C-Met receptor and its ligand hepatocyte growth factor (HGF) are overexpressed in a variety of osteosarcoma cell lines and osteosarcoma pathological samples. It is suggested that c-Met/HGF plays an important role in the development of osteosarcoma. This study aimed to explore the anticancer effect of the c-Met-targeted drug crizotinib on osteosarcoma (OS) cells. The effects of crizotinib on the proliferation of osteosarcoma cells (SaOS2, MG-63 and MNNG) at different concentrations were detected by CCK8. Human osteosarcoma cell line MG-63 was used as an in vitro model to evaluate the effects of 2.5 μM crizotinib, 5.0 μM crizotinib and DMSO on cell apoptosis, cell cycle, migration and invasion. The expression of the c-Met signaling pathway in osteosarcoma cells was detected by western blot. The results showed that crizotinib inhibited the proliferation of cell lines in a concentration-dependent manner. Crizotinib significantly increased the number of apoptotic cells compared with the control group. Compared with the control group, crizotinib increased G0/G1 phase cells and decreased S phase cells. Compared with the control group, crizotinib inhibited the migration and invasion of osteosarcoma cells and decreased the expression of c-Met/Gab1/STAT5. This study will provide a promising therapeutic target and theoretical basis for the clinical application of crizotinib in osteosarcoma.
Osteoarthritis (OA), a progressive disease of the joints, affects millions of people worldwide. In the present study, we investigated the effects of oridonin, a diterpenoid isolated from Rabdosia rubescens, on IL-1β-induced inflammation using human osteoarthritis chondrocytes. The results showed that oridonin significantly suppressed IL-1β-induced MMP1, MMP3, and MMP13 production. IL-1β-induced NO and PGE2 production, as well as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression were also attenuated by oridonin. Western blot analysis demonstrated IL-1β-induced NF-κB activation was reduced by oridonin. Furthermore, the expression of PPAR-γ was increased by oridonin in a concentration-dependent manner. PPAR-γ antagonist could reverse the anti-inflammatory activity of oridonin. The results suggested that oridonin could be a candidate agent for the treatment of OA.
Objective To evaluate one-stage repair of leg open fracture with soft tissue defect by fixation plus skin flap.Methods From January 2011 to January 2018,34 patients with leg open fracture plus soft tissue defect were treated at Department of Orthopedics,The Second Affiliated Hospital to Harbin Medical University.They were 31 males and 3 females,aged from 13 to 54 years(average,35 years).According to the Gustilo classification,the defects were type ⅢA in 31 cases and type Ⅲ B in 3 cases.The wound size ranged from 6 cm × 3 cm to 25 cm × 10 cm.All patients were treated with emergency fracture fixation and flap repair by one stage after thorough debridement.Internal bone plating was performed in 32 patients and external fixation at a single arm in 2 patients.All defects were repaired with a free anterolateral thigh flap.Flap survival,vascular crisis and bone infection were followed up postoperatively.Results All the free flaps survived in the 34 cases with no vascular crisis.These patients were followed up for 6 months to 3 years (average,10 years).The flaps healed well,with good appearance and soft texture.No bone infection occurred at the defective sites.After postoperative rehabilitation,the knee extension ranged from 160° to 180°,knee flexion from 90° to 110°,ankle dorsal extension from 90° to 110°,and ankle plantar flexion from 100° to 120°.Conclusion Soft tissue defects in leg open fracture can be effectively treated by flap repair after thorough debridement at the same time of fracture fixation,because the primary wound healing can avoid infection and lead to fine survival of the flap.
Glucocorticoids (GCs) are widely used to treat a variety of autoimmune diseases, but long-term use can lead to osteoporosis. To elucidate the mechanism of osteoporosis caused by glucocorticoids and to find effective protective drugs/foods, osteoblasts treated by prednisolone acetate were studied and salvianolic acid B (Sal B) was added to osteoblasts. The results showed that Sal B increased the activity of ALP and stimulated the expression of ALP that had been suppressed by prednisolone acetate. To further study the mechanisms of the protective effect of Sal B on osteoblasts treated with prednisolone acetate, the effects of gene expression involved with bone formation and differentiation were studied. The results show that the mRNA and protein expression of Runx2, Osx, OCN, IGF-I, Col-I and HO-I was up-regulated by Sal B. In conclusion, by stimulating the osteoblast activity and the expression of genes related to bone formation and differentiation, Sal B had a protective effect on osteoblasts that had been treated with prednisolone acetate.
