Identification of irradiated fruits is of paramount importance to address the limitation of irradiation technology because of varying national and international regulations. Thermoluminescence (TL) analysis was carried out to identify oranges irradiated with γ-ray, electron beam and X-ray. Non-irradiated samples exhibited background TL signals, but all the irradiated samples showed defined TL glow curves characterized by a prominent peak at 158–163 °C. Characterizations of the irradiated standard minerals showed that feldspars were the major contributors to the TL emission and stable TL signals revealed a successful detection of irradiated oranges even after a prolonged storage.
배 농축액의 유가식 배양을 통해 영양원 없이 산도 12% 이상의 고산도 식초를 제조하면서 발효단계(Stage 1-4) 및 초기 알코올농도(6-9%)에 따른 품질특성 변화를 확인하였다. 환원당, 유리아미노산, 총 페놀 함량, 총 플라보노이드 함량 및 라디칼 소거능은 1차 초산발효한 일반산도 식초에 비해 2차 초산발효한 고산도 식초에서 증가하였고, 이는 유가식으로 첨가된 feeding 알코올의 영향으로 사료되었다. SPME/GC-MS 분석을 통한 20여종의 휘발성분은 식초의 산도에 따라 함량의 차이를 나타내었고, 초기 알코올농도의 증가에 따라 자극적인 향의 acid류 함량이 다소 증가하였다. 이상의 결과, 초기알코올 농도 6-7% 조건에서 유가식 배양으로 제조된 고산도 배식초는 일반산도 식초에 비해 우수한 품질을 나타내었고, 이에 관능특성 및 대규모 생산에 대한 연구가 필요한 것으로 사료되었다. High-acidity vinegar was manufactured using pear concentrate by fed-batch fermentation without additional nutrients, and the physicochemical properties and volatile components were investigated at different fermentation stages (Stages 1-4) and at various initial alcohol concentrations (IAC; 6-9%). The levels of reducing sugar, free amino acids, total phenolic content, total flavonoid content, and radical scavenging ability increased slightly during Stage 4 (high-acidity vinegar), which was affected by alcohol feeding. The contents of approximately 20 types of volatile compounds differed between the moderate- and high-acidity vinegar samples, as determined by solid-phase microextraction/gas chromatography-mass spectroscopy. The level of acetic acid in high-acidity vinegar increased according to the initial alcoholic content applied. The high-acidity vinegar produced by fed-batch culture at an IAC of 6-7% showed improved physicochemical and volatile properties as compared to the moderate-acidity vinegar.
Imported Valencia oranges were evaluated to determine the relationship between the fruit size and its quality. The orange size was classified into three groups on a commercial basis: small (140~160 g/113±5 fruit/box), medium (190~220 g/88±5 fruit/box), and large (250~280 g/72±5 fruit/box). The physicochemical and sensory properties were analyzed to evaluate the orange quality. No significant difference in the peel thickness and flesh ratio was detected across the fruit sizes. The juice yield of the medium-sized orange and the TSS/TA ratios of the medium-sized and large oranges showed the highest value, respectively (p<0.05). The large orange had the highest vitamin C content, which was positively correlated with both its DPPH and ABTS radical scavenging abilities (p<0.05). In the sensory evaluation of the fruits and their juice, the scores for sourness and overall preference were significantly higher in the medium-sized and large oranges than in the small ones. The total soluble solids, total acidity, TSS/TA ratio, and reducing sugar content were significantly related to the sensory properties. Finally, the quality and sensory properties were considered superior in the medium-sized and lager Valencia oranges. Further studies on the effects of the variety and origin of Valencia oranges on their quality and sensory properties are required.
Gamma-irradiated (0–10kGy) dried mushrooms (Lentinus edodes) powders were mixed at different ratios (1–10%) in the non-irradiated samples and investigated using photostimulated-luminescence (PSL), electron spin resonance (ESR) and thermoluminescence (TL) techniques. The PSL results were negative for all samples at 1% mixing ratio, whereas intermediate results were observed for the samples containing 5% or 10% irradiated component with the exception (positive) of 10% mixing of 10kGy-irradiated sample. The ESR analysis showed the presence of crystalline sugar radicals in the irradiated samples but the radiation-specific spectral features were absent in the mixed samples. TL analysis showed the radiation-specific TL glow curves; however, the complicated results were observed at 1% mixing of 2 and 5kGy-irradiated samples, which required careful evaluations to draw the final conclusion about the irradiation status of the samples. TL ratios could only confirm the results of samples with 5% and 10% mixing of 10kGy, and 10% mixing of 5kGy-irradiated components. SEM-EDX analysis showed that feldspar and quartz were major contaminating minerals, responsible for the radiation-specific luminescence characteristics.
