Methods:Whole blood and serum samples were collected from 86 patients with HBV-ACLF and 42 patients with chronic hepatitis B (CHB). Serum alanine aminotransferase (ALT), total bilirubin (TBil), creatinine (Cr), albumin (ALB) cholinesterase (CHE) and international standardized ratio (INR) were detected by routine method . The CXCL10 mRNA level of peripheral blood mononuclear cells was detected by fluorescence quantitative PCR. The genotype and allele of rs56061981 were detected by direct DNA sequencing. The genotype and allele frequencies between the ACLF and CHB groups were compared. The levels of ALT, TBil, INR, ALB, CHE, model for end-stage liver disease (MELD) and CXCL10 mRNA were compared between the two groups according to whether the T allele was carried.Results:There was no significant difference in sex, age, alcohol history, smoking history, HBV genotype, HBeAg status and HBV-DNA level between CHB group and HBV-ACLF group (P > 0.05). The frequency of CT+ TT genotype and the frequency of T allele at the rs56061981 locus of the CXCL10 gene in group ACLF were significantly higher than that in the CHB group ( χ2=4.83, P=0.03 and χ2=4.95, P=0.03). The difference is significant Genotype CT+ TT is a risk factor for CHB to develop into HBV-ACLF ( OR=2.897, 95% CI: 1.09~7.68). Allele T is a risk factor for CHB to develop into HBV-ACLF ( OR=2.746, 95% CI: 1.10~6.89). The plasma INR and MELD scores of ACLF individuals carrying T alleles were significantly higher than those carrying C alleles (t=2.63, P=0.013 and t=2.7, P=0.011). The difference is significant. Albumin (ALB) and cholinesterase (CHE) was significantly lower than those carrying C alleles (t=2.67, P=0.01 and t=3.545, P=0.001). The difference is significant. But there was no significant difference in serum TBL and ALT between the two groups. Conclusions:The polymorphism of CXCL10 gene rs56061981 is significantly correlated with the susceptibility and severity of ACLF. Allele T may be a susceptible gene of ACLF, and it can also be used as a predictor of the severity of the disease.

p60 is a subunit of katanin involved in microtubule-severing. Previous studies of p60 were primarily focused on microtubule regulation and cell cycle regulation. More recent research has demonstrated that katanin p60 possesses a function in prostate cancer bone metastasis; however, its role in breast cancer bone metastasis remains unclear. In the present study, immunohistochemistry was used to analyze the expression of katanin p60 in primary and bone metastatic breast cancer. The role of up- and downregulated katanin p60 was investigated using cell proliferation, and migration experiments. Overall, katanin p60 was highly expressed in breast cancer bone metastatic tissue compared with primary tumor tissue. In breast cancer cells, overexpression of katanin p60 inhibited cell proliferation, but promoted cell migration, whereas silencing katanin p60 expression promoted cell proliferation but inhibited cell migration. Overall, the present study indicated that katanin p60 serves a role in cell proliferation and migration, and thus may be a novel therapeutic target for prevention of breast cancer metastasis.
Objective To analyze the correlation between clinicopathologic features and pelvic lymph node metastasis in patients with endometrial carcinoma.Methods 100 cases of endometrial cancer were selected as the research objects.Correlation between clinical pathological characteristics and pelvic lymph node metastasis was analyzed.Results There were 19 cases of endometrial cancer patients with lymph node metastasis,the main factors were pathological type,pathological stage,cervical involvement and muscle invasion,vascular invasion and so on (P < 0.05).Conclusion The pathological types of the patients are non endometrial carcinoma,muscular invasion and the degree of vascular invasion,the degree of pathological stage is low and cervical involvement is associated with lymph node metastasis in patients with endometrial cancer.
Polysaccharide content isolated from Liriope Spicata Lour. var. Prolifera Y. T. Ma was determined by the anthrone sulfuric acid method, the three factors including material liquid ratio, extraction temperature and time were studied via orthogonal design and the optimized polysaccharide extraction parameters were acquired as follows:material liquid ratio was 1:30 (mg/mL), extraction temperature was 80℃, extraction time was 120 min. According to the optimized condition, polysaccharide extraction rate reached the highest value 46.03%.
