Hintergrund/Einleitung: IM862 ist ein Dipeptid (L-Glutamyl-L-Tryptophan) mit antiangiogener Wirkung und potentieller antitumoraler Aktivität.
Epithelial–mesenchymal transition (EMT) endows cells with migratory and invasive properties, a prerequisite for the establishment of endometriotic lesions. However, the role EMT might play in the pathophysiology of endometriosis is still unknown. Therefore, we examined five recognized markers for EMT in endometrium and endometriosis: E-cadherin, N-cadherin, Twist, Snail and Slug.
Ziele: Quantifizierung der Effekte verschiedener Angiogeneseinhibitoren auf die Vaskularisation experimenteller Pankreaskarzinomen mittels dynamischer, kontrastverstärkter MRT. Methode: 109 Lewis-Ratten wurden Fragmente des Pankreaskarzinoms DSL-6A/C1 in den Pankreasschwanz implantiert. 3 Wochen später erfolgte der Therapiebeginn mit Bevacizumab oder Suramin, die Kontrollgruppe blieb unbehandelt. 24 Stunden, 1 Woche und 4 Wochen (n=38, 27 und 44) nach Therapiebeginn erfolgte die Albumin-(Gd-DTPA) -verstärkte MRT zur Ermittlung der Tumorgefäß-Permeabilität (KPS, µl/min/100ml) und des Plasmavolumens (fPV, %) anhand eines Zweikompartimentmodells. Ergebnis: Die Avastin-Gruppe zeigte 24h nach Therapiebeginn signifikant geringere KPS-Werte als die Kontroll- und Suramin-Gruppe (0,0019±0,0077; 0,0565±0,0462 und 0,0640±0,0620; p<0,001). KPS war nach 1 bzw. 4 Wochen in der Avastin-Gruppe signifikant kleiner als in der Kontroll-Gruppe (0,0364±0,0368 und 0,1115±0,08800; p<0,01 bzw. 0,0815 ±0,0735 und 0,02623±0,04620; p<0,05).
Einleitung: Die spezifische Hemmung des Epidermal Growth Factor-Rezeptors (EGFR) ist ein wirksamer Therapieansatz bei einer Reihe von Tumoren. Wir untersuchten die Wirkung des EGFR-Antikörpers Cetuximab auf 3 humane Magenkarzinom-Zelllinien in vitro sowie in vivo als Monotherapie und in Kombination mit Carboplatin in einem murinen Magenkarzinom-Modell.
Hintergrund: Die Aufrechterhaltung der Makro- und Mikrozirkulation ist von entscheidender Bedeutung für die Heilung von gastrointestinalen Anastomosen. Kolloide und Kristalloide finden in der Volumentherapie weite Verbreitung. Ziel dieser Studie war es, die Effekte von humanem Albumin, Hydroxyethylstärke (HES 130/0,4) sowie physiologischer Kochsalzlösung auf Mikrozirkulation und Wundheilung von Kolonanastomosen in Ratten zu evaluieren.
The transcription factor Snail is a key regulator of epithelial to mesenchymal transition (EMT), a process vital for cancer progression. The study investigated an EMT-independent role of Snail for cell motility of macrophages in inflammation and wound healing. Snail expression was evaluated in tissue samples from humans and rats. End-to-End colonic anastomoses were performed in rats. The anastomotic region was excised after various time points up to two weeks. Human samples were evaluated from patients with infected wounds and acute appendicitis or chronic inflammatory bowel disease (IBD). Snail expression in the macrophage cell line THP-1 was investigated after stimulation with TGF-β, LPS, IL-8, and MMP-3. Snail expression in the anastomotic region was significantly higher than in control colon tissue of rats. Double staining for the macrophage marker CD68 and Snail confirmed migrating macrophages as the Snail expressing cells. Snail expression was depending on time point and location: the amount of Snail-positive cells was maximally upregulated (18 fold) 48 hours after performing the anastomoses and decreased after two weeks to a 7 fold amount in comparison to control tissue. Snail-positive macrophages were 22 fold increased at the site of anastomoses, 14 fold at 1 cm, and 5 fold at 4 cm distance of the anastomoses. Acute human inflammation (wound infection, appendicitis) was characterized by a significantly increased number of Snail-positive macrophages. Snail-mRNA expression in IBD was upregulated 12 fold (Crohn’s disease) and 5 fold (ulcerative colitis) in comparison to control bowel tissue. Infiltrating macrophages were identified as Snail expressors. With regard to signal transduction pathways of Snail induction, only TGF-β displayed a time-dependent stimulation of THP-1 macrophages, with a 3 fold upregulated Snail after 48 hours, resulting in 50% increased migration. This study demonstrates for the first time an EMT-independent function of Snail. Snail seems to activate the migration of macrophages in wound healing, acute and chronic inflammation, depending on time point and location.
