选取45个微卫星位点,对爆发马立克氏病的4只患鸡的淋巴细胞和正常的组织及3号鸡的肿瘤组织,提取核酸,利用45个所选微卫星引物进行PCR扩增,变性聚丙烯酰胺凝胶电泳检测微卫星不稳定性的变化.研究结果表明,在4只患鸡中有39个微卫星位点发生微卫星不稳定性的变化,其中3号鸡发生变化概率最高,证实同个体的淋巴细胞和肿瘤组织同位点发生MSI变化的符合率达到75.6%.
A strain of degrading feather keratin bacterium with high efficiency was isolated from environment by adopting the culture medium taking feather meal as sole carbon and nitrogen source.This strain was identified as Bacillus pumilus by morphology,physiological and biochemical experiments and 16SrDNA gene analysis,and named as the Bacillus pumilus WHK4.The degradation rate of feather keratin reached to 85.76% after 48h fermentation.The results indicated that WHK4 was an appropriate strain for exploring microorganic fermentation system to degrade feather keratin,and had a great potential to be applied in producing high quality protein feed.
2008年9月,安徽省明光市某散养鸡场发生了一种以皮肤肿块、眼睛肿大、鸡冠病变为特征的疾病.笔者对该病例进行病理剖检,制备常规石蜡切片进行病理组织学检查,根据马立克氏病病毒IPC4基因序列设计1对特异性引物,建立PCR检测方法,综合诊断该病为马立克氏病.
近年来,在南京及安徽省周边地区雏鹅频发病毒性肝炎,根据病鹅的临诊症状、病理变化以及流行病学调查、治疗性诊断、实验室检查结果,初步认为是由鸭肝炎病毒1型引起。
[Objective] The basis of clinical medicine was provided through the investigation of the pathogen of main symptoms of the diseases such as pericarditis,hepatitis and peritonitis in one chicken farm in Anhui Province.[Method] The pathogen of these diseases,cultured after isolated from samples collected,was identified based on the morphological observation,physiological and biochemical tests and 16S rRNA sequence analysis and the clinical drug treatment was conducted according the results of its sensitivity test.[Results] The cultured pathogen was matched with the E.coli in the morphological,physiological and biochemical characteristics,16S rRNA sequence.The test result of the drug revealed that the pathogenic bacteria would be highly sensitivity to Jubizhi,amikacin,ADM and others;moderate sensitivity to cefradine and low sensitivity to nitrofurantoin and gentamicin.The disease would be under control in 3 days after the treatment of Jubizhi.[Conclusion] It was effective way of the clinical treatment of control of chicken E.coli to isolate and identify the pathogen,and to test its sensitivity.