Objective To investigate the correlations of expression of interferon inducible protein 10 ( IP-10) mRNA in peripheral blood mononuclear cells ( PBMCs) with serum levels of HBsAg and HBV DNA in patients with HBV-related acute on chronic liver failure ( HBV-ACLF ) , and to assess its value in predicting disease prognosis .Methods Eighty patients with HBV-ACLF, 60 patients with chronic hepatitis B (CHB), and 25 healthy subjects were enrolled during October 2013 and February 2015.IP-10 mRNA in PBMCs was measured by real time quantitative polymerase chain reaction ( RT-qPCR) , and the difference in IP-10 mRNA expression among three groups was compared by ANOVA .Serum levels of HBsAg and HBV DNA were also measured in patients with HBV-ACLF, and Pearson correlation test was performed to analyze the correlations of IP-10 mRNA with HBsAg and HBV DNA levels .t test was used to analyze the differences in IP-10 mRNA, HBsAg and HBV DNA levels between fatal and surviving cases after 3-month entecavir therapy in HBV-ACLF group.Receiver operating characteristic curves ( ROC) was used to evaluate the prognostic value of IP-10 mRNA, HBsAg and HBV DNA in HBV-ACLF patients .Results IP-10 mRNA level in HBV-ACLF group was 0.998 ±0.186, which was higher than those in CHB patients and healthy controls (0.641 ±0.083 and 0.412 ±0.062, t=3.841 and 16.661, P<0.01).Pearson correlation analysis showed that IP-10 mRNA level was negatively correlated with HBsAg level in HBV-ACLF patients (r=-0.576, P<0.01), but positively correlated with HBV DNA level (r=0.547, P<0.01).After 3-month entecavir therapy , IP-10 mRNA level in surviving cases of HBV-ACLF group was 0.894 ±0.164, which was lower than that in fatal cases ( 1.103 ±0.177, t =-4.328, P <0.01 ); HBsAg level in surviving cases was higher than that in fatal cases (3.303 ±0.565 vs.2.605 ±0.844, t =3.251, P<0.01).The area under ROC of IP-10 mRNA in evaluating prognosis of HBV-ACLF was 0.820, which was higher than those of HBsAg (0.663) and HBV DNA (0.570).Conclusions IP-10 mRNA in patients with HBV-ACLF is over-expressed and is correlated with HBsAg and HBV DNA levels .It may be used for predicting the prognosis of patients with HBV-ACLF.
目的:探讨丹参川芎嗪注射液对慢性阻塞性肺疾病急性加重合并呼吸衰竭患者肺功能及炎症因子的影响。方法选择2011年6月至2014年12月就诊的老年慢性阻塞性肺疾病急性加重合并呼吸衰竭患者116例作为研究对象,随机分成两组,各58例。对照组:给予有创无创序贯机械通气和营养支持、祛痰止咳、扩张支气管、抗生素等基础治疗。观察组患者在接受给予有创无创序贯机械通气和营养支持、祛痰止咳、扩张支气管、抗生素等基础治疗的基础上,加用丹参川芎嗪注射液治疗。结果观察组总有效率96.55%高于对照组总有效率81.04%,差异具有统计学意义( P<0.01)。治疗后,观察组血清 IL-6(16.3±4.6) pg/ml、TNF-α(55.3±12.4) pg/ml 及 CRP (7.3±2.0) mg/L 都低于对照组血清 IL-6(25.1±7.2)pg/ml、TNF-α(79.6±20.5)pg/ml及CRP(22.5±4.7)mg/L,差异具有统计学意义(P<0.01)。治疗后,观察组患者的PaCO2(42.5±5.6)mmHg低于对照组患者的PaCO2(52.8±7.7)mmHg,且观察组患者 PaO2(86.7±22.5)mmHg 高于对照组患者 PaO2(75.1±16.0)mmHg,差异具有统计学意义(P<0.01)。治疗后,观察组患者的PEF(4.91±0.87)L/s、FEV1(2.81±0.60)L、FVC(3.12±0.56)L高于对照组患者的 PEF(3.99±0.64)L/s、FEV1(2.11±0.52)L、FVC(2.15±0.39)L,差异具有统计学意义(P<0.01)。结论丹参川芎嗪注射液治疗老年慢性阻塞性肺疾病急性加重合并呼吸衰竭,能够减轻患者的炎症反应,改善患者的血气指标,促进患者肺功能的恢复。
Objective To investigate the IgG expression in laryngeal squamous cell carcinoma (LSCC),compare it with IgG of B lymphocytic origin,and explore its possible clinical significance.Methods IgG expression in LSCC cells was detected with immunohistochemistry and in situ hybridization.Heavy chains and light chains of IgG were detected with laser guided microdissection coupled with RT-PCR.VDJ rearrangement of IgG heavy chain variable region 1 was assessed with sequencing and BLAST.The correlation between IgG expression and the pathological param-eters was assessed by statistics.Results Igγ,Igκand Igλwere detected in the same cell of LSCC,characteristic of peripheral distribution at cancer nests.IGHG1 mRNA expressions were co-localized with Igγin the cancer cells.The mRNAs of IGHG1,Igκand Igλwere detected in LSCC cells by RT-PCR.VDJ rearrangement occurred when the heavy chains were synthesized,but it presented a monotonous pattern and a small quantity of point mutations,the ratios of which were 2.03%-3.67%.IgG expressions were much stronger in the cases of low differentiation,Ⅲ-Ⅳ stages or lymph node metastasis.Conclusion IgG is expressed in LSCC cells,which has unique VDJ rearrangement different from IgG of B lymphocytic origin.IgG derived from LSCC cells is significantly associated with stages,grades and lymph node metastasis.
