This study evaluated the relationships between sow parity, prepartum faecal score, farrowing and umbilical cord traits, and reproductive performance. Data were obtained from sows of different parities on commercial farms with records of litter characteristics, farrowing intervals and duration, prepartum faecal scores, and umbilical cord artery and vein diameters. Fourth- and sixth-parity sows produced more total piglets and live-born healthy piglets than first-parity sows, while piglets born to second- to fourth-parity sows had higher live birthweights. Parity did not influence the number of piglets born weak or perinatal mortality rates. Faecal scores of 3.00–3.99 and >4.00 three days prepartum were associated with increased numbers of healthy piglets and reduced numbers of stillborn piglets. Short farrowing intervals (<10 minutes) were numerically associated with higher numbers of live-born piglets and fewer stillborn piglets per litter, although these differences were not statistically significant, and a tendency towards an increased number of piglets born weak was observed. Prolonged farrowing duration was associated with increased perinatal mortality. A higher umbilical cord vein diameter was associated with increased live birthweights and fewer piglets born weak, whereas umbilical artery diameter showed no clear association with reproductive performance. Stillbirths tended to increase with umbilical cord vein diameters of 3.6–4.0 mm. These results indicate that sow parity, prepartum faecal score, farrowing traits, and umbilical cord vein traits are closely associated with reproductive outcomes and perinatal mortality. Optimising gastrointestinal function during the prepartum period and improving farrowing management may contribute to enhanced piglet viability and birthweight. This study evaluated the effects of supplementing a reduced-energy diet with a compound enzyme on sow performance, nutrient utilisation, and faecal characteristics. Thirty healthy multiparous Danish hybrid sows with similar parities and expected farrowing dates were randomly assigned to a control diet, a negative control diet (50 kcal/t less net energy), or a multi-enzyme diet (negative control diet + 150 g/t of a compound enzyme product). The three experimental diets were fed from day 85 of gestation until weaning. During lactation, feed intake tended to be higher in sows fed the control and multi-enzyme diets than in those fed the negative control diet. Body condition parameters and reproductive performance traits (litter weight, birthweight, and numbers of live-born, healthy, and stillborn piglets) did not differ among the dietary treatments. Compared with the negative control group, sows receiving the multi-enzyme diet showed improved faecal characteristics during the periparturient period and lactation, whereas a higher incidence of constipation was observed in sows fed the negative control diet. Apparent digestibility of crude protein and dietary fibre was higher in the multi-enzyme group than in the control and negative control groups, while fat digestibility was higher in the control and multi-enzyme groups than in the negative control group. Piglets from the negative control group tended to show a higher incidence of diarrhoea during lactation. In conclusion, supplementing a reduced-energy diet with a compound enzyme improved nutrient digestibility and faecal characteristics in sows, whereas effects on reproductive performance and piglet growth were limited under the conditions of this study. IntroductionDietary fibre in late gestation may affect sow physiology, colostrum quality, gut microbiota, and piglet performance. This study examined whether increasing fibre levels enhances sow health and reproductive outcomes.MethodsForty pregnant Landrace × Yorkshire sows (parity 2 or 4) were allocated to either a low-fibre (LF, 5.2%) or high-fibre (HF, 7.7%) diet from day 85 of gestation until parturition, after which all sows received a common lactation diet. Sow performance, constipation, nutrient digestibility, colostrum quality, cord blood parameters, and gut microbiota profiles were assessed. Piglet growth performance was recorded until weaning. Correlation analyses were conducted to examine associations between bacterial taxa and physiological indicators.ResultsHF sows showed improved piglet vitality (p < 0.05), higher faecal scores, and lower constipation rates (p < 0.05). Colostrum protein, total solids, IgA, and IgM concentrations were increased in HF sows (p < 0.05). Cord blood glucose and IL-10 were reduced, whereas eNOS was elevated (p < 0.05). Digestibility of crude fibre, ADF, and NDF increased, while protein digestibility decreased (p < 0.05). Gut microbiota analysis indicated increased abundances of Lactobacillus, Lachnospiraceae_XPB1014_group, and Methanobrevibacter, and decreased Treponema, Prevotellaceae_UCG-001, and Escherichia-Shigella. Piglets from HF sows exhibited greater average daily gain and weaning weight (p < 0.05). Beneficial associations were observed for Lactobacillus, Terrisporobacter, and Lachnospiraceae_XPB1014_group with reproductive traits; Phascolarctobacterium and X-Eubacterium_ruminantium_group correlated with piglet growth; while Treponema and Rikenellaceae_RC9_gut_group were negatively associated with constipation.DiscussionHigher dietary fibre in late gestation improved gastrointestinal function, colostrum quality, and piglet growth, likely through enhanced fibre digestibility and favourable modulation of gut microbiota. These findings support increased fibre supplementation to optimise sow and piglet health.
