OBJECTIVE:To investigate whether peritumoral electroacupuncture (EA) suppresses tumor growth viaCD8+ T cell infiltration mediated by sympathetic nervous system (SNS) regulation. METHODS:Utilizing a mouse model of triple-negative breast cancer (TNBC), the SNS was activated through chronic restraint stress (CRS); chemical sympathetic denervation was induced by 6-hydroxydopamine (6-OHDA). The anti-tumor and immune regulatory effects of EA were evaluated by multidimensional methods, including in vivobioluminescence imaging, flow cytometry, immunofluorescence, and enzyme-linked immunosorbent assay. RESULTS:Peritumoral EA decreased the volume and weight of tumors within 22 d after the implantation. Intratumoral CD8+ T cells significantly increased after EA. C-C motif chemokine ligand 5, as a chemokine that recruits effector T cells to the tumor microenvironment, also up-regulated. Meanwhile, EA reduced intratumoral sympathetic innervation and norepinephrine level. Subsequently, we found CRS significantly accelerates tumor growth and reduces the density of CD8+ T cell infiltration. However, EA reversed the tumor-promoting effect and immune suppression caused by CRS effectively. Furthermore, chemical sympathetic ablation with 6-OHDA revealed that the anti-tumor effect of EA was dependent on sympathetic integrity, as the ablation impaired both tumor-suppressing effect and CD8+ T cell recruitment mediated by EA. CONCLUSION:EA enhances anti-tumor immunity through SNS emphasizing the effective remodeling of the tumor immune microenvironment in mice with TNBC by neuro-immune interactions.
Background: Angiogenesis significantly contributes to breast cancer progression. While glyoxalase I (GLO1) has been extensively studied in endothelial cells related to cardiovascular and metabolic diseases, its role in breast cancer-associated endothelial cells (BCECs) remains unclear. Moreover, Additionally, a comprehensive analysis of GLO1 inhibitors is lacking. This study aims to systematically evaluate the pharmacological profiles of GLO1 inhibitors, identify those with potential effects on BCECs, and validate these findings experimentally. Materials and methods: Seventeen GLO1 inhibitors were analyzed using SwissADME and pkCSM for their pharmacological profiles. Target identification employed SwissTargetPrediction and SuperPred, and intersected with differentially expressed genes (DEGs) from the GSE80506 dataset to construct a BCECs-relevant target network. This network underwent enrichment analysis, expression correlation analysis, and molecular docking. Experimental validation was conducted using cellular assays and an aortic ring assay in mice. Results: Among 515 targets associated with the 17 GLO1 inhibitors, 36 were linked to BCECs regulation. Enrichment analysis highlighted the cell cycle as a pivotal pathway, with key targets including CCNA2, CCNE1, CDC25A, CDC25B, CDK6, CHEK1, PLK1, and TTK. Molecular docking indicated that Glyoxalase I inhibitor 6 plays a significant role in regulating these targets. Experimental assays demonstrated that this inhibitor arrested cells in the G0/G1 phase by modulating CCNA2 and CCNE1, suppressed BCECs proliferation, migration, and angiogenesis, and promoted cell death. Conclusions: GLO1 inhibitors exhibit significant regulatory effects on BCECs. Notably, Glyoxalase I inhibitor 6 effectively inhibits cell cycle progression, proliferation, migration, and angiogenesis in these cells, suggesting a promising therapeutic approach for targeting tumor-associated endothelial cells in breast cancer.
