1Efficacy and Safety of Seawater-Based Nasal Irrigation on Nasal Congestion in Infants and Toddlers with Common Cold: a Randomized Controlled Trial
Oliver Chen, Constance Prime, William Hooper, Thomas Vanderzande, Marita Alegre, Anna Rynkiewicz, Anetta Karpińska-Nowak, Annahita Ghassemi

Nasal congestion associated with the common cold is a frequent source of distress in infants and toddlers. While hypertonic seawater solutions offer a physiological mechanism for decongestion via osmosis, robust clinical evidence supporting their use as an adjunct to standard care in this specific pediatric population is essential to validate their therapeutic value. To evaluate the efficacy and safety of a hypertonic seawater nasal spray enriched with copper and manganese combined with standard of care compared to standard of care alone in alleviating nasal congestion, improving sleep quality, and accelerating symptom recovery in infants and toddlers aged 3–48 months experiencing the common cold without high fever (excluding acute rhinosinusitis). In a randomized, open-label, controlled parallel study, 220 participants received standard care (which included ample hydration, at-home rest, and the use of paracetamol as needed) with or without the nasal spray. Outcomes, including symptom severity, recovery rate, sleep quality, secondary infections, and caregiver satisfaction, were assessed via questionnaires over 10 days. Participants using the nasal spray experienced a significant (p < 0.05) reduction in nasal congestion from the evening of Day 0 through Day 5, with immediate symptom relief observed within 5–15 min of application. By Day 10, recovery rates were higher in the nasal spray group (75.3 https://clinicaltrials.gov/search?cond=NCT05365789 ).

2Effects of Alkali Metal Chloride and Carbonate Salts on Micellization of a Nonionic Surfactant
    Steven T. Adamy

Despite a substantial number of studies on the effects of salts on micellization behavior in surfactant systems reported in the literature, studies including carbonate salts are not easily found. The effects of salt concentration on nonionic surfactant critical micelle concentrations (CMC) are well known, for example via the linear dependence of the log of CMC on salt concentration. In this study, measurements of CMC of an ethoxylated alcohol surfactant in the presence of chloride and carbonate salts of Li+, Na+, K+, Rb+, and Cs+ are presented. It was found that CMC reduction behaviors were different between chloride and carbonate salts. Studies of conductivity showed much higher degrees of salt-surfactant interaction in carbonate systems. Light scattering studies showed slightly larger micelle sizes in carbonate systems, suggesting higher aggregation numbers or interaction of the salts with the micelle. The differing outcomes in chloride and carbonate systems may have been due to differing degrees to which the salts affect the aqueous bulk phase and the extent to which the salt interacts with the surfactant.

3Limosilactobacillus Reuteri 3613-1 Delays Onset of Unconfirmed Urinary Tract Infections in Otherwise Healthy Women
Valentine Turpin, Charles Kakilla, Jessica Foote, Oliver Chen, William Hooper, Wafaa Ayad, Annahita Ghassemi, Noah Zimmermann, Kieran Rea, Amy Wescott

Urinary tract infections (UTIs) impose a substantial burden on women’s health, and probiotics have emerged as an alternative strategy to support urogenital wellbeing. This study evaluated the antimicrobial properties of Limosilactobacillus reuteri 3613-1 and its ability to improve UTI outcomes in women with a history of recurrent uncomplicated UTIs. In vitro assays demonstrated that L. reuteri 3613-1 inhibited the growth of Escherichia coli isolates and proved superior inhibition of Gardnerella vaginalis and Candida albicans compared with a comparator L. reuteri strain, supported by confirmed reuterin production and genomic profiling. A randomized, double-blind, placebo-controlled clinical trial (n = 130) assessed daily supplementation with L. reuteri 3613-1 for 24 weeks. While the proportion, frequency, and intensity of confirmed UTIs did not differ significantly between groups, L. reuteri 3613-1 delayed the onset of the first UTI, reaching significance in participants with suspected while unconfirmed UTIs. Vaginal pH and vaginal microbiome composition remained stable and comparable between groups across the intervention. The product was safe and well tolerated. Overall, L. reuteri 3613-1 shows promise as a probiotic candidate with antimicrobial activity and potential to delay symptom onset in women susceptible to recurrent UTIs, warranting further investigation in larger studies.

