网络直播凭借丰富的内容和较强的互动性,已经成为社会生活娱乐的重要组成部分,网络直播也形成了新时代特有文化现象,显示了传统媒体无可取代的优势.当代大学生是伴随网络成长的一代,作为互联网常驻"居民",也是网络直播文化的主要受众,网络直播文化会直接冲击大学生尚不成熟的价值观,影响到大学生社会主义核心价值观的确立.本文结合直播特点,分析直播对大学生价值观的影响,并提出在新媒体背景下如何引导学生确立社会主义核心价值观的建议.
Diabetic nephropathy (DN) is one of the most severe complications that can occur in patients with diabetes, and without effective and timely therapeutic intervention, can gradually progress to renal failure. Previous studies have focused on investigating the pathogenesis of DN; however, the role of dual-specificity phosphatase 6 (DUSP6) in DN is not completely understood. Therefore, the present study aimed to investigate the role of dual-specificity phosphatase 6 (DUSP6) in DN. DN model mice were established and the expression levels of DUSP6 in the kidney tissues and high glucose (HG)-induced murine podocytes (MPC5 cells) were determined using immunohistochemistry, reverse transcription-quantitative PCR and western blotting. In addition, the levels of reactive oxygen species (ROS) and inflammatory cytokines in MPC5 cells were analyzed using commercial assay kits or ELISA kits, respectively, and flow cytometric analysis was performed to analyze the rate of cell apoptosis. The present study indicated that DUSP6 expression levels were significantly decreased in DN model mice compared with control mice, and in HG-induced MPC5 cells compared with normal glucose-induced MPC5 cells. DUSP6 overexpression enhanced MPC5 cell viability and increased protein expression levels of cell markers, such as synaptopodin and nephrin, compared with the negative control group. DUSP6 overexpression also reduced the levels of ROS and inflammatory cytokines, including interleukin (IL)-1 beta, IL-6 and tumor necrosis factor-alpha secreted by MPC5 cells under HG conditions. Moreover, compared with the HG group, cell apoptosis was inhibited by DUSP6 overexpression under HG conditions, which was further indicated by decreased expression levels of cleaved caspase-3 and Bax. Thus, these findings indicated that DUSP6 mediated the protection against HG-induced inflammatory response.
水果采摘作为水果生产过程中的一个重要环节,具有季节性强和劳动密集的特点,所占用的劳动力数量较大且整体自动化程度较低.为提高水果采摘效率,设计了一种基于视觉的橘类水果识别系统,基于水果的颜色特征、霍夫圆检测原理进行成熟橘类水果的识别,使用深度摄像头实现目标的定位.以蜜桔为研究对象,分别在正常光线无遮挡、正常光线有遮挡以及外部光源补光的情况下开展试验研究.试验结果表明,提出的基于视觉的橘类水果识别系统能够为水果识别与定位技术提供技术支撑和可靠数据,为实现水果智能化采摘样机的研制奠定基础.
糖尿病肾病(DN)是糖尿病患者最常见且最严重的并发症之一.在糖尿病患者中,约三分之一可发展为DN,进而发展为终末期肾病(ESRD),是导致糖尿病人群死亡率的主要原因[1].有关DN病因和发病机制的阐述涉及诸多因素,普遍认为是由遗传和环境因素共同引发的一系列复杂病理生理事件[2].通过在细胞和分子水平上对DN病因学的深入探究发现,炎症是其致病的一种重要病理生理机制,只是该炎症区别于传统意义上以红、肿、热、痛为主要特征的经典炎症,因其具有慢性、轻微等特征,通常被称为“微炎症”[3].
目的:探讨PHZ处理对P.bANKA感染小鼠前炎性细胞因子分泌的影响.方法:建立PHZ处理的P.bANKA感染的C57BL/6鼠疟模型,计数红细胞感染率;感染后第0、3和5天制备脾细胞悬液,ELISA检测脾脏中INF-γ和TNF-α分泌水平的变化.结果:与PbANKA感染组小鼠相比,P.b+PHZ组小鼠虫血症增高明显,生存期明显缩短;于感染后第3天和5天TNF-α明显减少;于感染后3天IFN-γ水平也显著下降.结论:PHZ处理,明显的增加了P.bANKA感染早期小鼠原虫血癌水平,生存期显著缩短.可能与贫血的P.y17XL感染早期C57BL/6小鼠分泌的TNF-α和IFN-γ水平降低,不能有效的杀死红内期疟原虫密切相关,确切机制还有待进一步研究.
