Five new alkaloids, ferupencines A-E, featuring a pyrrol-oxazine core with a C3- C4O2N framework, were isolated from the secondary metabolites of the endophytic fungus Penicillium sizovae residing in Ferula sinkiangensis. Their structures were elucidated through comprehensive 1D and 2D NMR spectroscopy, along with mass spectrometric analysis, experimental and calculated (ECD) spectra, and other complementary methods. The absolute configuration of compound 1 was determined via X-ray crystallography, supported by experimental and calculated electronic circular dichroism (ECD) data. Notably, this is the first report of these compounds being isolated from endophytic fungi. The cytotoxicity of ferupencines A-E (1-5) was evaluated against three human tumor cell lines: HeLa, SGC-7901, and SHG-44. Compound 2 exhibited moderate cytotoxic activity against HeLa and SGC-7901 cells, with IC50 values of 18.6 μM and 14.2 μM, respectively. Additionally, compound 5 showed an IC50 value of 23.8 μM against HeLa cells, indicating moderate cytotoxic potential.
This study aimed to explore the correlation between rhizosphere soil microorganisms of wild Arnebia euchroma and the content of medicinal components to provide guidance for the selection of the ecological planting base. The total DNA of rhizosphere soil microorganisms of wild A. euchroma was extracted, and the microbial community structure of rhizosphere soil microorganisms was analyzed by IlluminaMiseq high-throughput sequencing technology. The content of total hydroxynaphthoquinone pigment and β,β'-dimethylacrylalkannin in medicinal materials was determined by high-performance liquid chromatography(HPLC). The physicochemical pro-perties of rhizosphere soil of wild A. euchroma in main producing areas were determined, and the correlation of soil microbial abundance with index component content and soil physicochemical properties was analyzed by SPSS software. The results showed that the species composition of rhizosphere fungi and bacteria in A. euchroma from different habitats was similar at the phylum and genus levels, but their relative abundance, richness index(Chao1), and community diversity(Simpson) index were different. Correlation analysis showed that the content of available phosphorus in soil was positively correlated with the content of total hydroxynaphthoquinone pigment and β,β'-dimethylacrylalkannin, and the abundance of five fungal genera such as Solicoccozyma and six bacterial genera such as Pseudo-nocardia and Bradyrhizobium was positively correlated with the content of medicinal components in medicinal materials. The abundance of Bradyrhizobium was significantly positively correlated with the content of β,β'-dimethylacrylalkanin. The abundance of fungi such as Archaeorhizomyces was significantly positively correlated with the content of available phosphorus in rhizosphere soil, and Bradyrhizobium was significantly negatively correlated with soil pH. Therefore, the abundance of fungi and bacteria in the rhizosphere of A. euchroma has a certain correlation with the medicinal components and the physicochemical properties of the rhizosphere soil, which can provide a scientific basis for the selection of ecological planting bases in the later stage.
A new compound from Ferula sinkiangensis has the potential to treat gastric cancer through apoptosis induction and cell cycle arrest.
目的:优化新疆阿魏种子中总倍半萜香豆素的提取工艺.方法:以乙醇体积分数、料液比、提取时间为自变量,在单因素试验的基础上,应用Box-Behnken中心组合设计建立数学模型,以总倍半萜香豆素提取率为响应值,进行响应面分析.结果:最佳提取工艺条件为80%乙醇加热回流提取40 min,料液比为1∶40.总倍半萜香豆素提取率预测值为6.60%,实际值为6.62%,相对误差为0.3%.结论:该优选工艺稳定可靠,为新疆阿魏资源的合理利用提供参考.
Two new sesquiterpene coumarins, namely, sinkiangenorin F (1) and 8-O-acetyl sinkiangenorin F (2) were isolated from the seeds of Ferula sinkiangensis. The structures of the new compounds, including the relative stereochemistry and the absolute configuration were elucidated through extensive spectroscopic methods. The two compounds were tested against the K562, HeLa, and AGS human cancer cell lines and showed cytotoxic activities with 50% inhibitory concentration values between 27.1 and 62.7μM.