4Organ Growth and Fermentation Profiles of Broilers Differing in Body Growth Rate
    Brad Gorenz,Maci L. Oelschlager,Julianna C. Jespersen,Chang Cao,Alexandra H. Smith,Roderick I. Mackie,Ryan N. Dilger

This study sought to determine the relationship among broiler performance, organ development, and indicators of microbiota colonization. A total of 1,200 two-day-old male Ross 308 broiler chicks, divided among 3 cohorts of equal size, were housed in battery cages, and allotted based on body weight. On study d 11, birds were weighed, and birds with BW gain within the 10th and 90th percentiles were assigned to the Slow and Fast groups, respectively. Birds (n = 30 for each group) selected on d 11 were provided water and a corn-soybean meal-based diet ad libitum while maintained individually through study d 25 (i.e., a 14-d growth period). Parameters regarding growth performance, organ and intestine weights and lengths, and intestinal volatile fatty acid concentrations were measured. All data were analyzed by one-way ANOVA using the Mixed procedure of SAS. Fast birds exhibited greater (P < 0.001) BW gain and feed intake than slow birds, but feed conversion ratio (FCR) did not differ (P = 0.19). Additionally, Slow birds had higher (P < 0.05) relative weights (% of BW) for nearly all organs on d 11 and 25, most notably the gizzard, proventriculus, pancreas, and liver. Conversely, intestinal sections were longer (P < 0.05) in the Fast birds. Measurement of gut histomorphology did not show any notable differences between growth rate groups in terms of villi height, crypt depth, or their ratio for either time-point (P > 0.05). In terms of volatile fatty acid concentrations of luminal contents, acetate concentrations were 10.2% higher (P < 0.001) in the ileum of the Slow birds compared with Fast birds on d 25. Overall, the findings suggest that total BW gain is influenced by the development of metabolically active organs, as supported by lower weight gain in Slow birds with relatively larger organ weights and shorter intestinal lengths than their Fast counterparts. The general lack of differences in fermentation end-product concentrations in luminal contents does not rule out influence of the microbiota on growth rate of broilers, which warrants further investigation.

5Evaluation of Long-Term Supplementation of a Bacillus Subtilis Direct-Fed Microbial and Enzymatically Hydrolyzed Yeast Cell Culture Product Used Alone or in Combination on Clostridia, Clostridium Perfringens, Escherichia Coli, and Salmonella Prevalence in Beef Steers.
    Erin R. Dehaan,Jesse Thompson,Warren C. Rusche, Mackenzie de Jesus,Elliot Block,Tom Rehberger,Zachary K. Smith

The objective was to determine the influence of long-term supplementation (258 d) of a direct-fed microbial (DFM) and/or yeast cell wall (YCW) product on bacterial populations in beef steers. Single-sourced Charolais x Red Angus steers (n = 256; body weight = 246 +/- 1.68 kg) were used in a randomized complete block design and blocked by location into one of four treatments: 1) fed no DFM and no YCW (Control); 2) fed only the DFM (DFM; Certillus CP B1801 Dry, 28 g/steer d(-1) ); 3) fed only the YCW (YCW; Celmanax; 18 g/steer d(-1) ); and 4) fed the DFM and the YCW (DFM+YCW). Steers were vaccinated for respiratory and clostridial diseases and treated for internal and external parasites at processing and individually weighed on days 1, 14, 42, 77, 105, 133, 161, 182, 230, and 258. To determine bacterial prevalence, fecal samples were collected on days 1, 14, 77, 133, 182, and 230 and environmental (pen area, feed, and water) samples were collected at the beginning of the week when cattle were weighed. No treatment x day interactions or treatment effects (P > 0.05) were observed between treatment groups at any sampling days for the bacterial populations. Samples on days 1, 133, and 182 had greater (P < 0.05) Clostridia levels compared to the other sampling points but were not different from each other. Clostridia levels were also greater (P < 0.05) on day 77 compared to days 14 and 230. Samples on days 77 and 230 had greater (P < 0.05) Clostridium perfringens levels compared to the other sampling points but were not different (P > 0.05) from each other. Samples on days 1 and 14 had lower (P < 0.05) total Escherichia coli levels compared to the other sampling points but were not different (P > 0.05) from each other. Escherichia coli levels on day 77 were higher (P < 0.05) compared to days 133, 182, and 230. Little Salmonella prevalence (1.5%) was observed throughout the study. This study had greater levels of Clostridia compared to small and large commercial feedlots in the Church and Dwight research database, but C. perfringens, total and pathogenic E. coli, and Salmonella prevalence were notably lower. Collectively, there were no appreciable treatment influences on bacterial populations. These data further indicate a low pathogenic bacterial challenge at the trial site, which could partially explain the lack of differences with DFM or YCW supplementation. The DFM and YCW used alone or in combination cannot be expected to show additional benefits when animals are relatively unstressed with a low pathogenic bacterial challenge.