Objective To analyse the pathological diagnosis,differential diagnosis and prognosis of the calcifying epithelial odontogenic tumour( CEOT). Methods The conventional HE staining, special staining and immunohistochemistry were used to analyse the clinicopathological features of 3 cases of the calcifying epithelial odontogenic tumour,and follow-up had been done. Results Two cases of CEOT occured in mandible,and 1 case in maxilla. Microscopically,the tumor consisted of epithelioid islet or sheets and fibrous stroma. In the center or outer periphery of the tumor mass,the substance with eosinophilic staining and uniform transparence was observed,and Congo red staining showed that the transparent substance was amyloid-like material,which was calcified in the manner of concentric circle. Immunohistochemistry showed that epithelial cells expressed P63 and HCK. Six years after surgery,the tumor in one patient was recurred,one patient without recurrence,and one patient was free of tumor. Conclusion Odontogenic calcifying epithelioma is a rare odontogenic tumor,wich has unique histopathological characteristics,with locally aggressive growth. It needs to be differentiated from the primary or metastatic bone squamous cell carcinoma,salivary gland malignant tumors including mucoepidermoid carcinoma and acinar cell carcinoma,and renal cell metastasis carcinoma.

Objective:To study the clinical and pathological features of adenomyoepithelioma of breast.Methods:Three cases of breast adenomyoepithelioma were studied by microscopy and immunohistochemistry,with review of related literature.Result:These cases were all female aging from 26 to 49 years,with an average ages of 30 years and all were benign.The tumor had distinct components of adenoepithelial and myoepithelial cells by microscopy,with the main structures derived of spindle and transparent myoepithelioma cells,which lead to hyperplasia of myoepithelioma.Adenoepithelial cells displayed CK18、EMA positive,while myoepithelial cells displayed SMA、S-100 and p63 positive.Conclusions:Adenomyoepithelioma of breast is a rare distinct benign entity,which is composed of hyperplasia of adenoepithelial and myoepithelial cells.
目的:探讨子宫腺瘤样瘤的发生、临床病理特点、免疫组化表达特征及鉴别诊断。方法:对20例子宫腺瘤样瘤进行临床病理、免疫组织化学观察。结果:20例子宫腺瘤样瘤占同期子宫切除标本的1.49%,肿瘤多位于子宫浆膜下、近浆膜下肌壁及子宫角;镜下见肿瘤由大小不等、形态不一的腺样及腔隙样结构组成,伴有间质平滑肌增生。免疫表型:AE1/AE3、vimentin和calretinin(+),EMA、CEA和CD31(-)。结论:子宫腺瘤样瘤是一种良性肿瘤,支持间皮起源,临床及病理检查均易误诊和漏诊。免疫组化检查可作为诊断及鉴别诊断的重要依据。
Objective To establish a real-time PCR based method for detection of survivin gene samples methylation with dye SYBR Green Ⅰ.Methods DNA samples from 25 pairs of gastric cancer tissue and matched normal gastric tissues were digested with mCpG-sensitive Hpa Ⅱand Msp Ⅰ,and detected by SYBR Green Ⅰ real-time PCR for survivin exon 1 specific primers. The concentration of digested DNA was monitored by real-time quantitative PCR with survivin intron 2 specific primers. A normal genomic DNA which served as a standard was diluted serially 10-fold to determine the sensitivity of real-time PCR. The products were further verified by agarose gel electrophoresis analysis.Results After HpaⅡ enzyme digestion,a peak of PCR product derived from methylated targeted gene was found in the melting curve at Tm (91.5±0.5) ℃,which was verified as 338 bp strap. The control gene (survivin intron 2) characteristic of Tm (79.5±0.5) ℃ was amplified in all digested samples,indicating no non-specific degradation occurred in the genome DNA. The sensitivity of this method was 100 copies/μL. The new established PCR system was applied to detecting 25 cases of lung cancer tissue samples. As a result,the frequency of survivin exon 1 demethylation was 96%.Conclusion As a reliable new method for detection of survivin exon 1 methylation,SYBR Green Ⅰ real-time fluorescent PCR assay is rapid,accurate,sensitive,timing and simple.
