Inhalt: Im Rahmen von Versuchen zur Stimulierung der Superovulation beim Rind mit Serumgonadotropin (PMSG 1500–3100 IE einmalig i.m.) sowie hypophysarem Schweine-FSH (2mal taglich uber 5 Tage jeweils 5 mg i.m.) wurden Hormonprofile fur LH, FSH, PMSG und Progesteron im peripheren Blut sowie konjugkrte astrogene im Urin radioimmunologisch ermittelt. Zwei Tage nach Behandlungsbeginn wurde die Luteolyse mit 0,5 mg Estrumate® i. m. eingeleitet. Die Versuche wurden an 14 Farsen und 2 Kiihen durchgefiihrt. Die Entnahme der Blutproben erfolgte in 6- bzw. 12-stiindlichem Abstand, Die hypophysaren Gonadotropine zeigten keine wesentlichen Unterschiede in den Verlaufskurven verglichen mit denen von normalen zyklischen Tieren, abgesehen davon, dafl der praovulatorische LH- oder FSH-Gipel bei gut reagierenden Tieren eher erfolgte, d. h. mit dem Beginn der Brunst zusammenfiel. PMSG wies nach der einmaligen Injektion eine lange Verweildauer auf und konnte 10 Tage nach der Applikation noch deutlich im peripheren Blut nachgewiesen werden. Wahrend der FSH-Stimulation stiegen die FSH- Werte kaskadenartig an. Die praovulatorischen FSH-Gipfel waren deutlich hoher verglichen mit denen nach PMSG-Stimulation. Bei gut reagierenden Tieren konnten klare Ustrogengipfelwerte vor Brunstbeginn gemessen werden. Die Progesteronprofile von gut reagierenden Tieren unterschieden sich von normalen zyklischen Tieren. Nach dem praovulatorischen LH-Gifpel stiegen die Progesteronwerte aufgrund der grofleren Anzahl von Corpora tutea schneller an und erreichen wesentlich hohere Absolutwerte. Dabei konnten bei fast gleicher Anzahl von C. 1. hohere Absolutwerte nach PMSG-Stimulation gemessen werden. Die Progesteronbestimmung 3 Tage nach Brunstbeginn kann als Entscheidungshilfe fur die Feststellung des Stimulationserfolges h eranger ogen w erden. Ausden Horm onpro film war nich t ersic h tlich, warum einige Tiere gar nicht oder nur schlecht auf die Stimulation reagieren. Contents: Superovulation in cattle: Hormonal profdes during superovulation with PMSG or pituitary FSH.: Hormonal profiles for LH, FSH, PMSG, progesterone in peripheral blood and oestrogens in urine during superovulation with PMSG (1500–3100 IU i. m.) or pituitary porcine FSH (5 mg i.m. 2 times daily for 5 days) were evaluated radioimmunologicaly. Two days after the begin of treatment luteolysis was induced by means of 0.5 mg Estrumat® i. m. The experiments were performed with 14 heifers and 2 cows. Blood samples were taken in 6 or 12 h intervals. Preovulatory LH and FSH peaks occurred coinciding with the onset o f oestrus in good responding animals instead of a few hours later, as in weakly responding animals or during normal oestrous cycle. PMSG could be still measured in peripheral blood 10 days after application. Preovulatory gonadotropin peaks are preceded in good responding animals by clear oestrogen peaks contrary t o weak responding ones. Progesterone concentrations of good responding animals increase faster after the preouulatory LH peak and reach higher absolute values compared t o bad responding ones or during normal oestrous cycle. Furthermore there was a clear difference in progesterone values between both stimulation methods. Progesterone concentrations after induction of superovulation with PMSG are significantly higher f o r about the same number of corpora lutea as after stimulation with pituitary FSH. For judgement of the success of stimulation the determination of progesterone can be considered as parameter. From the hormonal profiles no conclusion can be drawn why some animals don't respond to the stimulation.
