With the aim of engineering multifunctional nanoparticles useful for cancer therapy, a diketopyrrolopyrrole-porphyrin based photosensitizer was here conjugated to a block copolymer (Pluronic F108), and used to stabilize in water lipidic cubic liquid crystalline nanoparticles (cubosomes), also loaded with the antineoplastic agent docetaxel. The physicochemical characterization by SAXS, DLS, and cryo-TEM demonstrated that the formulation consisted of cubosomes, about 150 nm in size, possessing a bicontinuous cubic structure (space group Pn3m). The cellular imaging experiments proved that these nanoparticles localized in lysosomes and mitochondria, while cytotoxicity tests evidenced a slight but significant synergistic effect which, after irradiation, increased the toxicity induced by docetaxel alone, allowing further reduction of cell viability.
Hintergrund: Die zunehmende Impfkritik in bestimmten Bevolkerungskreisen wird in Zusammenhang mit der zunehmenden Verbreitung von Komplementarmedizin gebracht. Homoopathie ist einer der starksten Vertreter unter der wachsenden Komplementar-Medizin. Besonders in der Padiatrie, wo die meisten Impfungen durchgefuhrt werden, wird Homoopathie vermehrt eingesetzt. Eine Umfrage aus dem Jahre 1996 zeigte erstmals, dass entgegen der offentlichen Meinung homoopathische Arzte in Deutschland keine entschiedenen Impfgegner sind, aber durchaus kritischer und individueller beim Impfen vorgehen. Methodik: In der vorliegenden Arbeit werden Impfeinstellung und Impfverhalten unter niedergelassenen Padiatern in ganz Deutschland untersucht. Hierbei werden Padiater mit und ohne Zusatzbezeichnung Homoopathie gegenubergestellt. Mittels einer reprasentativen Querschnittuntersuchung wurden im Jahre 2006/2007 50 % (N=3019) aller niedergelassenen Padiater aus ganz Deutschland per Post (Zufallsstichprobe) zum Thema Impfen befragt. Erganzend wurden zeitgleich 245 homoopathisch orientierte Padiater in die anonymisierte Befragung eingeschlossen. Das Instrument wurde zuvor in einer Pilotstudie unter 200 deutschen Padiatern evaluiert. Die Auswahl der Padiater erfolgte uber Adressverzeichnisse der Kassenarztlichen Vereinigungen und Arztekammern der verschiedenen Bundeslander, sowie zusatzlich uber den Deutschen Zentralverein fur homoopathische Arzte (DZVhA) und der Karl und Veronica Carstens (KVC)-Stiftung um eine moglichst grose Vergleichsgruppe an homoopathischen Padiatern zu erhalten. Die Rucklaufe der Zufallsziehung, der zusatzlich homoopathischen Padiater und der Pilotstudie wurden fur die Auswertung gepoolt. Fur einen Vergleich wurden mittels 6 (Teil)-Fragen des Erhebungsinstrumentes Padiater mit und ohne Schwerpunkt-/Zusatzbezeichnung Homoopathie differenziert. Letztere wurden zusatzlich unterschieden in Padiater, die Homoopathie oder eine andere alternative/komplementare Medizin anwenden oder rein konventionelle Medizin betreiben. Das Impfverhalten wurde im Hinblick auf das Einhalten der STIKO-Empfehlungen anhand von 3 Fragen des Erhebungsinstrumentes als Mittelwert-Score operationalisiert:1. Impfen Sie nach dem empfohlenen Impfkalender der STIKO? 2. Wie wurden Sie eigene minderjahrige Kinder Impfen? 3. Ab welchem Alter empfehlen Sie in der Regel die erste Durchfuhrung von Impfungen? Um die Impfeinstellung zu operationalisieren, wurden die skalierten Positionierungen zu 8 vorgegebenen Meinungen zum Impfen zu einem Mittelwert-Score verrechnet. Der Einfluss folgender relevanter Variablen auf Impfeinstellung und Impfverhalten wurde mit einer multivariablen linearen Regression untersucht: Alter, Geschlecht, Ort der Niederlassung, Anteil an Privatpatienten und Zusatzbezeichnung Homoopathie. Fur einen Vergleich mit den Studienergebnissen von 1996 wurde ein Impfkritiker-Index erstellt. Ergebnisse und Diskussion: Mit insgesamt 3464 angeschriebenen niedergelassenen Padiatern (uber 50 % aller niedergelassenen Padiater aus jedem Bundesland) handelt es sich um die bisher groste bundesweite Studie zum Thema Impfen. Die Responserate ist mit insgesamt ca. 67 % uberdurchschnittlich hoch, was ein groses Interesse zum Impfthema vermuten lasst. Vor dem Hintergrund