Bone cement materials have some disadvantages, including slow degradation and no biological activity, which greatly weakens their clinical application. Therefore, the search for a multifunctional bioactive bone cement has become urgent. In this study, a novel bone cement sample of calcitonin gene-related peptide (CGRP)/chitosan-strontium (Sr)-calcium phosphate cement (CPC) was developed. The structure and morphology were observed by scanning electron microscope (SEM). The cytotoxicity and proliferation of CGRP/chitosan-Sr-CPC were also measured. The expression of CGRP receptor 1 was measured using an immunofluorescence assay. Real-time polymerase chain reaction (PCR) and enzyme-linked immunosorbent assay (ELISA) were employed to quantify the VEGF mRNA and protein levels, respectively. Finally, the ability of the material to improve angiogenesis was assessed by using human umbilical vein endothelial cells (HUVECs) tube formation assay. The results showed that CGRP/Chitosan-Sr-CPC had the characteristics of a good orthopedic material without showing cell cytotoxicity to HUVECs. Meanwhile, CGRP/chitosan-Sr-CPC could release CGRP and enhance the proliferation of HUVECs via CGRP receptors. Moreover, CGRP/chitosan-Sr-CPC significantly upregulated the expression of the VEGF gene and protein in HUVECs, which might help improve the angiogenesis microenvironment. Besides, CGRP/chitosan-Sr-CPC could significantly improve angiogenesis of HUVECs. Objective To explore the effects of calcitonin gene-related peptide (CGRP) on proliferation of vascular endothelial cells (VECs) and osteogenic differentiation of bone mesenchymal stem cells (BMSCs) of osteoporotic rats in vitro,and cell/ molecular mechanism of promoting osteoporotic fracture healing.Methods 10 3-month-old SD rats were employed to make osteoporosis model by taking bilateral ovariectomy.Micro-CT analysis was used to determine the success of modeling at 6 months of postoperation.BMSCs and VECs were isolated from osteoporotic rats by blood adherence mcthod.10-7-10-10 mol/L CGRP were added to the culture medium.CCK-8 method was used to examine the cell proliferation.Effect of CGRP on osteogenic differentiation of BMSCs:Alkaline phosphatase (ALP) activity,and reverse transcription polymerase chain reaction (RT-PCR) were used to detect the expression of ALP,Collagen Ⅰ,BMP-2,Osteonectin and RunX2 gene.Effect of CGRP on vascularization of VECs:Vascular cell endothelial growth factor (VEGF) protein secretion and gene expression level were tested by ELISA and RT-PCR.Resuits BMSCs and VECs of osteoporotic rats were isolated successfully.The optical density (OD) values of CCK-8 method increased in a dose-dependent manner,for BMSCs at 7 d,the control group of OD value was 0.58±0.02,the CGRP group 10-7-10-10 mo]/L was to 0.80±0.03,0.79±0.03,0.74±0.03,0.69±0.03,difference was significant;for VECs,the control group of OD value was 2.20±0.01,the CGRP group 10-7-10-10 mol/L was to 4.12±0.04,3.90±0.02,3.56±0.04,3.12±0.04,difference was significant.Effect of CGRP on osteogenic differentiation of BMSCs:Alkaline phosphatase (ALP) activity,and reverse transcription polymerase chain reaction (RT-PCR) were used to detect the expression of ALP,Collagen Ⅰ,BMP-2,Osteonectin and RunX2 gene.Effect of CGRP on vascularization of VECs:Vascular cell endothelial growth factor (VEGF) protein secretion and gene expression level were tested by ELISA and RT-PCR,difference between the control group and CGRP group were all significant.Conclusion 10-7 mol/L,10-10 mol/L of CGRP can directly promote the proliferation of the BMSCs and VECs of osteoporotic rats,promote the osteogenic differentiation of BMSCs,improve the expression of VEGF gene and protein secretion,promote angiogenesis.
Objective:To explore the correlation of the serum total cholesterol level with mortality in elderly male patients with coronary heart disease. Methods:Two hundred and eight elderly male patients with coronary heart disease were divided into the group A (serum total cholesterol <4. 16 mmol/L),group B(4. 16 mmol/L≤serum total cholesterol <4. 68 mmol/L),group C(4. 68 mmol/L≤serum total cholesterol <5. 20 mmol/L) and group D(serum total cholesterol >5. 20 mmol/L) according to the serum total cholesterol levels. The serum triglyceride level,physical examination,age,body mass index and mortality in all patients were analyzed. Results:The differences of the serum total cholesterol levels between four groups were statistically significant(P < 0. 01). The mortalities in group A and D were higher than that in group B,and the mortality in group D was higher than that in group C(P<0. 05). The differences of the mortalities in group A & group D and group B & group C were not statistically significant(P >0. 05). The differences of age,body mass index and triglyceride level between four groups were not statistically significant(P>0. 05). The mortality in elderly male coronary heart disease patients with serum total cholesterol levels <4. 16 mmol/L or >5. 20 mmol/L increased. Conclusions:The serum total cholesterol level in elderly male patients with coronary heart disease is closely correlated to the mortality. The high or low serum total cholesterol levels are the risk factors of the death of the elderly male patients with coronary heart disease.
