Subclinical ketosis (SCK) is a prevalent metabolic disorder that occurs during the transition to lactation period. It is defined as a high blood concentration of ketone bodies (beta-hydroxybutyric acid f ≥ 1.2 mmol/L) within the first few weeks of lactation, and often presents without clinical signs. SCK is mainly caused by negative energy balance (NEB). The objective of this study is to identify single nucleotide polymorphisms (SNPs) associated with SCK using genome-wide association studies (GWAS), and to predict the biological functions of proximal genes using gene-set enrichment analysis (GSEA). Blood samples were collected from 112 Holstein cows between 5 and 18 days postpartum to determine the incidence of SCK. Genomic DNA extracted from both SCK and healthy cows was examined using the Illumina Bovine SNP50K BeadChip for genotyping. GWAS revealed 194 putative SNPs and 163 genes associated with those SNPs. Additionally, GSEA showed that the genes retrieved by Database for Annotation, Visualization, and Integrated Discovery (DAVID) belonged to calcium signaling, starch and sucrose, immune network, and metabolic pathways. Furthermore, the proximal genes were found to be related to germ cell and early embryo development. In summary, this study proposes several feasible SNPs and genes associated with SCK through GWAS and GSEA. These candidates can be utilized in selective breeding programs to reduce the genetic risk for SCK and subfertility in high-performance dairy cows.
This study aimed to investigate the correlation between the linear type traits of mother cows (dams) and the birth weight(BW) of the progeny. The study used data collected between 2002 and 2018 by the Korea Animal Improvement Association (KAIA). This study evaluated the data specifically with respect to 19 traits. A statistical model was established to estimate the correlation between the maternal linear classification traits and the progeny BW. The significance of each statistical variable was analyzed using the generalized linear model (GLM) method in the SAS package 9.1.3 software, while the genetic parameters were estimated using the BLUPF90 Family program. The BW of Hanwoo cows increased by approximately 0.4 kg each year; however, the BW heritability was moderate (0.23). The linear classification traits showed values of 0.34, 0.37, 0.11, 0.45, 0.31, and 0.40, for ST (Vertical distance from the apex of the rump to the ground), BL (Horizontal distance between anterior shoulder point to the posterior extremity of the pin bone), RA (The slope from the rump to the pin bone), RL (Horizontal distance from the anterior rump to the tip of the pin bone), PBW (Lateral width of pin bone), and FS (Final score), respectively, with moderate heritability for all traits, except RA. The BW of the progeny showed phenotypic correlations of 0.06, -0.01, 0.03, and 0.04, and genetic correlations of 0.09, 0.03, 0.02, and -0.04 with ST, BL, RL, and PBW, respectively. The average parity in Hanwoo cows increased from 2.5 in 2016 to 2.8 in June 2020. There is a low genetic correlation between the maternal linear classification traits and the BW of the progeny. An increase in the parity of dams and a greater accumulation of progeny BW data and test records could lead to the accurate evaluation of the BW as a breeding objective trait.
In Korea, Korea Proven Bulls (KPN) program has been well-developed. Breeding and evaluation of cows are also an essential factor to increase earnings and genetic gain. This study aimed to evaluate the accuracy of cow breeding value by using three methods (pedigree index [PI], pedigree-based best linear unbiased prediction [PBLUP], and genomic-BLUP [GBLUP]). The reference population (n = 16,971) was used to estimate breeding values for 481 females as a test population. The accuracy of GBLUP was 0.63, 0.66, 0.62 and 0.63 for carcass weight (CWT), eye muscle area (EMA), back-fat thickness (BFT), and marbling score (MS), respectively. As for the PBLUP method, accuracy of prediction was 0.43 for CWT, 0.45 for EMA, 0.43 for MS, and 0.44 for BFT. Accuracy of PI method was the lowest (0.28 to 0.29 for carcass traits). The increase by approximate 20% in accuracy of GBLUP method than other methods could be because genomic information may explain Mendelian sampling error that pedigree information cannot detect. Bias can cause reducing accuracy of estimated breeding value (EBV) for selected animals. Regression coefficient between true breeding value (TBV) and GBLUP EBV, PBLUP EBV, and PI EBV were 0.78, 0.625, and 0.35, respectively for CWT. This showed that genomic EBV (GEBV) is less biased than PBLUP and PI EBV in this study. In addition, number of effective chromosome segments (Me) statistic that indicates the independent loci is one of the important factors affecting the accuracy of BLUP. The correlation between Me and the accuracy of GBLUP is related to the genetic relationship between reference and test population. The correlations between Me and accuracy were -0.74 in CWT, -0.75 in EMA, -0.73 in MS, and -0.75 in BF, which were strongly negative. These results proved that the estimation of genetic ability using genomic data is the most effective, and the smaller the Me, the higher the accuracy of EBV.
