AIM:To explore the mechanism of 3-nitropropionic acid (3-NPA) preconditioning that induces cerebral ischemic tolerance in rats by affecting the expression of brain-type glucose transporters (GLUT1 and GLUT3) at mRNA and protein levels in cerebral tissues.METHODS: The male SD rats were used in the experiments and divided randomly into sham operation group (sham group, n=4), control group of 3-NPA preconditioning (3-NPA group, n=4), cerebral ischemia group (M group, n=16) and 3-NPA preconditioning group (IPC group, n=16). M group and IPC group were further divided into 4 subgroups according to the different reperfusion time(4 h, 12 h, 24 h and 48 h). All rats were killed at the corresponding time points. The cerebral tissues in the ischemic side (left) and coronal intermediate 1/3 of cortex were collected. The protein levels and mRNA expression of GLUT1 and GLUT3 were determined by Western blotting and RT-PCR. RESULTS: Compared with M group, the ischemic reperfusion and 3-NPA preconditioning induced the upregulation of GLUT1 and GLUT3 at protein levels with significant differences (F=5.848, P<0.05 and F=6.295, P<0.05, respectively), especially after ischemia-reperfusion for 48 h. The mRNA expression of GLUT1 in IPC group began to increase at 4 h, peaked at 48 h after reperfusion, with significant difference as compared to M group at the corresponding reperfusion time points in each group or sham group. In contrast, the mRNA expression of GLUT3 in IPC group increased at 24 h, and was the highest at 48 h as compared to cerebral ischemia group at the corresponding reperfusion time points or sham group.CONCLUSION: 3-NPA preconditioning increases the expression of GLUT1 and GLUT3 at protein and mRNA levels to maintain the energy supply in brain tissues, indicating a cerebral protective mechanism.

AIM: To explore the effect of high purified human chorionic gonadotropin (hCGs) and glycodelins isolated from amniotic fluid and serum of pregnant women on E-selectin mediated endothelial cell adhesion. METHODS: The hCGs and glycodelins in human amniotic fluid and serum collected from pregnant women were purified by using chromatography. The binding of ~ 51 Cr-labelled HepG2 cells to IL-1β stimutated human umbilical vein endothelial cells (HUVECs) in the presence of increasing concentrations of hCGs and glycodelins was quantified. The effect of hCG and glycodelin on E-selectin-mediated endothelial cell adhesion was observed. RESULTS: HCGs isolated from amniotic fluid and serum of pregnant women inhibited the E-selectin mediated adhesion of HepG2 cells to HUVECs in a dose-dependent manner, the IC_ 50 was 3.2×10~ -6 mol/L and 6.3×10~ -6 mol/L, respectively and the relative inhibition was 1.5×10~4 and 1.1×10~4, respectively, compared to sialyl-Lewis~x. The IC_ 50 of amniotic fluid glycodelin and serum glycodelin was 3.2×10~ -6 mol/L and 6.3×10~ -6 mol/L respectively, and the relative inhibition was 7.2×10~2 and 3.7×10~2, respectively, compared with sialyl-Lewis~x. CONCLUSION: Pregnancy-associated glycoproteins, hCG and glycodelin, are the powerful inhibitors for E-selectin-mediated endothelial cell adhesion.
AIM: To investigate the influence of hepatic portal interdicting on gastric mucosa of the liver cirrhosis rats and the protective effect of famotidine on gastric mucosa. METHODS: Fifty Wistar rats were randomly divided into 5 groups; group A (healthy rats), group B (healthy rats with interdicted hepatic portal), group C (liver cirrhosis rats without interdicted hepatic portal), group D (liver cirrhosis rats with interdicted hepatic portal) and group E (liver cirrhosis rats without interdicted hepatic portal and use of famotidine); The gastricfluid pH, gastric mucous content (GMC), gastric mucosa blood flow (GMBF) and the damage index of gastric mucosa (DIGM) of the every group were observed. RESULTS: There is no significant difference in the above parameters between group A and group B (P0.05). Compared with group A, GMBF of group C, D, and E were reduced (P0.01), pH, GMC of group C, D and E were also reduced (P0.05); DIGM of group C, D and E were increased (P0.01). All the changes of group D were more obvious than those of group C (P0.01); While there was no significant difference in all the parameters between group E and group C (P0.05). CONCLUSION: Hepatic portal interdicting can aggravate the gastric mucosa damage of the liver cirrhosis rat, and famotidine can protect against such gastric mucosa injury through improving the microcirculation of gastric mucosa.

