Stephania tetrandra is a medicinal plant with ecological, germplasm, and economic value whose wild resources are increasingly constrained by overexploitation and climate change. To support conservation planning and sustainable cultivation, we quantified current and future potential habitat suitability across China using an ensemble species distribution modeling (SDM) framework and translated the outputs into climate-based priority areas for protection, germplasm safeguarding, monitoring, and phased cultivation trials. Occurrence records were compiled from multiple sources and preprocessed via cleaning and spatial thinning to reduce sampling bias. Current predictors were derived from WorldClim (1970-2000) and complemented with topographic and edaphic variables; future climates were represented by CMIP6 projections for the 2050s, 2070s, and 2090s under SSP1-2.6, SSP2-4.5, and SSP5-8.5. Multiple algorithms were trained in a consistent cross-validation workflow and filtered using AUC (ROC) and TSS before generating a weighted ensemble (EMwmean). Current projections indicate a well-defined suitability core in the humid subtropical monsoon region south of the Yangtze River. Nationally, high-, moderate-, and low-suitability areas were estimated at 51.90 & times; 104 km2, 22.95 & times; 104 km2, and 31.05 & times; 104 km2, respectively. Future impacts are dominated by suitability-grade reallocation rather than a collapse of total suitable extent. Under SSP5-8.5 in the 2090s, high suitability declines to 13.32 & times; 104 km2 (approximate to 74% reduction), accompanied by contraction of stable habitat (48.95 & times; 104 km2) and expansion of loss areas (33.64 & times; 104 km2), while gains remain limited (4.30 & times; 104 km2). Extrapolation diagnostics (Multivariate Environmental Similarity Surface, MESS; Most Dissimilar Variable, MoD) highlight elevated uncertainty in northwestern arid/high-elevation and strongly seasonal transition zones. Environmental-space niche overlap decreases moderately (Schoener's D = 0.51-0.67), indicating niche displacement and a narrowing suitability window. These results represent potential climatic habitat suitability rather than guaranteed future occupancy. They support prioritizing in situ protection and germplasm safeguarding in areas that are currently highly suitable and remain comparatively stable under future climates, while treating marginal gain zones as candidates for monitoring and carefully phased cultivation or introduction trials.
To explore the distinctness, uniformity, and stability(DUS) testing technology for Angelica dahurica and develop its DUS testing guidelines, this study conducted systematic research on the morphological and growth characteristics of A. dahurica throughout its whole growth period. The research was based on 428 A. dahurica lines from 48 sampling sites across China, with unified standardized field planting, regular and fixed-point observation, classified statistics of phenotypic characteristics, and comprehensive data analysis. Ultimately, 49 test characteristics were identified, including 36 basic characteristics and 13 optional characteristics. Classified by attribute, they consisted of 4 qualitative characteristics, 3 pseudo-qualitative characteristics, and 42 quantitative characteristics. Classified by organ and growth stage, the characteristics covered 2 cotyledon characteristics, 19 leaf characteristics(including basal leaves and cauline leaves), 2 plant characteristics, 3 saccate leaf sheath characteristics, 5 fruit characteristics, 9 root characteristics, 5 stem characteristics, 3 flower characteristics, and 1 growth period characteristic. Through the evaluation of characteristic discriminability and stability, five grouping characteristics were screened out, namely "basal leaf: anthocyanin coloration on the back of the leaf sheath" "basal leaf: anthocyanin coloration at the attachment site of the petiolule" "flowering period" "plant: height" "main root: arrangement pattern of lenticel-like protuberances". These can serve as important bases for the preliminary screening and classification of A. dahurica varieties. Meanwhile, 20 standard varieties with typical phenotypes were identified to provide a unified reference for characteristic observation. In addition, the guidelines also specify the scope of application, requirements for propagation materials, growth stages, observation periods, observation methods, DUS judgment criteria, and other content. This study fills the gap in the field of DUS testing technology for A. dahurica, and provides a scientific basis and technical support for DUS testing, resource identification and description, variety breeding of A. dahurica varieties, as well as management and protection of new A. dahurica arieties.