Die epitheliale mesenchymale Transition (EMT) beschreibt die Umwandlung epithelialer Zellen zu einem migratorischen Phänotypen. Die EMT ist für die die Metastasierung und Invasion von Tumoren entscheidend. Die Transkriptionsfaktoren Twist, Snail und Slug sind Induktoren der EMT durch Herunterregulation des Adhäsionsmoleküls E-Cadherin. Twist wiederum induziert die Hochregulation von N-Cadherin, dass mit einem hohen invasiven Potential in Tumoren assoziiert ist. Endometriose ist eine gutartige Erkrankung ist, kann aber auch invasive und metastatisch wachsen. Es sollte daher untersucht werden, ob die EMT in der Pathogenese der Endometriose möglicherweise eine Rolle spielt.
The effect of antiangiogenetic monotherapy on experimental pancreatic cancer is limited. Therefore this study evaluated the potential of a combination of Gemcitabine with the antiangiogenetic agent IM862. We investigated nude mice after orthotopic implantation of human pancreatic cancer cells (AsPC-1, poorly differentiated and HPAF-2, moderately differentiated). 72 animals were randomized in 2 control and 4 treatment groups that received either Gemcitabine (125 mg/kg, once a week i. p.) alone or both Gemcitabine and IM862 (100 mg/kg, daily i. p.). Tumor volume (TU-Vol), dissemination (D-Score) and microvessel density (MVD) were analyzed 14 weeks after therapy or death. All control animals developed extensive tumor growth, infiltration, and distant metastases, while there was significantly reduced tumor growth in the Gemcitabine-only group. Gemcitabine therapy with IM862 was most effective in HPAF-2 tumors and inhibited primary tumor growth and metastasis almost completely. However, the effects of Gemcitabine on survival were comparable with the results of the combination treatment.
Epithelial to mesenchymal transitions are vital for tumor growth and metastasis. Several inducers of EMT are transcription factors that repress E-cadherin expression such as Snail, Slug and Twist. In this study we aimed to examine the expression of these transcription factors in pancreatic cancer. The expression of Snail, Slug, and Twist, was detected by immunohistochemistry in tissue samples from patients with pancreatic ductal adenocarcinoma. Four human pancreatic cancer cell lines (Capan-1, HPAF-2, MIAPaCa-2, and Panc-1) were analyzed by RT-PCR, real-time PCR and western blotting. An orthotopic nude mouse model of pancreatic cancer was applied for in vivo experiments. 78 % of human pancreatic cancer tissues showed an expression of Snail, and 50 % of the patients displayed positive expression of Slug. Twist showed no or only weak expression. Snail expression was higher in undifferentiated cancer cell lines (MIAPaCa-2, Panc-1) than in more differentiated cell lines (Capan-1, HPAF-2) and correlated positively with the metastatic potential of the cancer cells in the animal model. Expression of Slug was detected in all cell lines with different intensities. Twist was not expressed. The transcription factors Snail and Slug are expressed in pancreatic cancer but not in normal tissue suggesting a role in the progression of human pancreatic tumors.
Suramin, a naphthyl urea derivative, inhibits growth of several tumours, among them ductal pancreatic adenocarcinoma, but its use as a single therapeutic agent is problematic due to its toxicity. Therefore, the aim of this study was to test the efficacy of a combination therapy of low dose suramin and docetaxel in pancreatic cancer. Rat ductal pancreatic adenocarcinoma cells (DSL6A) were incubated in the presence of increasing concentrations of suramin (in µg/mL: 0; 5; 10; 100) or docetaxel (in µg/mL: 0; 0.003; 0.015; 0.03; 3) or a combination of both drugs. Cell proliferation and viability were assessed after 72 hours. Docetaxel as well as suramin inhibited cell proliferation and viability in a dose-dependent way (p < 0.01). The highest concentrations used as monotherapy inhibited cell growth by 74 % (suramin) and 95 % (docetaxel), respectively. A combination of both drugs was more effective than each of the single agents given alone. Therefore, one can conclude that the addition of low dose suramin (in itself without any relevant effect on cell proliferation) enhances docetaxel efficacy in vitro. Further studies using an orthotopic tumour model of rat ductal pancreatic adenocarcinoma are necessary in order to confirm whether this combination therapy is of relevance in a preclinical therapeutic setting.