einer weitestgehend vollstandigen und aktuellen Praxis-Adress-Datenbasis kann diese Studie als deutschlandweit reprasentativ angesehen werden. Die Ergebnisse dieser Arbeit sind trotz leichter methodisch bedingter Einschrankungen in der Konstruktion der Stichprobe einmalig und aussagekraftig. Padiater mit und ohne Zusatzbezeichnung Homoopathie unterscheiden sich im Hinblick auf ihre Impfeinstellung und ihr Impfverhalten (orientiert an den STIKO-Empfehlungen) signifikant. Die uberwiegend positive, aber kritische Haltung zum Impfen unter homoopathischen Padiatern ahnelt den Ergebnissen von P. Lehrke von 1996. Das Vorurteil „Homoopathen seien Impfgegner“ konnte fur Kinderarzte aussagekraftig widerlegt werden. Mit ca. 70 % hat die Mehrheit der homoopathisch arbeitenden Padiater in Deutschland eine positive Einstellung zum Impfen. Sie sind aber auch mehrheitlich individuelle Impfkritiker. Die starkste Impfkritik ist unter rein klassischen Homoopathen zu finden, welche ca. 47 % der Padiater mit Zusatzbezeichnung Homoopathie ausmachen. Die Zusatzbezeichnung Homoopathie hat von allen relevanten Variablen den grosten Einfluss auf eine impfkritische Einstellung und entsprechendes Verhalten. Abweichungen von den STIKO-Empfehlungen betreffen aber uberwiegend spatere Impfungen (uberwiegend 1–3 Monate), sowie neuere Impfungen wie gegen Pneumokokken, Varizellen und Meningokokken. Als haufigster Grund fur Abweichungen von den STIKO-Richtlinien wurde der Wunsch der Patienten angegeben. Da impfkritische Eltern eher Padiater mit komplementarmedizinischem Zusatzangebot auswahlen, beeinflussen sie auch die Erfahrungen und das Impfverhalten des Arztes. Es kann davon ausgegangen werden, dass sich das Patientenklientel von Homoopathen und Nicht-Homoopathen systematisch unterscheidet. Unterschiedliche Erfahrungen durch den Umgang mit Ungeimpften und den Moglichkeiten homoopathischer Behandlungen konnten die Veranderungen der Impfeinstellung hin zu mehr Impfkritik im Lebenslauf von Homoopathen erklaren. Homoopathen geben mit Abstand am haufigsten (uber 60 %) eine Veranderung in ihrer Einstellung zum Impfen im Lebenslauf an, Nicht-Homoopathen in nur ca. 10–22 %. Bezuglich der Impfentscheidung der Patienten hat wiederum die personliche arztliche Empfehlung als wichtigste Vertrauensperson einen grosen Stellenwert. Erfahrungen und Einstellung beeinflussen sich wechselseitig. Basierend auf der Zufallsstichprobe haben ca. 10 % aller niedergelassenen Padiater Deutschlands die Zusatzbezeichnung Homoopathie. Es gibt Hinweise, dass die Zusatzbezeichnung Homoopathie auf Grund der Patientennachfrage zugenommen hat. Das kann die haufige Anwendung von Homoopathie unter Padiatern ohne Zusatzbezeichnung Homoopathie (ca. 40 %) erklaren. Entsprechend dem marktwirtschaftlichen Prinzip des Gesundheitssystems wird die Homoopathie trotz wissenschaftlich strittiger Studienlage zunehmend von den Kassen vergutet, u.a. um konkurrenzfahig zu bleiben und Kosten vor allem im Bereich chronischer Krankheiten zu reduzieren. Ausblick: Ein Arzt sollte sich der Wirkung seiner Impf-Einstellung auf seine Patienten bewusst sein und bei seiner Impfberatung die Erwartungen und auch die Impfkritik seiner Patienten berucksichtigen. Die Komplexitat und Vielseitigkeit des Impfthemas sollte den Impfentscheidern (Patienten) nicht vorenthalten werden sondern eine eigenverantwortliche Impfentscheidung (wie gesetzlich vorgesehen) fordern. Eine fundierte, neutrale und individualisierte Impf-Beratung sollte daher fester Bestandteil der medizinischen Ausbildung sein und in der Umsetzung auch entsprechend honoriert werden, unabhangig von der Impfentscheidung der Patienten. Damit konnen Verunsicherungen durch zT. emotionalisierende und polarisierende Informations-Medien (Internet, soziales Umfeld, Hebammen) vorgebeugt werden. Bei zunehmender Verbreitung von Komplementarmedizin und speziell der Homoopathie sollten das Thema Komplementarmedizin in der medizinischen Ausbildung fest integriert werden und entsprechend den Moglichkeiten zur Heilungsunterstutzung in der Forschung und klinischen Anwendung den entsprechenden Stellenwert bekommen.
