According to the World Health Organization, as of January 3, 2020 to September 13, 2023, there were approximately 23 million confirmed cases of COVID-19 reported in the Russian Federation, about 400 thousand of which were fatal. Considering the high rate of mutation of the RNA-containing virus genome, which inevitably leads to the emergence of new infectious strains (Eris and Pyrola), the search for medicinal antiviral agents remains an urgent task. Moreover, taking into account the actively mutating receptor-binding domain, this task requires fundamentally new solutions. This study proposes a candidate immunoliposomal drug that targets the S protein of SARS-CoV-2 by the monoclonal neutralizing antibody P4A1 and ensures the penetration of a highly active ribonuclease into the virus-infected cell, which degrades, among cellular RNA, viral RNA too. We demonstrate a more than 40-fold increase in the neutralizing activity of the developed drug compared to the free monoclonal neutralizing antibody.
We studied the antitumor activity of the combined use of local proton irradiation in two modes (10 and 31 Gy) with preliminary intra-tumoral injection of two types of bismuth nanoparticles differing in surface coating: coated with the amphiphilic molecule Pluronic-F127 or Silane-PEG (5 kDa)-COOH polymer. Nanoparticles were used in doses of 0.75 and 1.5 mg/mouse. In two independent series on experimental tumor model (solid Ehrlich carcinoma), bismuth nanoparticles of both modifications injected directly into the tumor enhanced the antitumor effects of proton therapy. Moreover, the radiosensitizing effect of bismuth nanoparticles administered via this route increased with the increasing the doses of nanoparticles and the doses of radiation exposure. In our opinion, these promising data obtained for the first time extend the possibilities of treating malignant neoplasms.
Obtaining genetically engineered NK cells is a developing area of immunotherapy. In this work, we analyzed the subset heterogeneity of NK cells subjected to retroviral transduction, taking into account the content of adaptive NK cell progenitors. It was shown that subsets KIR2DL2/DL3+, as well as CD57–KIR2DL2/DL3+NKG2C+, can be modified with greater efficiency than the corresponding subsets that do not carry the KIR2DL2/DL3 and NKG2C markers. After genetic modification, the CD57–KIR2DL2/DL3+NKG2C+ cells began to express CD57 de novo, acquiring the adaptive NK cell phenotype.
The synthesis and investigation of a new bioconjugate for the targeted therapy of breast cancer is carried out. The ZHER2 scaffold polypeptide specific for the extracellular domain epitope of the HER2 transmembrane receptor, which is conjugated to a carrier molecule of human serum albumin (HSA) with the DOTA chelating agent attached to it, is used as the targeting agent. The radiolabeling reaction of the ZHER2–HSA–DOTA molecule is carried out with the 177Lu radionuclide, as well as with 212Pb, obtained using the developed laboratory generator. The radiochemical purity of the 177Lu-labeled preparation was 57 ± 10%, and that of the 212Pb-labeled preparation was 72 ± 5%. The degree of dissociative stability of these compounds was 73 ± 8% for 177Lu and more than 90% for 212Pb. In vitro tests of the cytotoxic activity of the obtained targeted radioactive compounds [177Lu]ZHER2–HSA–DOTA and [212Pb]ZHER2–HSA–DOTA in tumor-cell lines show a significant level of proliferation inhibition of the proliferation of human breast-cancer cells SK-BR-3 and BT474 with overexpression of the HER2/neu tumor marker in contrast to MCF-7 breast ductal adenocarcinoma cells with a low level of HER2/neu expression.
The information about methods of surface preparation for dentures made from ceramic based on zirconium dioxide for better adhesive fixation is presented in the first part of this article. Micromechanical methods of preparation for cementation of the surface of a zirconia dental prosthesis are discussed in the first part of this article. The second part of the article provides an overview of scientific research on chemical methods for preparing a surface made of zirconia and the problem of the bond strength of polymer cement with a prepared surface made of zirconia.
Upconversion nanoparticles (UCNPs) are a promising nanoplatform for bioreagent formation for in vivo imaging, which emit UV and blue light under the action of near-infrared radiation, providing deep tissue penetration and maintaining a high signal-to-noise ratio. In the case of solid tumor visualization, the UCNP surface functionalization is required to ensure a long circulation time, biocompatibility, and non-toxicity. The effective UCNP accumulation in the solid tumors is determined by the disturbed architecture of the vascular network and lymphatic drainage. This work demonstrates an approach to the UCNP biofunctionalization with endogenous polysialic acid for in vivo bioreagent formation. Bioreagents possess a low level of nonspecific protein adsorption and macrophage uptake, which allow the prolongation of the circulation time in the bloodstream up to 3 h. This leads to an intense photoluminescent signal in the tumor.
