"形势与政策"课作为高校思想政治理论课的重要组成部分,肩负着宣传党和国家重大方针政策、宣讲新成就和新形势的使命和任务,是培养大学生树立正确形势观政策观、掌握科学分析时事方法的必修课程.创新"形势与政策"课教学方式,形成领导干部宣讲政策、名师专家解读时事、劳动模范引领价值等多元授课主体进行专题式教学模式,可以有效提高"形势与政策"课的政治性、理论性和实效性.
基于CRISPR/Cas系统识别和切割靶标DNA或RNA的特性不仅开发出精准、高效的基因编辑技术,也设计出革新的高灵活性和灵敏度的病原体检测技术.该文介绍并阐明CRISPR/Cas系统的不同作用机制和最新分类,着重综述"2"类CRISPR系统在诸多病原体检测中的研究进展,讨论分析了 CRISPR/Cas系统在病原体检测领域存在的优点以及面临的挑战.
CRISPR/cas9基因编辑系统可高效的实现目标基因靶向敲除,是当前基因功能研究的强有力工具.然而目前该系统主要针对编码蛋白质基因的靶向编辑,靶向非编码基因,尤其是miR-NAs的基因少有报道.由于miRNA序列的简短性和多样性,从基因功能缺失的水平上研究其功能、成熟过程等还有一定的困难.我们基于CRISPR/cas9基因编辑系统,特异有效的敲除了HEK293T细胞中的miRNA-155和miRNA-182的种子区或发夹序列区,同时发现cas9与gRNA不同配比的转染剂量显著影响目标miRNA的敲除效率,其中cas9∶gRNA的转染剂量在1∶4时敲除效率最高.我们的研究阐明了 CIRSPR/cas9系统在靶向敲除miRNAs基因的可行性,为研究miRNA在特定细胞中的生理学功能提供技术基础.
Methylation plays an important regulatory role in embryo development and organization.Somatic nuclear transfer embryos often show the abnormal mode of DNA methylation,DNA methyltransferase inhibitors can inhibit the levels of DNA methylation,so as to improve the efficiency of SCNT.The application of DNA methyltransferase inhibitors in animal somatic cell nuclear transfer was taken an overview in this paper.
Apoptosis is a process that cells autonomicly go to death,so that to maintain homeostasis and adapt to living environment.Apoptosis involves a series of gene activation,expression and regulation.It has been observed that there are the occurrence of apoptosis during preimplantation embryos development,but if the degree of apoptosis is beyond the normal range,and it is not conducive to the development of embryos.This paper provides a review of the current state of knowledge on apoptosis during preimplantation embryo development,in order to provide a theoretical basis for improving the quality of embryo in vitro culture.
