We aim to understand the structural basis of prion strains and their pathogenicity mechanisms. Structures of purified authentic ex vivo prions are studied by high-resolution cryoEM. Various modalities of fluorescence microscopy, including correlative light and electron cryo-microscopy (cryoCLEM) will be developed to monitor prion infection pathways in cells, including identification of their interaction partners. This will involve propagation of intrinsically fluorescent prions using genetic code expansion (GCE) technology.