본 연구는 저장성이 낮은 양파를 이용하여 양파의 발효특성을 조사하였다. 양파 발효의 화학적 특성(잔류 당 함량, pH, 산도 및 알콜 함량)과 관능적 특성(색상, 향, 맛 및 전반적인 기호도)은 반응표면분석을 통해 모니터링 되었다. 알콜 함량에 대한 반응표면분석 결과, 회귀식의 $R^2$가 0.9620으로 1% 이내에서 유의성이 인정되었으며, 예측된 정상점은 최대점으로 발효온도 $29.27^{\circ}C$, 발효시간 103.43 hr 및 초기 당도 $27.52^{\circ}Brix$였으며, 최대점까지는 초기 당 함량과 발효시간이 경과할수록 증가하였다. 전반적인 기호도에 대한 회귀식의 $R^2$는 0.9060으로 5% 이내에서 유의성이 인정되었으며, 예측된 정상점은 안장점으로, $39.27^{\circ}C$, 57.28 hr 및 $22.14^{\circ}Brix$의 조건에서 최대값 6.64점으로 예측되었다. 알콜 함량과 전반적인 기호도 모두를 만족시켜 주는 조건을 구하기 위해 반응표면 중 최적 반응표면만을 superimposing 하여 양파 발효주의 최적 발효조건을 설정한 결과, 발효온도 $28{\sim}32^{\circ}C$, 발효시간 80~90 hr 및 초기 당도 $20{\sim}25^{\circ}Brix$로 예측되었다. Central composite design along with response surface methodology (RSM) was applied to improve the fermentation process in onion (Allium cepa) wine production. The effects of different fermentation parameters (time, temperature, and initial sugar content) were found to be significant with respect to the physicochemical and sensory properties of wine. The maximum score for the alcoholic content was obtained at $29.27^{\circ}C$ fermentation temperature, 103.43 h fermentation time, and $27.52^{\circ}Brix$ initial sugar content. The maximum score for overall palatability was obtained at $39.27^{\circ}C$ fermentation temperature, 57.28 h fermentation time, and $22.14^{\circ}Brix$ initial sugar content. The coefficients of determination ($R^2$) were 0.9620 and 0.9060 for alcoholic content and overall palatability, respectively. The ranges of the optimum fermentation conditions ($28{\sim}32^{\circ}C$, 80~90 hr, and $20{\sim}25^{\circ}Brix$) were obtained by superimposing the response surfaces with regard to the alcoholic content and overall palatability of onion wine.
총 산도 10% 이상의 시판 고산도 식초를 원료(사과농축액, 맥아엑기스, 액상포도당 및 현미당화농축액)와 산도(2배 및 3배)에 따라 구입하여 이화학적 품질 및 항산화 특성을 확인하였다. 총산 함량은 2배 식초(12.10~13.41%)와 3배 식초(18.15~18.36%) 사이 유의적인 차이를 나타내었고, 식초의 당도 및 환원당 함량은 각각 7.00~10.80°Brix 및 1.32~3,885.90 mg% 범위로 확인되었으며 일정한 경향은 관찰되지 않았다. 고산도 식초의 유리당으로 fructose 및 glucose가 주로 확인되었고, 2배 식초의 경우 3배 식초에 비해 종류와 함량 측면에서 비교적 우수한 것으로 분석되었다. 식초의 품질지표인 acetic acid는 모든 식초에서 7,171~14,762 mg% 함량으로 분석되었고, oxalic acid의 경우 시판 고산도 식초에서는 확인할 수 없었다. 맥아엑기스를 사용
국내 다소비 건포류인 오징어채, 쥐포, 멸치를 대상으로 감마선 조사에 의한 이화학적 품질 특성 및 조사여부 확인 시험을 실시하였다. 모든 시료에서 조사선량의 증가에 따라 기계적 색도의 큰 변화는 관찰되지 않았지만 전반적 색차는 다소 증가하였다. 갈색도의 경우 오징어채와 쥐포는 수용성 갈색도가, 멸치는 지용성 갈색도가 높게 확인되었고, 쥐포의 갈색도는 조사선량에 따라 다소 증가하였다. TBA 값은 지방의 함량이 비교적 높은 오징어와 멸치의 경우 조사선량에 따라 증가하는 경향을 나타내었다. 광자극발광(PSL) 분석 결과, 멸치를 제외한 모든 조사시료에서 음성(-)과 중간값(M)을 나타냄에 따라 스크리닝 판정에 적합하지 않는 것으로 확인되었다. 그러나 열발광(TL) 분석에서는 모든 조사시료가 특유의 peak를 나타내었고, 강도 또한 선량에 비례적으로 증가하였으며, TL ratio 역시 기준 값에 만족하여 TL에 의한 조사여부 확인이 가능하였다. 관능평가 결과, 모든 시료에서 조사선량 간 유의적인 차이는 나타나지 않았다. Dried fish products, such as seasoned cuttlefish, anchovy, and filefish, were analyzed to investigate the physicochemical quality and luminescence properties following gamma-irradiation. The overall color difference slightly increased in all irradiated samples, showing significant changes in Hunter's L, a, and b color values. Cuttlefish and filefish showed higher values in hydrophilic browning, whereas anchovy showed a higher value in lipophilic browning. The brown color intensity of filefish showed a dose-dependent increase. The thiobarbituric acid (TBA) value significantly increased in irradiated samples of cuttlefish and anchovy, which have relatively high lipid contents. Irradiated samples, except anchovy, were not suitable for screening using photostimulated luminescence (PSL) because only negative or intermediate values were observed. Thermoluminescence (TL) measurements were suitable for all fish samples, i.e., irradiation-specific glow curves were obtained, signal intensity increased, and the TL ratio fulfilled the criterion. There was no significant difference among the irradiated samples with regard to the sensory properties.
