Objective: In order to obtain information of functional genes,a normalized cDNA libray from Flue-cured tobacco variety of Nanjiang-3(Nicotiana tobacum cv.Nanjiang-3) was constructed.Method: RNA was extracted from leavies and flowers of Nanjiang-3 according to RNeasy Plant Mini Kit manipulation,the first cDNA was synthesized by reverse transcriptase.Double cDNA obtianed through long-distance PCR(LD-PCR) amplification method as template,the normalized cDNA library of Nanjiang-3 was constructed by the SMART technology which based on the double strand specific nuclease(Duplex-Specific Nuclease,DSN).Result: The normalized cDNA library was constructed,which contained 1.26×106 independent clones,its recombination ratio was 97.47%,average insertion size was above 1.2kb.Random selected 48 clones were tested by PCR,then 20 positive clones were sequenced and BLAST,the results showed that many genes and expression sequence tags(ESTs) were contained in the cDNA pool.Conclusion: Basis material information of research for genetic resources and gene function of Nanjiang-3 were provided by the cDNA library.
A new flue-cured variety Nanjiang 3 was developed from a mutant of Honghua Dajinyuan using the systematic breeding method. It had wide adaptability with moderately resistant to brown spot, black shank and granville wilt, yet slightly subject to damage from weather fleck, and its leaves were easy to cure. This variety produced good appearance quality, acceptable nicotine content and suitable chemical components. It yielded about the same as K326 with equal smoking quality, and was met with the demands of the raw materials in Chinese-style cigarettes. Its leaf yield and value were higher than those of K326 by 15.71% and 33.93% respectively. Nanjiang 3 has high production value.
According to the introduction procedure of tobacco variety,K326LF was introduced from Brazil to Guizhou by Guizhou Tobacco Corporation in 1998.Introduction test,variety regional test and productive demonstration were conducted in 1998-2003.The results showed that K326LF had the same resistance to major diseases as K326,its cured leaf yield and value were 182.10 kg/ha and 1 252.95 yuan/ha,respectively,which was higher than those of K326.The main chemical components of flue-cured tobacco leaves and smoking quality met the raw material requirement in Chinese-style cigarettes.K326LF is a promising variety in China at present.
Mutants of tobacco are very important materials in tobacco functional genomics.In this paper,the roles of toabacco mutants in genes were reviewed,followed by the construction of tobacco mutant pools,the related methods and its future application to tobacco functional genomics.
flue-cured tobacco varieties introduced newly were evaluated comprehensively by fuzzy analysis on 13 traits. The results indicated that the comprehensive characters of variety RGH51 were more excellent than K326, expressing on it's high yield, output value, average price and the rate of high-grade tobacco, and lower occurence rate of brown spot disease. The varietiy PVH08 was a good one but less than RGH51. It was testified to be better in yield, output value, average price, the rate of high-grade (tobacco), and the contents of K_2O and nicotine, and lower occurence rate of the root knot nematode disease. It should be taken part in the next production-text. The varieties PVH01, PVH02, PVH03, PVH05, PVH06 and RGH12 were of common varieties on their comprehensive characters and were suggested to study their traits under different environments. The varieties PVH09 and RGH04 needed to conducting further test.
1998~1999年从巴西Profigen公司引进的12个烤烟品种,经过云南出入境检验检疫局室内检疫和隔离种植,没有发现烟草霜霉病,2000~2001年以K326为对照进行田间引种观察试验和抗病性鉴定,设云南和贵州两个点.两年的试验结果表明,12个新品种的生育期与对照K326基本相近,除了RGHl2、RGH51、NC55和K236感TMV外,其它品种抗TMV.各新品种都抗黑胫病,对照K326中抗黑胫病.RGH51中抗南方根结线虫病,K326抗,其它品种都感南方根结线虫病.NC55中感青枯病,其它品种中抗青枯病.各品种都感赤星病.综合各经济性状来看,PVH09、RGH04、RGH51、PVH03、PVH01表现较好,PVH06、PVH05、NC55、PVH07等表现中等,PVH08和K326表现较差.

应布菲金(Profigen)巴西公司邀请,以中国烟叶生产购销公司周义和为团长,由中国烟草育种研究南北方中心、贵州省烟草科学研究所、东北烟草试验站组成的6人的烟草种子技术培训团,2001年1月4日至2月6赴巴西进行烟草种子技术培训.在巴西的28天,参观考察了英美烟草公司(BAT)在巴西的苏兹·克鲁斯公司(SOUZA CRUZ)、大陆公司(Meridional)、Profigen巴西种子公司及其杂交种和常规种繁种基地,访问了20多个农户,还考察了巴西主产烟区南大河州、桑塔·卡拉莉那州和巴拉那州,对巴西烟草生产概况、烟叶收购、加工、销售、出口、烟草种子生产加工、新品种的培育等内容进行实地考察.现就着重介绍布菲金巴西公司烟草育种和种子的生产、加工、销售等情况.
贵烟11号是以76D-1×77E-1的6代中间材料为母本,以NC89为父本杂交选育而成的烤烟新品种。几年多点试验示范表明,该品种主要化学成分含量适宜,内在质量与贵州省主栽品种K326相当,适合卷烟配方使用。平均亩产154.23kg,均价6.50元/kg,亩产值1002.50元。贵烟11号与对照品种K326相比,亩产量提高12.26%,均价提高7.26%,亩产值提高20.41%,上等烟增加9.18个百分点。抗黑胫病,抗爪哇根结线虫病和气候斑点病,中抗赤星病和青枯病,耐CMV和TMV能力比K326强。具有优质、稳产、经济效益高、适应性广和易烘烤等优点。1997年1月通过全国烟草品种审定委员会审定,定名为贵烟11号。
