The Eukaryotic Pathogen, Vector and Host Informatics Resource (VEuPathDB, https://veupathdb.org) represents the 2019 merger of VectorBase with the EuPathDB projects. As a Bioinformatics Resource Center funded by the National Institutes of Health, with additional support from the Welllcome Trust, VEuPathDB supports >500 organisms comprising invertebrate vectors, eukaryotic pathogens (protists and fungi) and relevant free-living or non-pathogenic species or hosts. Designed to empower researchers with access to Omics data and bioinformatic analyses, VEuPathDB projects integrate >1700 pre-analysed datasets (and associated metadata) with advanced search capabilities, visualizations, and analysis tools in a graphic interface. Diverse data types are analysed with standardized workflows including an in-house OrthoMCL algorithm for predicting orthology. Comparisons are easily made across datasets, data types and organisms in this unique data mining platform. A new site-wide search facilitates access for both experienced and novice users. Upgraded infrastructure and workflows support numerous updates to the web interface, tools, searches and strategies, and Galaxy workspace where users can privately analyse their own data. Forthcoming upgrades include cloud-ready application architecture, expanded support for the Galaxy workspace, tools for interrogating host-pathogen interactions, and improved interactions with affiliated databases (ClinEpiDB, MicrobiomeDB) and other scientific resources, and increased interoperability with the Bacterial & Viral BRC.
In this article, a motion planning framework for autonomous driving is explored to achieve unmanned last-mile delivery vehicle application in complicated urban scenario. This approach can dramatically improve the intelligence, driving safety, driving robustness, and scalability of autonomous vehicles. In this framework, a specific High-Definition (HD) map representation was proposed for last-mile delivery applications, and a Route Planning layer generates a kinematically feasible reference line with smoothness condition based on Routing and HD Map components. Then a Scenario Planning layer considers routing results and both static and dynamic obstacles into account to select a corresponding scenario to execute, such as cruise on-road scenario, cross intersection scenario, parking scenario, and so forth. Finally, a Trajectory Planning layer which is classified with trajectory generation and optimization modules is described. In the trajectory generation part, a rough path-speed profile and corresponding decisions, such as nudge, stop, yield, and so forth, are generated. Then, the rough path-speed profile is postprocessed by optimization algorithms in the trajectory optimization part. On the basis of the real road test results from thousands of times for JD.com's real autonomous delivery vehicle operations and approximate 115,475 km of fully autonomous driving mode in an urban scenario, the proposed motion planning framework demonstrates the efficiency of autonomous driving, improves the driving quality and reduces the manual intervention.
Background Synovial macrophages play important roles in the formation and progression of osteoarthritis (OA). This study aimed to explore the biological and clinical significance of macrophage-associated genes (MAGs) in OA. Methods The OA synovial gene expression profiles GSE89408 and GSE82107 were obtained from the GEO database. Single-sample gene set enrichment analysis (ssGSEA) and GSEA were employed to decipher differences in immune infiltration and macrophage-associated biological pathways, respectively. Protein–protein interaction (PPI) network analysis and machine learning were utilized to establish a macrophage-associated gene diagnostic signature (MAGDS). RT-qPCR was performed to test the expression of key MAGs in murine models. Results OA synovium presented high levels of immune infiltration and activation of macrophage-associated biological pathways. A total of 55 differentially expressed MAGs were identified. Using PPI analysis and machine learning, a MAGDS consisting of IL1B, C5AR1, FCGR2B, IL10, IL6, and TYROBP was established for OA diagnosis (AUC = 0.910) and molecular pathological evaluation. Patients with high MAGDS scores may possess higher levels of immune infiltration and expression of matrix metalloproteinases (MMPs), implying poor biological alterations. The diagnostic value of MAGDS was also validated in an external cohort (AUC = 0.886). The expression of key MAGs was validated in a murine model using RT-qPCR. Additionally, a competitive endogenous RNA network was constructed to reveal the potential posttranscriptional regulatory mechanisms. Conclusions We developed and validated a MAGDS model with the ability to accurately diagnose and characterize biological alterations in OA. The six key MAGs may also be latent targets for immunoregulatory therapy.
我国向来以礼仪之邦著称,礼仪文化在悠久的历史长河中占据十分重要的地位.拥有良好的礼仪素质,不仅仅是对学生自身气质和修养的展现,同时也是对社会文明形象的展示.加强初中阶段的礼仪教育,是当前亟待解决的问题.初中阶段的学生对礼仪的学习并没有树立公德意识和恭敬意识,不利于自身综合素养的发展.因此,本文主要基于在教材中发掘礼仪素材,引导学生树立正确的价值观念,加强教学礼仪环境的创设等内容进行分析,从而实现初中道德与法治教学过程中礼仪教育的渗透.