Objective To study the application of modified dynamic Kirschner wire-rubber traction external fixator in the treatment of intra-articular fracture or dislocation of the proximal interphalangeal joint.Methods From January 2014 to June 2017,23 patients (32 fingers) with intra-articular fracture or dislocation of the proximal interphalangeal joints were treated by modified dynamic Kirschner wire-rubber traction external fixator after closed reduction under C arm X ray machine guidance.Results After 8 to 12 weeks,the fractures healed and the joint function recovered to the greatest extent.All the patients were follow-up for 3 to 15 months.No obvious complications such as adhesion and joint stiffness occurred.According to TAM functional evaluation,the results were graded as excellent in 19 cases and good in 4 cases.Conclusion The modified dynamic Kirschner wire-rubber traction external fixator is exquisite and simple.It does not produce iatrogenic blood supply damage during the treatment of intra-articular fracture or dislocation of the proximal interphalangeal joint.It is conducive to fracture healing,supports early functional exercise,protects joint function to the greatest extent,and closed reduction can promote early fracture healing.
Licochalcone A (Lico A), a flavonoid found in licorice root (Glycyrrhiza glabra), has been reported to have anti-inflammatory activity. In this study, we evaluated the anti-inflammatory effects of Lico A on IL-1β-stimulated human osteoarthritis chondrocytes and investigated the possible mechanism. Results demonstrated that Lico A treatment significantly inhibited PGE2 and NO production induced by IL-1β. IL-1β-induced iNOS and COX-2 expression were also inhibited by Lico A. Lico A inhibited MMP1, MMP3, and MMP13 production in IL-1β-stimulated chondrocytes. Lico A also inhibited IL-1β-induced phosphorylation of NF-κB p65 and IκBα. Meanwhile, Lico A was found to upregulate the expression of Nrf2 and HO-1. However, Nrf2 siRNA reversed the anti-inflammatory effects of Lico A. In conclusion, our results suggested that Lico A showed anti-inflammatory effects in IL-1β-stimulated chondrocytes by activating Nrf2 signaling pathway.
目的:探讨中国汉族人群中载脂蛋白Eε4等位基因与急性脊髓损伤(ASCI)预后的相关性.方法:采用限制性片段长度多态性聚合酶链反应(PCR-RFLP)方法,检测2012年5月-2015年10月就诊于哈尔滨医科大学附属第二医院脊柱外科的50例(男性患者35例,女性患者15例)颈段(C3-C8)脊髓损伤的中国汉族患者载脂蛋白E ε4等位基因的遗传学状态,并以ASIA脊髓损伤神经学分类标准为患者神经功能恢复情况进行评估,分析中国汉族人群中载脂蛋白Eε4等位基因与急性脊髓损伤预后的相关性.结果:在运动方面,载脂蛋白Eε4阴性的患者恢复情况显著强于载脂蛋白Eε4阳性的患者恢复情况(mean 6.1vs 3.7,P=0.04).而在针刺觉方面,载脂蛋白Eε4阴性的患者恢复情况显著弱于载脂蛋白Eε4阳性的患者恢复情况(mean 4.0 vs 6.1,P=0.03).在轻触觉方面,载脂蛋白Eε4阴性和阳性患者的恢复情况没有显著的差异(mean 7.4 vs 5.9,P=0.28).结论:中国汉族人群中载脂蛋白Eε4等位基因与ASCI的预后密切相关:载脂蛋白Eε4阳性不利于患者运动的恢复,利于患者针刺觉的恢复.
Thymoquinone, an active ingredient isolated from Nigella sativa, has been reported to have anti-inflammatory effects. However, the anti-inflammatory effect of thymoquinone on IL-1β-stimulated osteoarthritis chondrocytes remains unclear. In this study, we designed to investigate the anti-inflammatory effects and elucidated the underlying mechanism of thymoquinone on IL-1β-stimulated human osteoarthritis chondrocytes. The effects of thymoquinone on inflammatory mediators COX-2, iNOS, NO, PGE2, as well as MMP-1, MMP3, MMP13 production were detected. The results demonstrated that thymoquinone concentration-dependently inhibited IL-1β-induced COX-2, iNOS, NO, and PGE2 production. Thymoquinone also suppressed IL-1β-induced MMP-1, MMP3, and MMP13 production. We found that thymoquinone significantly inhibited IL-1β-induced NF-κB activation and IκBα degradation. In addition, thymoquinone was found to suppress IL-1β-induced mitogen-activated protein kinases (MAPKs) activation. In conclusion, thymoquinone inhibited IL-1β-induced inflammatory mediator production by inhibition of NF-κB and MAPKs signaling pathways in osteoarthritis chondrocytes. Thymoquinone may be a potential agent in the treatment of osteoarthritis.

