OBJECTIVETo investigate the effect of tetrandrine (Tet) on the expression of bax, bcl-2, and transforming growth factor-β2 (TGF-β2) mRNA in cultured human fibroblasts of Tenon's capsules (TCFS) and explore its possible mechanism.METHODSThe third passage of TCFS cultured in vitro were exposed to 1×10(-5) mol/L Tet for 24 h, and real-time fluorescence quantitative PCR was used to detect the changes in the expressions of bax, bcl-2, and TGF-β2 mRNA.RESULTSThe expression level of bax mRNA was obviously higher, while bcl-2 and TGF-β2 mRNA levels were significantly lower in Tet-treated TCFS than those in the control cells (P<0.05).CONCLUSIONTet can inhibit the proliferation of TCFS possibly by reducing the expressions of bcl-2 and TGF-β2 mRNA, enhancing the expression of bax mRNA and inducing cell apoptosis, suggesting its potential in preventing fibrous scar formation after glaucoma filtration surgery.
目的研究喉罩通气全麻对乳腺癌手术患者呼吸和循环功能的影响。方法选取乳腺癌根治手术患者40例,随机分为气管导管组(T组)和喉罩组(L组),记录两组各时间点的循环和呼吸功能监测指标。结果两组在间歇通气10min和30min时MV、VT、SpO2的差异无统计学意义(P>0.05);在60min时L组的Ppeak和PETCO2高于T组(P<0.05),L组置入以后各时点的MAP和HR较T组降低(P<0.05)。结论喉罩通气全麻用于乳腺癌手术对呼吸循环系统影响小,安全有效。
结膜下瘢痕组织增生是抗青光眼滤过术后失败的主要原因,其主要病理过程包括局部成纤维细胞增殖、胶原基质产生及血管新生[1].一些抗瘢痕药物,如皮质类固醇、丝裂霉素C等已用于临床青光眼滤过性手术,对改善滤过泡、提高手术成功率起到一定的作用,但也有一些副作用及并发症,有些药物还具有严重的毒性作用.本实验旨在观察汉防己甲素(Tet)滴眼液对兔眼滤过模型术区滤过泡瘢痕形成的影响,进一步探讨其作为青光眼滤过术后辅助用药的可能性及作用机制,旨在为临床运用提供实验依据.1 材料与方法1.1 主要仪器与试剂手术显微镜(苏州,SOM2000B),Tono-Pen眼压计(Model XL,mentor,Inc美国),LKBV型超薄切片机(Leitz公司),Olympus双筒显微镜(日本Olympus公司),透射电子显微镜(日本日立H-600型),全自动图像分析系统(Kontron IBAS 2,O德国),彩色图像摄录输入仪(Axioplan2 imaging德国).Tet滴眼液(天津第一中心医院药学部配制,pH值:6.1、渗透压:294mmol/L),丝裂霉素C(mitomycin C,MMC),手术用平衡盐注射液(BSS),复方妥布霉素滴眼液等眼药水及消毒液(中山眼科中心药剂室).
