Purpose: MicroRNAs have been identified as crucial regulators of carcinogenic pathways. Micro environmental alterations such as hypoxia and acidosis frequently accompany tumor progression and metastasis. Thereto expression profiles of 24 different microRNAs, exhibiting oncogenic as well as tumor suppressor functions, were investigated under the influence of these two exogenic stimuli in endometrial cancer (EC) in vitro.
Zielsetzung: Mehr als zweitausend unterschiedliche MicroRNAs (miRNAs, miR) wurden bisher beschrieben. Sie steuern verschiedenen regulatorischen Funktionen hinsichtlich des menschlichen Transkriptoms. miRNAs spielen eine wichtige Rolle in fast allen biologischen Prozessen, inklusiv der Genregulation und der Kontrolle von Zellentwicklung. Aberrante miRNA-Expressionsmuster korrelieren stark mit der Progression von Pathologien wie Krebserkrankungen. Vor kurzem wurde in diversen Studien versucht, die Korrelation zwischen miRNA Expression und Ovarialkarzinomen zu untersuchen. In unserem Projekt analysierten wir die Expression von verschiedenen miRNAs in Ovarialkarzinom Zelllinien unter unterschiedlichen Bedingungen wie die kontrovers diskutierte HIPEC-Therapie (Hypertherme Intraperitoneale Chemoperfusion).
Zielsetzung: MicroRNAs (miRNAs, miR), eine Klasse von nicht-kodierenden RNA Moleküle, spielen eine essentielle Rolle in fast allen zellulären Signalwege der Tumorigenese, Tumorprogression, Zellinvasion und Metastasierung, Apoptose und auch der therapieinduzierten Resistenzentwicklungen. Die Hemmung von Östrogen-Signalwegen stellt eine potenzielle und attraktive Möglichkeit der Behandlung von Ovarialkarzinomen dar. Allerdings gilt die Entwicklung von Resistenzen gegen antihormonelle Therapien als eine besondere Herausforderung. Weiterhin stellen die zielgerichteten Therapien mit Cediranib, als VEGFR (vascular endothelial growth factor receptor)-Inhibitor, und Olaparib, als PARP (Poly ADP-ribose polymerase)-Inhibitor, innovative Ansätze in der Behandlung von serösen Ovarialkarzinomen dar. In unserer Studie wurde die Expression von unterschiedlichen miRNAs in Ovarialkarzinom-Zelllinien unter den obengenannten aufgeführten Therapiemodalitäten untersucht.

This study focused on the putative biomarker properties of 24 microRNA types, investigated in three breast cancer (BC), endometrial cancer (EC) and ovarian cancer (OC) in vitro models, respectively, to determine their diagnostic potential.
Einleitung/Zielsetzung: Elafin ist ein epidermaler Serin-Protease-Inhibitor mit potenter inhibitorischer Aktivität gegen Humane Neutrophile Elastase (HNE) und Proteinase 3 (PR3) sowie vielfältigen anti mikrobiellen, anti-inflammatorischen und immunmodulatorischen Eigenschaften. HNE spielt eine etablierte Rolle bei der Brustkrebsentwicklung und gilt als unabhängiger prognostischer Marker. Elafin konnte als wichtiger endogener HNE-Inhibtor zunehmend mit Brustkrebs in Zusammenhang gebracht werden. Eine vermehrte Expression von Elafin konnte bei Basal-like Mammakarzinomen nachgewiesen und mit einem schlechteren Outcome assoziiert werden. Andererseits zeigte exogenes Elafin wachstumshemmende Effekte bei luminalen Mammakarzinomzelllinien.
Einleitung/Zielsetzung: Die Anwendbarkeit blut-basierter microRNAs zur Detektion von Mammakarzinomen hat sich in neueren Studien bestätigt und eröffnet ein neues Feld für zirkulierende microRNAs als potentielle Brustkrebs-Biomarker. Anhand dieser Pilot-Studie wurden erstmalig spezifische Expressionsmuster Urin-basierter microRNAs hinsichtlich ihrer Anwendbarkeit als innovative Biomarker zur Brustkrebsdetektion untersucht.
