The relic Lake Mogilnoe, separated from the Barents Sea by a sand and pebble dam, is located in the high Arctic on the Kildin island (Murmansk region). This lake is a classic example of a meromictic basin of marine origin. The data obtained during the 2018 expedition showed changes in the hydrochemical regime of the lake that have occurred over the past 20 years. Sulfide concentration in the monimolimnion of the lake was as high as 140 mg/L. A tendency for salinization of the surface waters to 7 g/L has been noted. The Lake Mogilnoe is characterized by a discrepancy between the halocline and thermocline levels. The chemocline zone in the lake is below the halocline level. In a narrow oxygen-containing layer between 3 and 7.5 m, aerobic microflora of the marine type and marine fauna were present. The bacterial plate was formed at the boundary of the sulfide layer at 8 m and mainly consisted of green sulfur bacteria (GSB). Brown-colored GSB species containing bacteriochlorophyll e were predominant. The previously formed concept of anaerobic phototrophic bacteria (APB) biodiversity based on morphological characteristics was modified using metagenomic data obtained by analyzing DNA from two samples of lake water in the chemocline zone, and was also supplemented by identifying new GSB species. Molecular diagnostic data confirmed the absolute dominance of the brackish species of GSB Chlorobium phaeovibrioides. This is the first isolation and identification of brown- and green-colored Prosthecochloris aestuarii morphotypes from Lake Mogilnoe, as well as of bacteriochlorophyll c-containing Prosthecochloris sp. The taxonomic position of Pelodyction phaem, which was constantly present in the Lake Mogilnoe, is discussed in detail. Despite the partial isolation of the ecosystem of Lake Mogilnoe from the Barents Sea, the main properties of the dominant GSB species and Prosthecochlori aestuarii turned out to be similar to those of the phylotypes living in lakes on the White Sea coast.
The composition and structure of the anoxygenic phototrophic bacterial (APB) community in the water column of meromictic Lake Bol’shie Khruslomeny during winter were investigated. The community developed at the depth of 4.25 m, and its activity in the ice-cover period was very low (6.2 µmol C L–1 day–1). The water in the zone of highest development of phototrophic bacteria was of an unusual lemon-yellow color, probably due to the production and accumulation of polysulfides. The near-bottom water was also of lemon-yellow color and was resistant to oxidation by the air oxygen. In the zone of peak APB development the content of BChl d from green-colored green sulfur bacteria was considerably higher than that of BChl e from brown-colored green sulfur bacteria: 77 and 23
Despite surprisingly a small number of protein-coding gene in mammalian genomes, a large variety of different RNAs is being produced. These RNAs are amazingly different in their number, size, cell localization, and mechanism of actions. Although new classes of short RNAs (sRNAs) are being continuously discovered, it is not yet obvious how many of the sRNAs are originated. Altogether, the research in the recent few years has identified an unexpectedly rich variety of mechanisms by which noncoding RNAs act, suggesting that we have identified probably only few of the many potential functional mechanism and more investigation will be needed to comprehensively understand the complex nature and biology of mammalian RNAome. Here, we focus on various aspects of the diversity of the biological role of these nonprotein-coding RNAs (ncRNAs), with emphasis, on functional mechanisms recently elucidated.
— In the present work, separation and quantitative assessment of bacteriochlorophylls d and e of green sulfur bacteria (GSB) in lake water was attempted. The method developed reveals the ratio of green- and brown-colored GSB in natural water by measurement of absorption spectra of the pigments in the extracts from environmental samples.
— The community of anoxygenic phototrophic bacteria (APB) from the water column of the meromictic Lake Trekhtsvetnoe (Kandalaksha Bay, White Sea, Russia) was studied in March 2012 and 2013 and in September 2013 and 2014. The community structure below the chemocline was shown to restore during three years after partial mixing resulting from seawater admixture into the lake in autumn 2011; a dense layer (at least 10 8 cells mL –1 ) of green-colored (g/c) sulfur bacteria (GSB) was formed. During winter, development of low numbers of brown colored (b/c) GSB was observed in the upper layer of green water. During summer seasons, b/c GSB were found to be located in the oxic zone above the green water layer, which was unusual for these organisms. The APB community was found to contain purple bacteria. Four APB strains were isolated from the upper part of the sulfide zone. The b/c and g/c GSB strains were phylogenetically close to each other and to the type species Chlorobium phaeovibrioides DSM 265 (99% similarity gene sequences). One strain of purple bacteria was phylogenetically related to the brackish sulfur bacteria Thiocapsa marina , while the other was related to freshwater bacteria Rhodopseudomonas palustris . The strains of sulfur bacteria were phylogenetically close to the chemocline bacteria from the stratified Lake Kislo-Sladkoe, also located in the coastal zone of the Kandalaksha Bay, White Sea.