A new sesquiterpene coumarin with a novel sesquiterpene carbon framework, Sinkiangenorin D, and ten known sesquiterpene coumarins were isolated from the seeds of Ferula sinkiangensis. The structures of these compounds, including the relative stereochemistry, were elucidated on the basis of spectroscopic data. All of the isolated compounds were tested against the AGS, HeLa, and K562 human cancer cell lines and showed cytotoxic activities with 50% inhibitory concentration values between 12.7 and 226.6 μM.
Morphology and molecular identification technology were used to identify 3 original plants of Fructus Elaeagni which was commonly used in Uygur medicine. Leaves, flowers and fruits from different areas were selected randomly for morphology research. ITS2 sequence as DNA barcode was used to identify 17 samples of Fructus Elaeagni. The genetic distances were computed by kimura 2-parameter (K2P) model, and the Neighbor-Joining (NJ) and Maximum Likelihood phylogenetic trees were constructed using MEGA5.0. The results showed that Elaeagnus angustifolia, E. oxycarpa and E. angustifolia var. orientalis cannot be distinguished by morphological characteristics of leaves, flowers and fruits. The sequence length of ITS2 ranged from 220 to 223 bp, the average GC content was 61.9%. The haplotype numbers of E. angustifolia, E. oxycarpa and E. angustifolia var. orientals were 4, 3, 3, respectively. The results from the NJ tree and ML tree showed that the 3 original species of Fructus Elaeagni cannot be distinguished obviously. Therefore, 3 species maybe have the same origin, and can be used as the original plant of Uygur medicineal material Fructus Elaeagni. However, further evidence of chemical components and pharmacological effect were needed.
Two new steroidal esters with an unusual framework, Sinkiangenorin A and B, a new organic acid glycoside, Sinkiangenorin C, and four known lignin compounds were isolated from the seeds of Ferula sinkiangensis. The structures of these compounds were established by spectroscopic analysis and single-crystal X-ray diffraction. All of the isolated compounds were tested against Hela, K562 and AGS human cancer cell lines. Sinkiangenorin C showed cytotoxic activity against AGS cells with an IC50 of 36.9 mu M. OBJECTIVE:We studied the diversity of endophytic fungi associated with Ferula sinkiangensis K. M. Shen. METHOD:Endophytic fungi from different years (1-2 years, 3-4 years and > 5 years) and different parts (root, stemand leaf) of Ferula sinkiangensis K. M. Shen were isolated by tissue expand method. Strains were classified by morphology and similarity of internal transcribed spacer (ITS) sequence by Clustal X method. Composition, diversity and preference of endophytic fungal community were analyzed by the isolation rate (IR), isolation frequency (IF), Shannon-Wiener biodiversity index (H'), Margalef Richness index (R). RESULTS:In total 140 endophytic fungi were isolated from F. sinkiangensis K. M. Shen and classified into 18 genera. Among the 140 isolates, Aureobasidium (25.7%), Alternaria (16.4%) and Phyllosticta (15.7%) were the dominant genera. The isolation results show that there were some notable differences between distribution and composition of the endophytic fungi isolated from different years and different parts of Ferula sinkiangensis K. M. Shen. Meanwhile, a certain degree of years and tissue preference were also obvious. CONCLUSION:The results obtained in this study will be helpful to exploit the endophytic fungal resources of Ferula sinkiangensis K. M. Shen, which can also provide a new way for the realization of the artificial breeding of Ferula sinkiangensis K. M. Shen.