Zielsetzung: Hypoxie fungiert als entscheidender Regulator im Tumorwachstum und induziert die transkriptionelle Aktivierung von Wachstumsfaktor-Signalwegen und Prozessen wie Angiogenese und Gewebsinfiltration. Cyr61, ein Mitglied der CCN-Familie, ist ein sezerniertes, matrizelluläres Protein, das wichtige Rollen in der Tumorprogression spielt und pro-angiogene Fähigkeiten zeigt. Die Expression von Cyr61 ist unter hypoxischen Bedingungen transkriptionell induziert und konnte mit dem Erreichen höherer Stadien im Tumorwachstum bei Brustkrebs in Verbindung gebracht werden. Neben der Hypoxie gab es bereits einige Hinweise darauf, dass eine post-transkriptionelle prä-mRNA-Prozessierung des Cyr61-Genes vermutlich auch dessen Expression regulieren kann, was jedoch bisher nicht näher untersucht wurde.
Introduction: CD44 is a transmembrane adhesion molecule involved in cell-cell and cell-substrate interactions. As a cell surface glycoprotein, CD44 may be shed or released into circulation by proteolytic enzymatic mechanisms. Alternative splicing of CD44 and aberrant levels of soluble CD44 protein in the serum of cancer patients has been correlated to tumor progression and metastasis. In this study we evaluated the clinical value of CD44 serum levels (sCD44) in patients with primary (non metastatic) breast cancer. Material and Methods: Concentrations of the soluble, variable isoforms sCD44std, sCD44v5 and sCD44v6 were determined with a sensitive ELISA and normalized against the total protein concentration (TP). Pre-operative serum samples from 84 patients and 107 healthy blood donors were analyzed. The results were correlated to histo-pathological parameters. Results: Sera of breast cancer patients showed elevated concentrations for sCD44v6 (p=0,0001) and for total protein (p=0,0001), whereas overall sCD44 (sCD44std) and sCD44v5 did not differ. Patients with lymph node metastasis showed elevated sCD44v6 serum concentrations in comparison to patients without lymph node involvement and healthy controls. Furthermore, high sCD44v5- and sCD44v6-concentrations above the 75%-percentile were correlated to large tumor size as well as lymph node metastasis. Conclusion: Our results suggest that soluble CD44v6 is upregulated in breast cancer patients. sCD44v6 seems to play an important role in tumor progression and metastasis in breast cancer. Determination of sCD44v6-concentrations may be helpful to identify subgroups of breast cancer patients with a high risk for lymph node metastasis.
Introduction: Tra2, member of the extended family of serine-argenine-rich (SR) splicing factors, regulates alternative splicing of exons with C/A-rich enhancer sequences. Besides the CD44 gene which undergoes extensive alternative splicing of at least 10 variable exons, other biological important genes, like VEGF, prolactin- estrogen- and insulin receptor, BRCA2 and Syk are alternatively spliced and contain these C/A-rich sequences. Recently, we demonstrated that alternative exon v4 inclusion is increased upon over-expression of the DNA and RNA binding protein YB-1 and Tra2 in a mouse model of breast cancer development. Material and Methods: In this study we investigated expression levels of TRa2 in a matched pair analysis of tumor samples and normal tissues of patients with invasive breast cancer by Western Blot and RT-PCR analysis. In addition, we analyzed the alternative splicing of the above mentioned tumor biological important genes in the same paradigm. Results: Western Blot and RT-PCR revealed a marked induction of Tra2-beta in adenocarcinomas of the breast in comparison to physiological tissue samples of the same patient. The splicing analysis showed in parallel an dramatically changed alternative splicing in the investigated genes with C/A-rich enhancers in tumors versus matched physiological breast tissue. Conclusions: Our results suggest that involvement of Tra2 in exon recognition and alternative splicing may be important to over-expression of alternatively spliced genes with potential functional consequences in invasive breast cancer leading to tumor progression and metastasis.