The physiochemical properties of commercial fruit vinegars were compared according to fermentation methods. Type A vinegars were synthesized through acetic acid fermentation while Type B vinegars were produced using both alcohol and acetic acid fermentation serially. There were differences from using these fermentation methods; Type A vinegars had a lower pH and slightly higher total acidity than Type B vinegars. The content of total sugar and reducing sugar were relatively higher in Type B vinegars, which showed a higher content of the free sugars (fructose, glucose, sucrose, and maltose). The intensity of brown color and Hunter`s a and b values were also high in Type B vinegars. In contrast, the content of organic acids was higher in Type A vinegars, which were mainly composed of acetic, tartaric, malic, and succinic acid. We were also able to estimate the fruit juice content of vinegars through its content of organic acids. Type B vinegars contained a higher total phenolics and flavonoids content than Type A vinegars, and showed a higher DPPH radical scavenging activity.
The quality of commercial cider vinegars was compared according to their acidity levels (low, general, double strength and triple strength). The pH, reducing sugar content and brown color intensity decreased, while the total acidity increased with the increasing acidity levels, which may be resulted from difference in formulation and manufacturing procedures. The free sugars were mainly composed of fructose and glucose, which were the highest in low acidity vinegar, followed by double strength, general, and triple strength acidity vinegars. Acetic acids and malic acids were identified as the major organic acids. The citric acid concentration was the highest in triple strength vinegars. The contents of total phenolics and flavonoids were the highest in low acidity vinegars, which indicated their dependence on the apple juice content. DPPH and ABTS radical scavenging abilities were the highest in low acidity vinegars, that showed high amounts of antioxidants.
Fresh (raw roots), white (dried), and red (steamed-drid) ginseng samples were gamma-irradiated at 0 to 7 kGy. Electron spin resonance (ESR) technique was used to characterize the irradiation status of the samples, targeting the radiation-induced cellulose radicals after different sample pretreatments. All non-irradiated samples exhibited a single central signal (g=2.006), whose intensity showed significant increase upon irradiation. The ESR spectra from the radiation-induced cellulose radicals, with two side peaks (g=2.0201 and g=1.9851) equally spaced (±3 mT) from the central signal, were also observed in the irradiated samples. The core sample analyzed after alcoholic-extraction produced the best results for irradiated fresh ginseng samples. In the case of irradiated white and red ginseng samples, the central (natural) and radiation-induced (two-side peaks corresponding to cellulose radical) signal intensities showed little improvement on alcoholic-extraction. The water-washing step minimized the effect of Mn(2+), but reduced the intensity of side peaks making them difficult to indentify. The effect of different origins was negligible, however harvesting year showed a clear effect on radiation-induced ESR signals.
