Rice blast, caused by the fungal pathogen Pyricularia grisea, is a serious disease affecting rice-growing regions around the world. Current methods for identification of blast-resistant germplasm and progeny typically utilize phenotypic screening. However, phenotypic screens are influenced by environmental conditions and the presence of one resistance gene can sometimes phenotypically mask other genes conferring resistance to the same blast race. Pi-z is a dominant gene located on the short arm of chromosome 6 that confers complete resistance to five races of blast. Using sequence data found in public databases and degenerate primer pairs based on the P-loop, nucleotide binding sites and kinase domain motifs of previously cloned resistance genes, we have developed PCR-based DNA markers that cosegregate with the gene. These markers are polymorphic in a wide range of germplasm, including the narrow crosses characteristic of applied rice-breeding programs. They can now be used as a low cost, high-throughput alternative to conventional phenotypic screening for direct detection of blast resistance genes, allowing rapid introgression of genes into susceptible varieties as well as the incorporation of multiple genes into individual lines for more-durable blast resistance.

Three RFLP maps, as well as several RAPD maps have been developed in common bean (Phaseolus vulgaris L.). In order to align these maps, a core linkage map was established in the recombinant inbred population BAT93×Jalo EEP558 (BJ). This map has a total length of 1226 cM and comprises 563 markers, including some 120 RFLP and 430 RAPD markers, in addition to a few isozyme and phenotypic marker loci. Among the RFLPs mapped were markers from the University of California, Davis (established in the F2 of the BJ cross), University of Paris-Orsay, and University of Florida maps. These shared markers allowed us to establish a correspondence between the linkage groups of these three RFLP linkage maps. In total, the general map location (i.e., the linkage group membership and approximate location within linkage groups) has been determined for some 1070 markers. Approaches to align this core map with other current or future maps are discussed.

An accelerated rice-breeding program was initiated in 1969 at the California Rice Experiment Station. The program is broad in scope, developing cultivars in all US market classes (long, medium, and short grains) and special purpose types (waxy and aromatics). This grower-funded rice-breeding program has released 27 new cultivars as well as improved germplasm lines. Statewide paddy rice yields have risen from 6.2 to 9.3 t/ha since 1978. The incorporation of semi-dwarfing genes, earlier maturity, and increased yield potential have contributed significantly to increases in grain yield. High experimental yields (>I1 t/ha) are routine and achieving increased increments of yield will become more difficult. After the initial shift to semi-dwarf cultivars, increasing efforts were directed toward improving adaptation to environmental stresses and grain quality. Screening and selection for cold tolerance in the form of seedling vigour for water seeding and resistance to cool temperature induced sterility at the reproductive stage are integral parts of the rice-breeding program. Progress is being made on incorporation into California rice cultivars of resistance to stem rot (Sclerotiurn oryzae Cattaneo) and aggregate sheath spot [Rhizoctonia oryzae-sativae (Swada) Mordue] from wild species and tolerance to rice water weevil (Lissorhoptus oryzophilus Kuschel). New restrictions and regulations of agronomic management practices may negatively impact rice production, creating problems in stand establishment, soil fertility, and weed, disease, and insect control. Breeding efforts to help minimise the adverse effect of these restrictions on yield and quality will increase in the future. Increased emphasis is being placed on improving milling yield and cooking and processing characteristics, and new laboratory methods are being explored to aid in evaluation and selection for grain quality.

Rice, Oryza sativa, plants regenerated from anther culture with and without in vitro selection pressure were evaluated for chalky seed. Progeny evaluated included 21 spontaneously doubled haploids selfed 4 times, progeny from plants regenerated from S-aminoethylcysteine resistant callus selfed 4 times and backcrosses of both types to the parental type. All lines with in vitro histories had higher seed chalkiness than the controls both in the intensity of chalkiness and in the number of seeds expressing the character. The full range of intensity and amount of chalkiness was expressed in the progeny. The average intensity of anther/tissue culture-derived progeny was 4–5, based on a scale of 1 (translucent) to 10 (fully opaque), and the average amount of chalkiness within plants sampled was 50–75 percent. The chalky characteristic is transmitted from parent to offspring into a range of identifiable F2 segregants.

plants having long, narrow, drooping leaves. Stems are straight-necked and yellow at maturity. Distance from the flag leaf to the spike ranges from 14 to 22 cm; the collar is closed and the basal internode is straight and 1 to 3 mm long. The rachis is tough with glabrous edges. Kline is resistant to spot blotch, caused by Cochliobolus sativus (Ito & Kurib.) Drechs. ex Dast., and barley yellow dwarf virus (BYDV). It is moderately resistant to leaf rust and scald, caused by Rhynchosporium secalis (Oud.) JJ. Davis. Kline is moderately susceptible to septoria glume blotch, caused by Leptosphaeria nodorum Muller, and is susceptible to powdery mildew and loose smut, caused by Ustilago nuda (Jens.) Rostr. Powdery mildew is not a serious disease in the Southeast and loose smut can be controlled by carboxin (Vitavax) seed treatment. This cultivar was named to honor the late Dr. David Kline, USDA-ARS, cereal pathologist, North Carolina State Univ., Raleigh. Dr. Kline pioneered in research on new fungicides to control small grain diseases after organic mercurial compounds were banned early in the 1970s. Breeder seed will be maintained by the Agronomy Dep., College Exp. Stn., Athens, GA 30602. Foundation seed will be maintained by the Georgia Seed Development Commission, 2420 South Milledge Avenue, Athens, GA 30605.