Background:Acupuncture has been widely used to relieve or cure various diseases. In this study, bibliometrics was used to comprehensively analyze the research focus and development trend of acupuncture regulating neuroplasticity, to provide new insights for future research. Method:In this study, the Science Citation Index-Expanded database in the Web of Science Core Collection was used to retrieve and download the publications about acupuncture regulating neuroplasticity from 2005 to 2024. CiteSpace, VOSviewer, "Bibliometric" software package and Microsoft Excel were used for bibliometric analysis, and the number of publications, countries/regions, institutions, authors, journals, references and keywords were analyzed. Results:A total of 264 publications, involving 905 institutions in 100 countries/regions. Overall, the publishing volume in the past 20 years has shown a steady upward trend. China has become the leading force in this research field, contributing the most high-quality publications. Guangzhou University of Chinese Medicine and Shanghai University of Traditional Chinese Medicine stand out as the most prolific institutions. Beijing University of Chinese Medicine and Harvard University have high academic influence. Among 1,586 authors, CZ Liu, XY Hua and JG Xu were identified as the most prolific authors. Frontiers in Neuroscience is the most published journal. In recent years, the mechanisms of acupuncture have attracted wide attention and become a hot topic. Conclusion:With the increasing research in this field, cooperation between different countries/regions, institutions and authors should be strengthened. AMPK, BDNF, brain-gut axis and brain-immune axis are the research hotspots and frontiers in this field. In the future, with the improvement of technical level, we should deepen the mechanism research, optimize clinical research and promote the standardization and individualization of acupuncture technology, which is expected to further reveal the scientific connotation of acupuncture regulation plasticity.
Emerging research suggested a potential role of immune cells in colorectal cancer (CRC) development. However, the causal relationship between immune phenotypes and CRC remains elusive. Hence, this two-sample Mendelian randomization (MR) study aimed to explore the causal association. In this study, a bidirectional, two-sample MR analysis and multivariate MR was conducted, leveraging public genetic data. Four types of immune phenotypes were employed. A comprehensive sensitivity analysis was carried out to validate the robustness, heterogeneity, and horizontal pleiotropy of the results, with Bonferroni correction applied for accurate interpretation. Objective To identify core targets of hepatocellular carcinoma(HCC)by using bioinformatics and specific algorithms,explore their relationships with immune cells,and investigate the causal relationships between immune cells and HCC through Mendelian randomization.Methods Relevant genes associated with the development of HCC were screened using the GEO and TCGA databases.Immune infiltration analysis was conducted using GSVA and CIBERSORT algorithms.A bidirectional Mendelian randomization analysis was then performed to explore the causal relationships between immune cells and HCC.Results A total of 284 HCC-related genes were identified,with 120 genes recognized within the protein interaction network.Immune infiltration analysis revealed significant correlations between key genes and immune cells.Mendelian randomization results indicated that HLA DR on CD33+HLA DR+CD14dim(OR=1.097,95%CI:1.002-1.201,P=0.045,PBonferroni=0.091)and CD8 on CD28+CD45RA+CD8+T cell(OR=1.123,95%CI:1.027-1.228,P=0.011,PBonferroni=0.022)were the risk factors for HCC.Conversely,HLA DR++monocyte absolute count was identified as a protective factor for HCC(OR=0.812,95%CI:0.702-0.938,P=0.005,PBonferroni=0.139).Conclusion The occurrence and development of liver cancer may be related to CDK1,CCNB1,and CDC20,showing a high degree of correlation with Th2 cells,T helper cells,Th17 cells,and DCs.Mendelian randomization shows that HLA DR on CD33+HLA DR+CD14dim and CD8 on CD28+CD45RA+CD8+T cells are associated with an increased risk of HCC.The risk of hepatocellular carcinoma is associated with a decrease in the level of HLA DR++monocyte absolute count.
Hepatocellular carcinoma (HCC), a common and lethal form of liver cancer, includes mitochondrial dysfunction in its pathogenesis. This study investigated the relationship between mitochondrial function-related genes and HCC progression. We systematically retrieved mitochondrial genetic data from the MitoCarta database and identified differentially expressed genes through Gene Expression Omnibus analysis. Weighted gene co-expression network analysis was subsequently employed to construct co-expression networks and identify key modules associated with HCC progression. We evaluated 113 machine learning algorithms to develop mitochondrial gene-based prognostic models. Gene set enrichment analysis further delineated the pathways and biological processes enriched in the module hub genes, offering mechanistic insights into HCC. Immune infiltration analysis using CIBERSORT highlighted the pivotal roles of M1 and M2 macrophages in HCC. Finally, therapeutic candidates targeting critical genes were explored using computational drug prediction, molecular docking, and molecular dynamic simulations, providing novel strategies for HCC-targeted therapy. Stepwise Logistic Regression with Gradient Boosting Machine was chosen as the optimal model (area under the curve [AUC] = 0.977). Moreover, 15 potential HCC biomarkers were identified, including PSMD4 (AUC = 0.888), TBCE (AUC = 0.879), and CKS1B (AUC = 0.860). Additionally, fluoxetine and paroxetine were predicted as potential HCC drugs and validated through molecular docking and dynamic simulations. This study highlights the prognostic significance of mitochondrial function-related genes in HCC and establishes a framework for developing innovative diagnostic and therapeutic interventions. Future research should prioritize clinical validation of these findings and evaluate the translational potential of the identified drug candidates in HCC.