건포도(Thailand산), 건망고(Philippine산), 건무화과(Iran산)를 0~5 kGy로 감마선 조사한 후 총균수 및 기계적 색차를 측정하고 조사여부 판별가능성을 확인하였다. 총 균수 측정 결과 3 kGy 이상으로 조사된 시료에서는 대부분 검출할 수 없는 수준으로 나타나 미생물학적으로 안전함을 알 수 있었다. 건조과일의 색차 측정에서, 건망고의 색 변화가 방사선조사에 가장 민감하였다. PSL 측정 결과, 건포도의 경우 비 조사시료는 음성으로, 조사시료는 중간 값으로 확인되었고 건망고의 경우 모든 시료가 음성으로 확인되었으며, 무화과의 경우 5 kGy 조사시료만이 양성으로 확인되어, 건조과일의 경우 PSL 측정법은 screening 방법으로의 적용가능성이 낮았다. TL 측정 결과 건무화과의 경우 조사시료는 150~200℃ 사이에서 특유의 발광곡선을 나타내었고, 신호 강도 또한 조사선량에 의존적으로 증가하여 TL 적용이 가능하였다. ESR 측정 결과 건포도와 건망고의 경우 조사시료에서 multi-component 신호를 보여주었고, 신호 강도 역시 조사선량에 따라 유의적으로 증가하여 ESR 적용이 가능하였다.
Photostimulated luminescence (PSL) and thermo-luminescence (TL) analyses were conducted to detect irradiated kiwifruits. Samples were irradiated with Co-60 gamma-rays at 0-2 kGy. The freeze-dried kiwifruit peel showed 309 photon counts (PCs) for nonirradiated samples that accounted for less than the lower threshold value (700 counts/60 s, negative) and above 9306 PCs for 1 and 2 kGy-irradiated samples, which was higher than the upper threshold value (5000 counts/60 s, positive). However, PSL signals of irradiated samples remarkably decreased after 6 weeks of storage. The TL measurement using minerals isolated from the whole kiwifruit surface revealed a glow curve (TL 1) with a low intensity at 200-300 degrees C in nonirradiated samples but with a higher intensity at around 180 degrees C in irradiated samples at 1 kGy or more. The TL ratios, integrated areas of TL 1/TL 2 that was measured after 1 kGy re-irradiation for the TL 1-tested minerals, were less than 0.1 in nonirradiated samples and higher than 0.1 in irradiated ones and could verify TL 1 results. The inorganic dust minerals used were mainly composed of feldspar and quartz.
The detection characteristics of gamma-irradiated soybeans produced in Korea and China were investigated by thermoluminescene (TL), electron spin resonance (ESR), and DNA comet assay. The TL glow curves were shown at around for irradiated soybeans, while that at for the non-irradiated one. The normalization with a re-irradiation step at 1 kGy could verify the above detection results. The Korean soybean showed higher glow curves than Chinese did. The ESR spectroscopy for husks of irradiated soybeans revealed specific signals (g = 2.02374, 1.98715) derived from cellulose radical, which intensities were proportional to irradiation does, with the higher peaks in Chinese sample than Korean one. The DNA comet for the non-irradiated sample showed no or little tails, while those for irradiated samples above 0.5 kGy were remarkably changed in their length, size, and concentration, thus resulting in distinguishing non-irradiated from irradiated samples. As a result, TL, ESR, and DNA comet determinations were found suitable for the detection of irradiated soybean at 0.5 kGy or more, and negligible differences were observed between Korean and Chinese origins in their detection characteristics.
This study was performed to investigate chemical and functional properties of Glechoma hederacea leaves in respect to its potential use as food material or as a medicinal herb. The chemical compositions on a dry basis were 20.38% in protein, 3.96% in fat, 59.58% in carbohydrate, 15.78% in ash, 5.36% in reducing sugar, 14.11% in total sugar and 0.26% in polyphenol, respectively. The free sugars were mainly composed of glucose, fructose and sucrose. In fatty acids compositions, linolenic acid showed the highest concentration at 45%, while the ratios of saturated to unsaturated fatty acids were 1 : 1.91. Seventeen kinds of total amino acids were determined, with the highest concentration (2,465.71 ㎎%) of glutamic acid. Among the free amino acids, proline showed the highest concentration (260.09 ㎎%), followed by glutamine, α-amino adipic acid, glutamic acid and valine. The contents of major minerals were 647.32 ㎎% in Na, 597.53 ㎎% in K and 239.75 ㎎% in Ca. The antioxidative activity of 10% water extract was similar to that of 50 ppm tocopherol. The nitrite scavenging ability reached the highest level at pH 1.2 and the lowest at pH 6.0.
