1Genome-Wide Identification and Expression Analysis of LOX Gene Family in Rice (oryza Sativa L.) under Abiotic Stress Conditions
Ahana Deb Tusti, Md. Nazmul Hasan, Jeba Faizah Rahman, Md. Hammadul Hoque, Anindita Chakraborty, Shamsul Haque Prodhan

Background Lipoxygenases ( LOX s) are enzymes that facilitate the oxygenation of polyunsaturated fatty acids (PUFAs) to create oxylipins through hydroperoxides that are involved in methyl jasmonate (MeJA) signaling pathway and are essential for seed germination, growth and development, defense mechanisms, and responses to stress. This study was designed for systematic analysis and exploration of expression patterns of the LOX gene family of rice. Results Twelve LOX genes were identified, and phylogenetic and structural analyses grouped them into 9-LOXs, Type I 13-LOXs, and Type II 13-LOXs subfamilies with conserved lipoxygenase domains, motif and gene structure patterns. These genes were dispersed unevenly across seven chromosomes. Analysis of gene duplication found four tandem and segmental duplication events predominantly driving their moderate expansion, with Ka/Ks ratios suggesting purifying selection. Syntenic analysis across three genomes indicated relatively conserved roles of orthologous gene pairs of this family. The cis-acting regulatory elements revealed 46 CRE motifs responsive to light, cellular development, hormones and stress. Notably, 151 putative miRNAs were identified as potential post-transcriptional regulators. PPI analysis highlighted 10 key nodes with high interaction degrees. GO enrichment classified the functions of OsLOX s into biological, metabolic and cellular processes. RNA-seq expression profiling demonstrated OsLOX s expression across diverse tissues, developmental stages and stress. RT-qPCR data analysis further validated that OsLOX genes exhibited temporally variable differential expression under multiple abiotic stress conditions including drought, salinity, saline-alkalinity, heat and cold, and in response to IAA hormonal treatment. Conclusion In conclusions, drought and cold stress significantly upregulated certain OsLOX s, enabling rice plants to confront these adverse conditions. This study will underscore the path for forthcoming in-vivo analyses to uncover the molecular processes of LOX genes involved in tolerance to stress in rice.

2Genotypic Evidence of Extended-spectrum Β-Lactamase and Carbapenemase-producing Shigatoxigenic and Intimin-producing Shigatoxigenic Multidrug-resistant Escherichia Coli from Diarrheic Calves: Hidden Zoonotic and Public Health Risk
Plabon Ketan Barua, Uaye Mya, Partha Paul, Snigdha Das, Mukta Das Gupta,Ashutosh Das

Shiga toxin-producing Escherichia coli (STEC) is a pathotype of E . coli associated with a wide variety of diarrhoea in neonatal calves, causing a global economic loss in the dairy industry with significant zoonotic risks via STEC and intimin-producing STEC, resulting in enteric and systemic illness, including diarrhoea, haemorrhagic colitis (HC), and haemolytic uremic syndrome (HUS) in humans. The prominence of multidrug-resistant (MDR) STEC from neonatal diarrhoeic calves is increasing public health risks and restricted treatment alternatives. The prevalence of STEC was investigated in 75 rectal swabs from diarrhoeic calves aged ≤ 12 weeks, collected before initiating antibiotic treatment. The E. coli isolation rate among diarrhoeic calves was 80% (60/75; 95% CI: 69.17–88.35). The presence of stx genes ( stx1 , stx2 ), intimin-producing eae gene, carbapenemase-producing genes ( bla NDM group and bla KPC group), and extended-spectrum β-lactamase genes ( bla TEM group, bla CTX−M group, and bla SHV group) was screened by polymerase chain reaction (PCR). The prevalence of pathogenic E. coli was 13.33% (10/75; 95% CI: 6.5–23.1). The STEC isolates were detected in 12% (9/75, 95% CI: 5.64–21.56). Of these pathogenic E. coli isolates, the STEC with both stx genes, stx1 only, and stx2 only genotypes were present in 40% (4/10) and 10% (1/10), respectively. Intimin-producing STEC isolates ( eae -STEC) were identified in 40% (4/10) among the pathogenic isolates, with the following genotypes: stx1 + eae (2/10, 20%), stx2 + eae (1/10, 10%), and stx1 + stx2 + eae (1/10, 10%). Only one (1/10, 10%) eae- encoded non-STEC isolate was found, called enteropathogenic E. coli (EPEC). All STEC (100%) isolates were tested positive for either ESBL or carbapenemase or both ESBL and carbapenemase in the phenotypic assays. ESBL-producing STEC isolates were genetically identified in 6.67% (5/75) with the following combinations: three isolates coharbored stx1 - bla TEM , two had stx1 - bla SHV , and stx2 - bla TEM , respectively. All intimin-producing STEC ( eae- STEC) isolates were both ESBL and carbapenemase producers, which were identified in 5.33% (4/75), with followings genotypic expression: stx1 + stx2 + eae / bla TEM + bla CTX−M + bla NDM + bla KPC in 1.33% (1/75), stx1 + eae / bla TEM + bla KPC in 1.33% (1/75), stx1 + eae / bla TEM + bla CTX−M +bla KPC in 1.33% (1/75), and stx1 + eae / bla TEM + bla CTX−M +bla KPC in 1.33% (1/75). ESBL and carbapenemase-producing eae -STEC isolates were more likely to be multidrug-resistant (MDR) than ESBL-STEC and EPEC isolates. The highest antimicrobial resistance rates were observed in ciprofloxacin and meropenem (100%), nitrofurantoin and cefoxitin (90%), ampicillin, streptomycin, and trimethoprim-sulfamethoxazole (80%), chloramphenicol (70%), and doxycycline, tetracycline, and cefotaxime (60%). In contrast, the lowest resistance rates were found in gentamycin (20%) and amoxicillin-clavulanate (40%). In terms of pathogenicity, only STEC isolates induced mild to moderate non-bloody diarrhoea, whereas intimin-producing STEC caused severe bloody diarrhoea with 100% mortality (4/4) ( p < 0.05) in neonatal calves. To the best of our knowledge, this is the first report on ESBL and carbapenemase-producing MDR STEC from diarrhoeic calves in Bangladesh.