目的 明确脐带间充质干细胞(UC-MSCs)对2型糖尿病(T2DM)db-/db-小鼠的临床疗效及安全有效的移植途径,旨在为临床应用提供实验与理论数据.方法 db/m和db-/db-小鼠各10只,每种小鼠各分为2组,db/m对照组,db/m+UC-MSCs组,db-/db-模型组,db-/db-+UC-MSCs组,5只/组.对照组和模型组给予1mL生理盐水腹腔注射;db/m+UC-MSCs和db-/db-+UC-MSCs组腹腔注射UC-MSCs为1×107个细胞/只鼠,总液体量为1mL.每周固定时间记录餐后血糖与体重,计算各组小鼠生存率.结果 与db-/db-模型组小鼠相比,腹腔途径移植UC-MSCs后,db-/db-小鼠的血糖水平明显下降,与非移植db-/db-小鼠比较差异有显著性,对db/m小鼠无影响;移植与非移植小鼠体重无明显变化.db-/db-+UC-MSCs组小鼠出现部分死亡,生存率为75%,其它组小鼠生存率均为100%.结论 UC-MSCs腹腔移植途径可有效控制T2DM小鼠血糖变化,对体重变化无明显影响.可为临床治疗提供参考.
目的 探讨弓形虫感染小鼠氧化应激状态对免疫功能的影响和香菇多糖(LNT)对小鼠脑保护作用及可能的作用机制.方法 流式细胞术检测各组实验小鼠脑组织中活性氧自由基(ROS)及脾脏中Th1/Th2细胞亚群.结果 感染后第4天,对照组小鼠出现死亡,于感染后第8天小鼠全部死亡;而LNT组小鼠感染后第6天才出现死亡,感染后第10天小鼠全部死亡;pre-6d LTN组小鼠在感染后第8天开始出现死亡,存活率为20%.对照组小鼠脾脏中ROS水平于感染后8d出现有意义升高(P<0.01);pre-LNT组和LNT组小鼠在感染后4和8d脾脏中ROS水平均出现有意义升高(P<0.05,P<0.01).各组小鼠脾脏中CD4+IFN-γ+(Th1)细胞数均于感染后4和8d均出现有意义升高(P<0.01);但与同天对照组小鼠相比,pre-LNT组和LNT组第4天和8天脾脏中Th1细胞水平均显著下降(P<0.05,P<0.01).对照组小鼠和LNT组小鼠脾脏中CD4+ IL-4+ (Th2)细胞数无明显变化(P>0.05);pre-LNT组小鼠在感染后4和8dTh2细胞水平均出现有意义升高(P<0.05);但与同天正常感染组小鼠相比,pre-LNT组第8天脾脏中Th2细胞水平均显著增加(P<0.05).结论 LNT能有效控制急性弓形虫感染早期ROS的产生及Th1细胞数量,对于感染性疾病急性进展期控制严重的炎症反应极其有效.
This study aimed to investigate the prevalence of facial infection with Demodex spp.among 291 Indian students and 314 native students of class 2013 in Jiamusi University,China.Samples were collected with the method of skin squeeze/scraping smear and subjected to microscopic examination within a drop of glycerol.Statistics were performed with SPSS 19.0 software.The results showed that in foreign students the positive rate of infection in males was 16.4% (34/207),and that in females was 9.5% (8/84),while in native students they were 49.0% (71/ 145) and 18.9% (32/169),respectively (x2 =43.14,P < 0.05 for males).There was a dominance of Demodex folliculorum infection followed by Demodex brevis infection and mixed infection in all the foreign students and the male natives,and a dominance of mixed infection in the female natives.In addition,in native students the infection rate in those with facial symptoms (68.2%) was significantly higher than those showing healthy facial skin (19.0%) (x2 =69.43,P < 0.05).
