以裸鼠为载体建立卵巢癌模型至今已有三十余年的发展,随着裸鼠实验技术的日臻完善,裸鼠模型已广泛应用于卵巢癌各种基础和临床研究.裸鼠移植瘤模型因其移植人瘤成功后保存原肿瘤特性,与原瘤组织对抗癌药物具有同样的敏感性,在探索肿瘤发生发展规律、开发新的治疗方法和药物等方面得到充分应用.卵巢癌裸鼠移植瘤模型按肿瘤来源可分为人卵巢癌切除标本实体瘤和人卵巢癌细胞株;按接种后所在部位分为皮下移植裸鼠模型,腹腔种植裸鼠模型,裸鼠体内原位移植模型[1].本文将对三种裸鼠移植瘤模型生物学特征应用和近年来模型检测可视化手段进行综述.

Objective:To explore the invade and migration of human ovarian carcinoma skov3 cell under by siRNA interference on the expression of EZH2 mRNA.Methods:The nude mouse ovarian cancer model was developed by placing human ovarian cancer cell SKOV3 fragments into the peritoneal cavity.Thirty healthy female nude mice were randomly and equally divided into 3 groups(n=10 each) :siRNA group(EZH2-siRNA group),negative control group(NC-siRNA group) and blank control group.Each animal was injected intraperitoneally,daily for 5 days Twenty four hours after last intraperitoneal injection,animals were sacrificed,and tumor tissue were collected and homogenized.Real time PCR and Western blot were used to detect the expression of EZH2 mRNA and the protein.Results:The expression levels of EZH2 mRNA and the protein were down regulated in siRNA group as compared with those in negative control group and blank control group(P0.05).Conclusion:EZH2 low expression can restrain the invade and migration of human ovarian carcinoma skov3 cell,suggest that EZH2 gene expression decreasing may be the new method for ovarian cancer gene therapy.
Objective:To explore the proliferation of human ovarian carcinoma SKOV3 cells under inhibition of zeste homolog 2 by lentivirus-based EZH2 RNAi.Methods:We designed lentivirus-based RNA interference(RNAi),synthesized and transfected SKOV3 cells to inhibit EZH2 expression.Cell proliferations and growth were evaluated with Cell Counting Kit 8(cck-8).Expressions of EZH2,PCNA mRNA and protein level were determined using RT-PCR and Western bolt respectively.Results:Comparing with blank group and empty transfection group,EZH2-shRNA transfection could suppress SKOV3 cells proliferation and growth(P<0.05),and downregulate EZH2,PCNA mRNA expression and protein levels(P<0.05,P<0.05) further.Conclusion:The lentivirus-EZH2 RNAi can restrain proliferation of SKOV3 cells,so silencing EZH2 may be a new way of gene therapy.
