Cholesterol absorption inhibition (CAI) represents an important treatment option for hypercholesterolemia. Herein, we report the design and evaluation of a series of substituted oxazolidinones as ligands for the Niemann Pick C1 Like 1 (NPC1L1) protein, a key mediator of cholesterol transport. Novel analogs were initially evaluated in a brush border membrane NPC1L1 binding assay; subsequently, promising compounds were evaluated in vivo for acute inhibition of cholesterol absorption. These studies identified analogs with low micromolar NPC1L1 binding affinity and acute in vivo efficacy of >50% absorption inhibition at 3mg/kg.
This paper describes the recent results of the GridShib and MyProxy projects to integrate the public key infrastructure (PKI) deployed for Grids with different site authentication mechanisms and the Shibboleth identity federation software. The goal is to enable multi-domain PKIs to be built on existing site services in order to reduce the PKI deployment and maintenance costs. An authorization framework in the Globus Toolkit is being developed to allow for credentials from these different sources to be merged and canonicalized for policy evaluation. Successes and lessons learned from these different projects are presented along with future plans.
In this paper we describe our work in progress to integrate the Shibboleth SAMLbased framework and Globus Toolkit’s PKIbased security infrastructure. The result will provide identity federation and attributebased policy enforcement for Grids that leverages the Shibboleth system being deployed on campuses. We provide an overview of both Shibboleth and the Globus Toolkit, present our motivating use cases, and describe our planned integration work.
