Colonic mucosal protection is provided by the mucus gel, mainly composed of mucins. Several factors can modulate the formation and the secretion of mucins, and among them butyrate, an end-product of carbohydrate fermentation. However, the specific effect of butyrate on the various colonic mucins, and the consequences in terms of the mucus layer thickness are not known. Our aim was to determine whether butyrate modulates colonic MUC genes expression in vivo and whether this results in changes in mucus synthesis and mucus layer thickness. Mice received daily for 7 days rectal enemas of butyrate (100 mM) versus saline. We demonstrated that butyrate stimulated the gene expression of both secreted (Muc2) and membrane-linked (Muc1, Muc3, Muc4) mucins. Butyrate especially induced a 6-fold increase in Muc2 gene expression in proximal colon. However, butyrate enemas did not modify the number of epithelial cells containing the protein Muc2, and caused a 2-fold decrease in the thickness of adherent mucus layer. Further studies should help understanding whether this last phenomenon, i.e. the decrease in adherent mucus gel thickness, results in a diminished protective function or not.
Les nouveau-nés de petit poids de naissance ayant souffert d’un retard de croissance intra-utérin (RCIU) sont une population à haut risque pour développer un « syndrome métabolique » (obésité, hypertension, insulino-résistance) à l’âge adulte. Ces bébés sont généralement nourris avec des laits « enrichis » en protéines afin de leur assurer une croissance de rattrapage. Or le risque de pathologies cardiovasculaires ou d’obésité à l’âge adulte est corrélé à la vitesse de croissance dans les toutes premières semaines de vie. Notre hypothèse est qu’une alimentation hyperprotéique prescrite en période néonatale chez des nouveau-nés RCIU pourrait avoir des conséquences néfastes sur la santé à l’âge adulte. Pour étudier les conséquences à distance d’une alimentation néonatale hyperprotéique, des ratons RCIU, nés de mères carencées en protéines durant la gestation (60 % de carence) ont été équipés au 5ème jour de vie (J5) d’une canule de gastrostomie posée de façon percutanée (modèle de ratons « pup in the cup »). Ils ont reçu, de J5 (pose de la canule) à J21 (jour du sevrage) et de façon randomisée, soit un lait « normoprotéique » (NP : 87,0 g/l, n = 9), soit un lait « hyperprotéique » (HP : 130,5 g/l, n = 7). La croissance des rats et les quantités ingérées ont été suivies régulièrement (de J5 à J160, rat adulte). Le lait HP ne modifie pas la croissance des ratons avant le sevrage. Le lait HP présente des effets à distance : (1) un surpoids des rats nourris au lait hyperprotéique vs. normoprotéique entre J50 et J65, (HP : 329 ± 14 g vs. NP : 307 ± 28 g pour des rats de J60, p = 0,079), lié à une augmentation des quantités ingérées d’aliments (de J50 à J60, HP : 28 ± 3 g/j vs. NP : 25 ±3 g/j, p = 0.079) ; (2) une augmentation, de la masse du pancréas (0,29 ± 0,15 g vs. 0,17 ± 0,06, p= 0.046) et de la masse de la graisse mésentérique (6,3 ±1,4g vs. 4,5 ± 2, 1, p = 0,088) respectivement chez des rats HP vs. NP. Ces données préliminaires suggèrent qu’un lait hyperprotéique, administré à des ratons RCIU durant la période néonatale, a des effets mémoire sur le poids et la masse grasse mésentérique, traduisant des désordres métaboliques. Des mesures de composition corporel le, glucose et insuline plasmatiques, nous permettront de préciser ces perturbations métaboliques.