This study reports two cases of rectal cancer patients who developed low anterior resection Syndrome (LARS) following rectal cancer surgery. Both patients presented with significant bowel dysfunction, including frequent defecation, urgency, fecal incontinence, and incomplete evacuation. Current treatments for LARS are limited by variable responses, high costs, and adherence issues, highlighting the need for practical, safe therapies with minimal side effects. The patients underwent a 6-month electroacupuncture treatment targeting Baliao points. Assessments were conducted using the LARS score, Wexner fecal incontinence score, and the EORTC QLQ-C30 scale. Results indicated a marked reduction in bowel frequency, significant relief of fecal incontinence symptoms, and improvement in overall health status and quality of life. In addition, emotional and cognitive functions were enhanced. These case reports suggest that electroacupuncture may be a valuable adjunctive treatment for managing LARS and improving patient emotional status and quality of life. Further high-quality research is necessary to evaluate the long-term efficacy of this treatment fully.
BackgroundProstate cancer (PCa) is the most common non-cutaneous malignancy in men globally. Sappan lignum, which exists in the heartwood of Caesalpinia sappan L., has antitumor effects; however, its exact mechanism of action remains unclear. This study elucidated the underlying mechanisms of Sappan lignum in PCa through network pharmacology approaches and molecular docking techniques. Moreover, the therapeutic effects of Sappan lignum on PCa were verified through in vitro experiments.MethodsThe constituent ingredients of Sappan lignum were retrieved from the HERB database. Active plant-derived compounds of Sappan lignum were screened based on gastrointestinal absorption and gastric drug properties. Disease targets for PCa were screened using unpaired and paired case datasets from the Gene Expression Omnibus. Intersection targets were used for gene ontology and Kyoto encyclopedia of genes and genomes (KEGG) pathway enrichment analysis. Core targets were identified through topological analysis parameters and their clinical relevance was validated through The Cancer Genome Atlas database. The affinity between the phytochemicals of Sappan lignum and core proteins was verified using the molecular docking technique. Validation experiments confirmed the significant potential of Sappan lignum in treating PCa.ResultsTwenty-one plant-derived compounds of Sappan lignum and 821 differentially expressed genes associated with PCa were collected. Among 32 intersection targets, 8 were screened according to topological parameters. KEGG analysis indicated that the antitumor effects of Sappan lignum on PCa were primarily associated with the p53 pathway. The molecular docking technique demonstrated a strong affinity between 3-deoxysappanchalcone (3-DSC) and core proteins, particularly cyclin B1 (CCNB1). CCNB1 expression correlated with clinicopathological features in patients with PCa. Experimental results revealed that 3-DSC exhibited anti-proliferative, anti-migratory, and pro-apoptotic effects on 22RV1 and DU145 cells while also causing G2/M phase cell cycle arrest, potentially through modulating the p53/p21/CDC2/CCNB1 pathway.ConclusionThis research highlights the promising therapeutic potential of Sappan lignum in treating PCa, with a particular focus on targeting the p53 pathway.