The aim of the present study is to examine the potential effect of dexamethasone (DEX) and taurine (TAU) on endoplasmic reticular stress (ERS) and inflammation. The macrophages were pre-treated with DEX or TAU, and the level of ERS and pro-inflammatory response was evaluated in LPS-activated macrophages. The expression of ERS marker proteins (GRP78 and CHOP) and the pro-inflammatory cytokines (IL-1β and IL-6) was analyzed by immunoblotting and ELISA of LPS-induced macrophages. At lower concentrations (<50 nM), DEX alone reduced the levels of ERS and pro-inflammatory markers. Similarly, TAU reduced the expression of LPS-induced ERS and pro-inflammatory markers. When treated with a combination of DEX and TAU, however, the macrophages showed even a greater level of reduction in ERS and pro-inflammatory responses. The RT-qPCR data indicate that the reduction of ERS markers is caused by the inhibition of their own transcriptional expression. The significant level of reduction can be interpreted as a strong synergistic effect (p < 0.01). In summary, the results strongly indicate that a single pre-treatment of DEX or TAU may attenuate ERS and inflammation in LPS-induced macrophages at the concentrations tested in this study. Most importantly, concurrent treatment of macrophages by both agents reduced the level of ERS and pro-inflammatory cytokines in a synergistic fashion.
Longevity is closely related to the survival rate of dairy cattle and refers to the period during which the cow has economic value, from first calving to culling. The purpose of this study was to analyze the culling patterns and survival rates of Korean Holstein cows and evaluate genetic characteristics related to parity and longevity of each lactation by using the test day milk yield collected in South Korea. The performance data of the dairy cattle were collected from 2004 to 2019 by the Nonghyup Dairy Cattle Improvement Center. The collected 1,702,304 records were used as pedigree data through the Korea Animal Improvement Association. The lactation period was divided into early-lactation (0–90 days: L1.1, L2.1, and L3.1), mid-lactation (91–299 days: L1.2, L2.2, and L3.2), and late-lactation (300 days-next parity: L1.3, L2.3, and L3.3). The heritability of longevity for the first, second, and third parity was 0.020, 0.028, and 0.039, respectively. In all parities, the heritability in late-lactation was higher than that in early- and mid-lactation. Most genetic correlations for survival in the first parity were higher than those in the second and third parities. The results of this study may serve as a basis for developing a more accurate model for evaluating longevity traits in South Korea.
This study was performed to investigate genetic relationship of age at first calving (AFC) with body development and reproduction capacity in female Hanwoo. The data sets of 52,299 reproduction records of 19,566 heads calved from 2011 to 2019 and conformation records of 19565 heads born from 1997 to 2017 were analyzed by separating them based on age at first calving. In the analysis, conformation traits included the body condition score (BCS), height (HT), body width (BW), hip bone length (HL), hip bone width (HW), and final score (FS). The heritability and genetic correlation of AFC with conformation traits and calving interval (CI) were analyzed. The heritability for AFC was 0.716, 0.087, and 0.097 for the before and after 24 months and combined data, respectively. The genetic correlation of AFC before 24 months with BCS was -0.778, -0.600 with BW, and 0.442 with HT. The genetic correlation of AFC after 24 months with HT was -0.826, -0.706 with BW, -0.623 with HL, -0.456 with HW, and -0.675 with FS. When the first calving age of young heifers approached 24 months, BCS and BW decreased, and HT increased. When first calving is delayed to after 24 months, the conformation traits become smaller, which indicates that conformation to some extent affects the delay in AFC. The genetic correlation between CI and AFC was -0.116, 0.307, and 0.250 for the before and after 24 month of AFC and combined data, respectively. When first calving date approached 24 months, CI was reduced. The obtained results suggest that it is important that first calving occurs at an appropriate age. Additional research is needed to perform proper genetic evaluation of first calving age in Hanwoo cattle in Korea.