The sustained monoculture and irregular planting practices rendered the cultivated Coptis chinensis more prone to various diseases compared to its wild counterparts. Rewilding the rhizomicrobiome of cultivated plants has emerged as a promising strategy to promote plant growth, but ancestral microbiota suitable for C. chinensis remain largely uncharted. The amplicon data analyses revealed that habitat transition strongly influenced the rhizosphere microbial communities. The rhizomicrobiomes of wild C. chinensis encompassed a more diverse array of ecological groups and exhibited a greater functional diversity compared to their cultivated counterparts. A higher proportion of beneficial fungi was observed in the rhizosphere of wild C. chinensis, while the cultivated plants had a higher population of pathogenic fungi. Furthermore, a well-documented plant-growth-promoting rhizobacterium genus, Paraburkholderia, was found to play an essential role in the resistance of the wild C. chinensis to potential disease caused by Ilyonectria. Two strains of Paraburkholderia (Paraburkholderia nemoris and Paraburkholderia phytofirmans) were isolated, and in vitro experiments confirmed that these isolates possess various growth-promoting properties and antagonistic activities against known pathogens for C. chinensis root rot. Both of the Paraburkholderia isolates could markedly promote the plant immune response and enhance the overall health of the cultivated C. chinensis. By a comprehensive comparison of the rhizosphere microbiome between wild and cultivated C. chinensis, the promising bacterial genus Paraburkholderia was identified as a beneficial microbe significantly promoting the growth of C. chinensis, providing pivotal insights for future endeavors aimed at engineering the rhizosphere microbiome of C. chinensis, as well as other medicinal herbs.
Geographical origins and varietal characteristics can significantly affect the quality of Citri Reticulatae Pericarpium (Chenpi), making rapid and accurate identification essential for consumer protection. To overcome the inefficiency and high cost of conventional detection methods, this study proposed a nondestructive approach that integrates hyperspectral imaging (HSI) with deep learning to classify Chenpi varieties and their geographical origins. Hyperspectral data were collected from 15 Chenpi varieties (citrus peel) across 13 major production regions in China using three dataset configurations: exocarp-facing-upward (Z), endocarp-facing-upward (F), and a fused dataset combining random orientations (ZF). Convolutional neural networks (CNNs) were developed and compared with conventional machine learning models, including partial least-squares discriminant analysis (PLS-DA), support vector machines (SVMs), and a multilayer perceptron (MLP). The CNN model achieved 96.39% accuracy for varietal classification with the ZF dataset, while the Z-PLSDA model optimized with second derivative (D2) preprocessing and competitive adaptive reweighted sampling (CARS) feature selection attained 91.67% accuracy in geographical origin discrimination. Feature wavelength selection strategies, such as CARS, simplified the model complexity while maintaining a classification performance comparable to that of the full-spectrum models. These findings demonstrated that HSI combined with deep learning could provide a rapid, nondestructive, and cost-effective solution for Chenpi quality assessment and origin traceability.
