The centrosome is composed of pericentriolar material (PCM) surrounding a pair of perpendicular cylinder centriole. Centrosome is known as major microtubule organizing center (MTOC) in mammalian cells and plays an important role in cell division. The centrosome is replicated in a semi-reserved form. The centrosome reduction occurs during the process of spermiogenesis and oogenesis. The Sperm and oocyte retain the part of the centrosome and reconstitute the functional integrity centrosome after fertilization. The defects of sperm centrosome will lead to maleinfertility, and the aging of oocyte also results in abnormal spindle structure due to the defects of centrosomal protein and causes abnormalities in fertilization and early embryonic development. The centrosome structure and function crucial in human fertilization and embryo development, and also show great significances in human assisted reproduction.
Objective Explore the change of IL-1β and IL-18 expression in pulmonary hypertension induced by monocrotaline.Methods Divide the mouses into two groups, control group and experimental group (n=10).Establish rats pulmonary hypertension model induced by monocrotaline.Detect the model by ultrasound, myocardial cells HE dyeing and tunnel test;ELISA was used to detect the serum biological markers NF-κB, COX2, IL-6, IL-1β, TNF-α and NO;Immunohistochemical was used to detect the expression level of IL-1β and IL-18 in the lung tissue;the protein change of NLRP3 in the lung tissue was detected by Western blot.Results Serum biological markers of NF-κB, COX2, IL-6, IL-1β, TNF-α and NO are significantly increased in PAH rats(P<0.05);The expression of IL-1β, IL-18 in the lung tissue increased obviously(P<0.05);The NLRP3 protein expression was significantly higher in experimental group.Conclusions Changes of NLRP3 effect increase expression of IL-1β and IL-18and which may play an important role in pulmonary hypertension induced by monocrotaline.
Objective to find a appropriate marker for the early diagnosis of pulmonary hypertension in serum .Methods 60 rats were randomly divided into normal control group (n=12) and experimental group (n=48) .The experimental group was injected with MCT to make the pulmonary artery hypertension model of rats while the control group was injected with to make the same a-mount of normal saline .NF-κB、TNF-α、COX-2、PGE2、MMP-1 and iNOS levels of serum were determined by ELISA in all groups . Results the results showed that the model was successful .The concentration of NF-κB、TNF-α、COX-2、PGE2、MMP-1、iNOS in 2 ,4 ,6 weeks had statistical significance compared to the control group (P<0 .05) .And the 2 ,4 ,6 weeks in the the experiment were in the the early stage of PAH development .We found that the specificity of NF-κB ,TNF-α,PGE2 ,iNOS was the most obvious in the the early .Conclusion NF-κB ,TNF-α,PGE2 ,iNOS ,in the rat PAH model can reflect the early pathological changes of PAH ,which has important value for the research of early PAH .
【Objective】 To detect and test the germicidal efficacy of water solution of sodiumpercarbonaxe(clean household oxygen-containing particles of Oxy-Clean particles) on staphyloccocus aureus sosenbach.【Methods】 The suspension quantitative bactericidal test was applied.【Results】 According to the test methods for evaluating daily chemical products in antibacterial and bacteriostatic efficacy of QB/T 2738~2005,the water solution of clean household oxygen-containing particles took effect with its density of above 0.2% for more than 15 minutes or with the density of above 0.5% for more than 5 minutes.The bactericidal rate reached more than 90%.It can be judged as an A1-level disinfector by its bacterioustatic efficacy on staphyloccocus aureus sosenbach.【Conclusion】 The water solution of sodiumpercarbonaxe(multi-functional washing oxygen-containing particles) has excellent germicidal efficacy on staphyloccocus aureus sosenbach.
【Objective】 To understand and evaluate the disinfection effect of Granular Oxygen-containing Cleaner for Household.【Methods】 Suspension quantitative method,field simulation experiment,field experiment and stability experiment were used to carry out experimental studies.Sterilization rate was used to evaluate the disinfection effect.【Results】 Suspension quantitative method showed 1.00% Granular Oxygen-containing cleaner for household could kill 100.00% staphylococcus aureus in 10 mins and pseudomonas aeruginosa in 5 min.The sterilization ability was stronger along with higher temperature,pH value and density.Organic had no effect on 1.00% Granular Oxygen-containing cleaner for household.Field simulation experiment showed 1.00% Granular Oxygen-containing cleaner for household could kill 100.00% staphylococcus aureus on the tabletop in 15 min,while 0.50% Granular Oxygen-containing cleaner for household could kill 100.00% pseudomonas aeruginosa on the tabletop in 10 min.1.00% Granular Oxygen-containing cleaner for household was daubed on chair,refrigeratory clapboard,tabletop for 10 mins,the average sterilization rate were 99.86%,99.49%,99.55%,respectively.Granular Oxygen-containing cleaner for household was put in 54℃ ncubator for 14 d the reduction rate of the effective constituent was 4.12%.【Conclusion】 The performance of ganular Oxygen-containing cleaner for household is steady.Granular Oxygen-containing cleaner for household has a good sterilizing effect on saphylococcus aureus,peudomonas aeruginosa and natural bacteria.It can be used for disinfection of tabletop,hair and refrigeratory clapboard.