Objective Milk fatty acid (FA) is a main nutritional component that markedly effects human health. Intentional modification of the FA profile has the potential to improve milk quality. This study aimed at the factors affecting elevated FA levels and the estimation of the genetic parameters for milk FAs in the Korean Holstein population. Methods Total 885,249 repeated test-day milk records including, milk yield, saturated fatty acids (SFA), polyunsaturated fatty acids (PUFA), monounsaturated fatty acids (MUFA), total unsaturated fatty acids (TUFA), fat and protein percentages were analyzed using CombiFoss FT+ system (Foss Analytical A/S, Denmark). Genetic parameters were estimated by the restricted maximum likelihood procedure based on the repeatability model using the Wombat program. Results The FA profile varies along with the lactation and the energy balance (EB). With the negative EB in early lactation, mobilization of body fat reserves elevates the desirable FA levels. As a result of that, milk quality is increased by means of nutritionally and usability aspects during the early lactation. Moreover, heritability estimates for SFA, MUFA, PUFA, TUFA were 0.33, 0.42, 0.37, 0.41 respectively. According to the parity wise heritability analysis, first parity cows had relatively lower heritability for SFAs (0.19) than later parities (0.28). Conclusion Genetic parameters indicated that FAs were under stronger genetic control. Therefore, we suggest implementing animal breeding programs towards improving the milk FA profile.
OBJECTIVE:The objectives of this study were to compare identified informative regions through two genome-wide association study (GWAS) approaches and determine the accuracy and bias of the direct genomic value (DGV) for milk production traits in Korean Holstein cattle, using two genomic prediction approaches: single-step genomic best linear unbiased prediction (ss-GBLUP) and Bayesian Bayes-B.METHODS:Records on production traits such as adjusted 305-day milk (MY305), fat (FY305), and protein (PY305) yields were collected from 265,271 first parity cows. After quality control, 50,765 single-nucleotide polymorphic genotypes were available for analysis. In GWAS for ss-GBLUP (ssGWAS) and Bayes-B (BayesGWAS), the proportion of genetic variance for each 1-Mb genomic window was calculated and used to identify informative genomic regions. Accuracy of the DGV was estimated by a five-fold cross-validation with random clustering. As a measure of accuracy for DGV, we also assessed the correlation between DGV and deregressed-estimated breeding value (DEBV). The bias of DGV for each method was obtained by determining regression coefficients.RESULTS:A total of nine and five significant windows (1 Mb) were identified for MY305 using ssGWAS and BayesGWAS, respectively. Using ssGWAS and BayesGWAS, we also detected multiple significant regions for FY305 (12 and 7) and PY305 (14 and 2), respectively. Both single-step DGV and Bayes DGV also showed somewhat moderate accuracy ranges for MY305 (0.32 to 0.34), FY305 (0.37 to 0.39), and PY305 (0.35 to 0.36) traits, respectively. The mean biases of DGVs determined using the single-step and Bayesian methods were 1.50±0.21 and 1.18±0.26 for MY305, 1.75±0.33 and 1.14±0.20 for FY305, and 1.59±0.20 and 1.14±0.15 for PY305, respectively.CONCLUSION:From the bias perspective, we believe that genomic selection based on the application of Bayesian approaches would be more suitable than application of ss-GBLUP in Korean Holstein populations.