当前,如何提高教学管理执行力已成为高校发展过程中迫在眉睫需要解决的问题.本文从执行文化角度分析了目前高校教学管理执行力不足的现象和原因,提出运用执行力文化改变教学管理执行不力的弊端.
Objective:To investigate the regulatory effect of dendritic cells (DCs) on Th17 cell differentiation and function during mouse infection with Plasmodium yoelii 17XL strain (Py17XL) and the underlying mechanisms.Methods:Twelve female BALB/c mice were randomly assigned into the infection group (Py17XL), the TLR4 blocking group (Py17XL + TLR4), TLR9 blocking group (Py17XL + TLR9), and TLR4 and TLR9 combined blocking group (Py17XL + TLR4+TLR9)(n=3 in each group). Mice in the Py17XL + TLR4 or the Py17XL + TLR9 group received intraperitoneal injection of 10 μg anti-TLR4 or 50 μg anti-TLR9 antibody (both 0.4 ml) to block DCs function at one day before infection. The Py17XL group received same volume of PBS. All groups were then given intraperitoneal injection of 1×10(6) red blood cells (RBCs) infected with Py17XL. The RBC infection rate was calculated on days 0, 3 and 5 after infection, and spleen cell suspension was prepared, in which the CD11c+TLR9+ and Th17 cells were counted by flow cytometry. The levels of IFN-γ and IL-10 in supernatant of spleen cell culture were determined by ELISA.Results:Flow cytometry showed that DCs were successfully blocked. On day 5 after infection, 28%,29%, 31% and 16.3% mice showed parasitemia in the Py17XL group, the Py17XL + TLR4 group, the Py17XL + TLR9 group, and the Py17XL + TLR4 + TLR9 group, respectively, and on day 7, the proportions were 43.3%, 47.5%, 32.5% and 8%. Mice in the Py17XL group and the Py17XL + TLR4 group all died, while those in other groups began to die from day 6. There was a slow rise of parasitemia rate in the Py17XL + TLR9 group and the Py17XL + TLR4 + TLR9 group compared with the Py17XL group, with a significant extension of survival to days 11 and 15. Results of flow cytometry showed that the proportions of Th17 cells were 1.2% and 1.44% in the Py17XL + TLR9 group and the Py17XL + TLR4 + TLR9 group on day 5, both sighificantly decreased compared with the Py17XL group (1.9%)(P < 0.05, P < 0.01). ELISA revealed that the levels of IFN-γ and IL-10 on day 5 in the Py17XL + TLR4 + TLR9 group [(232.4 ± 15.5) pg/ml and(1791.2 ± 58.2) pg/ml, respectively] were significantly higher than those in the Py17XL group[(90.7 ± 50.1) pg/ml and (962.6 ± 409.0) pg/ml](P < 0.05, P < 0.01).Conclusions:The differentiation and function of Th17 cells are regulated by DCs during Py17XL infection. Blockade of DCs decreases parasitemia and extends lifetime of mice. Further studies are needed to clarify the exact mechanisms.
目的 探讨柚皮素或柚皮苷对人子宫颈癌Hela细胞增殖及凋亡的影响.方法 柚皮素或柚皮苷(0.250、0.125、0.063 g/L)作用于人子宫颈癌Hela细胞后,MTT比色法检测Hela细胞增殖活性和半数抑制浓度(half maximal inhibitory concentration,IC50);流式细胞术检测柚皮素对Hela细胞凋亡的影响.结果 柚皮素能抑制Hela细胞增殖,且呈剂量和时间依赖性,24、48和72 h IC50值分别为0.24、0.11和0.07 g/L;柚皮苷无抑制Hela细胞增殖的作用.柚皮素组(0.22 g/L、0.11 g/L、0.05 g/L,作用48 h)人子宫颈癌Hela细胞凋亡率高于对照组,差异有统计学意义(P=0.000239681,P=0.00012746,P=5.09328E-05,P<O.01).结论 柚皮中黄酮类提取物具有抑制人子宫颈癌Hela细胞增殖作用,其中柚皮素作用明显,部分作用机制与诱导肿瘤细胞凋亡有关.柚皮苷无抗人子宫颈癌作用.