ABSTRACT A synopsis is presented of hormonal profiles (peripheral plasma values of LH, FSH, progesterone; urinary oestrogens) during the oestrous cycle of cattle. After ovulation (day one) progesterone levels begin to increase from about day 4, reach a plateau around day 10, and decrease to basal concentrations starting on days 17 to 18. Corresponding with the progesterone decrease, urinary oestrogens show a main increase reaching its maximum one day before ovulation. LH levels are fairly constant from ovulation until day 5, small but significant peaks can be observed between days 6 and 10 in some cows (6 out of 21) and levels then decrease from day 11 to 13. A gradual additional increase begins on day 17, resulting in the preovulatory LH peak 25.7 ± 6.9 h (n = 75) before ovulation. This peak has a duration of 7.4 ± 2.6 h (n = 89) and occurs any time of the day but preferentially around 8 a. m. and 8 p. m. respectively. Likewise related are the two main periods for ovulation. During oestrus, (average duration of 16.9 ± 4.9 h (n = 28) the electrical resistance of the vaginal mucus decreases and the lowest values always correspond with the LHpeak. FSH levels show a wavelike pattern with peaks around days 4, 8, 12–13, 17, 18 and also coinciding with the pre-ovulatory LH peak. The sharp decrease in FSH between the two peaks on days 17 and 18 correlates with the decrease of progesterone during luteolysis. Obviously there is a parallelism between FSH elevations and follicular growth (determined by means of pelviscopic observations).
A simple and reliable method for the determination of progesterone in milk fat by radioimmunoassay has been developed. Application of the assay under field conditions has shown that all samples diagnosed " questionable" in the former milk progesterone test (MPT) could be characterized as positive or negative. Thus the total figure of animals daignosed correctly rose from 72.3% to 85.8%. The borderlines (ng progesterone / 10 mcl milk fat) set up for this assay were (1) pregnant and (1) nonpregnant (Day 20) and (.3) in estrus and (.3) not in estrus ( Day 0). Within the application for diagnosis of pregnancy, 81.4% were diagnosed correctly as pregnant while 96.3% were diagnosed correctly as not pregnant. The latter figure is somewhat lower than in the MPT (100%), but under the practical conditions the error lies within the range of false diagnosis due to problems with sample identification.

Durch intramuskulare Injektion von 0,5 mg des Prostaglandinanalogs ICI 80 996 beim Rind während der Gelbkörperphase des Zyklus kann nach 15–24 Stunden ein Abfall der Progesteronwerte unter 1 ng/ml Blutplasma und damit eine nachhaltige Luteolyse erreicht werden. Die sich anschließende Brunst ist ovulatorisch und feral. Das Präparat wird ohne Nebenwirkungen vertragen. 2. Durch zweimalige Injektion (d. h. je 1 mal 0,5 mg i. m.) im Abstand von 11 Tagen kann dieser Luteolyseeffekt zu Brunstsynchronisationen herangezogen werden; dabei ist eine Zykluskontrolle oder Brunstbeobachtung nicht erforderlich. 3. in 8 Feldversuchen konnte bei 213 Tieren ein Synchronisationserfolg von 90,6 % und, nach KB in der 1. induzierten Brunst, ein Konzeptionsergebnis von 54,7 % (bzw. von 60,1 %, wenn man die Auswertung auf die erfolgreich synchronisierten Tiere beschränkt) erzielt werden. Die KB bei 119 nicht behandelten Kontrolltieren, die in 5 dieser 8 Versuche zur Verfugung standen, war zu 60,3 % erfolgreich. 4. In einem Problembestand konnte die Fertilitätslage durch das biotechnische Vorgehen der Prostaglandinanwendung unmittelbar nicht verbessert werden 5. Die palpatorischen Ovarbefunde zum Zeitpunkt der KB ergaben, daß Tiere mit aprungbereiten Follikeln'zu 65 %, mit kleinen, nicht reifen Follikeln zu 50,8 % und mit bereitsgesprungenen Follikeln zu 15,8 % konzipierten. 6. Die bei einem Teil der Versuche eingesetzte zusdtzliche Behandlung mit 1,0 bzw. 1,5 mg des Gonadotropin-Releasinghormons (LRH-Hoechst 471) i. m. zum Zeitpunkt der Besamung zeigte bezüglich des Konzeptionsergebnisses eine positive Tendenz.