Absorption and fluorescence spectra for living cells of green sulfur bacteria inhabiting the anaerobic zone of two meromictic lakes separated from the White Sea have been studied. The spectral-optical properties of pure cultures of green-colored and brown-colored species of green sulfur bacteria Chlorobium phaeovibrioides have been compared, and the content of bacteriochlorophyll molecules in one bacterial cell of each species has been estimated. The method of separating the contributions of different groups of green sulfuric bacteria to bacteriochlorophyll fluorescence was applied for a mixture of two species of bacteria with different pigmentation. The depth distributions of fluorescence intensity and concentration of bacteriochlorophylls for microorganisms inhabiting the Trekhtzvetnoe and Elovoe lakes at the Kandalaksha Gulf of the White Sea were plotted.
Optical characteristics of water in stratified lakes of the White Sea are of particular interest in connection with the observation of thin colored layers resulting from massive development of anoxygenic phototrophic bacteria around the chemocline. While optical properties of chlorophyll are widely used in remote sensing, spectral characteristics of bacteriochlorophylls (BChl) for natural microbial communities have been little studied. In this work, spectral study of green sulfur bacteria of four water bodies of the Kandalaksha Gulf of the White Sea was carried out. Absorption and fluorescence spectra were measured for water samples taken in March 2017 from different depths and compared with spectra of monocultures isolated from the same water bodies earlier. It has been shown that the BChl fluorescence in the living cells of green sulfur bacteria has two overlapping emission bands: in the region of 740–770 nm (BChl d and e ) and at 815 nm (BChl a ). The wavelength of the maximum of the first band depends on the ratio of the concentrations of green- and brown-colored forms of bacteria containing different types of BChl. A new method for separating the contributions of two types of bacteria is proposed, based on the deconvolution of the BChl fluorescence spectrum into three bands whose parameters are determined from the spectra of monocultures. The BChl content at various water depths has been estimated and the percentage ratio of different types of phototrophic bacteria has been determined.
The community of anoxygenic phototrophic bacteria (APB) in the water column of Lake Kislo- Sladkoe (Kandalaksha Bay, White Sea), which has recently become separated from the sea, was investigated in March?April 2012, March?April 2013, and in September 2013. The lake, which was previously considered meromictic, was in fact mixed and was strongly affected by the sea. In winter the lake is sometimes washed off with seawater, and this together with the seasonal cycles of succession processes determines the succession of the community. The consequences of the mixing in autumn 2011 could be observed in the APB community as late as autumn 2013. Green-colored green sulfur bacteria (GSB) usually predominated in the chemocline. In winter 2013 stagnation resulted in turbidity of water under the ice, which was responsible for both predominance of the brown GSB forms and the changes ratio of the species of purple sulfur bacteria (PSB) in anoxic water layers. Production of anoxygenic photosynthesis in the lake was at least 240 mg C m-2 day-1 in September and 0–20 mg C m–2 day–1 in March—April, which corresponded to 40 and 69%, respectively, of oxygenic photosynthesis. Okenone-containing purple sulfur bacteria, strain TcakPS12, were isolated in 2012 from lake water. The ells of this strain form filaments of not separated cells. Strain TcakPS12 exhibited 98% similarity with the type strains of Thiocapsa pendens DSM 236 and Thiocapsa bogorovii BBS, as well as with the strains AmPS10 and TcyrPS10, which were isolated from Lake Kislo-Sladkoe in 2010.
В 1979 г. был описан род галофильных пурпурных серобактерий (ПСБ) Lamprobacter с единственным видом Lpb. modestohalophilus. Для палочковидных бактерий рода Lamprobacter характерно присутствие в клетках газовых вакуолей в неподвижной стадии роста культуры, а также изредка возникающие подвижные клетки, лишенные газовых везикул. Бактерии содержали бактериохлорофилл а и каротиноид окенон. Названия этого рода и вида в 1988 г. включены в список одобренных названий микроорганизмов. Типовой штамм Lpb. modestohalophilus RO1T был утрачен, и, как следствие, данных анализа нуклеотидных последовательностей гена 16S рРНК этого вида не было получено. В 1998 г. на основании анализа гена 16S рРНК, был создан новый род Halochromatium, объединивший подвижные экстремально галофильные виды Chromatium-подобных бактерий. Представители этого рода не содержат в клетках газовые вакуоли. Несмотря на фенотипические отличия родов ПСБ Lamprobacter и Halochromatium филогенетические границы для этих таксонов не были определены. Таксономическая ситуация осложнилась после того как морская бактерия, по морфофизиологическим признакам принадлежащая к роду Lamprobacter, была описана как новый вид рода Halochromatium с видовым названием Hch. roseum. В нашей работе приводятся аргументы в пользу сохранения двух фенотипически и филогенетически различных родов Lamprobacter и Halochromatium. Предлагается считать Lpb. modestohalophilus типовым видом рода Lamprobacter. На современном уровне исследованы свойства двух штаммов Lamprobacter modestohalophilus, один из которых (штамм Sivash) предложен в качестве неотипового штамма данного вида. Предлагается также сохранить род Halochromatium, оставив в нем экстремально-галофильные виды Hch. salexigens и Hch. glycolicum, в то время как слабо галофильный Halochromatium roseum предложено перенести в род Lamprobacter, создав новую комбинацию Lamprobacter roseus comb. nov.