Objective To observe the expression offibroblast growth factor4 (FGF-4) in polydactyly of the thumb induced by cytarabine and investigate the pathogenesis of polydactyly. Methods Sixty-six 2 to 3 months old Sprague Dawley rats were caged together daily from 6 pm to 8 am next morning by a male:female ratioof 1:2.The day that a vaginal plug was found was considered as embryonic day 0.5 ( E0.5).30 pregnant rats were assigned to the experimental group, while another 30 were assigned as controls.At day E10.5 intraperitoneal injection of cytarabine ( 100 mg/kg) was carried out in the experimental rats to induce polydactyly and syndactyly in the pupa.Normal saline was injected to the control rats.E10.5 to E12.5 embryos were removed from the pregnant female rats and limb buds were taken out and fast frozen.mRNA was extracted from the limb buds.The expression of FGF-4 mRNA was quantified using RT-PCR. Results In the experimental group FGF-4 expression in the limb buds was significantly higher and lasted much longer than the control group (P <0.01).Conclusion The rat embryo's polydactyly of the thumb can be induced by cytarabine.FGF-4 expression in the limb buds in the polydactyly embryo is increased.
目的 研究5-氟尿嘧啶对体外培养的人类瘢痕疙瘩成纤维细胞MMP-1,MMP-2/TIMP-1,TIMP-2mRNA表达的影响,探讨5-氟尿嘧啶对瘢痕疙瘩的作用机制.方法 组织块法培养人瘢痕疙瘩及正常皮肤组织成纤维细胞,用不同浓度的5-氟尿嘧啶作用于体外培养的人瘢痕疙瘩成纤维细胞,检测MTT值;利用半定量RT-PCR分析用药前后MMP-1,MMP-2,TIMP-1,TIMP-2 mRNA表达变化.结果 5-氟尿嘧啶在体外能明显降低人瘢痕疙瘩成纤维细胞TIMP-1mRNA的表达;对MMP-1,MMP-2,TIMP-2mRNA的表达无显著影响.结论 5-氟尿嘧啶可能通过降低TIMP-1的表达,从而使MMP-1/TIMP-1相对比例增加,达到促进胶原降解,减少瘢痕形成的作用.
Vascular endothelial growth factor-B(VEGF-B) is an important member of the VEGF family,the third factor of the family discovered in 1996.The biological functions of VEGF-B have become a hot topic of studies.VEGF-B is expressed differently in different tissues of the human body.By binding with its specific receptor,it can promote angiogenesis,stimulate neurogenesis,act as a trophic factor,protect neuros from apoptosis,and regulate energy metabolism.This review focuses on the progress in resent researches on the biological functions of VEGF-B.
Objective:To study the clinical manifestations,diagnosis and treatment of glomus tumors in finger.Methods:The clinical data of 25 patients of the glomus tumors in finger were aralyzed retrospectively.Results:All of the patients had the same complaints that were severe painful on touching and with exposure to coldness in the finger tip.Radiographs can depict bone defection in some several patients.Glomus tumors could show high signal intensity on T2-weighted spin-echo MR images and strong enhancement after contrast medium injection.Complete excision,including the capsule,was the most effective treatment.The whole patients were cured after surgery,and the symptoms didn′t recur in 12-18 months after surgery.Conclusions:Pinpoint pain in the finger,paroxysmal pain and hypersensitivity to cold are the characteristics of glomus tumors.X-ray and magnetic resonance imaging(MRI) can contribute to diagnosis.The criticality of the cure is complete excision by surgery.
