Traditional tracking often have an online updating process, which often leads to heavy computing burden. We propose an off-line tracking re-detection framework, which is composed of a Siamese convolution network and a correlation filtering tracker. The Siamese convolution network is trained on a large-scale video data to obtain high object discrimination ability, to be a re-detection network. We use this Siamese network to re-detect the tracking results of correlation filtering tracker. In the tracking process, we do not have to update detection network online due to its high object discrimination ability. This mechanism reduces the computational burden of the algorithm and achieves high tracking speed. We also propose a hybrid judgment mechanism to make the re-detection network get better tracking decisions, as well as to re-detect tracked objects after tracking challenges occurring. Experiment results on large-scale data sets demonstrate that the proposed tracking framework outperforms the compared methods and high tracking speed.
研究显示高达90%的肝癌患者在发病进程中遵循"肝炎→进展期肝纤维化→肝硬化→肝癌"这一发病的规律,从这样的发病规律我们可以得出肝癌的发生在肝纤维化时期就已经埋下伏笔的结论,即中医所说"毒伏肝络".就目前对肝癌的治疗仍缺乏行之有效的手段.然而"毒伏肝络"机制能为肝癌的早发现、早治疗提供新的思路.
The APOBEC proteins play significant roles in the innate and adaptive immune system, probably due to their deaminase activities. Because APOBEC1 (A1) and APOBEC3 (A3) are absent in the chicken genome, we were interested in determining whether chicken APOBEC4 (A4) possessed more complex functions than its mammalian homologs. In this study, chicken A4 (chA4) mRNA was identified and cloned for the first time. Based on bioinformatics analyses, the conserved zinc-coordinating motif (HXE … PC(X)2–6C) was identified on the surface of chA4 and contained highly conserved His97, Glu99, Pro130, Cys131 and Cys138 active sites. The highest expression levels of constitutive chA4 were detected in primary lymphocytes and bursa of Fabricius. Newcastle Disease (ND) is one of the most serious infectious diseases in birds, causing major economic losses to the poultry industry. In vitro, Newcastle Disease Virus (NDV) early infection induced significant increases in chA4 expression in the chicken B cell line, DT40, the macrophage cell line, HD11 and the CD4+ T cell line, MSB-1, but not the fibroblast cell line, DF-1. In vivo, the expression levels of chA4 were up-regulated in several tissues from NDV-infected chickens, especially the thymus, testicles, duodenum and kidney. The high level expression of exogenous chA4 displayed inhibitory effects on NDV and reduced viral RNA in infected cells. Taken together, these data demonstrate that chA4 is involved in the chicken immune system and may play important roles in host anti-viral responses.
APOBEC家族成员中AID蛋白具有DNA/RNA编辑酶功能,同时具有抑制病毒增殖的作用,但具体机制尚不明了.本研究首先根据NCBI公布的鸡全基因组序列进行生物信息学分析,预测鸡AID的基因序列并通过设计PCR引物,成功在鸡法氏囊cDNA样品中扩增出鸡AID基因(chAID).通过建立并优化chAID的实时定量RT-PCR(rRT-PCR)检测方法,对鸡体内AID基因的表达以及其分布情况进行检测;同时通过Western blot试验对chAID蛋白在鸡细胞中的表达进行检测.试验显示,chAID基因能够在鸡细胞以及组织脏器中正常表达.新城疫病毒(NDV)感染鸡原代成纤维细胞、淋巴细胞后能够引起内源性chAID基因水平的上调;SPF鸡感染NDV后,chAID基因在不同组织脏器中均出现上调现象,以免疫器官最为显著.为了进一步验证chAID基因对NDV增殖的影响,进一步检测在细胞中过表达chAID基因对NDV增殖的影响.研究结果表明:重组chAID蛋白具有抗NDV病毒活性,本研究为进一步研究鸡chAID奠定了基础.