Fragestellung: Die chronische Erkrankung Endometriose (EM) tritt bei 7–15% der Frauen in der reproduktiven Lebensphase auf. EM ist als gutartige Erkrankung definiert, teilt jedoch einige Charakteristika mit malignen Tumoren wie potentiell invasives und metastasierendes Wachstum. In den letzten Jahren konnte gezeigt werden, dass für invasives Wachstum und Metastasenbildung die EMT (epithelial- mesenchymal transition) funktionell von großer Bedeutung ist. In der EMT spielt TWIST, ein embryonaler Transkriptionsfaktor, eine wichtige Rolle, indem er u.a. die Expression der Adhäsionsmoleküle E- Cadherin und N-Cadherin beeinflusst. Dieser Mechanismus reguliert die Zellmigration. Eine TWIST- Überexpression wurde bereits in verschiedenen malignen Tumoren wie z.B. dem Mamma- Ca und Magen- Ca nachgewiesen. In EM-Gewebe konnte nachgewiesen werden, dass E- und N- Cadherin ähnlich wie in Carcinomgeweben moduliert sind. Bisher wurde kein regulierender Mechanismus in EM-Gewebe identifiziert. Daher wurde nun erstmals die TWIST- Expression in EM- Läsionen untersucht.
Einleitung: Das Magenkarzinom zählt noch immer zu den häufigsten tumorbedingten Todesursachen, wobei die Inzidenz der Karzinome im gastroösophagealen Übergang (Kardia) signifikant zunimmt. Bislang ist in der Literatur kein Tiermodell für diese Erkrankung beschrieben.
SLT-VEGF Fusionsprotein reduziert Primärtumorwachstum und Disseminierung des experimentellen Pankreaskarzinoms, was in einem verbesserten 14-Wochen-Überleben der Versuchstiere resultiert. Die reduzierte mikrovaskuläre Gefäßdichte deutet darauf hin, daß dieser Effekt in erster Linie durch einen toxischen Effekt von SLT-VEGF auf Tumorendothelzellen vermittelt wird. Die eingesetzten Therapien waren nicht mit systemischen Nebenwirkungen wie Gewichtsverlust assoziiert.
Einleitung: Angiogenesehemmung ist eine neue Therapie für das Pankreaskarzinom. Aktivierte Endothelzellen der Tumorgefäße überexprimieren im Gegensatz zu ruhenden Endothelien Rezeptoren für VEGF, den Schlüsselfaktor der Neoangiogenese.
Background. Vascular endothelial growth factor (VEGF), a key mediator of angiogenesis, is overexpressed in pancreatic cancer. This study evaluated VEGF production in pancreatic cancer cells and the effect of VEGF antisense on growth and angiogenesis of human pancreatic cancer in a nude mouse model.Methods. In vitro: VEGF in cell culture supernatant of pancreatic cancer cells (AsPC-1, poorly differentiated; HPAF-2, moderately differentiated) was assessed by enzyme-linked immunosorbent assay. In vivo: A VEGF antisense oligonucleotide (AS-3) was synthesized. One-mm(3) fragments of subcutaneous pancreatic cancer donor tumors were implanted into the Pancreas of nude mice also receiving AS-3 (10 mg/kg/day) or vehicle intraperitoneally for 14 weeks. Primary tumor volume, metastasis, and VEGF in plasma and ascites were determined at autopsy. Microvessel density was analyzed in CD31-stained tumors.Results. In vitro: Both pancreatic cancer cell lines secreted VEGF protein (AsPC-1, 4200 +/- 40 pg/10(6) cells; HPAF-2, 8120 +/- 60 pg/10(6) cells). In vivo: AS-3 reduced tumor volume in the HPAF-2 group (860 +/- 140 vs 3830 +/- 590 mm(3)) and metastatic spread in both groups (AsPC-1, 65 +/- 0.8 vs 16.7 +/- 0.9 points; HPAF-2, 2.5 +/- 0.2 vs 8.3 +/- 1.5 points). Tumor volume was not different in the AsPC-1 group (1050 +/- 80 vs 1400 +/- 150 mm(3)). Survival was increased in the AsPC-1 group. Plasma levels of VEGF and microvessel density in tumors were significantly reduced in treated animals. Only control animals (50%) developed ascites with high VEGF concentrations.Conclusions. Human pancreatic cancer cells secrete VEGF at biologically relevant high levels. AS-3 therapy normalizes plasma VEGF and decreases neoangiogenesis, thereby reducing tumor growth and metastasis and improving survival. AS-3-treated animals developed no ascites, suggesting decreased vascular permeability by reducing VEGF expression in pancreatic cancer cells.