Summary Peripheral plasma levels of testosterone, LH and FSH were determined three times weekly by RIA in 4 bulls from birth through maturation and at 15 min. intervals in 6 males during short periods (8–48 hrs) at different ages (4 weeks—34 months). After an initial period of values below 1 ng./ml. plasma, the subsequent rise in plasma testosterone was characterized by a biphasic pattern. In 3 animals a first increase occurred after the 20th, 22nd or 28th week of age (mean values 2.2–8.4 ng./ml.), followed by a plateau or decrease. A second increase, occurred after the 39th, 51st and 54th week of age, respectively (mean values: 1.9–11.9 ng./ml.). In the 4th bull testosterone values exhibited a similar but delayed pattern and were generally lower during the whole trial. LH-levels remained constant in all animals between 0.6–2.7 ng./ml. during the experiment without any tendency to increase with age. FSH increased coincidentally with testosterone in 3 of the bulls, but showed a wave-like trend in the other bull. No changes in LH- and FSH-short term variations were found in relation to age. Testosterone values remained constantly below 1 ng./ml. in the 4 and 8 week old animals and started to exhibit short term variations after 20 weeks of age with a rather constant tendency of high values around 09.00, 16.00 and 22.00 h. An LH-peak preceded the observed increases in testosterone in 16 out of 21 observations. Zusammenfassung Testosteron, Luteinisierungshormon (LH) und Follikelstimulierendes Hormon (FSH) im peripheren Plasma von Bullen: Werte von der Geburt bis nach der Pubertät und Kurzzeitvariationen Testosteron, LH und FSH wurden mit Hilfe radioimmunologischer Methoden im peripheren Plasma von 4 Bullen vom Zeitpunkt der Geburt bis zur vollendeten Geschlechtsreife gemessen (Experiment I) und in 15 minütigem Abstand bei weiteren 6 Tieren im Alter von 4 Wochen bis 34 Monaten über einen Zeitraum von 8–48 Stunden (Experiment II). Experiment I: Nach einer ersten Periode von Testosteronkonzentrationen unter 1 ng./ml. Plasma (Periode I) verlief der darauf folgende Anstieg in 2 Phasen. Bei 3 Tieren wurde ein erster Anstieg (Periode II) nach der 20., 22., bzw. 28. Lebenswoche (Durchschnittswerte 2,2–8,4 ng/ml) und danach ein Plateau oder Abfall (Periode III) beobachtet. Dem folgte ein 2. Anstieg (Periode IV) nach der 39., 51. bzw. 54. Lebenswoche (Durchschnittswerte 1,9–11,9 ng/ml). Beim 4. Bullen zeigte sich ein ähnliches jedoch verzögertes Sekretionsmuster mit durchschnittlich niedrigeren Testosteronkonzentrationen. Die LH-Werte lagen bei allen Tieren konstant zwischen 0,6–2,7 ng/ml und zeigten keine Tendenzen, mit zunehmendem Alter anzusteigen. FSH stieg parallel zum Testosteronanstieg bei 3 der untersuchten Tiere an und zeigte einen nicht korrelierbaren wellenförmigen Verlauf bei dem 4. Bullen. Experiment II: Die episodischen LH- und FSH-Schwankungen wiesen keine Beziehungen zum Alter der Tiere auf. Testosteron lag konstant unter 1 ng/ml Plasma bei den 4 und 8 Wochen alten Tieren; mit dem Alter von 20 Wochen begannen die Konzentrationen im Verlauf eines Tages im peripheren Plasma zu schwanken, wobei — mit der Ausnahme einer Untersuchungsperiode — die Tendenz zu hohen Werten um 0900, 1600 und 2200 erkennbar war. Ein LH-Peak ging in 16 aus 21 Beobachtungen dem Testosteronanstieg voraus. Résumé Testostérone, hormone lutéinisante (LH) et hormone folliculostimulante FSH dans le plasma périphérique chez des taureaux: taux de la naissance à la puberté et variations pulsatiles On a mesuré la testostérone, LH et FSH avec des méthodes radioimmuno-logiques dans le plasma périphérique de 4 taureaux depuis la naissance à la maturité sexuelle (expérience I) et chez 6 animaux âgés de 4 semaines à 34 mois à des intervalles de 15 minutes durant une période de 8–48 heures (expérience II). Expérience I: Après une période de concentration en testostérone inférieure à 1 ng/ml plasma (période I), on a constaté une augmentation en 2 phases. Une première augmentation a été constatée chez 3 animaux après 20, 22 et 28 semaines (valeurs moyennes 2,2–8,4 ng/ml) (période II). Un plateau ou une chute s'en suivit (période III). Une seconde augmentation se manifesta après 39, 51 et 54 semaines (valeurs moyennes 1,9–11,9 ng/ml) (période IV). On a obtenu un échantillonnage semblable mais plus rapide avec des valeurs moyennes de testostérone plus basses chez le quatrième taureau. Les valeurs de LH restèrent constantes entre 0,6–2,7 ng/ml chez tous les animaux et