Xinhui Chenpi, the dried mature fruit peel of Citrus reticulata ‘Chachi’ (Cha-Zhi-Gan, CZG), is a geo-authentic materia medica widely used in traditional Chinese medicine. Despite its distinctive aroma, the influence of genetic background on volatile metabolite accumulation has not been fully understood. In this study, 11 volatile metabolites, including methyl 2-(methylamino)benzoate and γ-terpinene, were found at significantly higher concentrations in Xinhui Chenpi than in other Chenpi resources. A complete telomere-to-telomere (T2T) genome assembly of CZG (326.6 Mb) was generated, and genome sequences of 33 mandarin accessions were compiled to elucidate the origin and domestication of CZG. Comparative metabolomic and transcriptomic analyses across five developmental stages of C. reticulata ‘Unshiu’ (Guo-Qing-Yi-Hao, GQYH) peels revealed significantly elevated levels of 20 volatile compounds and differential expression of 2 525 genes in CZG peels. Notably, higher levels of methyl 2-(methylamino)benzoate were associated with the upregulation of CreBSMT1, CreCHD1 and CrePAL1, whereas increased γ-terpinene levels correlated with the upregulation of CreGPPS2, CreMTSL61, and CreMTSL3. Integration of metabolomic and transcriptomic data enabled the construction of a regulatory network for volatile metabolite biosynthesis in CZG. A NAC transcription factor, CreNAC8, was identified as a key regulator of both methyl 2-(methylamino)benzoate and γ-terpinene accumulation. Transient overexpression and virus-induced gene silencing (VIGS) assays confirmed that CreNAC8 regulated the accumulation of γ-terpinene and methyl 2-(methylamino)benzoate in CZG fruit peels through the regulation of CreMTSL61 and CreCHD1, respectively. These findings offer novel insights into the molecular mechanisms underlying the characteristic aroma formation of Xinhui Chenpi.
The formation of high-quality Chinese medicinal materials is a micro-evolutionary process of multiple genes involving quantitative inheritance under environmental stress. Atractylodes lancea is a traditionally used medicinal plant in China that is broadly distributed and possesses a considerable amount of essential oils. However, to date, limited research has been conducted to characterize the genetics and metabolites of A. lancea shaped by natural variation. Hence, we assembled a high-quality genome of A. lancea, featuring a contig N50 of 1.18 Mb. We further integrated population resequencing of A. lancea and conducted analyses to characterize its genetic diversity, population evolution, and rewiring of volatile metabolites. The natural variation effect exerted significant pressure on A. lancea from different geographic locations, resulting in genetic differentiation among three groups. Correlation analysis of metabolites in A. lancea revealed significant natural variations of terpenoids, heterocyclic compounds, ketones, and esters. We also found that 427 metabolites displayed noteworthy divergence due to directional selection. Additionally, our genome-wide association studies on the metabolome for medicinal quality traits identified several candidate genes, such as AlZFP706 and AlAAHY1, exhibiting significant correlations with atractylodin and hinesol levels, respectively. Overall, this study provides an intricate genomic resource for A. lancea, thereby expanding our understanding of the effect of natural variation on metabolites and facilitating the genetic improvement of its medicinal properties.
Rubus chingii and R. chingii var. suavissimus are unique dual-purpose plant resources, with significant nutraceutical, pharmaceutical, and economic value, as well as promising prospects for further development. To investigate the genetic structure and evolutionary characteristics of these two varieties, this study conducted plastome sequencing using the Illumina HiSeq XTen sequencing platform. Subsequently, the study performed assembly, annotation, and characterization of the genomes, followed by a comparative plastome and phylogenetic analysis using bioinformatics techniques. The results revealed that the plastomes of R. chingii and R. chingii var. suavissimus exhibited a tetrad structure, comprising a large single-copy region(LSC), a small single-copy region(SSC), and two inverted repeat regions(IRs). The study identified a total of 56 simple sequence repeats(SSRs) after comparative analysis, predominantly consisting of A and T. Furthermore, the structure of the IR boundary genes in both varieties was found to be highly conserved, with only minor nucleotide variations. Additionally, the study identified three highly variable regions: rps16-trnQ-psbK, trnR-atpA, and trnT-trnL, which held promise as potential identification marks for further development and utilization. Phylogenetic analysis results obtained by the maximum likelihood and Bayesian inference methods demonstrated a close clustering of R. chingii and R. chingii var. suavissimus(100% support), with their closest relatives being R. trianthus. This study, focusing on plastome-level genetic distinctions between these two varieties, lays a foundation for future species protection, development, and utilization.