Hintergrund: Monotherapie mit anti-angiogenen Substanzen oder Chemotherapeutika ist nur bedingt beim experimentellen humanen Pankreaskarzinom effektiv. Das Ziel dieser Studie war, das therapeutische Potential der kombinierten Angiogenesehemmung und Chemotherapie in einem orthotopen Nacktmausmodell des humanen Pankreaskarzinoms zu evaluieren. Methoden: Jeweils 5 Millionen Zellen der beiden humanen Pankreaskarzinom-Zellinien AsPC–1 (schlecht differenziert) und HPAF–2 (mittelgradig differenziert) wurden subkutan in Nacktmäuse injiziert. 1 cmm große Fragmente aus den resultierenden subkutanen Donortumoren wurden orthotop in das Pankreas von 72 anderen Nacktmäusen implantiert. Diese Tiere wurden in Kontroll- und 2 Behandlungsgruppen randomisiert: die kombinierte Gabe des Angiogeneseinhibitors IM862 (100mg/kg, täglich intraperitoneal) und von Gemcitabine (125mg/kg, zweimal pro Woche intraperitoneal) begann entweder 3 Tage (Prophylaxe) oder 6 Wochen (Therapie) nach der Tumorinduktion. Die Therapie wurde für maximal 14 Wochen, oder bis zum Tod der Tiere fortgesetzt. Bei der Autopsie wurden das Primärtumorvolumen (TU-Vol) und ein Disseminierungsscore (D-Score) aus lokaler Infiltration und Fernmetastasierung bestimmt. Um auch Mikrometastasierung zu erfassen, wurden HE-Schnitte von allen Organen angefertigt und mikroskopisch untersucht. Das Gewicht der Tiere wurde wöchentlich bestimmt. Ergebnisse: Alle Kontrolltiere entwickelten ausgeprägte Primärtumoren, lokale Infiltration der Nachbarschaft und Fernmetastasierung. Die Effekte der Kombinationstherapie nach 6-wöchiger Therapieverzögerung (Therapiegruppe) und frühzeitigem Therapiebeginn (Prophylaxegruppe) sind tabellarisch zusammengefasst. Die eingesetzte Kombinationstherapie war nicht mit systemischen Nebenwirkungen wie Gewichtsverlust assoziiert. Schlussfolgerungen: Die kombinierte Therapie mit dem Angiogeneseinhibitor IM862 und Gemcitabine zeigt therapeutisches Potential in einem klinisch relevanten Tiermodell des humanen Pankreaskarzinoms. Die Effekte waren am deutlichsten in mittelgradig differenzierten HPAF–2 Tumoren ausgeprägt, deren Wachstum und Metastasierung im prophylaktischen Ansatz nahezu komplett gehemmt wurde.
Background: A previous study demonstrated that Suramin, a naphthyl urea derivative, reduces the proliferation of human pancreas cancer cells in vitro. This was accompanied by reduced secretion of the proangiogenic key factor VEGF. The aim of the present study was to systematically evaluate the effect of Suramin on tumor growth, metastasis and angiogenesis in vivo using a clinically relevant orthotopic immuno-competent rat model of ductal pancreatic cancer. Methods: 107 cells of the rat ductal pancreatic cancer cell line DSL-6A/C1 were subcutaneously injected into 2 Lewis rats. 1 cmm fragments of the resulting subcutaneous donor tumors were orthotopically implanted into the pancreas of 16 other Lewis rats. The animals were randomized into 2 groups: the therapy group (Suramin 60 mg/kg, weekly ip.) and the control-group (vehicle ip.). Intravital microscopy was performed 4 weeks after tumor induction. The volume of the primary tumor and local infiltration, as well as systemic metastasis (score) and microvascular density were determined at autopsy. Results: The animals of the control group developed large primary tumors (1978,5 ± 231,2 cmm) and metastasis (9,0 ± 1,1 pts.). In contrast, half of the animals in the therapy group displayed no tumor growth, the other half showed only small tumors (4,1 ± 1,9 cmm; p < 0,05) without distant spread. Control animals showed typical tumor vessels with a vessel density of 421 ± 39 cm/scm, whereas vessel density of primary tumors in treated animals was reduced to 266 ± 31 cm/scm (p < 0,05). The weight of the animals was similar in both groups (controls: 265 ± 12 g; therapy: 259 ± 16 g). Conclusion: Sumarin reduces primary tumor growth, dissemination, and angiogenesis in the early stage of tumor development in an immuno-competent rat model of ductal pancreatic cancer.