In this study, students majoring in Clinical Medicine were enrolled to explore the effect of discussion-based teaching method in the teaching of medical parasitology. One hundred and fifty-six students (with an entry year of 2011) in classes 1-3 received the discussion-based teaching while 153 students in classes 4-6 received traditional teaching. The effect of teaching was evaluated in terms of final examination score and questionnaire, and compared between the groups. The final examination score of students receiving the discussion-based teaching (86.1±6.6) was significantly higher than those receiving the traditional teaching(74.2±8.3)(P<0.05). The discussion-based teaching method was graded as “excellent” by 89.1%(136/156)of the students, and was considered to be superior to the traditional teaching by 96.8%(151/156)of the students. The results indicate that the discussion-based teaching method can enhance interactions between participants, change the ways of thinking, and provide inspirations for learning and exploration.
脑卒中是一种常见的神经内科疾病,它包括脑梗死、脑出血等疾病,具有高发病率、高致残率、高死亡率和高复发率的特点.我国民众对其发病机制缺乏了解,缺乏对脑卒中发病危险因素的认识,使得该种疾病在我国的发病率始终保持在较高水平.为了加强民众对脑卒中的认识,自2009年6月开始,卫生部开展了全国范围内的"脑卒中筛查与防治工程",这一重大课题是降低脑卒中发病率、预防脑卒中复发的重大举措,是我国一项重大的国民健康促进工程.医学生是未来医学工作的接班人,提高医学生对脑卒中的认识,具有非常重要的意义.
目的 探讨疟疾感染过程中调节性T细胞(Treg)对DCs免疫功能的抑制作用与机理.方法 建立P.y17XL感染和Tregs消除BALB/c小鼠模型,计数红细胞感染率;感染后第0、3和5d制备脾细胞悬液,FACS检测脾脏中DCs亚群数量变化;DCs表面分子MHC-Ⅱ、CD80和CD86的表达水平及分泌IL-10的DCs数量变化.结果 与正常感染组小鼠相比,Tregs消除组小鼠于感染后第3 d DCs亚群,MHC-Ⅱ,CD80和CD86的表达均明显增加,感染后第5 d DCs亚群数量、MHC-Ⅱ和CD80表面分子的表达均明显减少;然而分泌IL-10的DCs数量于感染后第5d明显增加,是同天感染鼠的3.5倍.另外,我们采用两种anti CD25mAb体内阻断Tregs,效果有着明显的差异.7D4能长期有效的阻断CD25表达,而PC61仅能短期内维持CD25低表达.结论 P.y17XL感染早期,BALB/c小鼠Tregs数量升高与DCs功能受损具有相关性.Tregs能抑制DCs的免疫功能,这一现象可能与Tregs调控DCs的亚群、表型和细胞因子分泌模式等方面有关.
目的 观察灵芝酸对氯化锂-匹鲁卡品致痫大鼠脑皮质GFAP和FGF-2蛋白表达的影响,探讨灵芝酸抗癫痫作用及其作用机制.方法 选取雄性SD大鼠44只,随机分为4组(n=11):正常对照组,癫痫模型组,灵芝酸治疗组,丙戊酸钠治疗组,每组腹腔注射给药后,观察各组大鼠行为学改变,Western-blotting法和免疫荧光法检测各组大鼠皮质区GFAP和FGF-2蛋白表达.结果 与正常对照组相比,癫痫组FGF-2的蛋白表达水平显著下降(P<0.01);GFAP的蛋白表达水平显著升高(P<0.01).与丙戊酸钠组相比,灵芝酸组FGF-2的蛋白表达水平显著升高(P<0.05);GFAP的蛋白表达水平显著降低(P<0.05).结论 灵芝酸对癫痫大鼠脑具有一定的保护作用,其抗癫痫机制与灵芝酸能够调节FGF-2和GFAP蛋白的表达水平有关.