Background The sustainable supply of medicinal plants is important, and cultivating and domesticating them has been suggested as an optimal strategy. However, this can lead to a loss of genetic diversity. Tripterygium wilfordii Hook. f. is a medicinal plant commonly used in traditional Chinese medicine, but its wild populations are dwindling due to excessive harvesting. To protect the species and meet the increasing demand, it is urgent to cultivate it on a large scale. However, distinguishing between T. wilfordii and T. hypoglaucum , two similar species with different medicinal properties, is challenging. Therefore, it is crucial to understand the genetic diversity and population structure of these species for their sustainable utilization. Results In this study, we investigated the genetic diversity and population structure of the two traditional medicinal semiwoody vines plant species, Tripterygium wilfordii and T. hypoglaucum , including wild and cultivated populations using chloroplast DNA (cpDNA) sequences and microsatellite loci. Our results indicated that the two species maintain a high level of genetic divergence, indicating possible genetic bases for the different contents of bioactive compounds of the two species. T. wilfordii showed lower genetic diversity and less subdivided population structures of both markers than T. hypoglaucum. The potential factors in shaping these interesting differences might be differentiated pollen-to-seed migration rates, interbreeding, and history of population divergence. Analyses of cpDNA and microsatellite loci supported that the two species are genetically distinct entities. In addition, a significant reduction of genetic diversity was observed for cultivated populations of the two species, which mainly resulted from the small initial population size and propagated vegetative practice during their cultivation. Conclusion Our findings indicate significant genetic divergence between T. wilfordii and T. hypoglaucum . The genetic diversity and population structure analyses provide important insights into the sustainable cultivation and utilization of these medicinal plants. Accurate identification and conservation efforts are necessary for both species to ensure the safety and effectiveness of crude drug use. Our study also highlighted the importance of combined analyses of different DNA markers in addressing population genetics of medicinal plants because of the contrasts of inheritance and rates of gene flow. Large-scale cultivation programs should consider preserving genetic diversity to enhance the long-term sustainability of T. wilfordii and T. hypoglaucum . Our study proposed that some populations showed higher genetic diversity and distinctness, which can be considered with priority for conservation and as the sources for future breeding and genetic improvement.
Angelica sinensis (Oliv.) Diels belongs to the Apiaceae family. The root of A. sinensis, is used in traditional Chinese medicine for its antioxidant and immune regulation properties. The main active compounds in A. sinensis include organic acids, phthalides and coumarins, and their biosynthetic pathways are the focus of international attention. A. sinensis is prone to early flowering and bolting, which negatively impacts production for several reasons, including germplasm degradation and quality instability in artificial cultivation. The identification of top-geoherbalism of A. sinensis has also become the focus of recent research, as it would allow selection for breeds with excellent medicinal quality and remarkable curative effects. Advances in sequencing technology and bioinformatic methodologies have enabled extensive molecular and genetic studies in A. sinensis. In this review, we summarize the latest molecular research advances related to A. sinensis, including biosynthetic pathways and regulation of active compounds, and molecular underpinnings of early bolting and flowering and top-geoherbalism. We discuss limitations of the current research and propose prospective topics in need of further exploration.
Leonurus japonicus, a significant medicinal plant known for its therapeutic effects on gynecological and cardiovascular diseases, has genetic diversity that forms the basis for germplasm preservation and utilization in medicine. Despite its economic value, limited research has focused on its genetic diversity and divergence. The avg. nucleotide diversity of 59 accessions from China were 0.00029 and hotspot regions in petN-psbM and rpl32-trnL(UAG) spacers, which can be used for genotype discrimination. These accessions divided into four clades with significant divergence. The four subclades, which split at approximately 7.36 Ma, were likely influenced by the Hengduan Mountains uplift and global temperature drop. The initial divergence gave rise to Clade D, with a crown age estimated at 4.27 Ma, followed by Clade C, with a crown age estimated at 3.39 Ma. The four clades were not showed a clear spatial distribution. Suitable climatic conditions for the species were identified, including warmest quarter precipitation 433.20 mm 1,524.07 mm, driest month precipitation > 12.06 mm, and coldest month min temp > -4.34 °C. The high suitability distribution showed contraction in LIG to LGM, followed by expansion from LGM to present. The Hengduan Mountains acted as a glacial refuge for the species during climate changes. Our findings reflected a clear phylogenetic relationships and divergence within species L. japonicus and the identified hotspot regions could facilitate the genotype discrimination. The divergence time estimation and suitable area simulation revealed evolution dynamics of this species and may propose conservation suggestions and exploitation approaches in the future.
