目的 优选蒙药寒水石化灰小剂的组方构成和炮制工艺,对比分析主要成分.方法 制备寒水石化灰小剂各组样品的含药血清和半仿生法溶出样品.四氮唑盐比色法(MTT)观察各组样本对人胃癌细胞MGC80-3增殖的影响;细胞集落形成实验观察碳酸钙组方和硫酸钙组方对细胞集落克隆形成率的影响.原子吸收法测定钙含量,质谱对比分析组方中主要成分.结果 寒水石化灰小剂1号样本组(500℃、焖煅1h,组方中含有查干榜嘎和硫酸钙)对人胃癌细胞增殖抑制作用最为显著,且克隆形成率低.1号样本溶出最少,但溶出液中钙含量高于对应生品.液相色谱-质谱(LC-MS)分析各样品的胡椒碱和没食子酸等成分含量随温度升高而逐渐降低.结论 寒水石化灰小剂的最佳炮制工艺为500℃、焖煅1h,最优组方是寒水石(硫酸钙)、查干榜嘎组方.
目的:探讨蒙药寒水石化灰小剂对S-180荷瘤小鼠肿瘤生长的影响及凋亡相关蛋白的调控作用。方法:建立S-180荷瘤小鼠模型,随机分为模型组,阳性对照组,环磷酰胺+寒水石化灰小剂低剂量联合给药组,寒水石化灰小剂高、中、低剂量组。各组连续给药后,称瘤重、脾重,计算抑瘤率、脾系数;ELISA法检测血清中免疫相关因子TNF-α、IL-6、IL-10的含量;取肿瘤组织常规HE染色镜下观察肿瘤组织;应用Western Blot方法检测凋亡相关蛋白Bcl-2、Bax和血管内皮细胞标志物CD34的表达。结果:与模型组相比,各给药组的瘤重均显著降低,抑瘤率分别为阳性对照组50.28%,联合给药组54.73%,寒水石化灰小剂高、中、低剂量组分别为31.07%、36.05%、13.14%。联合给药组和寒水石化灰小剂高、中剂量组IL-6含量均显著高于模型组和阳性对照组;联合给药组和寒水石化灰小剂高、中剂量组IL-10含量均显著低于模型组(P<0.05,P<0.01)。模型组病理切片观察发现模型组肿瘤组织血管丰富,坏死率约为10%-15%,联合给药组,寒水石化灰小剂高、中剂量给药组血管相对较少,纤维化明显,坏死率明显高于模型组;肿瘤组织的Bcl-2蛋白表达显著降低,而Bax蛋白表达显著增加,Bcl-2/Bax比值显著降低(P<0.05)。结论:寒水石化灰小剂可能是通过调节免疫,调控凋亡相关蛋白的表达,抑制肿瘤血管生成相关蛋白表达,发挥抑制肿瘤生长的作用;同时,寒水石化灰小剂与环磷酰胺有协同抗肿瘤作用。
炎性细胞因子与胃癌的发展过程密切相关,促炎细胞因子和抗炎细胞因子及其相关的信号通路通过不同途径,对胃癌的发生、发展以及预后等阶段起到了不同作用.本文综述了幽门螺杆菌、IL-1β、TNF-α、IL-8、TGF-β、IL-10、IL-18和T淋巴细胞在胃癌组织或血液中的表达及功能,并将这些炎性细胞因子及其相关信号通路作为胃癌的治疗靶点也做了进一步总结.
目的 通过抗惊厥、镇静与烫伤治疗实验,研究合成的药用硫化汞纳米粉的主要药效,旨在探讨药用硫化汞纳米化对其药效学的影响.方法 (1)烫伤作用:取小鼠30只,随机数字表法分成3组,90℃水造成浅二度烫伤模型,模型组仅涂抹麻油,阳性对照组涂抹京万红,实验组涂抹硫化汞纳米粉膏剂,观察各组小鼠烫伤部位的结痂情况.(2)镇静作用:取小鼠50只,随机数字表法分为5组,模型对照组灌胃给予蒸馏水,阳性对照组腹腔注射地西泮片,实验组给予药用硫化汞纳米粉,连续给药7天,并于末次给药后,依次给各组小鼠腹腔注射戊巴比妥钠,记录睡眠潜伏期、睡眠发生率及睡眠时间.(3)抗惊厥作用:取小鼠50只,随机数字表法分为5组,模型对照组灌胃给予蒸馏水,阳性对照组腹腔注射给予地西泮片,实验组为药用硫化汞纳米粉.连续给药七天,并于末次给药后,依次给各组小鼠注射戊四唑,制造惊厥模型.观察小鼠惊厥发生率.结果 (1)烫伤作用:药用硫化汞纳米粉对伤口具有显著收敛作用(P≤0.01)(2)镇静作用:药用硫化汞纳米粉高、中、低剂量组均能显著减低睡眠潜伏期(P≤0.01)(3)抗惊厥作用:药用硫化汞纳米粉高、中、低剂量组惊厥率和死亡率明显较低(P≤0.01).结论 合成的药用硫化汞纳米粉具有治疗烫伤、镇静与抗惊厥的药理作用.
介绍色谱法、光谱法、核磁共振波谱、质谱及其联用技术,薄层扫描法、原子吸收分光光度法、硫酸-苯酚法、毛细管电泳柱头场强富集技术、免疫反应等几种现代分析方法在中成药含量测定中的应用。
This study was aimed to preliminary explore the anticancer activity, immunomodulatory effects and antimi-crobial activity of Calcitum ashing agent. Serum pharmacological method was used to prepare the serum containing Calcitum ashing agent. MTT method was used to observe the cell proliferation rate of S180 ascites tumor cells and human esophageal cancer cells cultured with the serum containing Calcitum ashing agent in vitro. In addition, this method was also used to observe the mouse spleen lymphocyte transformation rate. The Oxford Cup method was used to determine the antibacterial activity on Calcitum ashing agent in vitro. The results showed that serum containing Calcitum ashing agent inhibited the cell proliferation rate of S180 and esophageal cancer cells. Meanwhile, it had a synergistic effect with 5-fluorouracil (5-FU) on the inhibition of human esophageal cancer cells. The serum contain-ing Calcitum ashing agent induced the reduction in the mice spleen lymphocytes transformation rate. But it inhibited the reduction in the spleen lymphocytes transformation rate induced by 5-FU. The decoction with the concentration higher than 0.25 g·mL-1 had significantly inhibited the proliferation of bacteria. It was concluded that Calcitum ash-ing agent can inhibit the cell proliferation of S180 ascites tumor cells and human esophageal cancer cell in vitro. It can significantly reverse the reduction in the mice spleen lymphocytes transformation rate induced by immunode-pressant. However, the antibacterial activity still requires further study.

目的:对不同产地的寒水石炮制前后的主成分及微量元素含量进行对比分析。方法:对四种寒水石生品进行性状鉴别和矿物鉴定,X射线衍射定性分析炮制前后主成分,乙二胺四乙酸二钠(EDTA)滴定法测定其中钙离子含量,二米光栅摄谱仪进行微量元素定量分析,原子吸收法测定样品中As、Sb、Bi、Hg。结果:寒水石炮制后主成分钙离子百分含量增加,炮制前后所含微量元素含量发生改变。结论:炮制使寒水石钙离子含量增加,而微量元素含量变化不确定,没有显著规律性。
