Sustained renin-angiotensin system (RAS) activation in asthmatic patients plays a crucial role in airway hyperresponsiveness and airflow limitation. Angiotensin II (Ang II), as a key peptide of RAS, contributes to the contraction of human airway smooth muscle by activating the RhoA/Rho-associated coiled-coil containing protein kinase 2 (ROCK2) signaling pathway. Angiotensin-(1-7) [Ang-(1-7)], is a component of the angiotensin I converting enzyme 2 (ACE2)-Ang-(1-7)-Mas axis which counteracts the detrimental effects of the ACE- Ang II-angiotensin type 1 receptor (AT1R) axis in vivo; however, whether Ang-(1-7) can inhibit the effect of Ang II in the contraction of human airway smooth muscle cells (HASMCs) is unknown. In our study, collagen gel lattices and immunofluorescence were used to evaluate the contraction of HASMCs induced by Ang II. Real-time PCR and western blot analysis were performed to confirm the regulatory mechanism and the participating signaling pathway. Ang II caused the contraction of HASMCs; this effect was reversed by Ang‑(1‑7). In addition, irbesartan and A779, which are inhibitors of AT1R and Mas, respectively, attenuated the effect of Ang II and Ang-(1-7). Furthermore, Y-27632, an inhibitor of ROCK2, attenuated the Ang II-induced contraction of HASMCs by blocking the RhoA/ROCK2 signaling pathway which is involved in this contraction, and thus may be a major regulator involved in the basal maintenance of contractility in HASMCs. These data demonstrate that Ang II induces the contraction of HASMCs and that this effect can be reversed by Ang-(1-7), partially through the downregulation of of the RhoA/ROCK2 signaling pathway.
Objective:To explore psychological health state of unwanted pregnant adolescents and effect factors,provide a basis for intervention.Methods:33 unwanted pregnant adolescents under 26 years underwent induced abortion,then they were evaluated by Self-report Anxiety Scale(SAS),Center for Epidemiologic Studies Depression Scales(CRS-D) and self-designed questionnaire.Results:The age of first sexual intercourse was 17~24 years and the average age was(20±2) years,30% of the adolescents had the history of induced abortion(five times),the average times was 1.57 times;the score of SAS(48.88±3.92) was higher than that of norm(29.78±0.46)(Z=3.995 7,P<0.001),the detection rate of positive symptoms was 45%;the score of CRS-D in the minorities(33.00±6.80) was lower than that in the Han nationality(41.26±2.89)(F=10.639,P<0.01);the score of depression in patients with senior high school education level was significantly higher than those in patients with junior high school,junior college and undergraduate education level(P<0.05,P<0.01,P=0.00);the score of anxiety in patients whose sexual partner was middle school students was higher than that whose sexual partner was undergraduate(P<0.05);the score of depression in patients whose education level was higher than that of sexual partner was higher than that in patients whose education level was equal to sexual partner(P<0.05).Conclusion:The psychological health state of unwanted pregnant adolescents is bad,particular in Han nationality and students;education levels of patients and sexual partners are main effect factors of psychological health of unwanted pregnant adolescents.
目的:探讨骨髓间充质干细胞(MSCs)在肠道辐射损伤修复中的作用。方法:体外培养C57BL/6雄性小鼠的骨髓MSCs,通过诱导其向脂肪细胞、成骨细胞分化的方法进行鉴定。将受鼠C57BL/6雌性小鼠随机分成照射对照组(组Ⅰ)、骨髓单个核细胞组(组Ⅱ)和骨髓MSCs+骨髓单个核细胞组(组Ⅲ),给予60Coγ射线全身致死量照射后,组Ⅰ不做处理,组Ⅱ2h内尾静脉移植C57BL/6雄性小鼠的骨髓单个核细胞,组Ⅲ移植骨髓MSCs和骨髓单个核细胞。观察各组小鼠4周的生存率、病理切片等指标,并用PCR检测雌性受鼠体内Y染色体的Sry基因,确定供体干细胞的植入情况。结果:组Ⅲ的生存率较其他两组显著升高(P﹤0.05),病理切片显示肠道损伤有明显修复,PCR检测显示修复的细胞来源为供鼠细胞。结论:骨髓MSCs联合骨髓单个核细胞移植有促进肠道辐射损伤修复的作用,其作用优于单纯的骨髓单个核细胞移植,为骨髓MSCs用于修复肠道辐射损伤提供了实验依据。
Objective To inquire into the reasons and relevant factors of unwilling gestation of adolescents and to offer basis of sexual health and medical care of adolescents. Method We use unregistered and self-stated questionnaire designed by ourselves. Content involves the general condition, sexual contraception knowledge, sexual ideas, sexual attitude and gestation reason of investigated objects, and so on. By using questionnaire we investigate 32 adolescents for unwilling gestation selected. Results Among investigated persons students contain 37.5 %, migrant workers contain 25 %. Gestation rate for the ages from 19 to 23 is the maximum. For first sexual practice the youngest age is 17. The first three places of knowledge resource of sexual contraception: 43.8 % learn from network media, 31.3 % from school, and 15.6 % from sexual fellows. For the recognition that emergency contraception and induced abortion may result in sterility, the correctness rate contains 68.8 %. Among unwilling gestation persons the sexual fellows of 90.6 %are their lovers, those of 6.3 % are common friends, and that of 3.1 % is one-night lover. 62.5 % consider that the occurrence of the sexual behavior is the inevitable result of love. 75 % have not adopted contraception. 25 %break down to avoid pregnancy. 96.9 % mean to need to study the knowledge of contraception. Conclusion Unwilling gestation of adolescents has already become unavoidable society problem; The lacks of sexual education is the main reasons. Sexual health education of adolescents must pay attention on self-protection education and impartation of knowledge of sexual contraception.
新生儿先天性梅毒是指患有梅毒的妊娠期妇女梅毒螺旋体通过胎盘经血液循环感染胎儿,使胎儿出生后即患病。先天性梅毒患儿出生后即可出现皮肤粘膜、骨骼及内脏受损的临床症状和体征,并可造成各脏器功能障碍或死亡,且易被临床医生误诊或漏诊。因此积极开展孕期梅毒筛查工作和孕期梅毒治疗是预防先天性梅毒的重要途径和有力措施。
轮状病毒(RV)肠道外感染的问题日益受到人们的关注.本研究试图探讨RV病毒血症的发生率、出现的时间规律与肠道外感染的关系,为病毒血症与肠道外感染的防治提供参考.
OBJECTIVE:To investigate the extraintestinal dissemination of rotavirus (RV) in immunodeficient mice.METHODS:Immunodeficiency mouse model was established by injection of cyclophosphamide into the abdominal cavities of normal mice, then to which RV was administered either orally or intra-abdominally. The pathological changes in the organs were observed by light microscopy and RV was detected by in situ hybridization and PCR.RESULTS:Small intestinal villi, gastric lamina propria and cardiac myocytes exhibited pathological changes in the mice with oral RV administration. Besides these changes, the mice with intra-abdominal RV injection showed changes in the liver and kidneys. The intestinal villi of the mice with oral RV were RV positive by in situ hybridization. Positive results of RV in in situ PCR detection were found in the intestinal villi, intestinal gland cells, epithelial cells of the proximal convoluted tubules and collecting tubes in the kidney of the mice taken RV orally, and in the intestinal villi, kidneys, liver, heart and pancrease of mice with intra-abdominal RV injection.CONCLUSION:Immunodeficiency may be the important factor for inducing and aggravating the infection and extraintestinal dissemination of RV.