An original degradable fluorinated surfactant, 3-hydroxy-2-(trifluoromethyl)propanoic acid (MAF-OH), was applied for the emulsion polymerization of vinylidene fluoride (VDF) to yield a latex of PVDF, consisting of typically 100 nm particle diameters. This surfactant was shown to be easily decomposed in subcritical water, releasing fluoride anions. The % reversed head-to head (H-H) addition is greatly minimized (2.4%) in the PVDF, prepared in the presence of MAF-OH surfactant.
We designed novel polymer-free cubic bicontinuous liquid crystalline dispersions (cubosomes) using monoolein as molecular building block, phospholipids as stabilizers, propylene glycol as hydrotrope. Their kinetic stability was evaluated by analysing the backscattering profiles upon ageing, and the most stable formulation was chosen as potential photosensitizers delivery vehicle for photodynamic therapy (PDT) of human skin melanoma cells. Morphological and topological features of such formulation alternatively loaded with Chlorin e6 or meso-Tetraphenylporphine-Mn(III) chloride photosensitizing dyes were investigated by cryo-TEM, DLS, and SAXS. Bioimaging studies demonstrated that Me45 and MeWo cell lines effectively internalized these cubosomes formulations. Particularly, photodynamic activity experiments proved both the very low cytotoxicity of the cubosomes formulation loaded with Chlorin e6 dye in the "dark" condition, and its significant cytotoxic effect after photoirradiation. The toxic effect recorded when the photosensitizer was encapsulated within the cubosomes was shown to be one order of magnitude higher than that caused by the free photosensitizer. This is the first report of biocompatible polymer-free cubosomes for potential application in both PDT and bioimaging of skin malignant melanoma.
Unusual temperature gaps have previously been observed in the turbulent drag reduction effectiveness of Dobon and Habon cationic surfactant systems containing two anionic counterions of different binding strengths. Here, we report drag reduction data for a cationic surfactant with a mixture of dodecyl sulfate and tosylate counterions showing a temperature gap. Cryo-TEM images of nanostructures at different temperatures for this system support an explanation for this gap, based on the counterions’ relative binding strengths.
Targeted liquid crystalline nanoparticles with a reverse hexagonal inner structure as diagnostic and therapeutic tools in oncology.
This study is the first report of the synthesis and self-assembly in water of an amphiphilic PVDF-b-PVA block copolymer.
For extremely asymmetric n-hexyl(n-decyl)phosphate (HDeP), n-hexyl(n-dodecyl)phosphate (HDoP), and n-hexyl(n-cetyl)phosphate (HCeP), the effect of the long-chains on the dynamic behavior of their aggregate structures in water was examined by cryo-TEM imaging, SANS, and X-ray diffraction techniques. The cryo-TEM images demonstrated the complex and dynamic behavior of the aggregates, and its dependence on the length of the long-chain. Application of the one-dimensional aggregate theory to the SANS results led to the conclusion that the existence of a limiting size also depended on the length of the main long-hydrocarbon chain and affected strongly the dynamic behavior of the aggregates, causing breakage of thread-like micelles or ribbon-like aggregates. The X-ray diffraction patterns of the lyotropic liquid crystalline samples of HDeP and HCeP were used to estimate the aggregate structures of this limited size.
A new PEG2000-b-PFPE1200-b-PEG2000 amphiphilic triblock copolymer that undergoes self-assembly into micelles in water was synthesized by copper(i)-catalyzed alkyne–azide cycloaddition (CuAAC).
The association of building blocks into supramolecular polymers allows the fabrication of diverse functional architectures at the nanoscale. The use of minimal assembly units to explore polymer dynamics and phase transitions significantly contributes to the application of polymer physicochemical paradigms in the field of supramolecular polymers. We present a minimal model that displays spontaneous coordinated structural transitions between micro- and nanostructures, hydrogels with nanoscale order, and single crystals. The simple amphiphilic 9-fluorenylmethoxycarbonyl-3,4-dihydroxyphenylalanine (Fmoc-DOPA) modified amino acid undergoes a noninduced transition from spherical assemblies into nanofibrils followed by sol-gel transition, nanotube formation via intermediate assembly, and crystallization within the gel. Notably, the transition kinetics is slow enough to allow both multistage and multiscale characterization of the supramolecular arrangement using electron microscopy, vibrational and circular dichroism spectroscopies, nuclear magnetic resonance, and x-ray crystallography. This minimalistic system is the first comprehensive model for a complete spontaneous structural transition between diverse states governed by distinct molecular interactions.