Objective To compare the therapeutic effect of GCV and α-interferon to infectious mononucleosis(IM)in children.Methods222 patients were divided into the ganciclovir(GCV)treatment group(n=59),the α-interferon treatment group(n=74)and the control group(n=89).Among these three groups there were no differences in age,sex and patients,condition.Children in the GCVgroup were given GCV 10 mg/(kg·d),q12 h,iv drip for 7 days,then 5 mg/(kg·d),qd,iv drip,the whole treatment course were 10-14 days.Patients in the α-interferon group were given α-interferon 5×104U/(kg·d),im,for 10-14 days.The control group were given other drugs(ribavirin,etc).Results①Between the GCV group and the control group,significant difference were found in the shorten of fever duration,the improvement of the angina,the diminution of the lymph node、liver and spleen and the time atypical lymphocyte decreased10%.And the same results were also found in the comparation between the α-interferon group and the control group.But there was no statistical difference between the GCV group and the α-interferon group.②In the GCVgroup granulocyte were found in 4pts,11 pts had fever in the α-interferon group,the side effects were recovered within3-7 days after drug withdrawal.ConclusionBoth GCV and α-interferon are effective in the treatment of IM;And they both had no serious side effect in IM treament.
OBJECTIVETo explore the effect of different culture conditions and hormone combinations on callus induction of Ferula sinkiangensis.METHODSUsing different seedling age of Ferula sinkiangensis young cotyledon, hypocotyl, radicle as explants, which were cultivated on differentiation medium with different hormone combinations, to study the effects on Ferulic callus by different explants and hormone combinations.RESULTSFerulic seedlings with no true leaf was more favorable to the callus, the optimum medium for hypocotyl induction was MS + 2,4-D (1.0 mg/L) + KT (1.5 mg/L), for radicle induction was MS + NAA (0.5 mg/L) + 6-BA (0.5 mg/L), and the best subculture medium was MS + NAA (1.5 mg/L) + 6-BA (2.5 mg/L).CONCLUSIONSNAA,2,4-D and 6-BA must have the inductive effects, 6-BA is better than the KT in the process of proliferation and the medium containing GA3 is conducive to the process of subculture embryogenic callus production.

Objective To investigate the effect of creatine phosphate(CP)on myocardial injure induced by marcellomycin in children with acute leukemia.Methods 40 cases of acute leukemia with myocardial injure induced by marcellomycin were assigned to 2 groups randomly.The CP group(n=20),and the FDP group(n=20).The CP group was treated with CP(5yrs 0.5 g/d,≥5yrs 1g/d iv),The FDP group was treated FDP 250 mg/kg/d iv drip.Both groups were examined with cardiogram and myocardial enzymes and pre and post-therapy.Results The level of myocardial enzymes in both groups was decreased singnificantly after therpy(P0.01),and the difference of pre-and post-therapy has no significance(P0.05)between the two observed groups.Neither the improvement rate of the cardiogram nor the overall effective rate was significantly difference between them(P0.05).Conclusion CP shows the same good effects as FDP on myocardial injure induced by marcellomycin.
目的 评价蒙洛英注射液对儿童高热的退热疗效和安全性.方法 选择体温超过39.0℃的发热儿童50例,予蒙洛英深部肌肉注射.结果 蒙洛英退热效果显著,其显效率为84%,总有效率为100%,不良反应少且轻微.结论 蒙洛英注射液在年龄大于或等于6岁儿童高热的临床治疗中退热效果好,不良反应少,安全可靠,值得临床应用.
目的 观察蒲地蓝消炎口服液治疗疤疹性咽峡炎的临床效果.方法 对62例疤疹性咽峡炎患儿随机分为对照组和治疗组.治疗组32例用蒲地蓝每天15~30mL,分3次口服,对照组30例给予利巴韦林针剂(湖北潜江制药有限公司)10mg·kg-1,1日1次,静滴3~5d.两组均给予相同的对症支持处理,并使用头孢氛苄颗粒防治细菌感染.观察发热、口腔、咽峡部疤疹消退情况,并记录患儿退热时间和疤疹消退时间.结果 治疗组治疗总有效率与对照组比较有显著性差异(P<0.01).结论 蒲地蓝消炎口服液治疗疤疹性咽峡炎效果好.
