Objective:To study the effect of β-amyloid(Aβ) on calcium homeostasis and endoplasmic reticulum calcium storage of hippocampal neurons in rats.Methods:A total of 60 adult male SD rats were randomly divided into six groups by body mass matching method, with 10 rats in each group.Three groups were injected Aβ 25-35 into the hippocampus(2 μL per side), and divided into low dose group(2.5 μg/μL), medium dose group(5.0 μg/μL) and high dose group(7.5 μg/μL) respectively, and the other 3 groups were set up as the normal saline group(2 μL 0.9% sodium chloride solution), sham-operated group(rats craniotomy without injection) and normal control group(normal feeding without any treatment). The rats were fed until 14 days after operation, and the behavior and state of the rats were observed and recorded, as well as the body weight and total food intake ratio.And the rat hippocampal cells endoplasmic reticulum pathological change were observed by using the electron microscope and light microscope, meanwhile the concentration of intracellular free Ca 2+ ions was detected by the laser scanning confocal microscope and the expression level of PS, SERCA and RyR mRNA and protein by real-time PCR and Western blot methods respectively.The experimental results were analyzed using SPSS 25.0 software for statistical analysis. Repeated measurement ANOVA and one-way ANOVA were used for multi-group comparison, and Dunnett test and Tukey test were used for further pairwise comparison. Results:(1) The body weight of rats in each group was analyzed by repeated measurement ANOVA, and the difference of time effect was statistically significant( F=153.15, P<0.001), but there was no significant difference between the intergroup effects and interaction effects( F=1.547, P=0.374; F=1.598, P=0.113). The body weight of high, medium and low dose Aβ 25-35 groups at 7, 14 days after injection had no significant difference compared with the control group(all P>0.05). The food utilization rates of the high, medium and low doses of Aβ 25-35 groups were(22.9±4.0)%, (23.0±4.2)% and(22.6±3.2)%, respectively, and there was no significant difference compared with the control group((23.7±5.0)%, P>0.05). Within 14 days after injection, listlessness and lethargy were observed in rats in the high dose Aβ 25-35 group.(2) Pathological observation results showed that the endoplasmic reticulum of rat hippocampal cells in the high dose and medium dose groups of Aβ 25-35 was expanded and swelled, and the mitochondria were swollen and deformed.(3) 14 days after Aβ 25-35 injection, the fluorescence intensity of free calcium in hippocampus of rats in high, medium and low dose groups were(820.43±6.89), (720.12±4.30) and (680.50±4.32), respectively, which were all higher than that in the control group(592.17±3.97)(all P<0.001). (4) RT-PCR and Western blot results showed that compared with the control group, high dose and medium dose Aβ 25-35 injection could up-regulate the expression of PS and SERCA mRNA and protein in hippocampal cells(all P<0.05), while down-regulate the expression of RyR mRNA and protein in hippocampal cells(all P<0.05). Conclusion:The deposition of Aβ 25-35 in hippocampal tissues can disrupt the homeostasis of calcium ions in hippocampal tissues, and then cause the increase of free calcium and its related proteins, thus playing the neurotoxic role.
Objective To analyzethe incidence and treatment of the tuberculosis patients in prison from 2012 to 2015;to understand theepidemic of tuberculosis in prisons and to provide the basis for the prevention of tuberculosis.Methods In 2012-2015,patients withtuberculosis were diagnosed through the prison medical examination or medical screeningaccording to the tuberculosis diagnostic criteria,based on sputum culture and chest X-ray.The basic information of the population with tuberculosis was surveyed.Patients were received chemotherapy and their prognosis was evaluated.Results In from 2012 to 2015,there were 98 confirmed patients with tuberculosis in prisons,which accounted for 1.69% of the prison population.Among these patients,31.6% (31/98) were smear-positivepulmonary tuberculosis;60.2% (59/98) were smear-negative pulmonary tuberculosis;8.20 % (8/98) were tuberculous pleurisy.From 2012 to2015,theyearlyincidence of tuberculosis in prisons was2.46%,1.83%,1.36%,and 1.19%,respectively.Among these patients,49.0% (48/98) were diagnosed by the entry examination;16.3% (16/98) were diagnosed by medical screening;34.7% (34/98) were diagnosed by clinical visit of the prisoners.The cure rate of tuberculosis in prisons were 85.7% from2012 to2015.The yearly cure rate was 81.3% (26/32),85.7% (24/28),86.9% (20/24),and 93.3% (14/15),respectively from 2012 to 2015.Conclusion The incidence of tuberculosis in prisons was declining;the cure rate of tuberculosis showed an increasing trend from 2012 to2015.These results indicate that the prevention and managementfor tuberculosisin the prison was effective.