4Seroprevalence and Risk Factors of Lumpy Skin Disease in Cattle Across Asia and Africa: a Systematic Review and Meta-Analysis, 2000-2025
Md Jahedul Hasan Rocky, Sumsoon Naher Tuly, Mohammad Mehedi Hasan, Farjana Akter, Tanvir Ahmad Nizami,Abdullah Al Faruq,Mahabub Alam,Md Ahasanul Hoque, Tofazzal Md Ra

Background: Lumpy skin disease (LSD), caused by lumpy skin disease virus, is an economically important transboundary cattle disease that has expanded from Africa into Asia in recent decades. Despite the substantial economic impact of LSD, sero-epidemiological evidence remains fragmented across Asia and Africa. Recurrent outbreaks and continuing geographic expansion highlight the need for pooled estimates that can guide surveillance, vaccination, and risk-based control. Objectives: Therefore, this systematic review and meta-analysis aimed to estimate the pooled seroprevalence of LSD in cattle across Asia and Africa from 2000 to 2025 and to identify study-level and animal-level factors associated with variation in seropositivity. Results: Twenty studies involving 13,356 cattle were included. The overall pooled seroprevalence of LSD was 31.2% (95% CI: 21.0-42.5), with substantial between-study heterogeneity (I² = 99.43%, p < 0.001). Pooled seroprevalence was higher in Asia (39.1%) than in Africa (22.2%) and was higher during outbreaks (44.0%) than in endemic settings (26.4%). By diagnostic method, the highest pooled estimate was observed for ELISA (36.1%), followed by virus neutralization test (33.5%) and indirect fluorescent antibody test (11.3%). Studies with smaller sample sizes showed higher pooled seroprevalence than larger studies. Meta-regression indicated that sampling year and sample size contributed significantly to heterogeneity. Risk-factor analysis showed higher odds of LSD seropositivity in cattle aged over two years (OR = 1.47; 95% CI: 1.01-2.15), females (OR = 1.32; 95% CI: 1.06-1.65), crossbred cattle (OR = 2.92; 95% CI: 1.41-6.05), and cattle managed under extensive rearing systems (OR = 1.63; 95% CI: 1.26-2.12). Conclusions: LSD seroprevalence in cattle remains substantial across Asia and Africa, although estimates vary markedly by region, study design, sample size, diagnostic method, and management factors. The findings support the need for harmonized serological surveillance, standardized reporting, risk-based vaccination, and targeted control strategies, particularly in high-risk production systems and regions with active transboundary spread.

5An Updated Checklist of the Amphibians of Bangladesh: Taxonomic Diversity, Distribution, and Conservation Status
    Inan Rayan, Monirul Islam

Bangladesh is situated at the confluence of the Indo-Himalayan and Indo-Burma biodiversity hot spots and is home to a rich but poorly known amphibian fauna. By synthesizing published literature, museum records, historical reports, online biodiversity databases, recent taxonomic revisions, newly described species, and verified field observations, this study provides an updated checklist of amphibians of Bangladesh. The revised checklist recognizes 96 native amphibian species in 2 orders, 9 families, and 37 genera, of which 94 species are anurans and 2 are gymnophionan species, and no member of the order Urodela has been recorded from the country. Of the species, 6.25% are endemics and 93.75% are native non-endemics. The most species-rich family is Dicroglossidae (26 species), followed by Rhacophoridae (24 species), Ranidae (16 species), Microhylidae (15 species), Megophryidae (7 species), Bufonidae (5 species) and Chikilidae, Ichthyophiidae and Hylidae (1 species each). Several taxa are still only supported by historical or controversial records and need further confirmation. The study also identifies possible high-risk areas for the predicted distribution of Batrachochytrium dendrobatidis, highlighting emerging conservation issues. This revised checklist provides a critical baseline for further taxonomic, ecological and conservation studies, and provides important information for biodiversity assessment and habitat management in Bangladesh.
