目的:探讨偏头痛受风热侵袭后机体发生痛觉敏化的病理机制及芎芷地龙汤对模型的治疗机理.方法:使用热风及硝酸甘油协同诱导建立大鼠偏头痛风热证模型,观察2h内搔头次数,用免疫组化法测定三叉神经脊束核GLU、NR2B、nNOS表达情况.结果:偏头痛模型组、偏头痛风热证组2h内搔头次数明显增多,与正常组、风热组、中药治疗组相比,差异有统计学意义(P<0.01);偏头痛模型组、偏头痛风热证组三叉神经脊束核尾部GLU表达、NR2B表达、nNOS表达均明显上调,与正常组、风热组、中药治疗组相比,差异有统计学意义(P<0.01).结论:风热对正常机体无影响,而对硝酸甘油致偏头痛模型病理状态有显著影响,说明风热外袭致偏头痛痛觉敏化的发生,芎芷地龙汤能明显改善偏头痛动物模型的行为学症状,减少动物的阳性反应次数,减少三叉神经颈复合体中Glu、NR2B、nNOS表达.
Objective To study the effect of Xiongzhi Dilong decoction(XDD)on vasoactive substance in migraine with wind heat syndrome.Methods To establish migraine with wind heat syndrome model by combining exterior pathogen (hot wind)and GTN ad-ministration,examine the frequency of scratching head within 2 h,and survey the content of thromboxane B2 (TXB2 ),6 Keto PGF1α,calci-tonin gene related peptide(CGRP),endothelin(ET)in blood plasma by radioimmunoassay technology.Results The content of TXB2 ,ET and CGRP in plasma of migraine model was significantly higher than normal group and wind heat group,and the content of 6 Keto PGF1αwas significantly lower than normal group and wind heat group.The content of TXB2 ,ET and CGRP in plasma of migraine with wind heat syndrome moldel persistently increased,and The content of 6 Keto PGF1αpersistently decreased,which is significant difference compared to normal group(P<0.01),and is significant difference compared to migraine model group(P<0.05). The content of TXB2 ,ET,CGRP and 6 Keto PGF1αin plasma of Chinese medicine treatment group is recovered,which was signifi-cant difference compared to migraine with wind heat syndrome model and migraine model group(P<0.05).Conclusion There was no significant effect on normal organism which sufferd from wind heat,But there was significant effect on migraine model induced by Nitroglycerin,The vasoactive substance of Migraine model which was invaded by wind and heat was disbalance severely.XDD could ameliorate remarkably disbalance state of vasoactive substance in wind heat syndrome migraine model.
Objective:To try to establish a novel migraine animal model representing wind-heat syndrome.Method:Establishing rat model by having animals blew hot wind and GTN administration,taking pictures of rats' tongues,auricles and conjunctivae,analyzing the gray scale of rat tongues to evaluate the TCM syndrome and demonstrating the syndrome by Xiong Zhi Di Long Tang.Result:The gray scale level of tongue in GTN groups were significantly higher than control group(P0.01);the gray scale level in medicated group was slightly lower than GTN groups(P0.05).Observed by naked eyes,auricles and conjunctivae showed more blood vessel network than the control group.Wind-heat did not cause any significant difference between groups.Conclusion:GTN administration can cause changes in the manifestation of rat tongues,auricles and conjunctivae.The wind-heat syndrome model needs to be further considered to grow mature.