This study aimed to evaluate the effects of silymarin supplementation during late gestation on reproductive performance, haematological parameters, antioxidant capacity, and gut microbiota composition in sows. Twenty parity-4 crossbred sows (Landrace x Yorkshire) were enrolled and randomly allocated in parity blocks to either a control group (CG; n = 10, basal diet) or a silymarin-supplemented group (SIL; n = 10, basal diet + 200 mg/kg silymarin). The experimental period extended from day 85 of gestation to the completion of farrowing. The results demonstrated that dietary silymarin significantly reduced the number of stillbirths (P < 0.05), without exerting a significant effect on the total number and proportion of live-born piglets (P > 0.05). No notable differences were observed in haematological parameters between the two groups (P > 0.05). However, catalase (CAT) activity and total antioxidant capacity (T-AOC) were significantly elevated in the silymarin group (P < 0.05), and superoxide dismutase (SOD) showed a tendency to increase (P = 0.078). High-throughput 16S rRNA sequencing revealed 1 671 unique feature sequences in the silymarin group and 1 073 in the control group, with 1 600 sequences shared between the two groups. A trend towards increased dominance was observed in the silymarin group (P = 0.082), while both the Shannon and Simpson indices tended to decline (P = 0.087; P = 0.082), suggesting a possible reduction in microbial diversity. Principal coordinate analysis (PCoA) of I3-diversity revealed significant structural differences in gut microbiota between the two groups. SIMPER analysis identified Terrisporobacter as the principal genus contributing to these differences. In conclusion, silymarin supplementation during late gestation may enhance reproductive outcomes in sows, potentially through modulation of gut microbial composition and enhancement of systemic antioxidant status.
This study aimed to investigate the effects of silymarin supplementation on sow health, reproductive performance, colostrum quality, immune function, and blood biochemistry during late pregnancy. Twenty parity-4 sows (Landrace × Yorkshire) were included in the study and randomly assigned within each parity group to a control diet (CG, basic diet, n = 10) and a Silymarin diet (SIL, basic diet + 0.2 g/kg SIL, n = 10), from the 85th day of gestation until weaning. Blood and milk samples were collected for analysis. The results demonstrated that silymarin supplementation significantly reduced the number of stillbirths and the stillbirth rate (P < 0.05). Additionally, silymarin supplementation tended to increase the mean body weight of newborn and healthy piglets (P = 0.10; P = 0.08) and improved the intrauterine growth restriction (IUGR) score (P = 0.07). Silymarin supplementation also significantly increased protein content in sow colostrum (P < 0.05) and showed a trend towards higher total solids and lactose content (P = 0.05; P = 0.09). Furthermore, silymarin supplementation significantly increased immunoglobulin G (IgG), IgA, and IgM levels in sow colostrum (P < 0.05), as well as proline content (P = 0.10). It also significantly increased endothelial nitric oxide synthase (eNOS) levels in umbilical cord blood (P < 0.05). Moreover, silymarin supplementation reduced interleukin-6 (IL-6) levels and increased interleukin-10 (IL-10) levels in umbilical cord blood serum (P < 0.05). In conclusion, silymarin supplementation in late-gestation sow diets has no adverse effects on sow or piglet health. Instead, it promotes placental vascular development, reduces inflammation, and decreases stillbirth and IUGR rates. It also enhances the average birth weight of viable piglets and modulates the composition of sow colostrum, enriching key nutrients, immunoglobulins, and amino acids.