Objective To observe the effects and reasons of thrombin on HIV envelope V3 area crown sequence in different strain by using cell-cell fusion experimental system and expend the range of strain.Methods In order to prepare theEnv-293T cell by transfecting 293T cells,plasmid pSVⅢ-ADA,plasmid pSM-06057c3 and plasmid pSM-03009c34 are selected.And these three kinds of plasmids must carry HIV-1 ADA,06057c3 and 03009c34 env genes.239T cell are transfected by plasmid pSVⅢ-ADA and pcTat,pSM-06057c3 and pcTat pSM-03009c34 and pcTat.The ratio of plamid and pcTat is 2∶ 1.After that there is a cell fusion experiment between the Env-293T cell and Magic5A cell in different Th concentrations and count the fusion cells and observe the effect of cell fusion.Results The treated cell can promote ADA and 06057c3 Env mediated fusion,whose V3 crown sequences are GPGRAF.Th can not promote 03009c34 Env mediated fusion,whose V3 crown sequence is GPGQAW strain.Conclusion Th can promote HIV meditated cell fusion,and the reason which might be related with the V3 crown GPGRAF sequences.
目前脑出血治疗缺乏特异、有效手段,其原因主要是脑出血后血肿周围组织损伤机制尚未完全阐明。大量研究证实炎症反应参与脑出血后的病理过程。本研究拟探讨脑出血患者外周血白介素( IL)-17、IL-23的表达及意义。
目的 通过检测呼吸道合胞病毒(RSV)感染宿主HeLa细胞Fas蛋白和FasL蛋白的变化,探讨翻白草油的抗RSV作用. 方法 用RSV感染宿主HeLa细胞,采用Reed Muench法求出RSV对细胞半数感染量(CCID50);将翻白草油与H eLa细胞共孵育,MTT法检测翻白草油对Hela细胞的毒性作用;选取最大无毒浓度的翻白草油和100 CCID50的RSV病毒进行抗病毒试验,在药物4种作用方式下(对RSV病毒的直接灭活作用,在生物合成阶段对RSV病毒的作用,对RSV病毒吸附的作用,药物预处理作用),采用免疫荧光和Western blot方法检测宿主HeLa细胞Fas/FasL蛋白的表达. 结果 RSV培养液对HeLa细胞中的毒力为105.8 CCID50/L;翻白草油对HeLa细胞的最小毒性浓度是0.10mg/L.翻白草油(除药物预处理作用方式外)Fas蛋白和FasL蛋白表达的荧光强度分别为1.851±0.022、1.830±0.044、1.801±0.038和1.832±0.034、1.767±0.022、1.816±0.024,RSV病毒对照组荧光强度分别为2.112±0.048和1.904±0.025,翻白草油(除药物预处理作用方式)组与病毒组比较,宿主HeLa细胞Fas蛋白和FasL蛋白的表达显著降低(P<0.05);翻白草预处理组Fas蛋白和FasL蛋白表达的荧光强度分别为2.098±0.075和1.882±0.055,与RSV病毒对照组比较差异无统计学意义(P>0.05). 结论 翻白草油在直接灭活阶段、病毒复制阶段、病毒吸附阶段的抗RSV作用与调节宿主HeLa细胞Fas/FasL蛋白的表达有关.

目的 探讨翻白草油对呼吸道合胞病毒(RSv)感染宿主HeLa细胞Bcl-2蛋白和Bax蛋白的影响作用.方法 RSV感染宿主HeLa细胞后,选取翻白草油进行干预,采用Western印迹检测Bcl-2蛋白和Bax蛋白的表达.结果 翻白草油(除翻白草油预处理作用方式)组与病毒组比较,宿主HeLa细胞Bax蛋白的表达明显降低(P<0.05),而Bcl-2蛋白的表达明显增加(P<0.05).结论 翻白草油在三种作用方式下(直接灭活、病毒复制、病毒吸附阶段)抗RSV作用可能与调节Bcl-2蛋白和Bax蛋白的表达有关.