BACKGROUND:Huaier (Trametes robiniophila Murr), a traditional Chinese medicine, is widely used in China as a complementary and alternative therapy to treat hepatocellular carcinoma (HCC). Past studies have shown that Huaier can arrest the cell cycle, promote apoptosis and inhibit the proliferation of cancer cells. However, how it regulates the metabolism of HCC is still unclear. OBJECTIVE:This study explores the metabolic-related function of Huaier in treating HCC with an in-silico approach. METHODS:A network pharmacology and bioinformatics-based approach was employed to investigate the molecular pathogenesis of metabolic reprogramming in HCC with Huaier. The compounds of Huaier were obtained from public databases. Oral bioavailability and drug likeness were screened using the TCMSP platform. The differential gene expressions between HCC and non-tumor tissue were calculated and used to find the overlap from the targets of Huaier. The enrichment analysis of the overlapped targets by Metascape helped filter out the metabolism-related targets of Huaier in treating HCC. Protein-protein interaction (PPI) network construction and topological screening revealed the hub nodes. The prognosis and clinical correlation of these targets were validated from the cancer genome atlas (TCGA) database, and the interactions between the hub nodes and active ingredients were validated by molecular docking. RESULTS:The results showed that Peroxyergosterol, Daucosterol, and Kaempferol were the primary active compounds of Huaier involved in the metabolic reprogramming of HCC. The top 6 metabolic targets included AKR1C3, CYP1A1, CYP3A4, CYP1A2, CYP17A1, and HSD11B1. The decreased expression of CYP3A4 and increased expression of AKR1C3 were related to the poor overall survival of HCC patients. The molecular docking validated that Peroxyergosterol and Kaempferol exhibited the potential to modulate CYP3A4 and AKR1C3 from a computational perspective. CONCLUSION:This study provided a workflow for understanding the mechanism of Huaier in regulating the metabolic reprogramming of HCC.
Background: This protocol aims to facilitate the evaluation of acupuncture in the treatment of low anterior resection syndrome (LARS) on the bowel in rectal cancer patients.Methods: This pragmatic pilot study was designed using a convergent parallel mixed methods design combining a single-arm trial and semi-structured qualitative interview.Results: Sixty patients with LARS will be recruited from out/inpatient departments. For evaluation of efficacy, the single-arm objective performance criteria will be used in the pilot study in which all eligible participants will receive electroacupuncture mainly on Baliao acupoints three times a week for four weeks. The LARS scale, Memorial Sloan Kettering Cancer Centre Bowel Function Index, and anorectal manometry will be used to assess symptoms and pressure changes. The European Quality of Life Five Dimensions Questionnaire and the European Organization for Research and Treatment of Cancer-Quality of Life Questionnaire-Core 30 will be used to evaluate quality of life. Semi-structured interviews will be conducted among twenty participants to understand their experience and feelings. The qualitative and quantitative data will be analyzed and summarized before comparative analysis. Qualitative themes derived from qualitative analysis will be ranked with the variables of quantitative statistics. Finally, we will answer the research question from multiple perspectives by comparing different types of evidence for the same dimension.Conclusion: This mixed method study design will potentially evaluate the feasibility and effects of electroacupuncture for LARS and gain an in-depth understanding of the attitudes, experiences, feelings, and acceptance among patients with LARS.