目的 观察双侧卵巢切除(OVX) SD大鼠不同时期血清骨生化标志物与骨小梁三维结构变化之间的关系.方法 分别于6月龄雌性SD大鼠假手术(Sham)组和双侧卵巢切除术(OVX)组术后2、4、8、12周随机取样,每组每个时间点处死4只,取血清测量雌二醇(E2)、骨碱性磷酸酶(BALP)、抗酒石酸酸性磷酸酶(TRAP)水平,取胫骨用显微CT(Micro-CT)和组织学HE染色观察胫骨骨小梁微结构变化.结果 术后2周,OVX组E2较Sham组开始下降,4周时差异有统计学意义(P<0.05),一直持续到12周,Sham组E2水平随时间变化不大;OVX组血清BALP在术后2周达峰值,然后迅速下降,12周时降至术前水平;OVX组血清TRAP活性在术后迅速升高,术后4周达峰值,然后下降,术后12周时仍高于Sham组及术前水平;Micro-CT分析胫骨干骺端相对骨体积(BV/TV)、骨小梁数目(Tb.N)、骨小梁分离度(Tb.Sp)、Conn.Dn、骨密度(BMD)在2周时与Sham组比较差异无统计学意义,第4周开始出现明显差异,12周差异更加明显,骨小梁厚度(Tb.Th)始终无明显差异;Micro-CT重建显示术后12周OVX组骨小梁较Sham组稀疏,缺失严重;HE染色显示术后12周OVX组大鼠胫骨干骺端骨骺板下骨小梁变细且排列稀疏,骨小梁结构不成熟,骨小梁间连接差,部分断裂,出现大量骨小梁盲端,小梁壁厚度不一致.结论 OVX术骨质疏松模型建立过程中大鼠成骨及破骨细胞活跃度先增高后降低,破骨细胞相对而言上升周期更长且下降后能维持在更高水平,比成骨细胞更活跃,骨吸收大于骨生成导致骨质疏松,主要体现为骨小梁BV/TV、Tb.N、Conn.Dn、BMD下降,Tb.Sp升高,组织学表现为骨小梁结构不成熟,骨小梁间连接差,部分断裂,出现大量骨小梁盲端.
目的:探讨血清总胆固醇( TC)水平与老年男性冠心病患者病死率的相关性。方法选择入该院治疗并随访成功的208例老年男性冠心病患者,按照血清 TC 的检测结果分为 A 组( TC<4.16 mmol/L )、B 组(4.16 mmol/L≤TC<4.68 mmol/L )、C 组(4.68 mmol/L≤TC<5.2 mmol/L)和D组( TC≥5.2 mmol/L)。同时对患者进行血清甘油三酯( TG)水平检测和体格检查,记录年龄、体质量指数和死亡情况。结果四组患者血清TC水平〔A、B、C、D组分别为(3.62±0.91)、(4.41±1.11)、(4.99±1.25)、(5.73±1.44)mmol/L〕两两比较具有统计学意义(P<0.05);A、D组患者的病死率(36.21%,42.50%)明显高于B、C组(12.73%,14.55%)(P<0.05);血清TC<4.16 mmol/L或>5.20 mmol/L均可增加老年男性冠心病患者的病死率。结论老年男性冠心病患者的血清TC水平和病死率有着紧密的相关性,血清TC水平过高或过低均为引起老年男性冠心病患者死亡的危险因素。
目的:探讨血清胱抑素C在检测糖尿病患者肾功能方面的临床价值。方法:对2013年2月至2014年2月我院收治80例糖尿病肾病患者的临床资料进行回顾性分析,将其作为实验组。选择同期在我院进行体检的80例健康人作为对照组。对两组患者进行血清胱抑素C(Cys C)、血肌酐(Scr)和尿素(BUN)检测,并对比分析两组患者的检测结果。结果:与对照组患者相比,实验组患者Cys C、Scr和BUN的指标明显较高,差异显著(P<0.05),有统计学意义。与进行Scr和BUN检测相比,实验组患者进行Cys C检测结果的阳性率较高,差异显著(P<0.05),有统计学意义。结论:血清胱抑素C的指标在检测糖尿病肾病患者肾功能方面具有重要的临床价值。
Objective Explore the clinical significance of the hepatic function examination result of hepatitis and hepatic cir hosis patients.Methods We have selected 64 hepatitis and hepatic cir hosis patients as the study object of the observation group and selected 60 persons proved healthy in the physical examination as the study object of the control group.We would analyze the level of the albumin(ALB),total bile acid(TBA),serum cholesterol(CHO)and serum cholinesterase(CHE)of them.Results The TBA level of the observation group is higher than that of the control group.The levels of ALB,CHO and CHE of the observation group are lower than that of the control group significantly.The comparisons show a significant dif erence ( <0.05).Conclusion Dif erent examination indexes of the hepatic function can reflect the degree of the hepatic injury accurately,which provide reference frame for the diagnosis and treatment of the hepatitis and hepatic cir hosis.