OBJECTIVE:To investigate the differentially expressed genes in rat in the process of regression of vascular calcification by using the suppression subtractive hybridization (SSH). METHODS:24 SD male rats which aged 6 weeks and specific pathogen free grade were selected and randomly divided into 3 groups (n = 8): control group, calcification group and regression group respectively. Vascular calcification model (vitamin D3 plus nicotine, VDN) were made from rats in calcification group and regression group, and rats in control group were intragastric administered with normal saline and lavaged with peanut oil. Rats were bred for 8 weeks in calcification group and control group, while rats in regression group were fed for 16 weeks. All rats were killed to measure concentration of calcium in the arterial tissue and examine the pathological lesion changes. Subtractive hybridization among vascular cDNA sequences from calcification group and regression group were established. The cDNA fragments which expressed higher or lower in regression group than those in calcification group were isolated. Differentially expressed genes with cDNA fragment were inserted into PMD18-T plasmid vector and transformed competent DH-5alpha, cDNA libraries of differentially expressed gene between calcification group and regression group were then constructed. Recombinant vectors were analyzed by colony PCR, positive genes were randomly selected for sequencing and analyzed by BLAST. 4 genes were randomly selected for RT-PCR certification combined with semi-quantitative analysis of DNA bands. RESULTS:VDN model of rats were successfully constructed. Concentration of tissue calcium in calcification group (15.34 mg/g +/- 2.51 mg/g) was significantly increased compared to that in control group (5.20 mg/g +/- 0.75 mg/g, P < 0.001), while in comparison with calcification group (15.34 mg/g +/- 2.51 mg/g), calcium in regression group was relatively lower (12.73 mg/g +/- 1.89 mg/g, P < 0.05). 28 up-regulated genes and 22 down-regulated genes were gained through sequencing and BLAST analysis among positive clones. RT-PCR validation indicated that 4 genes such as prdx3 and Ank2 had increasedly expressed in regression group than those in calcification group, the average fold change was 1.7. CONCLUSION:Rat vascular calcification tissue had characteristic of active regression. Genes in relation to pyrophosphoric acid synthesis, glutamate signal peptides, anti-oxidant and ant-apoptosis were up-regulated, at the same time many genes related to ossification and oxidation activity were down-regulated in the process of calcification regression. Increased expression of calcification suppressor genes accompanying decreased expression of calcification promoting genes might be the intrinsic mechanisms which initiated the active regression of calcified tissues.
Objective:To investigate the correlation between plasma anti-endothelial cell antibody(AECA)and the diffuse proliferative lupus nephritis.Methods:39 patients with SLE were divided into two group,one group with diffuse proliferative lupus nephritis and the other without it.The positive rate and concentration of AECA in the two groups were detected using indirect immunofluorescence technique.Results:AECA positive rate was 38.5% in all 39 lupus nephritis patients.The group with diffuse proliferative lupus nephritis had higher AECA positive rate and plasma concentration than another group without diffuse proliferative lupus nephritis,but the difference was no significance.The area under the ROC curve of AECA for diffuse proliferative lupus nephritis was 0.63(95% Confidence Interval 0.45~0.81,P=0.20).Conclusion:Positive rate of AECA was relatively higher in patients with diffuse proliferative lupus nephritis.AECA was not a good predictive factor for diffuse proliferative lupus nephritis.
目的 检测软骨脱细胞基质(ACM)对软骨细胞的生物学作用,探讨其作为软骨组织工程支架的可行性.方法 切取健康的雄性日本大耳白兔膝关节的软骨,经低温粉碎成软骨颗粒,通过多步骤酶方法将软骨颗粒行脱细胞处理,并用含20%胎牛血清的DMEM研磨制成ACM液.再切取兔膝关节的软骨,制成碎屑后分离培养软骨细胞.取培养第2、3代的软骨细胞,分成对照A组和B、C、D3个实验组(分别加入0.5、1.0、5.0ms/ml的ACM液).采用MTT法检测软骨细胞的活力,测定糖胺多糖的含量,以及H-胸腺嘧啶(3H-TdR)和3H-脯氨酸(3H-Pro)掺人法测定软骨细胞的增殖情况和胶原的合成能力.结果 ACM扫描电镜观察软骨巢内软骨细胞消失.软骨细胞在ACM液中生长良好,形态正常.B、C、D与A组相比,软骨细胞的生长活力分别增加了7.73%、13.50%、16.5%(P<0.05或P<0.01).糖胺多糖的含量在B组与A组相比其差异无统计学意义(P>0.05);而C、D组比A组增加了51.15%、62.68%(P<0.05或P<0.01).软骨细胞3H-TdR和3H-Pro掺入后,在B、C、D组中细胞的增殖及合成的胶原与A组相比,其差异均有统计学意义(P<0.01).结论 ACM能够提高软骨细胞的生长活力,增加糖胺多精的含量,刺激软骨细胞的增殖和胶原的合成,是可供选择的组织工程支架材料之一.
