The phenomenon of passive cutaneous anaphylaxis in guinea pigs was shown to be applicable in the laboratory determination of residual antigenicity in fractionated and chemically treated castor-seed proteins. Six protein fractions obtained by paper-strip electrophoresis of castorseed protein were shown to be antigenic.
Rats fed a low-methionine, low-choline diet containing 1% of gallic acid developed fatty livers. Added choline or methionine prevented the increased liver fat. Pyrogallol, in contrast, did not cause fatty livers. These results are in agreement with the metabolism studies in that gallic acid, but not pyrogallol, is O-methylated in the animal body.
Rats ingesting diets containing 0.5% biphenyl developed nephrotoxic effects including focal tubular dilation in approximately 60 days. Rats returned to control diet for 60 days after ingesting a dietary level of 0.5% biphenyl for 165 days showed a regression of kidney lesions with scar formation. A more sensitive criterion of the renal effects of biphenyl was polyuria, which is probably a defense mechanism response to the irritant action caused by the insoluble biphenyl metabolites passing through the kidney. No evidence of either polyuria or histopathologic damage was observed from the ingestion of 0.1% biphenyl in the diet for 120 days.
The castor seed antigen CB-1A, prepared by the basic lead acetate method of Spieset al., has been subjected to column chromatography on diethylaminoethyl cellulose. Antigenic differences between fractions were found by the, Schultz-Dale technique, which indicated the possible existence of six antigenic or allergenic components.
A water-soluble, heat-stable protein component of castor seed meal was subjected to paper-strip electrophoresis in buffers of different chemical composition, pH values, and ionic strengths. It was shown that phosphate buffer at pH 7.4 to 8.0 and an ionic strength of approximately 0.05 gave a sharp resolution of castor seed proteins into bands which would bind bromophenol blue. Spies' Allergen CB-1A was shown to be resolved into six or more components at pH 8.0. Each major component band was found to be antigenic by passive cutaneous anaphylaxis in guinea pigs that were sensitized with rabbit antiserum to the crude castor seed protein preparation. Five bands were shown to be allergenic to humans.

Dihydroquercetin, which is closely related to quercetin in chemical structure, has been shown to be nontoxic when fed to albino rats at a dietary level of 1 per cent for long periods of time. Chromatographic examination of the urine of two human volunteers before and after the ingestion of 2 grams of dihydroquercetin demonstrated the conversion of the compound to 3,4-dihydroxyphenyl-acetic, m-hydroxyphenylacetic, and 3-methoxy-4-hydroxyphenylacetic acids. These same metabolites are excreted following oral administration of quercetin, or DOPA, to rats, rabbits, or humans.

Data are given on the continued oral ingestion of dried cells of Bacillus megaterium and brewers' yeast by rats for four hundred days. No toxic effects were observed in rats ingesting diets containing up to 20 per cent of B. megaterium or brewers' yeast as judged by growth response, hemoglobin values, organ weights, and histopathological examination of the visceral organs.
The metabolic fate of orally administered rutin or its aglycone, quercetin, has been investigated in rabbits. Evidence for the absorption and metabolism of these compounds is based upon the urinary excretion of degradation products. One of these products has been isolated and identified as 3,4‐dihydroxyphenylacetic acid. The amount isolated accounts for at least 2 5 per cent of the quercetin administered.

Data are given on the toxicity of quercetin and quercitrin after intravenous administration in rabbits and rats after oral ingestion of diets containing as much as 1 per cent of quercetin or quercitrin for approximately 410 days. The results indicate a low order of toxicity as judged by growth, amount of food consumed, hematological findings, organ weights, and microscopical examination of stained section of visceral organs.
Summary and ConclusionsLong‐term feeding experiments with albino rats involving concentrations of lauryl gallate as high as 0.5% in the daily diet produced no deleterious effects as judged by food intake, rate of growth, gross observations at time of autopsy, and histopathological examination of tissues. Diets containing 1.0% lauryl gallate definitely inhibited growth of male rats and retarded growth of females slightly. Levels of 2.5 and 5% lauryl gallate caused no histopathological changes in various organs but decreased the food intake to a point where starvation was an obvious factor in death of the animals. Lauryl gallate produced skin irritation but did not produce skin sensitization. The property of skin irritation might be of importance to workers engaged in the production and handling of lauryl gallate. There is no evidence to indicate that ingestion of slight amounts, such as might be encountered in edible fats and oils protected against oxidation, would produce harmful effects.