A previous 1H-NMR method allowed the quantification of ephedrine alkaloids; however, there were some disadvantages. The cyclized derivatives resulted from the impurities of diethyl ether were identified and benzene was selected as the better extraction solvent. The locations of ephedrine alkaloids were confirmed with 2D NMR. Therefore, a specific 1H-NMR method has been modified for the quantification of ephedrine alkaloids. Accordingly, twenty Ephedrae Herba samples could be classified into three classes: (I) E. sinica-like species; (II) E. intermedia-like species; (III) others (lower alkaloid contents). The results indicated that ephedrine and pseudoephedrine are the major alkaloids in Ephedra plants, but the concentrations vary greatly determined by the plant species and the collection locations.
Coptis huanjiangensis, a new species of Ranunculaceae distributed in the valleys of Jiuwanshan National Natural Reserve in Huanjiang county (Guangxi, China), is described and illustrated for the first time based on morphological and plastome sequences data. It differs from C. chinensis, C. deltoidei and C. omeiensis mainly by having notably longer petiole, scape, bigger leaf blade with lobes obviously remote and robust rhizomes without stolons. Phylogenetic analyses support that C. huanjiangensis is sister to C. omeiensis and C. deltoidei.
Bactericera gobica is the major pest of Goji berry plants and causes severe damage. Psyllids mainly use the antennal sensilla to recognize olfactory cues necessary to find host plants and mates. However, the structure and function of the antenna and the antennal sensilla of B. gobica remains previously unexplored. Here, we identify the external and internal morphology of the antennal sensilla of B. gobica using both scanning electron microscopy (SEM) and transmission electron microscopy (TEM). We found seven types of sensilla on the filiform antennae, including apical setae (LAS, SAS), sensilla basiconica (SB1, SB2), sensilla campaniform (SCA), sensilla chaetica (ChS1, ChS2), cavity sensilla (CvS1, CvS2), antennal rhinaria (AR1, AR2), and sensilla trichodea (ST). Five of these sensilla types-apical setae, sensilla basiconica, sensilla chaetica, cavity sensilla, and antennal rhinaria-may have olfactory functions based on their porous surfaces and internal dendritic outer segments (DOS). We also found several differences between the two sexes of B. gobica in the sensilla array and internal structure. ChS and DOS in the protrusions of AR were more abundant in males than females. Altogether, we comprehensively revealed the fine structure and probable function of B. gobica antennae and identified differences in the distribution and structure between psyllid sexes. Our findings provide important insights for future studies on defining the olfactory function of psyllid antenna using electrophysiological methods.
目的:探究雅连根际土壤微生物群落结构特征及与雅连生物碱含量的相关性,揭示雅连根际土壤微生物群落结构与雅连品质的关系.方法:以二至五年生雅连及其根际土壤为材料,基于Illumina MiSeq平台,利用高通量测序技术对不同生长年限雅连根际土壤细菌16S rRNA基因和真菌ITS序列测序并进行数据分析;利用HPLC检测雅连生物碱含量.结果:各年生雅连根际土壤样本微生物的所有分类水平具有显著丰度差异;不同生长年限雅连6种生物碱总量由高到低依次为:四年生>五年生>三年生>二年生,且四年生雅连的6种生物碱总量显著高于其他各年生雅连;将雅连根际土壤中丰度前15的微生物门类与各年生雅连生物碱指标进行Spearman分析表明,细菌标志菌属MBNT15属和分支杆菌属Mycobacterium与雅连多种生物碱含量呈显著相关,真菌标志菌属端毛孢属Seimatosporium与雅连多个生物碱含量呈显著相关.结论:该研究揭示了雅连根际土壤微生物群落的种类与资源,发掘了与雅连生物碱含量相关的标志根际土壤微生物,为进一步研究影响雅连品质形成的核心功能微生物菌群提供了参考.