Objective The objective of this study was to investigate the genetic components of daily milk yield and to re-rank bulls in South Korea by estimated breeding value (EBV) under heat stress using the temperature-humidity index (THI). Methods This study was conducted using 125,312 monthly test-day records, collected from January 2000 to February 2017 for 19,889 Holstein cows from 647 farms in South Korea. Milk production data were collected from two agencies, the Dairy Cattle Genetic Improvement Center and the Korea Animal Improvement Association, and meteorological data were obtained from 41 regional weather stations using the Automated Surface Observing System (ASOS) installed throughout South Korea. A random regression model using the THI was applied to estimate genetic parameters of heat tolerance based on the test-day records. The model included herd-year-season, calving age, and days-in-milk as fixed effects, as well as heat tolerance as an additive genetic effect, permanent environmental effect, and direct additive and permanent environmental effect. Results Below the THI threshold (≤72; no heat stress), the variance in heat tolerance was zero. However, the heat tolerance variance began to increase as THI exceeded the threshold. The covariance between the genetic additive effect and the heat tolerance effect was −0.33. Heritability estimates of milk yield ranged from 0.111 to 0.176 (average: 0.128). Heritability decreased slightly as THI increased, and began to increase at a THI of 79. The predicted bull EBV ranking varied with THI. Conclusion We conclude that genetic evaluation using the THI function could be useful for selecting bulls for heat tolerance in South Korea.
Objective The objective of this study was to estimate the genetic parameters for milk β-hydroxybutyrate (BHBA), acetone (Ac), fat protein ratio (FPR), and energy balance (EB) using milk test day records and investigate the effect of early lactation FPR and EB on milk ketone body concentrations. Methods Total 262,940 test-day records collected from Korea Animal Improvement Association during the period of 2012 to 2016 were used in this study. BHBA and Ac concentrations in milk were measured by Fourier transform infrared spectroscopy (FTIR). FPR values were obtained using test day records of fat and protein percentage. EB was calculated using previously developed equation based on parity, lactation week, and milk composition data. Genetic parameters were estimated by restricted maximum likelihood procedure based on repeatability model using Wombat program. Results Elevated milk BHBA and Ac concentrations were observed during the early lactation under the negative energy balance. Milk FPR tends to decrease with the decreasing ketone body concentrations. Heritability estimates for milk BHBA, Ac, EB, and FPR ranged from 0.09 to 0.14, 0.23 to 0.31, 0.19 to 0.52, and 0.16 to 0.42 respectively at parity 1, 2, 3, and 4. The overall heritability for BHBA, Ac, EB and FPR were 0.29, 0.32, 0.58, and 0.38 respectively. A common pattern was observed in heritability of EB and FPR along with parities. Conclusion FPR and EB can be suggested as potential predictors for risk of hyperketonemia. The heritability estimates of milk BHBA, Ac, EB, and FPR indicate that the selective breeding may contribute to maintaining the milk ketone bodies at optimum level during early lactation.
Photoperiod is the most important environmental cue for the regulation of flowering time, a highly important agronomic trait for crop productivity. To help elucidate the photoperiodic control of flowering in Brassicaceae, we performed microarray experiments using species-specific oligo-arrays with the long day (LD) plant Arabidopsis thaliana and the photoperiod-independent plant rapid cycling Brassica rapa (RCBr). Enrichment analysis of the gene ontologies of differentially expressed genes (DEGs) did not uncover clear differences in gene expression between photoperiod-dependent and -independent plants. Most genes that were up-regulated under LD conditions in Arabidopsis were also up-regulated in RCBr. In addition, most genes associated with light signaling and the circadian clock showed similar expression patterns between Arabidopsis and RCBr, implying that most components known to be key regulators in the photoperiodic flowering pathway are not responsible for the photoperiod independence of RCBr. Nonetheless, we identified one clock-associated gene, PSEUDO-RESPONSE REGULATOR9 (PRR9), as a candidate gene explaining the photoperiod independence of RCBr. The mechanism underlying the role of PRR9 in photoperiodic control and genomic polymorphisms should be further explored using different B. rapa species.