Objective To study the mechanism of analgesic effect of Xiongzhi Dilong Decoction [Decoction of Chuanxiong (Rhizoma Ligustici Chuangxiong),Baizhi (Radix Angelicae Dahuricae),Dilong (Lumbricus)] on the migraine model rats. Methods The 48 healthy male SD rats were randomized into 4 groups:normal saline group,model group,Sumatriptan group,and Chinese medicine group,with 12 in each. The migraine models were made by subcutanous injection of nitroglycerin,and the brain tissue samples were collected 4h and 6h after medication intervention. The immunohistochemical staining technique was then conducted to determine the expression of celluar oncogene fos (c-fos) and 5-hydroxytryptamine (5-HT) in SPN 5 and PAG. Results Expression in SPN 5:A reduction of c-fos expression in the Chinese medicine group was observed,with a decrease in expression from 4h to 6h. There was statistical difference in comparing with that of the model group (P0.01). An increase of 5-HT expression in the Chinese medicine group was noticed,with a slight decrease in expression from 4h to 6h. There was statistical difference in comparing with that of the model group (P0.05,P0.01). Expression in PAG:c-fos expression in the Chinese medicine group was reduced,with decrease in expression from 4h to 6h. There was statistical difference in comparing with that of the model group (P0.01). The increase of 5-HT expression in the Chinese medicine group was more significant,and the positive cell was more at 6h than at 4h. There was statistical difference in comparing with that of the model group (P0.01). Conclusion Xiongzhi Dilong Decoction is able to reduce the c-fos expression induced by nitroglycerin,increase the 5-HT expression,hence depressing excitability in neurons of SPN 5 by stimulating the descending 5-HT analgesic pathway. This effect can last for a long period of time,to make the pain threshold increased and the transmission of nociceptive information inhibited.
Objective To observe the changes of multiple organ function in rats with liver-qi stagnation syndrome.Methods A rat liver-qi stagnation model was induced by independent chronic bandage feeding.Changes of behaviors in rats were evaluated by open-field test.Serum motilin(MTL) and interleukin-2(IL-2) levels were measured by radioimmunoassay.Changes in body mass index,thymus and spleen index,gastric empting and intestinal peristalsis were observed.Results The amount of horizontal and vertical movements was smaller,the growth of body mass index was slower,the gastric empting time was shorter,while the thymus index,serum motilin and IL-2 level were lower in rats with liver-qi stagnation syndrome than in controls(P0.05).Conclusion Liver-qi stagnation syndrome can lead to changes of multiple organ function in rats.
Objective:To observe the acupuncture treatment on depression patients with two TCM syndromes,one is stagnation of liver-qi and deficiency of the spleen(GP),the other is the syndrome of double deficiency of the heart and the spleen(XP).Methods:We have selected 12 patients of GP and 8 patients of XP.Two groups were selected different points before acupunture treatment,every subject were checked using Hamilton rating scale for depression(HAMD)and measuring scale of TCM syndrome as a clinical evaluation.Results:The GP group showed conspicuous therapeutic effect but the XP group didn't.Conclusion:In the end of the 3 weeks' treatment ,performanced on above acupuncture points ,conspicuous therapeutic effects were found within the GP group but not within the XP group.
Objective To study the influences of Chaihushugan Powder on the behaviors and serum levels of adrenocorticotropic hormone (ACTH) and cortisol (CORT) in rat model of liver-depression syndrome. Methods All rats were divided into the blank control group, liver-depression model group and Chaihushugan Powder group. The rat model of liver-qi depression syndrome was established by using die bandage combining with independent feeding. The changes of ethological indexes of open-field behaviors in rats were observed, and the levels of ACTH and CORT were detected by using radioimmunoassay. Results Chaihushugan Powder improved the horizontal and vertical movements, decreased the stopping time in middle position, and reduced the levels of ACTH and CORT in rat model of liver-qi depression syndrome. Conclusion Chaihushugan Powder had an antagonized effect on liver depression, and the mechanism may be related to that it can regulate the levels of ACTH and CORT.
Objective To study the influences of Chaihushugan Powder on the ethological changes and levels of serum gastrin(GAS) and plasma motilin(MTL) in rat model of liver depression syndrome.Methods All rats were divided into the blank control group,the model group of liver depression syndrome and the Chaihushugan Powder group.The method combining die bound and lonely feeding was used to establish the rat model of liver depression syndrome.The changes of ethological indexes of immobility time in forced swimming test were observed after the administration of Chaihushugan Powder,and the levels of serum GAS and plasma MLT were detected by using radioimmunoassay.Results Chaihushugan Powder reduced the immobility time in forced swimming test,and increased the levels of serum GAS and plasma MTL in the rat model of liver depression syndrome.Conclusion Chaihushugan Powder has the effect of antagonizing liver depression and the mechanism may be related to that it can regulate the levels of serum GAS and plasma MTL.