To study the effects of β-amyloid (Aβ25-35) on calcium homeostasis related genes such as Sorcin and RyR2mRNA in primary cultured rats hippocampal neurons.Methods After 8 days of primary cultured,hippocampal neurons were exposure with different doses (1 μmol/L,10 μmol/L,and 20 μmol/L,respectively) of Aβ25-35,and 0 μmol/L dose as control group.After cultured 24h and 48h,observed hippocampal neuronal morphology by transmission electron microscope,detected the Sorcin and RyR2 gene relative expressions of the cultured hippocampal neurons by real-time PCR.Results With the increase of Aβ25-35 dose and exposure time,morphological changes of primary cultured hippocampal neurons were observed,showing the percentage of apoptosis neurons increased in the Aβ25-35 exposure groups.There were more apoptosis neurons in the Aβ20μmol/L group than other groups.The results of Real-time PCR showed Aβraised the expression of Sorcin mRNA and Ryr2 mRNA.The Sorcin mRNA relative expression were (1.42 ±0.03)and (1.63 ±0.02)when cultured 24h and 48h respectively in 10μmol/L Aβ2s-35 group;the Sorcin mRNA relative expression were (2.11 ± 0.05) and (2.23 ± 0.05) when cultured 24h and 48h respectively in 20μmol/L Aβ25-35 group;these results had statistically difference (P < 0.05)compared with control group.The RyR2 mRNA relative expression were (1.64 ± 0.03) and (1.95 ± 0.03) when cultured 24h and 48h respectively in 20μmol/L Aβ25-35 group;These results also showed statistically difference compared with control group.Conclusion Aβ25-35 exhibits a neurotoxicity effect by inducing the apoptosis in primary cultured hippocampal neurons and inhibiting the growth of hippocampal neurons.It can induce AD by regulating the expression of calcium related proteins such as Sorcin and RyR.
Objective To study the effects of β-amyloid (Aβ25-35) on calcium homeostasis and mitochondrial permeability transition pore (MPTP) in primary cultured rat hippocampal neurons,and explore the mechanism of neurotoxicity of Aβ25-35.Methods After 8 days of primary culture,hippocampal neurons were exposure with different doses(1 μmol/L,10 μmol/L,and 20 μ mol/L,respectively) of Aβ25-35,and 0 μmol/L dose as control group.After cultured 24 h and 48 h,observed hippocampal neuronal morphology,intracellular free calcium concentration,activity of Ca2+-ATPase,mitochondrial membrane potential,and MPTP protein expression by Western Blotting method.Results With the increase of Aβ25-35 dose and exposure time,morphological changes of hippocampal neurons in primary culture were observed,showing a diffuse swelling,deformation,vacuoles,and cell edge fuzzy;The concentration of free calciumions of Aβ25-3520 μmol/L group were (31.04 ± 4.16) and (32.80 ± 0.43) compared the control group with the (20.95 ± 4.04) and (22.23 ± 0.49) 24 h and 48 h after the exposure of Aβ25-35,the concentration showed a gradual upward trend (P < 0.05).The activity of Ca2 +-ATPase Aβ25-3520 μmol/L group were (2.14 ±0.01) and (1.10 ± 0.05) U/mgprol,compared the control group with the (5.17 ± 0.08) and (4.57 ± 0.06) (P < 0.05) 24 h and 48 h after the exposure of Aβ25-35.The membrane potential of mitochondrial decreased significantly in the ap groups compared with the control group.(P < 0.05),The membrane potential of mitochondrial Aβ25-35 20 μmol/L group were (20.34 ± 7.05) and (19.05 ± 6.15),compared the control group with the (38.47 ±0.72) and (40.07 ± 1.26) (P <0.05) 24 h and 48h after the exposure of Aβ25-35.With the increase of exposure dose and time,the activity of Ca2 +-ATPase and mitochondrial,and there was a significant dose-response relationship.The results of WB of MPTP showed that the protein expression was increased with the increase of dose.Conclusion Aβ25-35 exhibits a neurotoxicity effect by disturbing the calcium homeostasis,and affecting mitochondrial function by means of activating of MPTP channels.
Objective:To analyze microelement content in the well water along Song-hua-jiang river.Methods: The 458 water samples were collected from 13 sampling points in Songhuajiang river basin,the content of microelements was measured by the atomic absorption spectrophotometer.Results: The zinc content of well water was low.There were only 2 samples beyond state hygienic standard,while 267 samples(58.08%) were not undetected.The manganese content of well water was high.There were 158 samples beyond state hygienic standard,the over-limit rate was 34.06%.The cadmium content of well water was below state hygienic standard.Conclusion: The zinc content of well water along Song-hua-jiang river are low.The manganese content of well water are beyond state hygienic standard.The cadmium content of water well are below state hygienic standard.
监狱医院对于保障服刑人员的生命健康权、促进其改造、稳定监管改造秩序以及履行监狱职能起到了积极的促进作用。相对于国家医疗保障体制的不断变革、健全、完善,监狱系统的医疗保障体制发展则显得相对滞后;监狱医疗的技术水平和保障能力虽有较大提高,但与各级领导和广大人民群众对监狱医疗的期望值还有一定的距离。