Ketogenesis is the production of ketone bodies, which provide energy when the body lacks glucose. Under ketogenic conditions, the body switches from primarily carbohydrate to fat metabolism to maintain energy balance. However, accumulation of high levels of ketone bodies in the blood results in ketosis. Treating ketosis with natural substances is preferable, because they are unlikely to cause side-effects. Momilactone B is an active compound isolated from Korean rice. Based on previous studies, we hypothesized that momilactone B could inhibit ketosis. We constructed an in vitro ketosis model by glucose starvation. We used this model to test the anti-ketosis effects of momilactone B. A primary target for treating ketosis is angiopoietin-like-3 (ANGPTL3), which modulates lipoprotein metabolism by inhibiting lipoprotein lipase (LPL), a multifunctional enzyme that breaks down stored fat to produce triglycerides. We showed that momilactone B could regulate the ANGPTL3-LPL pathway. However, a strong anti-ketosis candidate drug should also inhibit ketogenesis. Ketogenesis can be suppressed by inhibiting the expression of 3-hydroxy-3-methylglutaryl-CoA synthase-2 (HMGCS2), a mitochondrial enzyme that converts acetyl-CoA to ketone bodies. We found that momilactone B suppressed the expression of HMGCS2 through the increased expression of STAT5b. We also elucidated the relationship of STAT5b to ANGPTL3 and LPL expression.
This study was conducted to estimate the genetic parameters of β-hydroxybutyrate (BHBA) and acetone concentration in milk by Fourier transform infrared spectroscopy along with test-day milk production traits including fat %, protein % and milk yield based on monthly samples of milk obtained as part of a routine milk recording program in Korea. Additionally, the feasibility of using such data in the official dairy cattle breeding system for selection of cows with low susceptibility of ketosis was evaluated. A total of 57,190 monthly test-day records for parities 1, 2, and 3 of 7,895 cows with pedigree information were collected from April 2012 to August 2014 from herds enrolled in the Korea Animal Improvement Association. Multi-trait random regression models were separately applied to estimate genetic parameters of test-day records for each parity. The model included fixed herd test-day effects, calving age and season effects, and random regressions for additive genetic and permanent environmental effects. Abundance of variation of acetone may provide a more sensitive indication of ketosis than many zero observations in concentration of milk BHBA. Heritabilities of milk BHBA levels ranged from 0.04 to 0.17 with a mean of 0.09 for the interval between 4 and 305 days in milk during three lactations. The average heritabilities for milk acetone concentration were 0.29, 0.29, and 0.22 for parities 1, 2, and 3, respectively. There was no clear genetic association of the concentration of two ketone bodies with three test-day milk production traits, even if some correlations among breeding values of the test-day records in this study were observed. These results suggest that genetic selection for low susceptibility of ketosis in early lactation is possible. Further, it is desirable for the breeding scheme of dairy cattle to include the records of milk acetone rather than the records of milk BHBA.
The objective of this study was to estimate the environmental factors affecting milk ${\beta}$-hydroxybutyric acid (BHBA) and acetone (Ac) concentrations in Holstein cattle. A total of 264,221 test-day records collected from the Korea Animal Improvement Association (KAIA) during the period of 2012 to 2014 were used in this study. Analysis of variance (ANOVA) was performed to determine the factors significantly affecting ketone body concentrations. Parameters considered in the model were season of test, season of calving, parity, lactation stage, and milk collecting time (AM and PM). According to the ANOVA, the $R^2$ for milk BHBA and Ac were 0.5226 and 0.4961, respectively. 'Season of test' showed a considerable influence on ketone body concentration. Least square (LS) means for milk BHBA concentrations was the lowest ($39.04{\mu}M$) in winter while it increased up to $62.91{\mu}M$ in summer. But Ac concentration did not significantly change along with 'season of test'. The means of milk BHBA and Ac concentrations were high at first lactation stage, low around second lactation stage, and then gradually increased. Cows milked in the morning had lower mean BHBA and Ac concentrations ($48.49{\mu}M$ and $121.69{\mu}M$, respectively) in comparison to those milked in the evening ($53.46{\mu}M$ and $130.42{\mu}M$, respectively). The LS means of BHBA and Ac slightly increased over parities. These results suggest that proper maintenance of milk collection, herd management programs, and evaluation of ketone body levels in milk should be considered for the efficient management of resistance to ketosis.