目的 通过Morris水迷宫测试阿尔茨海默病(AD)小鼠空间学习记忆能力,检测AD小鼠脑海马中神经免疫炎性因子的蛋白阳性表达强度及脑组织中神经免疫炎性因子的mRNA水平,探讨音乐疗法对AD小鼠的免疫治疗作用机制.方法 选取野生C57BL/6和转基因APP/V717雌性小鼠作为研究对象,实验分为对照组(C57BL/6野生雌性小鼠,n=10)、AD模型组(APP/V717转基因雌性小鼠,n=10)和音乐治疗组(APP/V717转基因雌性小鼠,n=10).三组均给予正常饮食,音乐治疗组每天早、中、晚各一次给予音量40~65 dB舒缓、轻柔性的音乐30 min,共治疗12 w.采用Morris水迷宫检测各组小鼠空间学习记忆能力;苏木精-伊红(HE)染色在普通光学显微镜下观察各组小鼠脑海马神经细胞损伤程度;免疫组织化学染色检测各组小鼠脑海马中神经免疫炎性因子IL-1β、IL-6、TNF-α的蛋白阳性表达强度;实时荧光定量PCR法检测各组小鼠脑组织中神经免疫炎性因子IL-1β、IL-6、TNF-α、5-羟色胺(HT)和去甲肾上腺素(NE)的mRNA水平.结果 (1)Morris水迷宫检测结果:与模型组比较,音乐治疗组空间学习记忆能力明显提高(P<0.05).(2)HE染色可见音乐治疗组小鼠脑海马神经细胞固缩坏死较模型组明显减轻.(3)免疫组织化学染色可见音乐治疗组小鼠脑海马中神经免疫炎性因子IL-1β、IL-6、TNF-α的蛋白阳性表达强度均较模型组明显降低(P<0.05).(4)实时荧光定量PCR检测可见音乐治疗组小鼠脑组织中神经免疫炎性因子IL-1β、IL-6、TNF-αmRNA水平均较模型组明显降低(P<0.05),5-HT和NE mRNA水平均较模型组明显升高(P<0.05).结论 音乐治疗能够提高AD小鼠的空间学习记忆能力,同时通过下调AD小鼠脑海马及脑组织中神经免疫炎性因子IL-1β、IL-6、TNF-α的蛋白阳性表达强度和mRNA水平,上调神经免疫炎性因子5-HT和NE mRNA水平,在提高机体神经免疫治疗方面作用显著.
副黏病毒磷蛋白,即P蛋白(phosphoprotein),富含丝氨酸和苏氨酸,在自然条件下磷酸化水平较高.副黏病毒P蛋白的磷酸化对病毒的转录和复制具有重要的作用,但新城疫病毒(Newcastle disease virus,NDV)P蛋白磷酸化位点及其功能依然尚不明确.本研究通过LC-MS/MS质谱对NDV La Sota病毒P蛋白的磷酸化位点进行了检测,鉴定了Ser56、Ser77、Thr78、Thr82、Ser164和Ser141六个磷酸化位点.通过在线分析,预测这些磷酸化位点的磷酸化激酶分别是PKC、CKII和GSK3.为了鉴定6个磷酸化位点的生物学功能,在已建立的表达GFP基因的NDV微型基因组质粒pTVT-LGT基础上,用荧光素酶报告基因(luciferase)替换GFP报告基因,建立了表达荧光素酶的NDV微型基因组系统.利用“丙氨酸扫描”对6个磷酸化位点进行突变后,在NDV微型基因组平台中进行功能鉴定.结果 显示,T78、T82和S164位磷酸化位点的缺失显著影响了病毒RNA依赖性RNA聚合酶复合体(RNA dependent RNA polymerase,RdRp)的复制,其生物学意义有待进一步的研究.
Newcastle disease virus (NDV) V protein is crucial for viral interferon (IFN) antagonism and virulence, determining its host range restriction. However, little information is available on the B cell epitopes of V protein and the subcellular movement of V protein in the process of NDV infection. In this study, the monoclonal antibody (mAb) clone 3D7 against genotype VII NDV V protein was generated by immunizing mice with a purified recombinant His-tagged carboxyl-terminal domain (CTD) region of V protein. Fine epitope mapping analysis and B-cell epitope prediction indicated that mAb 3D7 recognized a linear epitope (152)RGPAELWK(159), which is located in the V protein CTD region. Sequence alignment showed that the mAb clone 3D7-recognized epitope is highly conserved among Class II genotype VII NDV strains, but not among other genotypes, suggesting it could serve as a genetic marker to differentiate NDV genotypes. Furthermore, the movement of V protein during NDV replication in infected cells were determined by using this mAb. It was found that V protein localized around the nucleus during virus replication. The establishment of V protein-specific mAb and identification of its epitope extend our understanding of the antigenic characteristics of V protein and provide a basis for the development of epitope-based diagnostic assays.
For high precision time synchronization demands of long term on orbit spacecraft,an autonomous high precision time management system is proposed,which combines Beidou navigation equipment clock generator and frequency synthesizer clock generator.Two clock generators can autonomously switch by states of navigation and localization,it eliminates error accumulation effect of frequency clock generator system and solves the problem for time precision falling dramatically when Beidou navigation equipment is unable to be located.By building error model for system taking an real application in spacecraft for example,to compute and analyze,which indicates that precision of the system time synchronization is prior to 37.8 μs.The result can be a reference for subsequent design of high precision time management system spacecraft.
