
Breast cancer is the most common type of malignant neoplasm in women. BRCA1 and BRCA2 are the most commonly mutated genes, but only up to 30% of hereditary breast cancer cases are attributed to alterations in these genes. A large proportion of genetic causes of hereditary breast cancer remains unknown. Thus, the search for new hereditary mutations and establishing a genetic alteration in each case of hereditary breast cancer is a clinically significant task; be the goal of our research. Next-generation sequencing (NGS) allows for simultaneous analysis of hundreds to thousands of genes at one time. We analyzed the genetic material of 49 patients of the northwest Russian population with clinical signs of hereditary breast cancer and identified new mutations associated with hereditary breast cancer. Research results show two missense mutations - SLIT3 p.Arg154Cys, CREB3 p.Lys157Glu, and truncating mutation - USP39 c.*208G>C. Research conclusion; The identified mutations can explain only a tiny fraction of hereditary breast cancer cases (0.7% to 1.1%). The next step to increase the practical value of the detected alterations should be the analysis of biological characteristics of tumors in carriers of these mutations that can potentially become a target for chemotherapy.
Toxoplasmosis is a disease that can infect animals and zoonotic in humans. In pregnant women (congenital toxoplasmosis), the risk and severity of symptoms of congenital toxoplasmosis are more severe if infection occurs early in pregnancy and asymptomatic. The only way to determine disease is by serological screening. Serological diagnosis techniques for toxoplasmosis include ELISA (Enzyme-linked Immunosorbent Assay) and ICT (Immunochromatography Assay Test). However, until now, research on toxoplasmosis with the ICT method is still minimal. This study aims to analyze the results of the IgM antibody examination Toxoplasma gondii using the ICT method with the ELISA method and calculate the sensitivity and specificity of the ICT method and the ELISA method in pregnant women. The research design was used analytic (cross-sectional), with the number of samples is 33 serums. Data from the ELISA method obtained from secondary medical record data and the remaining serum samples were examined by the ICT method by researchers at the Harapan Kita Hospital Laboratory. In the ICT method, two samples (6.1%) were positive, and 31 samples (93.9%) were negative. In the ELISA method, results obtained similar to the ICT method, two samples (6.1%) positive and 31 samples (93.9%) with negative results. Toxoplasmosis examination can be done using the ICT method as the initial serological screening stage, but it not recommended as a diagnosis enforcer.
Dengue hemorrhagic fever (DHF) is one of the diseases caused by Aedes Aegypti mosquitoes. DHF is still a public health problem, including in Indonesia, due to the high mortality rate caused by this mosquito. The risk caused by the bite of the Aedes sp mosquito is quite dangerous, including acute fever, bleeding, shock, and even death, so that efforts are needed to control it. One of the natural ingredients that can use to eradicate Aedes Aegypti mosquitoes without causing negative impacts on the environment is the Cananga flower and Zyzigium Leaves. This study purposes of determining the optimum concentration of Cananga Flowers and Zyzigium Leaves Ethanol extract of which was having biolarvaside effect to Aedes Aegypti larvae. The method used a simple experiment with Posttest Only With Control Group Design. The data result of the research analyzed with Logit and Wilcoxon test. Achievement of those tests was LC50 Zyzigium leaf extract on contact for 24 hours was 0.609-0.935, and LC99 was between 1.064-1.973 while Cananga flower extract obtained LC50 of 0.228-0.261 and the LC99 was 0.446 to 0.558. Wilcoxon test for compared bioalarvaside effect results between Cananga Flower and Zyzigium Leaf received a significance value less than α (0.001<0.05), so it can ascertain that the Cananga Flower extract was stronger biolarvaside than Zyzigium Leaf extract.
Breast cancer is the most common cancer in women throughout the world, with new cases and deaths which continue to increase. Soursop leaves (Annona muricata L) have been used extensively in traditional medicine, including cancer. Acetogenin, alkaloids, and phenols contained in soursop leaves are known to have anti-cancer effects. Among them, acetogenin has the most dominant role and reported to have a cytotoxic effect on various cancer cell lines. This study aims to determine the cytotoxic activity of soursop leaf ethanol extract on T47D breast cancer cell line. Measurement of cytotoxic activity was carried out by the MTT method, and the viability percentage of T47D cells was calculated based on the absorbance values in the treatment, cell control, and media control groups of each replicate. The correlation between extract concentration and viability percentage of the T47D cell line was outlined in the regression equation to obtain the IC50 value. IC50 values of 109.91 ± 3.04 with R values 0.975 and R2 0.9508 obtained. R values close to 1 indicated a strong correlation between extract concentration and the percentage of living T47D cells. Meanwhile, the amount of R2 suggested that the level of AMEE had a 95.08% influence on the rate of cell viability, and the other 4.92% influenced by factors other than the AMEE dose. These results indicated that the ethanol extract of soursop leaves has a cytotoxic effect and has the potential to inhibit T47D cell proliferation in vitro.