AIM: To study the expression and significance of osteopontin (OPN) in endometrial carcinoma and cervical cancer. METHODS: Immunohistochemical S - P assay was used to detect the expression of OPN in paraffin - embedded sections of 30 cases of endometrial carcinoma, 20 cases of cervical cancer and 30 cases of normal control tissues. The relationship between OPN expression and clinical- pathological characteristics was evaluated. RESULTS: The positive immunostaining rates of OPN in endometrial carcinoma (70% ) and cervical cancers (55% ) were significantly higher than that in the normal secretive and proliforative endometrium (50% and 10%) and normal cervical epithelium (10%), respectively (P<0.01). The positive immunostaining rate of OPN in sopiamous cell carcinoma and adenocarcinoma of the cervix was 53.3% and 60% , respectively, there was no significant difference between the two groups (P>0.05). The positive immunostaining rate of OPN in stage Ⅲ and stage Ⅱand G3 and G2 of endometrial carcinoma was significantly higher than that in stage Ⅰ and type G1 of endometrial carcinoma. The positive immunostaining rate of OPN in stage Ⅱ b and type G3 of cervical cancer was significantly higher than that in stage Ⅰa and type G1 of cervical cancer. CONCLUSION: OPN is significantly highly expressed in both endometrial carcinoma and cervical cancer, and its expression is closely related to the stage and grading of these malignant tumors.
AIM: To investigate the relationship among MCP-1 and monocyte chemoattract protein activity (MCA) and pathogenesis of lung cancer. METHODS: 173 patients were involved in the study and divided into three groups: group A: lung cancer group (60 patients); group B: benign lung disease group (55 patients) and group C: healthy control group (58 patients). MCP-1 level and MCA in bronchoalveolar lavage fluid (BALF) were measured. RESULTS: The concentration of MCP-1 and MCA in BALF in group A were much higher than those in group B and group C (P0.01), but the increase in MCA were not in direct proportion to the increase in MCP-1 in the patients with lung cancer. CONCLUSION: Monocyte chemoattract activity of the patients with lung cancer may be insufficient.
Objective: To explore the effect of HCGs isolated from human amniotic fluid, serum, and urine of pregnant women on cell adhesion mediated by E-selectin. Methods: The adhesion properties of 51 Cr-labelled HepG2 cells, which carry the ligands of membrane-bound sialyl-Lewis x epitope for E-selectin, were detected to recombinant E-selectin by means of adding different concentrations and sources of HCGs as potential inhibitors, and meantime, the effect of HCGs isolated from amniotic fluid, serum and urine in E-selectin mediated cell adhesion was assessed. Results: HCGs isolated from human amniotic fluid and serum of pregnant women inhibited the adhesion of HepG2 cells to E-Selectin in a dose-dependent manner, the relative inhibition was 2.4×10 4 and 4.5×10 3, respectively, compared with sialyl-Lewis x. But HCG isolated from urine of pregnant women did no effect. Conclusions: HCGs isolated from human amniotic fluid and serum of pregnant women is an effective ligand for E-selectin.
AIM: To investigate the effect of hypoxia on glyceraldehyde-3-phosphate dehydrogenase (GAPDH), β-actin and endothelin-converting enzyme (ECE)-2 mRNA levels in cultured mouse brain astrocytes (AC). METHODS: AC from neonatal mouse brain was incubated for 24 h in serum-free medium under hypoxic or normoxic conditions. The amount of transferred RNA was estimated using ethidium bromide stained 28S rRNA and 18S rRNA. The levels of tested mRNA were evaluated by Northern blot RNA hybridization. RESULTS: The GAPDH mRNA was up-regulated to 503.0% of the normoxic controls in hypoxic AC (n=10. P0.01), while the level of β-actin mRNA in hypoxic AC was down-regulated to 50.3% of control levels (n=6. P0.01). In contrast, the ECE-2 mRNA level was not affected by hypoxia. CONCLUSION: The results indicate that GAPDH and β-actin are not suitable for the internal standards for quantitative mRNA expression analysis in AC after hypoxia or ischemia. ECE-2 may function as an alternative in such circumstance.