The internationalization of traditional Chinese medicine(TCM) is one of the strategic development objectives in China, which has been incorporated into the national strategy as an important part of the Belt and Road Initiative development strategy. As the basis and prerequisite of TCM development, Chinese materia medica(CMM) has a direct impact on the internationalization of TCM. The International Organization for Standardization(ISO) is a global organization composed of national standardization bodies, and the ISO standards impact the world's economy, trade, communication and cooperation. Based on a brief introduction to ISO/Traditional Chinese Medicine Technical Committee(ISO/TC 249), this study elaborates the necessity of establishing ISO standards for CMM and analyzes the current status and challenges faced by the formulation of international standards for CMM. Finally, this study puts forward the development strategy of international standards for CMM. Specifically, efforts should be made to develop top-level design with international market demands as the guidance and improve the quality of standards to accelerate the transformation of domestic high-quality standards into international standards. Moreover, measures should be taken to give full play to the positive role of enterprises in the formulation of standards, vigorously cultivate compound talents for international standardization of TCM, and constantly strengthen international cooperation. The experience and thinking are of guiding significance for the scientific, efficient and reasonable formulation of high-quality ISO standards for CMM in the future.
The genus Coptis belongs to the Ranunculaceae family, containing 15 recognized species highly diverse in morphology. It is a conspicuous taxon with special evolutionary position, distribution pattern and medicinal value, which makes it to be of great research and conservation significance. In order to better understand the evolutionary dynamics of Coptis and promote more practical conservation measures, we performed plastome sequencing and used the sequencing data in combination with worldwide occurrence data of Coptis to estimate genetic diversity and divergence times, rebuild biogeographic history and predict its potential suitable distribution area. The average nucleotide diversity of Coptis was 0.0067 and the hotspot regions with the highest hypermutation levels were located in the ycf1 gene. Coptis is most likely to have originated in North America and Japanese archipelago and has a typical Eastern Asian and North American disjunct distribution pattern, while the species diversity center is located in Mid-West China and Japan. The crown age of the genus is estimated at around 8.49 Mya. The most suitable climatic conditions for Coptis were as follows: precipitation of driest quarter > 25.5 mm, annual precipitation > 844.9 mm and annual mean temperature -3.1 to 19 °C. The global and China suitable area shows an upward trend in the future when emission of greenhouse gases is well controlled, but the area, especially in China, decreases significantly without greenhouse gas policy interventions. The results of this study provide a comprehensive insight into the Coptis evolutionary dynamics and will facilitate future conservation efforts.
Ligustri Lucidi Fructus, the sun-dried mature fruit of Ligustrum lucidum, is cool, plain, sweet, and bitter, which can be used as both food and medicine, with the effects of improving vision, blacking hair, and tonifying liver and kidney. It takes effect slowly. However, little is known about the genetic information of the medicinal plant and it is still a challenge to distinguish Ligustrum species. In this study, the complete chloroplast genome of L. lucidum was obtained by genome skimming and then compared with that of five other Ligustrum species, which had been reported. This study aims to evaluate the interspecific variation of chloroplast genome within the genus and develop molecular markers for species identification of the genus. The result showed that the chloroplast genome of L. lucidum was 162 162 bp with a circular quadripartite structure of two single-copy regions separated by a pair of inverted repeats. The Ligustrum chloroplast genomes were conserved with small interspecific difference. Comparative analysis of six Ligustrum chloroplast genomes revealed three variable regions(rbcL-accD, ycf1a, and ycf1b), and ycf1a and ycf1b can be used as the species-specific DNA barcode for Ligustrum. Phylogeny analysis provided the best resolution of Ligustrum and supported that L. lucidum was sister to L. gracile. This study clarified the genetic diversity of L. lucidum from provenance, which can serve as a reference for further analysis of pharmacological differences and breeding of excellent varieties with stable drug effects.