To determine the possible detrimental effects of multiple methotrexate doses has on the ovarian reserve and to determine the beneficial effects of dehydroepiandrosterone supplementation. The rats (n:24) divided into three groups; Group 1: control group, Group 2: dehydroepiandrosterone and methotrexate group (6mg/kg dehydroepiandrosterone dissolved in 0.1 ml sesame seed oil subcutaneously for ten days and 1mg/kg intramuscular methotrexate at the 1st, 3rd, 5th and 7th days) and Group 3: methotrexate group (1mg/kg intramuscular methotrexate at the 1st, 3rd, 5th and 7th day). The groups compared in regards to their histopathological ovarian damage scores and AMH values. It established that multiple methotrexate applications had a considerable effect on reducing vascular congestion in the ovarian tissue. Both in groups 2 and 3 AMH values found to be significantly lower. When this decline in the ovarian reserve examined comparatively; while both the group 2 and 3 reported having a considerable and continuous reduction in the AMH levels correlative to the control group; the primordial, primary and total follicle counts shown to stay statistically the same in the group 2 (p<0.0167). The dehydroepiandrosterone supplementation, while did not have a significant effect on reducing ovarian damage induced by methotrexate, also was not shown to provide any considerable increase in AMH levels. However, prolonged usage of dehydroepiandrosterone by contributing to the antral follicle pool considered promising in increasing the AMH levels conceivably
This study aims to analyze the effect of ethanol extract of black cumin seeds (Nigella Sativa) on iNOS expression in the kidney of mice model preeclampsia. Design This study uses an experimental design with a posttest only control group design. Using a mice model preeclampsia. This study was divided into 6 groups: negative control, positive control (preeclampsia model), model + black cumin seed extract 500 mg/KgBB/day, 1000 mg/KgBB/day, 1500 mg/KgBB/day, and 2000 mg/KgBB/day days, after surgery on mice then examined iNOS Kidney expression by methods Immunohistochemical. Data from observations were analyzed by one way ANOVA test. The result there is an effect of ethanol extract of black cumin seeds on renal iNOS expression in mice model preeclampsia (p <0.05) at doses of 500mg, 1000mg, 1500mg, and 2000mg on decreasing renal iNOS expression. The conclusion Ethanol extract of black cumin seeds (Nigella sativa) can reduce iNOS Kidney expression in Mice Preeclampsia.
Tuberculosis is an infectious disease caused by Mycobacterium tuberculosis. The diagnosis of pulmonary TB in general still relies on microscopic examination with a sputum smear for Acid Fast Basil (AFB). Microscopic examination to detect M. tuberculosis has high specificity, but the sensitivity is 35-70%. The sputum culture method is the gold standard for the diagnosis of pulmonary TB but requires a long time, which is 6-8 weeks. In recent years ICT (Immuno Chromatography Test) has been introduced to identify potential and useful TB antigens to help diagnose pulmonary TB. Antigens detected were ESAT-6, CFP-10, CFP-21, and MPT-64. This study aims to compare the results of the examination of specimens of suspect TB sputum with the ICT method and AFB. This study based on observational with analytic design cross-sectional on 56 samples of Stored Biological Material suspects TB that were examined by the AFB method and the ICT method (Cocktail Antigen). Statistical test results Fisher Exact (α = 0.05) showed no difference between microscopic examination of smear sputum and ICT (Cocktail Antigen) TB (p = 1,000). Nevertheless, the use of ICT reagents for the diagnosis of pulmonary TB still needs to be further investigated, especially to find out the exact causes of false positive and false negative reactions.
Vector-borne diseases are still a health problem in some tropical countries. One vector-borne disease is Dengue Hemorrhagic Fever (DHF). DHF never decreases and even tends to continue to increase, and many cause deaths in children, 90% of them attack children under 15 years. DHF cases in 2018 amounted to 65,602 cases, with 467 deaths (CFR = 0.71%). The behavior of holding water in various places such as tubs and tendons indirectly creates a breeding place for Aedes aegypti mosquitoes. The use of chemicals as insecticides can cause the death of non-target animals, environmental pollution, and the occurrence of vector resistance to insecticides. Therefore it is necessary to do other methods including the use of vegetable insecticides. Sugar apple leaves can be used to kill Aedes aegypti larvae. Sugar apple leaves will obtain throughout the year. The purpose of this study is to know the ability of young and old sugar apple leaves to kill Aedes aegypti larvae. This type of research is a posttest only control group design. The study sample was part of the Aedes aegypti larvae on the final instar III. The experiment to kill Aedes aegypti larvae use two types of sugar apple leaf extract, namely young and old sugar apple leaves with nine treatments four replications. Each treatment consisted of 25 larvae. The research results show There was no significant difference in the number of dead larvae using either extracts from young sugar apple leaves or old sugar apple leaves. LC90 of sugar apple leaf extract was between 0.05632 to 0.08324% and the effective residual age at LC90 (0.06568%) with the death of Aedes aegypti larvae was 92% over 24 hours (1 day).