BACKGROUND There are many studies on the RIFLE classification to evaluate the occurrence rate and/or outcome of acute kidney injury (AKI) in ICU patients, but there are no studies on the RIFLE classification to evaluate the outcome of AKI in lupus patients. METHODS This retrospective study analysed the short-term outcomes of 79 diffuse proliferative lupus nephritis patients according to the RIFLE classification. RESULTS A total of 46% of patients were No AKI, 23% AKI-R, 16% AKI-I and 15% AKI-F according to the maximum RIFLE class reached on the first day of admission. The percentage of progression of AKI to the more severe RIFLE class was 6% for AKI-R, 23% for AKI-I and 75% for AKI-F (P < 0.0001), and there was an increased odds ratio (OR) of progression rate with more severe RIFLE category (OR 7.7, 95% CI 2.3-25.7, P < 0.001). The recovery rate at the end of a 24-week follow-up was 100% for AKI-R, 92% for AKI-I and 33% for AKI-F (P < 0.0001). The mean time to recovery for the groups AKI-R, AKI-I and AKI-F was 4, 11 and 20 weeks, respectively (P < 0.0001). The area under the ROC curve for progression to chronic kidney disease (CKD) was 0.96 (95% CI 0.91-1.0, P < 0.001). CONCLUSION The RIFLE classification is predictive of progression and short-term prognosis of AKI in diffuse proliferative lupus nephritis.
Objective To investigate relations of proliferation of hypertrophic scar fibroblast inhibited by 5-FU and expression of the cyclinD1,CDK4 and TGF-β1 and to learn about the mechanism of 5-FU inhibition on the proliferation of hypertrophic scar fibroblast.Methods After treated with different concentration of 5-FU,inhibition rate,and cyclinD1,CDK4,TGF-β1 expression of hypertrophic scar fibroblasts were detected by MTT assay and semi-quantitative RT-PCR respectively.Results The effect that 5-FU inhibited hypertrophic scar fibroblast proliferation became more and more significant along with the density increase of 5-FU,and the expression of the cyclinD1,CDK4,TGF-β1 mRNA could be inhibited significantly.Conclusion The mechanism of 5-FU on inhibiting fibroblast of hypertrophic scar may be related with regulation of the expression of the cyclinD1,CDK4 and TGF-β1.
Objective To investigate the expression of cyclinD_1, cdk4 and p53 in cultured fibroblasts human hypertrophic scar before and after treatment with 5-FU, and to learn about the mechanism of 5-FU′s inhibiting effects on the proliferation of hypertrophic scar. Methods After treated with 5-FU on different concentrations, inhibiting rate, the expression of cyclinD_1, cdk4, p53 in cultured fibroblasts from the hypertrophic scar and normal skin were detected by MTT assay and semi-quantitative RT-PCR respectively. Results 5-FU can significantly inhibit the proliferation of the fibroblasts from hypertrophic scar and decrease the expression of cyclinD_1 and cdk4 of cultured fibroblasts, but there was no significant effect on the mRNA expression of p53 in cultured fibroblasts. Conclusion The mechanism of 5-FU on inhibiting fibroblast of hypertrophic scar may be related with the inhibiting the expression of cyclinD_1 and cdk4, but may not be related with the expression of p53.
OBJECTIVE:To investigate the closing method of wound after removal of the traditional pedicled abdominal flap.METHODS:According to the design, the pedicled abdominal flaps were cut and lifted, and then the incision were extended from both sides on base of the flap to anterior superior iliac spine, respectively. After separating on superficial fascia, two flaps were obtained. The wound of donor site was closed completely by these two pedicled flaps. Twelve patients with skin defects on hands or forearms were treated using the reformed method of traditional pedicled abdominal flap.RESULTS:All of the 12 reformed pedicled abdominal flaps survived, and only one had local necrosis on the distal part of the abdominal flap, about 1.5 cm x 2.0 cm.CONCLUSION:This new design could provide a good method to close the abdominal wound after removal of pedicled abdominal flap.