n'ont pas augmenté avec l'âge. FSH augmenta parallèlement à la testostérone chez 3 animaux alors qu'il n'y a pas eu de corrélation chez le quatrième taureau. Expérience II: Les variations épisodiques de LH et FSH n'ont pas eu de rapport avec l'âge des animaux. La testostérone demeura constamment endessous de 1 ng/ml plasma chez les animaux de 4–8 semaines. Les concentrations commencèrent à varier dès la vingtième semaine au cours d'un jour dans le plasma péripherique et, à l'exception d'une période d'analyse, la tendance à des valeurs élevées fut remarquée à 0900, 1600 et 2200. Une pointe LH précéda l'augmentation de la testostérone dans 16 des 21 observations. Resumen Testosterona, hormona luteinizante (HL) y hormona estimulante del folículo (FSH) en plama sanguíneo periférico de toros: niveles desde el nacimiento hasta el final de la pubertad y variaciones a plazo breve Se midieron la testosterona, HL y FSH con ayuda de métodos radioinmunológicos en el plasma sanguíneo periférico de 4 toros desde el instante del nacimiento hasta consumada la pubertad (experimento I) y en distancias de 15 minutos en otros 6 animales, de edadas comprendidas entre 4 semanas y 34 meses, por un espacio de tiempo de 8–48 horas (experimento II). Experimento I: Tras un primer periodo de concentraciones de testosterona inferiores a 1 ng/ml plasma (periodo I), el aumento subsiguiente discurrió en 2 fases. En 3 animales se observó un aumento primero (periodo II) tras la semana 20°, 22° resp. 28° (valores medios 2,2–8,4 ng/ml) y después una meseta o descenso (periodo III). Siguió a esto un aumento 2° (periodo IV) tras la semana 39°, 51° resp. 54° (valores medios 1,9–11,9 ng/ml). Se apreció en el toro 4° un modelo secretor semejante, aunque retardado con concentraciones medias inferiores de testosterona. Los valores de HL se hallaban constantes en todos los animals entre 0,6–2,7 ng/ml, no mostrando tendencia alguna a aumentar con la edad. FSH aumentó paralela al incremento de testosterona en 3 de los animales examinados, evidenciando un curso ondulante no correlacionable en el toro 4°. Experimento II: Las fluctuaciones episódicas de HL y FSH no evidenciaron ninguna relación para con la edad de los animales. La testosterona se hallaba constante, siempre inferior a 1 ng/ml de plasma en los animales de 4 y 8 semanas de edad; con la edad de 20 semanas comenzaron a oscilar los valores en el curso de un día en el plasma periférico, reconociéndose — con la excepción de un periodo experimental — la tendencia a elevados alrededor de 0900, 1600 y 2200. Una cúspide de HL precedió en 16 de 21 observaciones al aumento de testosterona.

Summary. The luteotrophic properties of endogenously secreted LH and prolactin were studied by measuring CL function after neutral-ization of circulating LH and prolactin by the administration of highly specific antisera against these hormones on the 11th and 12th day of the oestrous cycle. In addition, the effect of a prolactin inhibitor (CB-154), which reduced prolactin concentration in peripheral blood by 80 to 90% was studied. A hysterectomized heifer carrying a persistent CL was treated with CB-154, a combination of CB-154 and prolactin antiserum and then with LH antiserum. In all animals treated with the LH antiserum, CL function ceased or was quantitatively inhibited. No change in circulating progesterone was seen after treatment with the prolactin antiserum or the prolactin inhibitor. It is concluded that LH is the dominant luteotrophic factor in the bovine species while prolactin has little or no activity.
Both hormones, LH and prolactin, have been discussed to stimulate and maintain Cl-function, not only in the small laboratory animal but also in the bovine. While there is strong evidence that LH exerts luteotropic properties in the latter species, contrarily and somewhat confusing results have been published concerning the role of prolactin.
The radioimmunoassay described for the determination of progesterone only involves extraction of the plasma andsubsequent performance of the protein-binding reaction. The antisera used were obtained from rabbits and directed against 11α- OH-progesterone hemisuccinate BSA. A high degree of reliability was obtained for the criteria specificity, sensitivity, accuracy and precision. Application of the method during the bovine oestrous cycle revealed that there is no preovulatory rise of progesterone in this species. The presence of progesterone in peripheral plasma of bulls was demonstrated.