Typhoid fever is a disease caused by Salmonella typhi infection. This disease is still a global problem. The purpose of this study was to develop a rapid detection method for Salmonella using molecular methods utilizing Inv A and Fim-C Genes. Two methods compared, which are Multiplex and monoplex PCR. The sample is in the form of the patient's blood, which is stored at 3 ml EDTA vacutainer tube consisting of 10 positive samples and five negative samples. The criteria for samples used previously have tested by widal/ immunoserology (titer more than 1/160). The results showed that multiplex PCR for detection of Salmonella using the Inv A gene and fim c gene not recommended because too many bands produced. The application of monoplex in the Inv A gene gives better results than the Fim C gene. In conclusion, the monoplex application on Inv A gene recommended than Fim C gene used to detect S. typhi in human blood samples.
Fat-infiltrated lymph nodes mostly seen on mammograms (MG) of the elderly and obese women. We analyzed the fat infiltration of lymph nodes for its potential use in estimating body mass index (BMI). 2D digital MG of 157 women assessed by one breast radiologist and one radiology resident. For each patient, the largest lymph node chosen as the index by analyzing the medial, lateral oblique (MLO) views. The width and length of the node and the hilum, the cortex, and the Hilo-cortical ratio (HCR) were measured. Breast density was categorized according to the ACR BI-RADS 4th edition and divided into two groups as dense and non-dense. Obesity was defined as BMI ≥ 30 kg/m2, overweight as BMI <30 and ≥ 25 kg/m2, normal weight as BMI <25 kg/m2. The association of lymph node dimensions and hilum dimensions, breast density, and age with BMI were analyzed. P <0.05 were considered as statistically significant. Good inter-observer agreements were found for most measurements, although the agreement of cortex width was low. The lymph node dimensions, hilum dimensions, HCR increased significantly with increasing BMI for the measurements of reader 1 (p<0.001 for all). In reader 2, only widths of lymph node and hilum related (p<0.05). Cortex width was not associated with BMI. There was a statistically significant relationship between BMI and mammographic density. Obese women had more non-dense breasts (p <0.001). Conclusion axillary lymph node fatty content was found related to BMI and could use as an indicator of obesity.
This study aimed to investigate the correlation of adrenal magnetic resonance imaging (MRI) mass size and 1 mg dexamethasone suppression test results in patients with adrenal incidentaloma diagnosis. Eighty-six patients (82.6% female) with incidentally discovered adrenal masses, diagnosed and treated in the Department of Endocrinology Bakirkoy Dr. Sadi Konuk Education and Research Hospital between June and August 2017, were included in the analysis. All the patients evaluated for their hormonal functionality. 24-h urinary excretion of cortisol did not correlate with tumor size. But there was a positive correlation between longer adrenal nodule diameter and metanephrine, and there was a negative correlation between shorter adrenal nodule diameter and renin levels. We found that smaller and clinically “silent” tumors often do not demonstrate subclinical hormonal activity.
Some people in South Kalimantan have used river water to fulfill their daily needs include washing the utensils. Wash the cutlery use river water can cause disease transmission. Squeezed lime has an antibacterial effect in inhibiting bacterial growth. Antibacterial dishwashing soap is effective in reducing pathogenic bacteria. This research is to know the difference of bacterial amount on the variation of contact time with soap. Know the difference of bacteria on the variation of concentration of lime soap and soap without lime. The study used the experimental method with pretest and posttest with control design. The independent variables are a lime soap with a concentration of 40%, 50%, 60% and variable of contact time 30 second, 60 second and 90 seconds. The dependent variable is the number of bacteria on the dinnerware of a dinner plate. The data obtained in this study is the data of measurement of the number of bacteria on the dinner plate. Data analyzed by Kruskal-Wallis test. MPN coliform examination of river water showed results >2400 CFU/ml sample. There were no significant differences in treatment with contact time variation of 30 seconds, 60 seconds and 90 seconds. The significance value is 0.796 (> 0.05). There were significant differences in the treatment of lime soap with concentration variations of 40%, 50%, 60%, and soap without lime. The value of significance is 0,000 (<0.05).