Le maintien de la masse protéique intestinale est une des conditions de l'intégrité de la muqueuse intestinale, elle-même indispensable pour permettre à l'intestin d'assurer ses fonctions de "barrière", et dépend de l'équilibre entre les vitesses de synthèse et de dégradation protéique. Or on connaît mal les facteurs qui régulent la synthèse protéique dans la muqueuse colique, particulièrement chez le nouveau-né. Le butyrate et les acides gras à chaîne courte issus du métabolisme des oligosaccharides prébiotiques sont les principaux carburants du colonocyte, et ont des effets trophiques sur le côlon adulte. Le but de cet étude était de déterminer 1) si les prébiotiques ont un effet trophique chez le nouveau-né, et 2) s'ils stimulent la synthèse protéique dans la muqueuse colique néonatale. Trente huit rats nouveau-nés porteurs d'une gastrostomie mise en place par voie percutanée ont reçu du 5e au 18e jour de vie (sevrage), une alimentation entérale exclusive à l'aide d'un substitut de lait de rat, supplémenté ou non, après ran-domisation, en galacto- et fructo-oligosaccharides (GOS/FOS). Au 18e jour de vie, ils ont reçu en phase nourrie une perfusion entérale de L-[U-13C] valine durant 2 h avant sacrifice. Les enrichissements isotopiques ont été mesurés par spectrométrie de masse couplée à la chromatographie gazeuse. Le turnover protéique du corps entier a été calculé à partir de la 13C-valine libre sanguine, et la vitesse fractionnelle de synthèse (FSR) protéique, mesurée par l'incorporation du traceur dans les protéines de la muqueuse, rapportée à l'enrichissement de la valine libre intracellulaire de la muqueuse. Les résultats des 2 groupes (Tableau) ont été comparés par test de t non apparié. Colonic mucosal protection is provided by mucous gel, mainly composed of secreted (Muc2) and membrane-bound (Muc1, Muc3, Muc4) mucins. Our aim was to determine the expression profile of secreted and membrane-bound mucins in experimental dextran sulfate sodium (DSS)-induced colitis. Acute colitis was induced in Balb/C mice by oral administration of 1.0% DSS (5 days) and chronic colitis was maintained by subsequent 0.15% DSS treatment (28 days). Clinical symptoms (mortality, weight gain), stool scores, and MPO activity confirmed the inflammatory state in the two phases of colitis. Muc2 gene expression was not modified by colitis, whereas Muc3 gene expression was increased (×2) only in the cecum and the distal colon of mice after acute colitis. Muc1 and Muc4 mRNA levels were more significantly increased in the cecum (×8–10) than in colonic segments (×4) after acute colitis. TFF3 involved in mucosal repair was up-regulated during colitis induction. These results indicate that Muc and TFF3 genes are regulated early in inflammation and suggest that their mRNA levels could be used as early markers of inflammation.
Inulin and fructo-oligosaccharides (FOS) are known to be fermented in the colon, producing high level of butyrate and stimulating the growth of lactate-producing bacteria.The butyrate-producing fermentation is suggested to stimulate the trophicity of the intestinal mucosa and maintain the integrity of the non-immune intestinal barrier.The mucus layer, lining epithelial cells, is suggested to contribute to the maintain of intestinal barrier.The aim of the present study was to investigate whether, a mixture of inulin and FOS (IF: Synergy1, Orafti) can protect the colonic mucosa from inflammatory damages or restore its integrity in established colitis, by modulating mucin production.Two separate experiments tested the preventive and curative properties of IF (4%) versus basal diet (BD) in dextran-sodium sulfate (DSS)-induced colitis in Balb/c mice.In the preventive conditions, mice received BD or IF during 21 days.From day 12, they also received 1.3% DSS for 9 days in drinking water.In the curative conditions, mice first recieved 1.3% DSS alone for 7 days and for the last 2 weeks received 0.2% DSS and BD or IF.In both experiments, DSS-treated mice were compared to healthy mice fed basal diet (BD) or supplemented IF diet (IF).We investigated fermentation parameters (cecal pH, short chain fatty acid [SCFA] concentration), inflammatory damages (macroscopic mucosal score, diarrhoea, MPO activity), and mucin barrier (Muc gene expression).In the preventive conditions, IF fermentation increased the SCFA production only in proximal colon of DSStreated mice (P≤0.05).IF diet reduced the inflammation and lowered MPO activity (P<0.05) of colonic tissue, without changing the macroscopic inflammation parameters.The significant (P≤0.05)increase of the expression of Muc genes (Muc2, Muc3, Muc4) induced by DSS treatment was not modulated by IF diet.In the curative conditions, IF diet stimulated the fermentation in distal colon of DSS-treated mice, but DSS increased the SCFA concentration in all caeco-colonic segements of DSS-treated mice with comparison to control mice (p≤0.05).However, IF diet did not modify any inflammation parameter.Only Muc 4 gene expression was changed by the injuries of mucosal induced by DSS treatment.A diet supplementated with a nutritional level of IF (4%), given in preventive conditions, reduced the severity of DSS-induced colitis , but failed in restoring the integrity of ceco-colic mucosa in already established colitis.Mucins did not seem involved in the preventive effect of IF diet.These results confirm the potential efficacy of some dietary fibre in preventing inflammatory bowel disease.