Significant progress has been made in the biological degradation of aflatoxin B1 (AFB1), which is the most toxic and carcinogenic mycotoxin commonly found in food and feed products; however, research in this area has currently focused on the effective methods for detoxifying AFB1 and the mechanisms underlying the degradation process. The relationship between AFB1 biotransformation and the metabolic feedback by the AFB1-degrading strain has rarely been reported. This study investigated the biotransformation pathway of AFB1 in a highly efficient degrading strain, Aspergillus niger FS10, and the metabolic regulation of FS10 during AFB1 degradation. We analyzed the AFB1 metabolic degradation products and evaluated its toxicity based on its structures by triple quadrupole-linear ion trap-mass spectrometry (Q-Trap-MS) coupled with LightSight™ Software. Four metabolic degradation products were obtained: Product 1 (C17H14O6), Product 2 (C17H14O7), Product 3 (C16H12O5), and Product 4 (C27H31N3O13, AFB2-GOH). Two biotransformation pathways were proposed based on the structures of these degradation products. Metabolomics data revealed that FS10 exhibited different degrees of bioremediation under low-, medium- and high- dose AFB1. The results also suggested the significant involvement of glutathione, a metabolite, in the AFB1 biotransformation pathway and its formation of AFB2-GOH with AFB1. The four products could probably be less toxic than AFB1, as reflected by changes in the AFB1 toxicity site. Therefore, the AFB1-degrading strain FS10 exhibits great potential for application in the detoxification of AFB1 in food and feed products.
A new, green, and cost-effective magnetic solid-phase extraction of aflatoxins and ochratoxins from edible vegetable oils samples was developed using polydopamine-coated magnetic multi-walled carbon nanotubes (PDA@Fe3O4-MWCNTs) as the absorbent. PDA@Fe3O4-MWCNTs nanomaterials were prepared by chemical co-precipitation and in situ oxidation and self-polymerization of dopamine and was characterized. Factors affecting MSPE and the adsorption behavior of the adsorbent to mycotoxins were studied, and the optimal extraction conditions of MSPE and the complexity of the adsorption process were determined. Based on this, the magnetic solid-phase extraction-high-performance liquid chromatography-fluorescence detection method (MSPE-HPLC-FLD) was established for determining six mycotoxins [aflatoxin B-1 (AFB(1)), AFB(2), AFG(1), and AFG(2), and ochratoxin A (OTA) and OTB)] in vegetable oils. The recovery was 70.15%similar to 89.25%, and RSD was <= 6.4%. PDA@Fe3O4-MWCNTs showed a high affinity toward aflatoxins and ochratoxins, allowing selective extraction and quantification of aflatoxins and ochratoxins from complex sample matrices.
针对快速谱峭度在低信噪比情况下诊断效果差的问题,提出了一种基于正交匹配追踪(OMP)算法的滚动轴承故障诊断新方法.该方法通过快速谱峭度图确定最优滤波器参数对信号进行滤波,利用故障信号在傅里叶稀疏基下的稀疏度已知的特点,对滤波信号的包络信号在傅里叶稀疏基下用OMP算法对轴承振动信号的包络信号进行重构,以减少噪声和其他无关成分的影响,最后对重构信号进行频谱分析获取轴承故障特征.通过轴承故障仿真数据、实验台故障轴承外圈和内圈试验数据的检验,验证了该方法的有效性和可行性.
Aflatoxin M1 (AFM1), a frequently occurring mycotoxin in milk and dairy products, causes acute and chronic poisoning. In this study, we developed a time-resolved fluorescence microsphere immunochromatographic test strip (TRFM-ICTS) combined with ultraviolet light for the rapid and quantitative detection of AFM1 in milk and its products. Polystyrene microspheres enclosing time-resolved fluorescent europium (III) [Eu(III)-TRFM] were prepared as a label and then conjugated to a monoclonal antibody. Pre-incubation was introduced for competitive recognition. Under optimal conditions, TRFM-ICTS had a linear range of 0.05-2.0 ng/mL with a 50% inhibitory concentration (IC50) of 0.204 ng/mL, which was nearly five-fold lower than those of ICTS in a traditional structure with traditional pretreatment (IC50 = 1.10 ng/mL). This test strip enhanced the sensitivity from 0.3 ng/mL (by using gold nanoparticles-strip method previously reported) to 0.019 ng/mL (by using this TRFM-ICTS method). The improved sensitivity may be due to the detection signals of immunoassay probes (with each fluorescence microsphere coupled with numbers of antibodies). TRFM-ICTS exhibited good recoveries ranging from 84.6% to 119.0% with a coefficient of variation (CV) lower than 8.31% in spiked samples, including milk powder, ultrahigh-temperature (UHT) and pasteurized milk. The results obtained using TRFM-ICTS were consistent with those obtained by ultrahigh-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) during testing of real milk samples. In addition, TRFM-ICTS has a longer storage period, which basically meets the needs of the commercial market. Owing to its high sensitivity and practicability, the proposed TRFM-ICTS can be used as a sensitive, simple, rapid, and versatile method for the quantitative detection of AFM1 in milk.