The pathogenesis of lung metastases from renal carcinoma is the deficiency of both the Lung and the Kidney, blockage of Sanjiao, and failure in eliminating Phlegm and Dampness.Renal carcinoma is originated in Kidney deficiency, in which deficiency of Kidney Yin negatively affects the Kidney Yang, which weakens the Qi transformation function of the Kidney.When the function of Qi, Blood, and body Fluid in the Sanjiao is abnormal, and the Lungs cannot be nourished by Qi, Blood, and body Fluid, resulting in insufficient Lung Qi and Yin over time.Deficiency of both Qi and Yin causes the Lung′s failure in clearing and descending, resulting in the inability of the body Fluid in the Lungs to disperse, and then the Phlegm, Water, and Dampness stay in the Lungs and get accumulated over time.The Kidney governs bones, and the Shaoyang governs the periosteum.Because renal carcinoma is originally caused by the loss of Kidney essence, which fails in nourishing the bones, and the bones that lose their nourishment are easily attacked by pathogenic Qi.At the same time, the power of Shaoyang to support the movement of Qi, Blood, and body Fluid comes from Kidney.If the kidney is deficient, the pivot of Shaoyang will be unfavorable, and Phlegm, Dampness and blood stasis will generate inward and stay in the fascia of the deficient bones, and get accumulated over time.Lung and bone metastasis of renal carcinoma is closely related to "Shaoyang belonging to Kidney and Kidney being connected to Lung so the two Zang are closely connected" and "Shaoyang governing the bone".Therefore, in treatment of renal carcinoma dredging and benefiting Shaoyang while nourishing the Kidney and strengthening the foundation should be paid special attention to.Clinically, it should be based on the whole, pay attention to the local area, by combining dredge method and tonic method, and the following priscriptions should be chosed: modified Liuwei Dihuang Pill combined with Chaihu Dayuan Drink, Haibai Baidong Decoction for lung metastases from renal carcinoma.Modified Guifu Dihuang Pills combined with Chaiqin Decoction and Yanghe Decoction were used for bone metastasis of renal carcinoma.In addition, emotional care and daily diet should also be emphasized.
OBJECTIVE To investigate the effect of the decoction of Fuzheng Jiedu Xiaoji formula (, FJXF) plus chemoembolization (TACE) on primary liver cancer (PLC) in patients, and study the underlying mechanism. METHODS Patients with PLC who met the inclusion criteria were randomized into case group and control group. The case group was treated with FJXF combined with TACE. The control group was treated with TACE alone. The short-term clinical effect was evaluated; liver biochemistry, liver function index and multidrug resistance-associated indicators were detected. RESULTS FJXF combined with TACE in the case group significantly increased the disease control rate than TACE alone in the control group (83.3% 61.1%). There was a reduction in the serum alpha-fetoprotein at 8 weeks after treatment in each group, while no difference between the two groups. The same trend can be observed for transaminase and direct bilirubin in both groups. In the case group, it showed a significant increase for albumin at 8 weeks after treatment, while no change in the control group. Multidrug resistance-associated indicators for multidrug resistance protein 1 and p-glycoprotein were upregulated in the case group but remained stable in the control group. CONCLUSIONS FJXF combined TACE had a better short-term effect than TACE alone in patients with PLC. The potential mechanism was probably associated with alleviated multidrug resistance induced by FJXF. Additionally, FJXF didn't increase the risk of liver damage in the combined therapy.
To analyze the research status of acupuncture and moxibustion for cancer at home and abroad in the past 45 years by using bibliometric and scientific knowledge map methods,and explore the development trends in future. The literature of acupuncture and moxibustion for cancer was retrieved from CNKI and Web of Science (WOS) till December 31, 2020 since the database establishment, and CiteSpace and VOSviewer software were used to perform visual map analysis through cooperation network, keyword co-occurrence, keyword timeline, keyword emergence and other methods. Totally, 1 585 literature in CNKI and 1 564 literature in WOS were included, and the annual publication amount showed a fluctuating upward trend. Cooperation between countries was centered on China and the United States, and there was relatively little cooperation among different institutions. The analysis of keyword and cited literature showed that researches focused on the control of acupuncture and moxibustion therapy on cancer complications and adverse reactions of western medicine. The main research types in WOS were systematic review and randomized controlled trial (RCT), while in CNKI was review, depth studies on mechanism of acupuncture and moxibustion for cancer were rare. The concern about the quality of life of cancer patients may become research emphasis in the field of acupuncture and moxibustion for cancer in future, and the research scope tends to integrative and holistic oncology.