目的:探讨基质金属蛋白酶MMP9、细胞粘附因子CD44v6在皮肤鳞状细胞痛(Squamous cell carcinoma,SCC)和基底细胞癌(Basal cell carcinoma,BCC)中表达的意义及相关性.方法:采用免疫组织化学法对82例石蜡包埋组织标本进行检测和统计分析.结果:①MMP9在皮肤SCC中阳性率(88.1%)显著高于皮肤BOC组(48%)和正常皮肤组(13.3%),CD44v6在正常皮肤中的阳性率(100%)显著高于皮肤SCC组(66.7%)和皮肤BCC组(20%);②MMP9的高表达率与皮肤SCC的分化程度呈负相关,CD44v6的高表达率与皮肤SCC的分化程度呈正相关;③MMP9和CD44v6在皮肤SCC和BCC中表达无明显相关.结论:CD44v6和MMP9与皮肤BCC和SOC的发生、分化密切相关.可作为研究和判断皮肤SCC和BCC生物学行为和预后独立的重要指标.
我院近期收治发生于脑膜的间叶性软骨肉瘤1例,结合有关文献对其临床病理特征分析如下. 1 病历摘要 男,29岁.因头痛20 d,双眼视物模糊半月收入我院神经外科.体检一般情况良好,颅神经检查正常,视力左眼1.0、右眼1.2,肌张力正常,无共济失调,病理反射未引出.血液常规检查未见异常.
Objective To investigate the expression of survivin variants and their relationship to each other as well as to clinic pathological factors in colorectal cancer.Methods survivin variant specific RT-PCR was developed to analysis their expression in 67 paired cancer and para-cancer tissues,and the expression of the wildtype survivin at the protein level was further verified by immunohistochemistry.Results All the cases tested expressed survivin mRNA,and immunohistochemistry obtained similar finding.The nonantiapoptostic survivin-2B was dominantly expressed in para-cancer tissues,whereas the antiapoptostic survivin-ΔEX3 was more frequently detected in cancer tissues.The expression of survivin-ΔEX3 mRNA was significantly associated with tumor metastasis.Conclusion The balance between antiapoptostic survivin iso-forms and nonantiapoptostic ones may play an important role in tumor genesis and tumor progression.
目的:探讨血管内皮生长因子(VEGF-C)及血管内皮生长因子受体-3(VEGFR-3)在少突胶质细胞瘤中的表达.方法:检测45例少突胶质细胞瘤的VEGF-C和VEGFR-3的表达.结果:VEGF-C和VEGFR-3阳性表达率分别为51%和41%.两者表达一致的符合率为71%.VEGF-C和VEGFR-3与少突胶质细胞瘤病理分级呈显著正相关.结论:少突胶质细胞瘤中有VEGF-C和VEGFR-3表达.VEGF-C和VEGFR-3的表达与少突胶质细胞瘤的病理分级相关.
对我院2006-10/2007-10超声诊断甲状腺结节性疾病57例分析如下. 1 临床资料 1.1 一般资料 本组男17例,女40例,平均年龄45岁.本组中共有75个结节,其中28例有多个结节,结节内有囊性变26个.术前患者均查T3(三碘甲状腺原氨酸)、T4(甲状腺素)及TSH(促甲状腺激素).
The purpose of this study was to quantify hepatitis B virus DNA by direct real-time PCR from serum without the need for DNA extraction. Crossing point (Cp) values were determined automatically using the second derivative maximum mode. Since serum samples from patients are inevitably haemolysed, lipaemic or icteric, the interference of endogenous substances from the serum in real-time PCR was evaluated. The result showed that, although serum protein quenched the intensity of fluorescence, the Cp value adopted to calculate the quantity of DNA copies remained unchanged. Importantly, real-time PCR from serum with or without DNA extraction reached a high level of concordance. This direct serum PCR method without the DNA extraction and gel electrophoresis allows for substantial labour and cost savings. In addition, it is also suitable for rapid DNA quantification during clinical diagnosis.