The type, tissue distribution and function of avian adenosine deaminase acting on double-stranded RNA (ADAR) have not been well characterized although recently it has been extensively studied for its roles in antiviral effect and virus evolution. In this study, ADAR2 mRNA was amplified from primary CEF cells previously stimulated with ConA, poly I:C and Newcastle disease virus (NDV). The eukaryotic plasmid expressing green fluorescent protein (GFP)-labeled ADAR2 was constructed to determine the antiviral effect of ADAR2. In the transfected DF1 cells, the GFP-labeled ADAR2 was visualized in cellular nuclei and growth of NDV was obviously inhibited, suggesting chicken ADAR2 might play a role in the anti-NDV activity. However, the detailed mechanism of ADAR2 mediated anti-NDV effect need to be carried out in the future.
Zinc protein ZEB1(zinc finger E-box binding homeobox 1) is a transcription repressor, which is highly conserved in different species. To determine the correlation of ZEB1 with Newcastle disease virus(NDV) infection, the gene of chicken ZEB1 was PCR-amplified from DF-1 cells and used for phylogenetic and bioinformatic structure analysis. The expression of ZEB1 in NDV-infected DF1 cells, CEF cells and SPF chickens were examined in Real-time PCR. The results showed that NDV infection introduced high expression of chicken ZEB1, suggesting ZEB1 protein might play a role in the life cycle of NDV. Nevertheless, higher expression of ZEB1 did not affect the growth and protein expression of NDV. Therefore, the biological role of Zinc protein ZEB1 need to be further studied.
目的:观察联合应用抗焦虑药物盐酸帕罗西汀和劳拉西泮治疗顽固性高血压患者治疗前后焦虑症状及血压控制情况。方法:收录诊断为顽固性高血压患者64例,随机分成两组,对照组32例,治疗组32例。所有患者均在常规抗高血压药物治疗及低盐饮食、健康教育的治疗,治疗组在常规治疗的基础上加用抗焦虑药物盐酸帕罗西汀20 mg/ d和劳拉西泮0.5 mg/ d 治疗4周,对照组常规治疗治疗4周。比较治疗前后患者收缩压(SBP)、舒张压(DBP)和汉密尔顿焦虑量表(HAMA)评分的变化。结果:抗焦虑治疗4周后,两组患者 HAMA、SBP、SDP 治疗后均比治疗前有所下降,治疗组比对照组血压下降更为显著,差异有统计学意义(P ﹤0.01);治疗后 SBP、DBP、HAMA 比较,治疗组优于对照组,差异有统计学意义(P ﹤0.05)。结论:应用抗焦虑药物治疗顽固性高血压可以有效地帮助患者降低血压,促进患者血压达标。
Objective To investigate Association of Adiponectin Gene Single Nucleotide Polymorphism with Gestational Dia-betes Mellitus.Method 150 cases of normal pregnant women in the people′s hospital of Shenzhen were selected,and 150 cases of gestational diabetes were selected.PCR -RFLP method was used to determine the distributions of allele and genotype frequencies of SNP +45T/G polymorphism and +276G/T.Results ①SNP +45T/G:genotype frequency distribution,GDM group com-pared with control group,the difference was statistically significant(P <0.05);G allele frequency in GDMgroup was higher than the control group,the difference was statistically significant(P <0.05).②SNP +276G/T:genotype frequency distribution,GDM group compared with control group,there was no statistically significant difference(P >0.05).G and t allele frequency in GDM group was higher than the control group,there was no statistically significant difference(P >0.05).Conclusion There is a certain relationship between the occurrence of +45T/G and GDMin adiponectin single nucleotide polymorphism;The occurrence of sin-gle nucleotide polymorphism +276G/T is not significantly correlated with the occurrence of GDM.
在对过程控制实验装置上某一液位测量信号分析的基础上,提出了兼顾平滑性和准确性的滤波性能指标.应用该指标分别编写了查询一阶惯性滤波法、中位值滤波法和平均值滤波法的最佳滤波参数程序.通过对3种方法最佳滤波参数的分析,确定了一阶惯性滤波法更适合过程控制实验装置的这一液位信号,该方法的实验室装置应用效果也证明了其有效性.
Objective To investigate the prevention of two different ways of hydration to aged contrast induced nephropathy after coronary angiography. Method 300 cases undergoing coronary angiography in elderly patients were randomly divided into experimental group and control group,150 cases in each group. The experimental group got intravenous infusion of normal saline at a speed of 1 ml/(kg·h),10 hours before coronary angiography. in addition,1 000 ml water was drinked within 2 h by the ex-perimental group. The control group only received conventional hydration before coronary angiography and normal saline 500 ml in less than 6 hours after it. We observed serum creatinine,cystatin C levels of the two groups before and 48 and 72 hours after coro-nary angiography. Results Serum creatinine and cystatin C levels were not statistically significant before coronary angiography be-tween the experiment group and the control group(P > 0. 05),but they were significantly lower in the experimental group than that of the control group both 48 and 72 hours after coronary angiography(P < 0. 05). Conclusion The way of hydration in the experimental group is better for renal function,so it can reduce contrast induced nephropathy,worthy of promotion.