Objective Based on the cancer genome atlas(TCGA) database, we conducted a systematic bioinformatics analysis of abnormal cancerous RNA binding proteins(RBPs) in patients with hepatocellular carcinoma(HCC), with the aim of identifying the prognostic markers and potential therapeutic targets. Methods The cancerous and adjacent liver tissue HCC RNA sequencing data was downloaded from TCGA database, and the functional enrichment analysis and visualization of interaction relationships among them were performed. The univariate and multivariate Cox regression analyses were subsequently applied to identify RBPs that were significantly related to the prognosis and a prognostic model was constructed. The predictive performance of the prognostic model was evaluated by survival analysis and receiver operating characteristic(ROC) curve and was verified in the test cohort. The human protein atlas online database was applied to verify the RBP levels in the prognostic model. Results A total of 82 differentially expressed RBPs were identified, including 55 up-regulated and 27 down-regulated(FDR< 0.05 and |log2 FC|>1); the further functional enrichment and interaction analyses showed that the differentially expressed RBPs were mainly related to regulation of mRNA metabolic process, RNA catabolic, mRNA catabolic process, and macromolecule methylation; five RBP genes, e.g., the LIN28B, SMG5, PPARGC1A, LARP1B, and ANG were identified as prognostic-related genes and were used to construct the prognostic model; the predictive ability of the prognostic model was verified in the test cohort; the ROC curve analysis showed that the prognostic model had good sensitivity and specificity; the independent prognostic analysis showed that the risk score could be an independent prognostic factor for patients with HCC. Conclusion The constructed prognostic prediction model by analyzing the differentially expressed RBPs in cancerous tissues in patients with HCC is reliable, which might be served as a prognostic biomarkers and therapeutic targets.
目的 系统评价艾灸治疗恶性肿瘤化疗后引起的白细胞减少症的疗效.方法 通过全面检索中国知网、万方、维普、Pubmed、Wed of Science等数据库中发表的艾灸治疗化疗引起的白细胞减少症临床随机对照试验文献,用Cochorane系统评价方法,依据纳入、排除标准筛选文献、整理资料,采用Rev Man5.4软件进行质量评价和Meta分析.结果 最终纳入18篇.Meta分析显示艾灸治疗化疗后白细胞减少症较常规升白药物疗效好,差异有统计学意义.结论 艾灸治疗化疗后白细胞减少症的疗效较好,操作方便,值得推广.
Background: Abnormal expression of RNA-binding proteins (RBPs) is closely related to tumorigenesis, progression, and prognosis. This study performed systematic bioinformatic analysis of RBPs abnormally expressed in colon adenocarcinoma (COAD) using the Cancer Genome Atlas (TCGA) database to screen prognostic markers and potential therapeutic targets. Methods: First, the gene expression data from COAD samples were used to screen out differentially expressed RBPs for functional enrichment analysis and to visualize interaction relationships. Second, RBPs that were significantly related to prognosis were screened through univariate and multivariate Cox regression analysis to construct a prognostic model. The prediction performance of the prognostic model was evaluated by survival analysis and receiver operating characteristic (ROC) curve analysis. It addition, it was verified in the test cohort. The Human Protein Atlas (HPA) online database was used to verify the expression levels of RBPs in the prognostic model. Results: The study identified 181 differentially expressed RBPs and analyzed their interaction and functional enrichment, which were mainly related to non-coding RNA processing, ribosome biogenesis, RNA metabolic processes, RNA phosphodiester bond hydrolysis, and alternative mRNA splicing. Five RBPs related to prognosis were used to construct a prognostic model, and its predictive ability was verified by the test cohort. ROC curve analysis showed that the prognostic model had good sensitivity and specificity. Independent prognostic analysis showed that risk scores could be used as independent prognostic factors for COAD. Conclusion: This study constructed a reliable prognostic model by analyzing COAD differentially expressed RBPs, facilitating the screening of COAD prognostic markers and therapeutic targets.