Классической основой адресной терапии являются моноклональные антитела, однако разработка альтернативных связывающих белков позволила использовать в качестве нацеливающих модулей белки неиммуноглобулиновой природы. Дарпины - каркасные белки на основе анкириновых повторов, обладают такими преимуществами перед антителами, как малый размер, стабильность в широком диапазоне температур и значений pH, низкая склонность к агрегации и легкость наработки в гетерологичных системах экспрессии. Различия в строении паратопа дарпинов и антител расширяют спектр молекул-мишеней, а легкость создания гибридных белков слияния позволяет получать биспецифические и мультивалентные конструкции. В представленном обзоре суммированы данные последних лет о разработке терапевтических и визуализирующих соединений на основе дарпинов.
Направленные токсины являются перспективными противораковыми агентами, позволяющими селективно уничтожать раковые клетки за счёт повышенного содержания на их поверхности онкоспецифических маркёров. Применение таких противораковых токсинов в медицине сдерживается в основном их высокой неспецифической токсичностью, в особенности гепатотоксичностью. В экспериментах на клетках человека мы показали, что удаление транслокационного домена токсина DARPin-PE40 приводит к уменьшению гепатотоксичности и неспецифической активности. Такой же эффект наблюдается и при инактивации в молекуле DARPin-PE40 сайта расщепления фурином. Одновременное удаление как транслокационного домена, так и сайта расщепления фурином показало наилучшие результаты. Такая модификация токсина может найти применение при создании противораковых направленных токсинов с повышенной селективностью.
Induction of direct cell death is one of the mechanisms of the antitumor effect of GD2-specific antibodies used for the therapy of high-risk neuroblastoma. The mechanisms of the cytotoxic signal triggered by antibody binding to GD2 ganglioside on the surface of the tumor cell remain insufficiently studied. Using inhibitor analysis we demonstrated that actin microfilaments are involved in the cell death induced by GD2-specific antibodies. Specifically, a strong antagonistic influence of cytochalasin D on the cytotoxic effect induced by GD2-specific antibodies was demonstrated in GD2+ tumor cell lines, which was expressed in at least 20% increase in cell survival and a significant decrease of the fraction of cells with fragmented DNA.
Periodontal diseases, especially those with polymicrobial etiology, are often associated with type 2 diabetes mellitus, proceeding more severely and affecting the course of diabetes mellitus. Recently, this feature has been associated with the ability of periodontopathogen microflora to cause not only a local infectious process in the oral cavity, but also to interact with the human immune system and induce various systemic effects. We investigated changes in the salivary cytokine profile of patients with chronic periodontitis, associated and not associated with type 2 diabetes mellitus. We observed a statistically significant decrease of MCP-1/CCL2, GM-CSF, IL-5, IL-6, and IFN- in the saliva of patients with chronic periodontitis associated with type 2 diabetes mellitus in comparison with patients with chronic periodontitis only. All of these cytokines are associated with macrophage activation. These data are an important contribution to the elucidation of the mechanism of periodontopathogens involvement in the manifestation of the systemic effects of type 2 diabetes.
Одним из принципиальных отличий многих типов опухолевых клеток от клеток нормальных тканей является транслокация на поверхность плазматической мембраны ряда внутриклеточных протеинов, в частности белка теплового шока 70 кДа (HSP70). Продемонстрировано, что такая необычная поверхностная локализация HSP70 распознается цитотоксическими эффекторами иммунной системы и приводит к усилению их цитолитического действия на данные клетки-мишени. Механизмы этого взаимодействия до конца не раскрыты, однако феномен поверхностной локализации HSP70 на раковых клетках может быть использован для разработки новых подходов к противоопухолевой иммунотерапии. В то же время известно, что присутствие HSP70 на клеточной поверхности не является универсальным признаком раковых клеток. Многие разновидности опухолевых тканей не экспрессируют мембрано-ассоциированные HSP70, что ограничивает потенциальные клинические возможности указанных подходов. В связи с этим в качестве одного из перспективных направлений противоопухолевой иммунотерапии можно рассматривать направленную доставку на поверхность раковых клеток экзогенных HSP70 с целью привлечения и активации цитотоксических эффекторов иммунной системы. Для осуществления адресной доставки HSP70 в опухолевые ткани можно использовать молекулярные конструкции, содержащие рекомбинантные мини-антитела к опухолеассоциированным антигенам, в частности, к HER2/neu-антигену и другим маркерам, высоко экспрессирующимся на поверхности широкого спектра раковых клеток. С целью оценки возможности и эффективности применения указанного подхода, в данном исследовании были созданы двухмодульные рекомбинантные конструкции, содержащие в первом модуле мини-антитела к HER2/neuантигену, а во втором модуле - молекулу HSP70 или ее фрагмент. Избирательное взаимодействие белковых модулей обеспечивалось состыковочным звеном, образуемым входящей в состав молекулярной конструкции парой барназа:барстар - гетеродимером с уникально высокой константой взаимодействия. Результаты, полученные при тестировании указанных препаратов в моделях in vitro, продемонстрировали селективное связывание разработанных конструкций с опухолевыми клетками, экспрессирующими HER2/neu-антиген, и достоверное стимулирующее действие этих препаратов на цитотоксическую активность клеток-эффекторов иммунной системы по отношению к соответствующим клеткам-мишеням.