AIM:To observe protective effect of L-glutamine on myocardial cell injury induced by endotoxin. METHODS:22 male SD rats,weighting(250±30)g,were randomly divided into three groups:control group( n= 7),endotoxin group( n= 7),glutamine/endotoxin group( n= 8). Heart were isolated from rats and perfused on Langendorff apparatus with Krebs- Henseleit(K-H)buffer(Saturation 95%O 2+5%CO 2)at a constant pressure(8.33 kPa)and temperature(37℃). The activity of superoxide dismutase (SOD)and malondialdehyde (MDA)content of efflux from coronary artery, monophasic action potential(MAP)and contractile force were measured at certain timepoints (0 min,20 min,50 min,80 min). RESULTS: Monophasic action potential and contractile force were markedly improved in Gln/ET group. The activity of SOD and MDA level in Gln/ET group restored closely to that in control group. CONCLUSION:L-gluta mine protects myocardial cells from injure induced by endotoxin.

AIM: To further investigate the role of centralcorticotropin-releasing hormone in stress-induced hyperthermia and lipopolysaccharide (LPS)-induced fever in the rat. METHODS: Test substances were administered intracerebroventricularly (icv) via a third ventricle cannula. Body temperature responses were monitored at 30 min intervals using colonic thermistor probes. Arginine vasopressin (AVP) level in the ventral septal area (VSA) determined by radioimmunoassay. RESULTS: In normal saline controls, rats were handled to take the colonic temperature, their body temperature significantly increased with a peak of (0.88±0.31)℃. The injection (icv) of α-helical CRH(9-41), a CRH-41 receptor antagonists, markedly attenuated the stress-induced hyperthermia within 90 min after injection of normal saline. LPS(300 ng, icv) stimulated a biphasic rise in the colonic temperature, the 3.5 h thermal response index (TRI3.5) and AVP levels in the VSA of LPS-treated rats were higher than those of control rats. The AVP responses to LPS were inhibited significantly by blockade of central CRH actions using α-helical CRH(9-41) (5 μg ,icv) administered 10 min prior to LPS, while α-helical CRH(9-41) (5 μg ,icv) resulted in exacerbated febrile responses to LPS(300 ng, icv). CONCLUSION: Central CRH plays an important role in stress-induced hyperthermia. The injection (icv) of α-helical CRH(9-41) enhances markedly LPS-induced fever in rats. CRH is a dual action molecule in LPS-induced fever, which itself mediates LPS-induced fever, at the same time, and limits the rise in body temperature during fever through actions of AVP in the VSA and glucocoticoids.
目的:近年来的研究表明,促肾上腺皮质激素释放激素(corticotropin releasing hormone, CRH)是一种重要的中枢发热介质.有资料表明,CRH在下丘脑发挥生理作用涉及cAMP依赖性和非cAMP依赖性两种不同的机制,但CRH发挥致热作用的中枢机制尚不明了.本研究观察了cAMP依赖性蛋白激酶信号传导通路在大鼠CRH性发热机制中作用.结果:第三脑室微量注射CRH(2.5 μg, 5.0 μg, 10 μg)引起大鼠结肠温度和下丘脑cAMP水平明显升高,下丘脑cAMP水平与大鼠2 h发热反应指数之间存在明显的正相关(r=0.994, P<0.01).事先向第三脑室微量注射腺苷酸环化酶抑制剂2',3'-dideoxyadenosine(DDA, 30 μg)或cAMP依赖性蛋白激酶抑制剂Rp-cAMPs(15 μg)均显著抑制中枢注射CRH引起的发热反应.结论:CRH性发热的中枢机制与CRH激活cAMP依赖性蛋白激酶信号传导通路有关.