Microbiological, biogeochemical, and isotopic geochemical investigation of Lake Kislo-Sladkoe (Polusolenoe in early publications) at the Kandalaksha Bay shore (White Sea) was carried out in September 2010. Lake Kislo-Sladkoe was formed in the mid-1900s out of a sea gulf due to a coastal heave. At the time of investigation, the surface layer was saturated with oxygen, while near-bottom water contained sulfide (up to 32 mg/L). Total number of microorganisms was high (12.3 × 106 cells/mL on average). Light CO2 fixation exhibited two pronounced peaks. In the oxic zone, the highest rates of photosynthesis were detected at 1.0 and 2.0 m. The second, more pronounced peak of light CO2 fixation was associated with activity of anoxygenic phototrophic bacteria in the anoxic layer at the depth of 2.9 m (413 μg C L−1 day−1). Green-colored green sulfur bacteria (GSB) predominated in the upper anoxic layer (2.7–2.9 m), their numbers being as high as 1.12 × 104 cells/mL, while brown-colored GSB predominated in the lower horizons. The rates of both sulfate reduction and methanogenesis peaked in the 2.9 m horizon (1690 μg S L−1 day−1 and 2.9 μL CH4 L−1 day-1). The isotopic composition of dissolved methane from the near-bottom water layer (δ13C (CH4) = −87.76‰) was significantly lighter than in the upper horizons (δ13C (CH4) = −77.95‰). The most isotopically heavy methane (δ13C (CH4) = −72.61‰) was retrieved from the depth of 2.9 m. The rate of methane oxidation peaked in the same horizon. As a result of these reactions, organic matter (OM) carbon of the 2.9 m horizon became lighter (−36.36‰), while carbonate carbon became heavier (−7.56‰). Thus, our results demonstrated that Lake Kislo-Sladkoe is a stratified meromictic lake with active microbial cycles of carbon and sulfur. Suspended matter in the water column was mostly of autochthonous origin. Anoxygenic photo-synthesis coupled to utilization of reduced sulfur compounds contributed significantly to OM production.
Lamprobacter, the genus of halophilic purple sulfur bacteria (PSB) with the single species Lpb. modestohalophilus was described in 1979. Rod-shaped Lamprobacter cells contained gas vesicles during the nonmotile growth phase; motile cells without gas vesicles were sometimes formed. Bacteria contained bacteriochlorophyll a and a carotenoid okenone. The names of this genus and species were included in the list of approved microbial names in 1988. Since the type strain Lpb. modestohalophilus RO1T has been lost, its 16S rRNA gene sequences have not been obtained. Based on analysis of the 16S rRNA genes, a new genus Halochromatium comprising the motile extremely halophilic Chromatium-like species was proposed in 1998. Members of this genus never contain gas vesicles. In spite of the phenotypic differences between the genera Lamprobacter and Halochromatium, phylogenetic boundaries between these taxa remained undetermined. Description of a marine bacteria formerly belonging to Lamprobacter according to its morphological and physiological properties as a new Halochromatium species, Hch. roseum, resulted in additional complication of the taxonomic situation. The present work provides evidence for the preservation of two phenotypically and phylogenetically different genera, Lamprobacter and Halochromatium, Lpb. modestohalophilus is proposed as the type species of the genus Lamprobacter. Characteristics of two Lpb. modestohalophilus strains were extensively investigated, and one of them (strain Sivash) was proposed as the neotype strain of the species. It was suggested to retain the genus Halochromatium as containing extremely halophilic species Hch. salexigens and Hch. glycolicum, while transfer of the weakly halophilic species Hch. roseum to the genus Lamprobacter is proposed, resulting in a new combination Lamprobacter roseus comb. nov.