本文主要研究了父母教养方式对大学生可就业能力是否产生影响,探讨了职业探索在两者之间的中介作用.通过随机发放问卷的方法,运用验证性因子分析法、主成分分析法和中介效应检验对收集的数据进行分析和处理.研究结果显示,不良的父母教养方式不利于大学生提高自身的可就业能力,科学合适的教养方式会正向影响大学生的职业探索能力,进而提升大学生的可就业能力.

We describe a “turn-on” magnetic fluorescent biosensor based on graphene quantum dots (GQDs), Fe3O4, and molybdenum disulfide (MoS2) nanosheets. It is used for rapid, efficient, and sensitive separation and detection of circulating tumor cells (CTCs). A facile approach (electrochemical synthesis method) for the preparation of photoluminescent GQDs functionalized with an aptamer [epithelial cell adhesion molecule (EpCAM) receptors] and a magnetic agent for one-step bioimaging and enrichment of CTCs is described. MoS2 nanosheets, as a fluorescence quencher, and the aforementioned aptamer@Fe3O4@GQD complex were assembled to construct “turn-on” biosensing magnetic fluorescent nanocomposites (MFNs). This system exhibits low cytotoxicity and an average capture efficiency of 90%, which is higher than that of other magnetic nanoparticles on account of the one-step CTC separation method. In addition, the MFNs could quickly identify and label CTCs within 15 min, surpassing other one-step and two-step marker detection methods. Furthermore, because of the presence of aptamers, the MFNs have specific capability to capture CTCs (both low- and high-EpCAM-expressing cells). In addition, high-sensitivity detection of up to ten tumor cells in whole blood was achieved. Therefore, the MFNs have great potential to be used as universal biosensing nanocomposites for fluorescence-guided tumor cell enrichment and bioimaging.
Nowadays polymeric insulators are widely applied, some internal defects could gradually appear due to various complex circumstances. In this paper, a novel ultra-wideband (UWB) antenna system for fault detection is presented. This technique makes use of the multi-mode transferring theory on dielectric waveguides, where the transmission signal level difference can be employed to detect internal defects in the insulating mandrel. An investigation by simulation is performed with regard to the interfacial mm-ordered cracks in transverse and longitudinal orientations, and approximately 20 ~ 30 dB and 20 ~ 40 dB differences have been observed over the same band 0 ~ 8 GHz, respectively.
针对滚动轴承早期故障特征难以识别的问题,提出了一种基于EEMD和改进OMP算法的滚动轴承故障诊断新方法.该方法首先对故障信号进行EEMD分解得到一系列IMF分量,根据相关峭度准则选取IMF分量进行重构;然后在稀疏分解阶段对信号截断分块构造矩阵,利用KSVD自学习训练方法得到包含冲击成分的冗余字典,再用基于信号分块思想的OMP算法进行字典原子的选取和稀疏系数的求解,以重构信号包络谱峭度最大为迭代终止条件,自适应确定迭代次数;最后对重构信号进行频谱分析,获取轴承故障特征.通过实验台故障轴承外圈和内圈试验数据的检验,验证了本方法的有效性和可行性.

In the past decade, online news consumption has been steadily growing. New articles are published every few minutes, and user preferences are also constantly changing. Traditional recommender systems update model at regular intervals, which cannot adjust recommendation list dynamically according to the changes of user preferences. In this paper, we propose a hybrid recommendation model which contains two key components: incremental update item-based collaborative filtering (CF) and latent semantic analysis based relative term frequency algorithms. The hybrid recommendation model adjusts recommendation list dynamically by updating similarity table of items incrementally in incremental update item-based CF module, moreover, combining collaborative filtering and content-based algorithm ensures the relevance of recommendation articles. Results show that our proposed hybrid recommendation model outperforms traditional recommender approaches.