Herein we provided the first proof of principle for in vivo fluorescence optical imaging application using monoolein-based cubosomes in a healthy mouse animal model. This formulation, administered at a non-cytotoxic concentration, was capable of providing both exogenous contrast for NIR fluorescence imaging with very high efficiency and chemospecific information upon lifetime analysis. Time-resolved measurements of fluorescence after the intravenous injection of cubosomes revealed that the dye rapidly accumulated mainly in the liver, while lifetimes profiles obtained in vivo allowed for discriminating between free dye or dye embedded within the cubosome nanostructure after injection.
Complexes were formed via the electrostatic interaction between 30-50 nm anionic liposomes and a star-shaped polyelectrolyte, poly{[2-(methacryloyloxy)ethyl]trimethyl ammonium iodide}, having cationic arms that radiate from a silicon-based central core. The complexation was investigated with attention given to assessing the capacity of the cationic stars for the anionic liposomes (both liquid and solid); the integrity of the complexed liposomes; and the stability of the resulting star/liposome complexes in aqueous salt solutions. We have found that by changing the content of anionic groups in the liposomal membrane as well as the phase-state of membrane, the stability of star/liposome complexes in aqueous salt media can be controlled. The liquid liposomes with 0.1 mol fraction of anionic palmitoy-loleoylphosphatidylserine (POPS1-), and solid liposomes with 0.1 mol fraction of POPS1-, retain their integrity when bound to the stars, with the resulting star/liposome complexes being stable in physiological solution, i.e. [NaCl] = 0.15 M. Multi-liposomal complexes containing up to 12 liposomes per star seem to hold promise as carriers for biologically active compounds. (C) 2016 Elsevier Ltd. All rights reserved.

To evaluate the potential of monoolein-based cubic bicontinuous liquid crystalline nanoparticles (cubosomes) as diagnostic tools in diabetes and pancreatic β-cell transplantation, fluorescent cubosomes were formulated, entrapping within the lipid bilayer a newly synthesized, potent, hydrophobic dye. Cryo-TEM, SAXS, and DLS were performed to assess the morphological and structural aspects of this formulation. Rat pancreatic β-cells (INS-1E) readily took up the cubosome nanoparticles, as revealed by in vitro internalization tests, and flow-cytometric analyses confirmed the persistence of the fluorescence for up to 24 hours. The cytotoxicity of cubosomes at various concentrations, evaluated with a real-time analysis of proliferation, revealed that the proliferation curves relative to the less concentrated formulation (5 μM in terms of monoolein content) were comparable to the untreated cultures. INS-1E cell cycle and apoptosis supported such results. In conclusion, this formulation, with dye content in the nanomolar range, seems to be useful for β-cell imaging.

We present here an innovative, fluorescent, monoolein-based cubosome dispersion. Rather than embedded within the monoolein palisade, the fluorescent imaging agent, namely dansyl, was conjugated to the terminal ethylene oxide moieties of the block copolymer Pluronic F108. We discuss the physicochemical and photophysical properties of this fluorescent Pluronic and of a cubosome formulation stabilized by a mixture of dansyl-conjugated and non-conjugated Pluronic, also including an anticancer drug (quercetin). Furthermore, we performed biocompatibility tests against HeLa cells to assess internalization and cytotoxicity features of this nanoparticles aqueous dispersion. Cryo-TEM, SAXS, and DLS analysis, proved the bicontinuous cubic inner nanostructure and the morphology of this fluorescent cubosome dispersion, while photophysical measurements and biocompatibility results basically validate their potential use for theranostic nanomedicine applications.
Cryogenic electron microscopy (cryo-EM) is a powerful tool for imaging liquid and semiliquid systems. While cryogenic transmission electron microscopy (cryo-TEM) is a standard technique in many fields, cryogenic scanning electron microscopy (cryo-SEM) is still not that widely used and is far less developed. The vast majority of systems under investigation by cryo-EM involve either water or organic components. In this paper, we introduce the use of novel cryo-TEM and cryo-SEM specimen preparation and imaging methodologies, suitable for highly acidic and very reactive systems. Both preserve the native nanostructure in the system, while not harming the expensive equipment or the user. We present examples of direct imaging of single-walled, multiwalled carbon nanotubes and graphene, dissolved in chlorosulfonic acid and oleum. Moreover, we demonstrate the ability of these new cryo-TEM and cryo-SEM methodologies to follow phase transitions in carbon nanotube (CNT)/superacid systems, starting from dilute solutions up to the concentrated nematic liquid-crystalline CNT phases, used as the 'dope' for all-carbon-fibre spinning. Originally developed for direct imaging of CNTs and graphene dissolution and self-assembly in superacids, these methodologies can be implemented for a variety of highly acidic systems, paving a way for a new field of nonaqueous cryogenic electron microscopy.
