Abstract The influence of 10-4 divalent cations on the uptake of labelled Zn in the concentration range 10-5 to 5 × 10-5 ᴍ into the interior of Chlorella fusca at 30 °C was measured during 75 min. The Zn adsorbed on the surface or contained in the free space was removed by washing with EDTA. Corrections were applied for the loss in concentration due to surface adsorption, etc. The results were consistent with competitive inhibition of Zn uptake by the foreign ions. No allosteric inhibition was found. From the Lineweaver-Burk diagram, for uninhibited Zn, Kᴍ was found as 7 × 10-6 ᴍ and νmax as 8.3 × 10-9 mol · min-1 · g-1 algae. The inhibitor constants were: Mg 1.3 × 10-5, Ca 4.3 × 10-5, Co 4.3 × 10-5, Ni 4.3 × 10-5, Cd 1.3 × 10-5 and Pb 9.4 × 10-7 ᴍ. The values given for Kᴍ and the inhibitor constant refer to the velocities of uptake after 45 min.
In experiments on the prebiotic formation of nitric oxides, anoxic mixtures of N2 and water vapour were sparked in contact with phosphate buffer solutions at various pH values. Nitrite was found in the aqueous phase, and nitrate grew from it, presumably by reaction with H2O2. In acid solutions, these anions were reduced and destroyed by Fe2+, and the same was true of nitrite in solutions kept at a pH value similar to that of the contemporary ocean (8.2) with HEPES buffer. Nitrate was not destroyed in short-term experiments, but as in sparking nitrate is formed only via nitrite, neither anion could accumulate. In further sparking experiments with alkaline sulphide, both nitrite and nitrate were reduced entirely. It is concluded that it is unlikely that the primeval ocean contained appreciable concentrations of nitrite or nitrate either at the reducing or at the redox-neutral stage.
Cadmium ist erst in der Industriegesellschaft in nennenswertem Ausmag in unserer Urnwelt erschienen; es wirkt auf viele Organismen als starkes Gift [1]. Wegen der chemischen )~hnlichkeit darf man annehmen, dab in Zellen Cd mit Zn konkurriert. Wir haben die Wechselwirkung der beiden Elemente bezfiglich des Wachstums von Chlorella geprfift, weft fiber die Mechanismen der Aufnahme yon Zn und anderen kationischen Spurenelementen durch diese Grfinalge Untersuchungen vorliegen [2]. Chlorella fusca (=pyrenoidosa, Stamm 2118 b) wurde asynchron und photoautotroph in einer thermostatierten S/iule [3] bei 30~ in Luft+3% CO2 (,,Nfihrgas") gezfichtet. Nfihrl6sungen von Ruppel [4], 10-Sm an ZnSO4, wurden verwendet, nur wurde der Phosphatpuffer durch/iquimolare KH2PO4L6sung ersetzt. Zur Bestimmung der Algenvolumina in Proben wurden Zentrifugenr6hrchen mit angeschmolzener Kapillare verwendet. Die Wachstumsversuche wurden bei 30 ~ in FlachgeffiBen aus Glas [5] unter Belichtung durchgeffihrt. Chlorella wurde in einer Suspensionsdichte von 1,2 ml gepackter Zellen/1 (ca. 0,25 g Trockengewicht/1) eingesetzt und zunfichst 40 h in Licht-Luft, aber ohne CO2, in den Nfihrl6sungen vorkonditioniert, in denen der Wachstumsversuch durchgeffihrt wurde. When in the primeval atmosphere ammonia approached exhaustion, bacteria resembling clostridia developed mechanisms for nitrogen fixation. The fixation was continued by the photosynthetic bacteria. In the later, oxidizing, atmosphere the combined activities of the nitrificants and the denitrificants could lead to a large-scale cyclic regeneration of free nitrogen. The possibility of a descent of the nitrificants from hypothetical photosynthetic bacteria, which used ammonia as electron donor, is discussed.
The uptake of labelled thallous ion into the interior of Chlorella fusca is enhanced by light. This is ascribed to active transport by way of the K (and Rb) pump. The Michaelis constants differ between the higher and the lower concentration ranges, but they are similar for Rb and Tl.
A flow method for the measurement of the uptake of labelled substances by plant cells has been developed. Accumulation in the medium of substances released by the plants is thereby avoided, and a more precise determination of the rate of uptake—also of transients—than through the usual, static methods is possible. The time function of the stimulation of the uptake of Rb by Chlorella fusca by light at 30° has been analyzed, and the component believed to represent active transport separated from other components that have shorter time constants; at 3° the active component is much reduced.