Carcass and price traits of 72,969 Hanwoo cows, bulls and steers aged 16 to 80 months at slaughter collected from 2002 to 2013 at 75 beef packing plants in Korea were analyzed to determine heritability, correlation and breeding value using the Multi-Trait restricted maximum likelihood (REML) animal model procedure. The traits included carcass measurements, scores and grades at 24 h postmortem and bid prices at auction. Relatively high heritability was found for maturity (0.41±0.031), while moderate heritability estimates were obtained for backfat thickness (0.20±0.018), longissimus muscle (LM) area (0.23±0.020), carcass weight (0.28±0.019), yield index (0.20±0.018), yield grade (0.16±0.017), marbling (0.28±0.021), texture (0.14±0.016), quality grade (0.26±0.016) and price/kg (0.24±0.025). Relatively low heritability estimates were observed for meat color (0.06±0.013) and fat color (0.06±0.012). Heritability estimates for most traits were lower than those in the literature. Genetic correlations of carcass measurements with characteristic scores or quality grade of carcass ranged from −0.27 to +0.21. Genetic correlations of yield grade with backfat thickness, LM area and carcass weight were 0.91, −0.43, and −0.09, respectively. Genetic correlations of quality grade with scores of marbling, meat color, fat color and texture were −0.99, 0.48, 0.47, and 0.98, respectively. Genetic correlations of price/kg with LM area, carcass weight, marbling, meat color, texture and maturity were 0.57, 0.64, 0.76, −0.41, −0.79, and −0.42, respectively. Genetic correlations of carcass price with LM area, carcass weight, marbling and texture were 0.61, 0.57, 0.64, and −0.73, respectively, with standard errors ranging from ±0.047 to ±0.058. The mean carcass weight breeding values increased by more than 8 kg, whereas the mean marbling scores decreased by approximately 0.2 from 2000 through 2009. Overall, the results suggest that genetic improvement of productivity and carcass quality could be obtained under the national scale breeding scheme of Korea for Hanwoo and that continuous efforts to improve the breeding scheme should be made to increase genetic progress.
The Korean native cattle, Hanwoo, is the most popular breed of beef cattle in Korea. However, the reproductive performance data are limited although reproduction is one of the most economically and biologically important in beef production. Therefore, this study was undertaken to investigate reproductive performance parameters including calving interval, parity for life time production. Data collected from 206,827 calvings were analyzed. There were no significant differences in calving interval and gestation days as parity increased from 2nd and 13rd parity cow, from spring to winter. However, we found a dramatic increase in calving interval after year 2000. About 1 month were increased per year ( y = 30.578x + 344.45 R2 = 0.9157). Interestingly, we observed that parities for life time can be affected by birth weight. Calves with 23 kg at birth showed highest parities, 3.4±2.0 times. In summary, this study provides valuable data on reproductive performance of Hanwoo and the data presented here can be used as a standard target for optimising and enhancing reproductive performance.
This paper sketches the strategies and designs for monitoring system of swine genetic improvement. The system should reflect every side of pig production. The system leads us to assess the efficiency of pig production and the scope of the system includes not only nucleus, multiplying and commercial herds, but also packing and processing sectors. For more accurate statistics, data for this monitoring system must be collected from all above mentioned areas, but not by random sampling. Futhermore, data analysis results including seedstocks and distribution information of genetic trend should be included in the system. The schema of knowledge database system could be employed in the system. The monitoring system in the final destination would unify the systems derived from various sources and provide any solution in swine industry including pig breeding.