Abstract Background: N6-methyladenosine (m6A) methylation modification can affect the tumorigenesis, progression, and metastasis of breast cancer (BC). Up to now, a prognostic model based on m6A methylation regulators for BC is still lacking. This study aimed to construct an accurate prediction prognosis model by m6A methylation regulators for BC patients.Methods: After processing of The Cancer Genome Atlas (TCGA) datasets, the differential expression and correlation analysis of m6A RNA methylation regulators were applied. Next, tumor samples were clustered into different groups and clinicopathologic features in different clusters were explored. By univariate Cox and Least Absolute Shrinkage and Selection Operator (LASSO) analysis, m6A regulators with prognostic value were identified to develop a prediction model. Furthermore, we constructed and validated a predictive nomogram to predict the prognosis of BC patients.Results: 19 m6A related genes were extracted and 908 BC patients enrolled from TCGA dataset. After univariate Cox and LASSO analysis, 3 m6A RNA methylation regulators (YTHDF3, ZC3H13 and HNRNPC) were selected to establish the prognosis model based on median risk score (RS) in training and validation cohort. With the increasing of RS, the expression levels of YTHDF3 and ZC3H13 were individually elevated, while the HNRNPC expressed decreasingly. By survival analysis and Receiver Operating Characteristic (ROC) curve, we found that the overall survival (OS) of high-risk group was significantly shorter than that of the low-risk group based on Kaplan-Meier (KM) analysis in each cohort. Univariate and multivariate analysis identified the RS, age, and pathological stage are independent prognostic factors. A nomogram was constructed to predict 1- and 3-year OS and the calibration plots validate the performance. The C-index of nomogram reached 0.757 (95% CI:0.7-0.814) in training cohort and 0.749 (95% CI:0.648-0.85) in validation cohort, respectively.Conclusions: We successfully constructed a predictive prognosis model by m6A RNA methylation regulators. These results indicated that the m6A RNA methylation regulators are potential therapeutic targets of BC patients.
肺癌是最常见恶性肿瘤之一,其发病率、死亡率高,且呈上升趋势.咳嗽是其最常见症状之一,也多为首发症状,且肺癌的咳嗽各具特点,临证时黄金昶教授通过抓主证的方法进行辩治,取得较好的临床效果.本文疏理肺癌患者常见的咳嗽类型,总结黄金昶教授治疗肺癌患者咳嗽的临床经验.黄金昶教授指出了肺癌患者临床最常见的九种咳嗽,包括因痒作咳,卧则咳嗽,动则咳剧,饮热呛咳,因痰而咳,剧烈阵咳,咳痰无力,脏结实咳,慢性咳嗽等,并分析相关病因病机,提出相应的治疗方案.黄金昶教授治疗肺癌咳嗽经验独到,疗效显著,值得学习与临床推广.
Objective:To explore the role of cyclophilin A(CyPA) in sensitization of lung adenocarcinoma to radiotherapy using CRISPR/Cas9 technology.Methods:A CyPA knockout human lung adenocarcinoma cell line H1975 was established by CRISPR/Cas9 technology.Groups included a control group(wildtype),CyPA knockout group,traditional Chinese medicine(TCM) extract with Fuzhengzengxiao decoction group,and TCM extract with Fuzhengzengxiao decoction+CyPA knockout group.Each group was exposed to radiation at doses of 0,2,4,6,and 8 Gy.After 24 h,MTT assays were used to determine the survival rate of lung cancer cells and calculate radiosensitivity.The qPCR was used to measure mRNA expression of DDIT3,CDKN1 A,and CDC25 A associated with DNA damage repair.Results:Without irradiation,Fuzhengzengxiao decoction reduced the survival rate of lung adenocarcinoma cells(P <.0001).After irradiation,TCM extract with Fuzhengzengxiao decoction,CyPA knockout,and TCM extract with Fuzhengzengxiao decoction+CyPA knockout groups had reduced survival rates(P <.0001) and radiosensitivity was increased significantly.Expression of DDIT3,CDKN1 A,and CDC25 A was upregulated after knockout of CyPA(P <.0001).Expression of DDIT3 and CDC25 A was increased after irradiation in wildtype cells treated with TCM extract with Fuzhengzengxiao decoction(DDIT3,P <.0001;CDC25 A,P=.0059).The TCM extract with Fuzhengzengxiao decoction+CyPA knockout group also had increased expression of DDIT3 and CDC25 A after irradiation(P <.0001).Conclusion:Fuzhengzengxiao decoction significantly decreases the survival rate of lung cancer cells,and its mechanism may be related to radiosensitization by decreasing expression of CyPA and inducing G1/S cell cycle arrest.