Genistin is an isoflavone of soybean, with estrogenic activity. This experiment was conducted to investigate its effect on reproductive performance, antioxidant capacity, and immunity in gestating sows. Seventy-two sows (Landrace × Yorkshire) were selected and randomly divided into two treatment groups (n = 36) based on their backfat thickness, parity and fed with basal diet or supplementation of 150 mg/ kg genistin to the basal diet based on DMI for the entire gestation period. Results showed that dietary genistin supplementation significantly increased the average number of live born per litter (p < 0.05), and tended to increase the number of healthy piglets per litter (p = 0.058), but decreased the average weight of live born per litter (p < 0.05). Dietary genistin supplementation significantly decreased the number of mummified and stillbirths per litter (p < 0.05). Moreover, the average daily feed intake (ADFI) and total feed intake of the gestating sows were also increased in the genistin-supplemented group (p < 0.05). Genistin significantly increased the serum concentrations of catalase (CAT), immunoglobulin A (IgA), IgG, and IgM at 35 days of gestation (p < 0.05). The serum concentrations of interleukin-10 (IL-10) and interferon-γ (IFN-γ) were also increased upon genistin supplementation (p < 0.05). However, genistin supplementation tended to decrease the serum concentrations of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and leptin at 85 days of gestation (p = 0.081 and p = 0.096, respectively). Interestingly, genistin supplementation decreased the transcript abundance of interferon-γ (IFN-γ) and placental imprinting gene H19, but significantly increased the transcript abundance of insulin-like growth factor I (IGF-I) and amino acid transporters such as the sodium-coupled neutral amino acid transporter 2 (SNTA2) and SNAT4 in the placenta (p < 0.05). These results suggested that dietary genistin supplementation during gestation can improve the reproductive performance of sows, which was probably associated with improving of antioxidant capacity and immunity, as well as changes of transcript abundance of critical functional genes in the placenta.

This experiment aimed to study the effects of guanidine acetic acid (GAA) on reproductive performance, lactation performance and blood biochemical indices of sows, as well as the performance of offspring piglets. A total of 20 sows (Landrace × Yorkshire, parity 4) were used. Half of the sows in each parity were fed a control diet (CG; basic diet, n = 10) or GAA diet (basic diet +1 g/kg GAA, n = 10) from the 85th day of gestation until weaning. The study results are presented as follows: Supplementation of GAA from late gestation to lactation did not adversely affect sow feed intake, backfat thickness, or blood routine indexes (p > 0.05). GAA supplementation showed a tendency to increase the number of healthy piglets and their birth activity (p = 0.06; p = 0.08), while significantly increasing the IUGR score of piglets (p < 0.05). GAA supplementation significantly increased colostrum protein content (p < 0.05) and tended to increase daily milk yield in sows (p = 0.07). GAA supplementation increased the level of immunoglobulin A in sow colostrum (p < 0.05) and showed a tendency to increase proline content (p = 0.10). GAA supplementation significantly decreased triglyceride content in sow cord blood (p < 0.05), with no significant effects observed on HDL-C, LDL-C, TC, and GLU (p > 0.05). GAA supplementation significantly increased eNOS levels in sow cord blood (p < 0.05), while showing no significant effects on IL-6 and IL-10 (p > 0.05). GAA supplementation did not significantly affect the α diversity of sow intestinal flora (ACE, Shannon, Chao1, Simpson, observed_otus, pielou_e, and good_cover), but PCoA analysis revealed differences in intestinal flora structure between groups. Additionally, GAA decreased the relative abundance of Sarciha and unidentified_ruminococcaceae and increased the relative abundance of Lactobacillus, Parabacteroides, and Pedobacter in the gut. GAA boosts nitric oxide synthase in sows’ umbilical cord blood, enhancing placental blood vessel development. This improves piglet health and vitality, increases beneficial gut bacteria (Lactobacillus, Parabacteroides, Pedobacter), and raises colostrum protein levels and lactation volume, leading to better piglet growth and performance.

