurring chronic kidney disease (CKD) and adult and senior cats without CKD were assessed to determine whether telomere shortening and nitrosative stress are associated with senescence in feline CKD. The histopathologic assessment of percent global glomerulosclerosis, inflammatory infiltrate, and fibrosis was performed. Senescence and nitrosative stress were evaluated utilizing p16 and iNOS immunohistochemistry, respectively. Renal telomere length was evaluated using telomere fluorescent in situ hybridization combined with immunohistochemistry. CKD cats were found to have significantly increased p16 staining in both the renal cortex and corticomedullary junction compared to adult and senior cats. Senior cats had significantly increased p16 staining in the corticomedullary junction compared to adult cats. p16 staining in both the renal cortex and corticomedullary junction were found to be significantly correlated with percent global glomerulosclerosis, cortical inflammatory infiltrate, and fibrosis scores. p16 staining also correlated with age in non-CKD cats. Average telomere length was significantly decreased in CKD cats compared to adult and senior cats. CKD cats had significantly increased iNOS staining compared to adult cats. Our results demonstrate increased renal senescence, telomere shortening, and nitrosative stress in feline CKD, identifying these patients as potential candidates for senolytic therapy with translational potential.
OBJECTIVE:Using 24-hour dietary recalls, compare Healthy Eating Index (HEI)-2005 scores of Expanded Food and Nutrition Education Program participants before and after 8-12 weekly lessons. DESIGN:Analysis of preexisting 24-hour dietary recalls information collected from October, 2012 through September, 2014. PARTICIPANTS:Participants with complete pre-post dietary data (n = 122,961); subset of those with complete demographic data (n = 97,522). MAIN OUTCOME MEASURES:Change in HEI-2005 scores (total and components). STATISTICAL ANALYSIS:Linear regression model fit separately for total HEI and 12 components. The response variable was changed in the HEI-2005 score; predictor variables included age, education, sex, and race/ethnicity. RESULTS:The mean total HEI scores were 51.1 (SD, 13.7) at entry and 56.5 (SD, 13.7) at exit, with a change of 5.4 (SD, 16.2). Nine of 12 component scores increased. Changes were greater as age increased, with increasing education, and in women. Hispanics had the greatest improvement (mean ± SE) in total HEI score (8.3 ± 0.1). CONCLUSIONS AND IMPLICATIONS:Although diet quality remained poor, participation in the Expanded Food and Nutrition Education Program resulted in improvement in dietary quality. The degree of improvements varied among demographic groups, but all groups improved.
Introduction: Combining an antiplatelet drug, clopidogrel, with the direct oral Factor Xa inhibitor, apixaban, could provide an effective antithrombotic strategy in dogs. Thus, a limited 3 thorn 3 phase I dose-escalation clinical trial in healthy dogs was conducted to evaluate bleeding (primary end-point) and pharmacodynamic (PD)/pharmacokinetic (PK) parameters (secondary end-point). Animals: Eleven beagle dogs, median body weight 10.2 kg (9.7-10.9 kg), were enrolled. Methods: Four doses of apixaban (three dogs/dose) administered for eight days. Clopidogrel dose was fixed at 18.75 mg per os (PO) q 24 h with escalation of apixaban dose at 5 mg PO q 12 h, 5 mg PO q 8 h, 10 mg PO q 12 h, and 10 mg PO q 8 h. Laboratory testing included fecal occult blood, coagulation parameters, Factor X activity, apixaban concentration, platelet aggregometry, and thromboelastography on days 1, 3, and 8. Results: Evidence of bleeding was not observed at any dosage. Dose-dependent changes in PD/PK parameters between baseline and 3 h post-medication were observed including a prolongation of prothrombin time, a prolongation of activated partial thromboplastin time, a decrease of Factor X activity level, and increased apixaban concentration.