目前我国旅游业发展中的生态文明导向已十分清晰,然而在不同旅游利益相关者层面的生态旅游实践却存在诸多困境.当前我国生态旅游存在政府行为失当、旅游企业不合理问题、居民参与度不高及旅游者生态旅游意识缺乏等诸多困境,从政府制度建设、旅游企业产品及基础设施建设、居民生态旅游教育、生态旅游者自我学习等方面探讨我国生态旅游发展的路径,以期为当前我国旅游生态文明建设提供一定参考.
Objective: To explore the expressions and significances of estrogen receptor( ER) and progesterone receptor( PR) in recurrent uterine leiomyoma. Methods: One hundred patients willing to undergo myomectomy in the hospital were randomly selected,including 50 patients undergoing myomectomy for the first time and 50 patients undergoing myomectomy for the second time. Fasting blood specimens were obtained in the morning before myomectomy; the specimens of uterine leiomyoma and normal cervical smooth muscle adjacent to uterine leiomyoma were abstracted during surgery; radioimmunoassay was used to detect the serum levels of estrogen and progesterone; ELISA was used to detect the contents of ER and PR in serum,uterine leiomyoma and normal cervical smooth muscle adjacent to uterine leiomyoma; immunohistochemical method was used to detect the positive expression intensities of ER and PR in uterine leiomyoma and normal cervical smooth muscle adjacent to uterine leiomyoma of patients undergoing myomectomy for the first time,uterine leiomyoma and normal cervical smooth muscle adjacent to uterine leiomyoma of patients undergoing myomectomy for the second time. Results: There was no statistically significant difference in the serum levels of estrogen and progesterone between uterine leiomyoma of patients undergoing myomectomy for the first time and uterine leiomyoma of patients undergoing myomectomy for the second time( P 0. 05); the contents and positive expression intensities of ER and PR in serum,uterine leiomyoma and normal cervical smooth muscle adjacent to uterine leiomyoma of patients undergoing myomectomy for the second time were statistically significantly higher than those of patients undergoing myomectomy for the first time( P 0. 01); the contents and positive expression intensities of ER and PR in all the patients increased from normal cervical smooth muscle adjacent to uterine leiomyoma to uterine leiomyoma,there were statistically significant differences( P 0. 05). Conclusion: Up-regulations of ER and PR in uterine leiomyoma and normal cervical smooth muscle adjacent to uterine leiomyoma of patients undergoing myomectomy for the second time may be induced by high hormone status because of local estrogen integrated to progesterone,it is speculated that ER and PR may be related factors of recurrent uterine leiomyoma.
目的:探讨妊娠合并急性胰腺炎的高危因素、诊断、治疗方法与预防。方法:回顾性分析13例妊娠合并急性胰腺炎患者的临床资料。结果:13例中均有上腹痛的表现,均有不同程度的血、尿淀粉酶的增高,13例中有胆囊炎或胆道结石病史6例,高脂血症4例,合并重度子痫前期1例;采用保守治疗10例,手术治疗3例;孕产妇死亡1例,胎死宫内1例。结论:并发胆囊炎或胆石症、高脂血症是妊娠期急性胰腺炎的高危因素,熟悉妊娠合并急性胰腺炎临床表现,早期诊断与治疗是预后的关键。
研究差异化结构框架下的软件故障检测问题。当前软件结构呈现差异化趋势越发明显,软件集成性使得整体结构的衔接关联性出现淡化。结构接口之间无明显衔接特征。传统的软件故障检测方法在进行差异化结构框架下软件故障定位时,由于缺少明确的接口衔接特征进行定位区域指示,造成软件故障信号归属区域无法认定,造成定位不准。提出采用差异结构融合算法的软件故障检测方法。利用信号融合技术,对软件故障检测信号进行有效的融合处理,从而为软件故障检测提供基础。利用分布式协同检测方法,对软件故障特征进行有效的特征归类检测,从而完成软件故障检测。实验结果表明,利用上述算法进行软件故障检测,能够极大地提高检测的准确性。
针对通用搜索引擎存在搜索不够快速、不够深入、不够准确的缺点,本文设计并实现了旅游信息搜索网络蜘蛛,给出了该网络蜘蛛对旅游网页的主题相关度进行预测和判断的算法.该算法保证了网络蜘蛛只采集旅游主题相关的网页,使得垂直搜索引擎在查询的准确率和效率上都有显著地提高.最后通过实现一个旅游信息垂直搜索系统,给出了其信息搜集模块、信息处理模块和信息搜索模块的设计与实现.