HeLa-Krebszellen wurden in einem Nährmedium gezüchtet, das am Kohlenstoff markiertes Leucin enthielt, sodann wurden die löslichen Proteine aus den Zellen durch Druck-Ultrafiltration gewonnen und elektrophoretisch in Papier oder Stärkegel aufgetrennt. Die Proteine im Pherogramm wurden nach Anfärbung densitometrisch sowie nach Verbrennung der Fraktionen zu Kohlendioxyd radiometrisch (Gaszählrohr) bestimmt. Beide Methoden ergaben breite Verteilungen, die sich von jener der Serumproteine außerordentlich stark unterscheiden. Dennoch ergab die immunologische Prüfung, daß HeLa-Zellen einige Antigene mit menschlichem Serum gemeinsam haben. Diese Prüfung wurde einerseits nach der immunoelektrophoretischen Technik, anderseits auch radio-immunologisch — nämlich durch Bestimmung der Verteilung des Radiokohlenstoffs zwischen Lösung und Niederschlag nach Reaktion radioaktiver HeLa-Proteine mit Antiserum gegen menschliches Serum — durchgeführt.
Bei der Züchtung von HeLa-Krebszellen in tritiumhaltiger Nährlösung während 48 h wurde festgestellt, dass die Mitoserate durch viel kleinere Strahlendosen nachweisbar herabgesetzt wird als die radiochemisch gemessene Rate der Proteinsynthese.
A METHOD has been described for detecting and measuring the autonomous production of well-defined proteins by cells in tissue culture1,2. The method is exceedingly sensitive and is applicable even when the cells are cultivated in a ‘natural’ nutrient medium, that is, a medium also containing serum; such media have an enormous excess of pre-existing proteins. The tissue is incubated in presence of a radioactive amino-acid and the whole of the soluble protein is extracted. Thereafter individual proteins are isolated from the mixture and burnt, and the carbon dioxide obtained tested for radioactivity with a gas counter (efficiency nearly 100 per cent)3,4. The identity of a fraction can be confirmed by treatment with specific antiserum, followed by determination of the distribution of the radiocarbon between precipitate and supernatant. Blank experiments show that after incubation of natural medium with radioactive amino-acid without cells practically no radiocarbon is found in protein.
Der Einbau von Radiokohlenstoff aus Glukose, Glykokoll, Alanin und Tyrosin in die löslichen Proteine von Hühnermesenchym-und (menschlichem) HeLa-Tumorgewebe bei Bebrütung mit Nährmedium wurde untersucht. Das Medium bestand aus einer Mischung vonGeyscher Salzlösung, Aszitesflüssigkeit, Nabelstrangserum und Extrakt von Hühnerembryonen. Die Proteine wurden durch Ultrafiltration unter Druck abgeschieden, durch Hochspannungselektrophorese in Stärke-Gel aufgetrennt und nach Verbrennung im Gas-Geiger-Zählrohr auf Aktivität geprüft. Auch gewebefreies Medium nimmt Radiokohlenstoff in Eiweiß auf, doch handelt es sich dabei, wie Kontrolle durch die Ninhydrinprobe ergab, nicht um echten Einbau von Aminosäureresten in peptidischer Bindung. Hingegen bewirkt das Gewebe echten Einbau. Dieser kann auf die Zahl oder Masse der Zellen bezogen werden und ist bei Mesenchym-und Tumorgewebe von der gleichen Größenordnung. Den größten Einbau an Radiokohlenstoff weisen die Albumin- sowie die γ-Globulinfraktion (beide durch das elektrophoretische Verhalten definiert) auf. HeLa nimmt mehr Kohlenstoff aus Glykokoll in γ-Globulin auf als Mesenchym. Da im Gegensatz zu Untersuchungen an Organismen oder Gewebeschnitten mit isoliertem Gewebe gearbeitet wird, besteht keine Möglichkeit der Störung durch Wechselwirkung von Geweben miteinander, insbesondere von Tumorgewebe mit gesundem Gewebe.
Zählrohre konnen mit reiner Kohlensaure unter Drucken von 10 cm bis 40 cm Hg gefüllt werden und geben unter diesen Bedingungen mit einem äußeren Löschkreis ausgezeichnete Konstanzbereiche. Die Bestimmung des Radiokohlenstoffs nach diesem Verfahren ist bequem und außerordentlich empfindlich.