目的:对尿酸结石的组成成分进行分析,为进一步探讨其成因提供依据。方法:从2007年7月至2009年6月,对837例尿石症患者的结石标本用傅立叶红外光谱法进行成分分析。探讨其中64例含尿酸成分的结石与性别、年龄及结石位置之间的关系。结果:6例(9.4%)为纯尿酸结石,尿酸混合结石58例(90.6%),多以尿酸与草酸钙混合(59.4%)。其中18例(28.1%)尿酸的比例在50%及以上(即为尿酸结石),46例(71.9%)少于50%(非尿酸结石)。单因素分析显示尿酸结石全部来自于男性(P=0.008),且患者平均年龄显著高于非尿酸结石(54.7岁vs46.6岁,P=0.039)。尿酸结石多来自于下尿路(P=0.007)。结论:红外光谱分析法是一种研究结石成分比较理想的分析方法。性别和年龄对尿酸结石形成有一定的影响,此项研究能够为以后更深入的研究尿酸结石的病因提供线索。
OBJECTIVE To investigate the perioperative management of severe obstructive sleep apnea hypopnea syndrome (OSAHS). METHODS Fifty-three patients with severe OSAHS received uvulopalatopharyngoplasty with uvula preservation and radiofrequency tongue base reduction. All the patients were treated with automated continuous positive airway pressure (CPAP) for 3-7 days before operation and automated antibiotic therapy administered in the oropharynx, with 24 h ECG monitoring postoperatively. Polysomnography were carried out before and 6 months after surgery. RESULTS The preoperative apnea hypopnea index (AHI) and lowest SaO(2) (LSaO(2)) were 58.4-/+5.1/h and 0.650-/+0.059, respectively, which were 15.5-/+3.2/h and 0.864-/+0.064 at 6 months after surgery, respectively, showing significant changes after surgery (P<0.01). Dyspnea occurred in 2 cases after operation, intraoperative bleeding in 1 case, primary bleeding in 2 cases and hypertension crisis in 1 case. CONCLUSION Severe OSAHS patients are subject to great surgical risk. Application of auto-CPAP before operation can significantly improve the patients' tolerance of surgery and anesthesia, and reduce the surgical risks and preoperative complications.
OBJECTIVE To assess the possible association between gene mutation of cytochrome P450 1A1(CYP1A1) in exon 7 A4889G locus and the susceptibility to endometriosis (EM). METHODS Allele specific-polymerase chain reaction method was used to analyze gene mutation in exon 7 A4889G locus of CYP1A1 in 76 patients with endometriosis and 80 healthy controls. RESULTS The frequency of allele G on A4889G locus of CYP1A1 gene showed a significant difference between the study cohort and the control group (Chi2=7.498, P<0.01), with an odds ratio of 1.957. Statistically significant difference in the frequencies of genotypes AA, AG and GG was observed between the two groups (Chi2=6.915, P<0.05). Individuals with homozygotes for G allele were at higher risk of suffering from EM when compared against those with homozygotes for A allele, the odds ratio being 3.437 (Chi2=5.430, P<0.05). CONCLUSION The above results suggest that gene mutation of CYP1A1 in exon 7 A4889G locus might be a genetic susceptible factor of endometriosis. The mutation allele of CYP1A1 gene appears to increase the risk of endometriosis.
OBJECTIVE To evaluate the possible association of the glutathione S-transferase M1 (GSTM1) gene polymorphism with the susceptibility to endometriosis in women of Han nationality in Guangdong Province. METHODS Polymerase chain reaction was used to identify the GSTM1 genotypes in 76 patients with endometriosis and 80 controls (surgical patients for gynecological problems other than endometriosis). RESULTS The frequencies of the GSTM1 null genotypes in patients with endometriosis and controls were 65.8% and 46.3%, respectively, showing a significant difference between the endometriotic cohort and the control group (X(2)=6.03, P < 0.05). Individuals with GSTM1 null genotype were exposed to risks for endometriosis 2.24 times that of subjects without these genotypes OR=2.24, 95% CI=1.17-4.27 . CONCLUSION GSTM1 gene deletion might bea risk factor for endometriosis in women of Han nationality who are native residents in Guangdong Province.
OBJECTIVE:To assess the possible association of the Msp I polymorphisms of cytochrome P4501A1(CYP1A1) gene with the susceptibility to endometriosis in women of Han Nationality in Guangdong Province.METHODS:Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was employed to analyze the 3 genotypes m1m1, m1m2 and m2m2 in 3'-flanking region of CYP1A1 in 76 patients with endometriosis and 80 healthy controls.RESULTS:The frequencies of genotypes m1m1, m1m2 and m2m2 were 30.3 %, 50.0 % and 19.7 % in patients with endometriosis while 42.5 %, 45.0 % and 12.5 % in the controls, respectively, showing no statistically significant difference in the frequencies of the three genotypes between the 2 groups. The frequencies of two alleles were of no significant difference between the patients and controls, either.CONCLUSION:Msp I polymorphisms of cytochrome P4501A1 in itself might not be associated with the susceptibility to endometriosis in women of Han Nationality in Guangdong Province.