Propionibacteria are cheese starters, also studied for their probiotic potential. An interesting feature is the ability of some strains to synthesize nitric oxide (NO) from nitrate reduction. NO production is hypothesized to be the mechanism by which some probiotics improve the mucosal barrier in rats. We therefore investigated the ability of two Propionibacterium acidipropionici strains (TL15 P TL223), which differ in their in vitro production of NO, to improve colitis in rats. Three groups of rats in which colitis was induced by trinitrobenzene sulfonic acid (TNBS), received intracolonic infusions of 9 g.L-1 NaCl (n=6), or 0.9x10(10) CFU.d(-1) of TL15 (n=7), or 1.4x10(10) CFU.d(-1) of TL223 (n=7), for 7 days. Following treatment, animal weight, food consumption, inflammatory score and myeloperoxydase (MPO) activity, together with changes in intestinal bacteria and SCFA concentrations, were investigated. As expected, both TL223 and TL15 supplements induced detectable counts of P. acidipropionici in colonic contents of rats (7.67 +/- 0.67 and 7.43 +/- 0.50 log eq. CFU.g(-1), respectively, as determined by real-time PCR). Food consumption and body weight of rats receiving propionibacteria were overall higher than those of rats with NaCl (P=0.007 and P=0.004). Moreover, both propionibacteria infusions similarly and dramatically reduced the gross score for inflammation (P=0.004) as compared with NaCl (0.6 +/- 0.3 and 0.7 +/- 0.5 vs. 4.8 +/- 1.5). A similar effect was observed for MPO activity (P=0.056), which reached 2.50 +/- 1.37 U.g(-1) of mucosal tissue for NaCl vs. 0.05 +/- 0.02 and 0.26 +/- 0.18 U.g(-1) for TL223 and TL15, respectively. This study shows for the first time the potential benefit of propionibacteria in colonic mucosa healing, but the mechanism involved needs to be elucidated.
Les acides gras a chaine courte (AGCC), principaux metabolites terminaux de la fermentation colique des glucides, constituent l'essentiel des anions organiques presents dans le colon. Leur production depend de plusieurs facteurs etroitement lies entre eux, dont les principaux sont la nature du substrat fermente, la composition et l'activite de la flore colique et les conditions environnementales regnant dans la lumiere intestinale. Les AGCC, principalement les acides acetique, propionique et butyrique, sont rapidement absorbes par la muqueuse colique selon differents mecanismes (passifs ou facilites). Le butyrate fournit plus de 70 % des besoins energetiques des colonocytes. Plusieurs etudes realisees chez l'homme et divers modeles animaux ont montre que les AGCC pouvaient modifier l'activite motrice gastrointestinale et colique, exercer un effet trophique sur l'epithelium intestinal et colique et augmenter egalement les defenses de la muqueuse intestinale. Contrairement a ce qui est observe sur les cellules normales, le butyrate est capable d'inhiber la proliferation des cellules cancereuses et d'agir comme agent differenciant. Les mecanismes d'action des AGCC sont encore mal compris. Leurs effets impliquent probablement plusieurs mecanismes, directs et indirects, aux niveaux cellulaire, hormonal et nerveux. Compte-tenu de leurs nombreux effets physiologiques, les AGCC pourraient avoir un effet prophylactique ou therapeutique, notamment comme antidiarrheiques et dans le traitement de certaines formes d'inflammation intestinale. Enfin, l'apport de butyrate dans des modeles experimentaux de cancer colique a montre des effets protecteurs qui meritent d'etre approfondis.
Three dietary fibres with different physicochemical properties were studied in healthy humans for their effects on small intestinal motility and postprandial hyperglycaemia. Duodeno-jejunal motor activity was evaluated electromyographically for 180 min in six subjects who had ingested a test meal composed of glucose alone or glucose with 15 g of wheat bran (WB), sugar beet (SB) or ispaghula (I) fibres. Glucose and insulin concentrations were determined during the same period. Each subject received each of the four test meals randomly during a 4 d period. Addition of SB or I to the glucose meal altered duodeno-jejunal motility. Both of these fibres inhibited stationary contractile activity and increased the propagation length and velocity of propagated activity, whereas addition of WB had no effect. These results could reflect the high water-holding capacity of SB and 1. Blood glycaemic response to the glucose meal was reduced by SB and I but remained unchanged with WB. Postprandial blood glucose levels were significantly correlated with the total motility index ( r 0·82) and stationary activity ( r 0·79). Taken together, these observations suggest that the contractile activity induced by dietary fibre in the small intestine probably plays a major role in delayed glucose absorption.