Capsaicin is an alkaloid isolated from peppers and is widely used as spice. Previous studies have shown that capsaicin has pleiotropic effects, such as anti-inflammatory, antioxidant and anticancer effects, but less attention has been paid to its effects on animal intestinal function and intestinal microflora composition. In this paper, the regulatory effect of capsaicin on animal intestinal function and intestinal microflora was reviewed, and its action mechanism in relieving the occurrence and development of animal intestinal inflammation was described, so as to provide a theoretical basis for the application of capsaicin in maintaining intestinal homeostasis.
As a foodborne pathogen of global importance, Salmonella enterica serovar Enteritidis (S. Enteritidis) is a threat to public health that is mainly spread by poultry products. Intestinal Enterobacteriaceae can inhibit the colonization of S. Enteritidis and are regarded as a potential antibiotic substitute. We investigated, in chicks, the anti-S. Enteritidis effects of Escherichia coli (E. coli) Nissle 1917, the most well-known probiotic member of Enterobacteriaceae. Eighty 1-d-old healthy female AA broilers were randomly divided into 4 groups, with 20 in each group, namely the negative control (group P), the E. coli Nissle 1917-treated group (group N), the S. Enteritidis-infected group (group S) and the E. coli Nissle 1917-treated and S. Enteritidis-infected group (group NS). From d 5 to 7, chicks in groups N and NS were orally gavaged once a day with E. coli Nissle 1917 and in groups P and S were administered the same volume of sterile PBS. At d 8, the chicks in groups S and NS were orally gavaged with S. Enteritidis and in groups P and N were administered the same volume of sterile PBS. Sampling was conducted 24 h after challenge. Results showed that gavage of E. coli Nissle 1917 reduced the spleen index, Salmonella loads, and inflammation (P < 0.05). It improved intestinal morphology and intestinal barrier function (P < 0.05). S. Enteritidis infection significantly reduced mRNA expression of angiotensin-converting enzyme 2 (ACE2) and solute carrier family 6-member 19 (SLC6A19) in the cecum and the content of Gly, Ser, Gln, and Trp in the serum (P < 0.05). Pretreatment with E. coli Nissle 1917 yielded mRNA expression of ACE2 and SLC6A19 in the cecum and levels of Gly, Ser, Gln, and Trp in the serum similar to that of uninfected chicks (P < 0.05). Additionally, E. coli Nissle 1917 altered cecum microbiota composition and enriched the abundance of E. coli, Lactobacillales, and Lachnospiraceae. These findings reveal that the probiotic E. coli Nissle 1917 reduced S. Enteritidis infection and shows enormous potential as an alternative to antibiotics.

Egg-laying rate is mainly determined by ovarian function and regulated by the hypothalamic-pituitary-gonadal axis; however, the mechanism by which the ovary regulates the egg-laying rate is still poorly understood. The purpose of this study was to compare the differences in the transcriptomes of the ovary of Lingyun black-bone chickens with relatively high and low egg-laying rates and screen candidate genes related to the egg-laying rate. RNA-sequencing (RNA-Seq) was conducted to explore the chicken transcriptome from the ovarian tissue of six Lingyun black-bone chickens with high (group G, n = 3) and low (group D, n = 3) egg-laying rates. The results showed that 235 differentially expressed genes (DEGs) were identified between the chickens with high and low egg-laying rates; among them, 209 DEGs were up-regulated and 26 DEGs were down-regulated. Gene Ontology analysis showed that the up-regulated 209 DEGs were enriched in 50 GO terms and the down-regulated 26 DEGs were enriched in 40 GO terms. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that up-regulated DEGs were significantly enriched in 25 pathways and down-regulated DEGs were significantly enriched in three pathways. Among the pathways, we found the longevity regulating pathway-multiple species pathway, Estrogen signalling pathway and PPAR signalling pathway may have an essential function in regulating the egg-laying rate. The results highlighted DEGs in the ovarian tissues of relatively high and low laying Lingyun black-bone chicken and identified essential candidate genes related to the egg-laying rate, thereby providing a theoretical basis for improving the egg-laying rate of Lingyun black-bone chicken.