Sargentodoxa cuneata is used as traditional Chinese medicine. In this study, we report its complete chloroplast genome by Illumina pair-end sequencing. The total chloroplast (cp) genome size was 158,094 bp in length, containing a pair of inverted repeats of 26,132 bp, separated by large single-copy and small single-copy regions of 86,508 bp and 19,322 bp, respectively. The chloroplast genome of S. cuneata encodes 113 different genes, including 79 protein-coding genes, 30 transfer RNAs, and 4 ribosomal RNAs. A total of 84 perfect chloroplast microsatellites were analyzed in the S. cuneata. The majority of the SSRs in this chloroplast genome are mononucleotides (66.67%). The reconstructed phylogeny revealed that S. cuneata was sister to the remaining Lardizabalaceae.
Prunus japonica is an ornamental and medicinal plant that is widely cultivated. The complete chloroplast genome of P. japonica was sequenced using Illumina Hiseq X Ten platform. The chloroplast genome was 158,080 bp in length, containing two short inverted repeat (IRa and IRb) regions of 26,385 bp, which was separated by a large single copy (LSC) region of 86,270 bp and a small single copy (SSC) region of 19,040 bp. The GC content of the whole chloroplast genome was 36.8%. The chloroplast DNA of P. japonica comprised 112 distinct genes, including 78 protein-coding genes, 4 ribosomal RNA genes and 30 transfer RNA genes. Phylogenetic analysis indicated that all species of Prunus formed a monophyletic group, P. japonica was closely related to P. hulimis.
Emergency of nanoparticulate drug delivery systems has improved the target, bioavailability, and curative effect of traditional Chinese medicine (TCM). However, the application of nano-preparation has been limited owing to the low content of active ingredients in part TCM. MicroRNAs (miRNAs) regulate plant growth, development, and response to environmental stresses at post-transcriptional regulation level by cleavage or translational inhibition. The molecular functions of miRNAs playing a role in synthesizing active comportments at medicinal plants have been widely researched. Dendrobium nobile is a perennial herb in the orchidaceae family. D. nobile protocorm can produce plant-specific metabolites at a short period. Therefore, it is a good substitute for producing metabolites. To understand the functions of miRNAs in D. nobile protocorm, Illumina sequencing of D. nobile protocorm (Dnp), D. officinale protocorm (Dcp), and D. nobile leaf (Dnl) were carried out. A total of 439, 412, and 432 miRNAs were identified from Dnp, Dcp, and Dnl, respectively. Some specific miRNAs were identified among them. Through combing GO and KEGG function annotation, miRNAs mainly involved metabolic pathways, plant hormone signal transduction, biological regulation, and protein binding. Acetyl-CoA acetyltransferase (AACT), mevalonate kinase (MK), 1-deoxy-D-xylulose 5-phosphate reductoisomerase (DXR), and 2-C-methyl-D-erythritol 4-phosphate cytidylyltransferase (HDS), synthesizing basic precursor isoprene pyrophosphate (IPP) in terpenoid backbone biosynthesis pathway, were predicted as potential targets of 6 different miRNAs. Twenty-six miRNAs participated in auxin, cytokinin, abscisic acid, jasmonic acid, and salicylic acid signal transduction pathway. This report provided valuable candidate genes in Dnp involved in terpenoid biosynthesis and plant hormone signal transduction pathway. At the same time, it can help accelerate the use of dendrobine into nano preparation.
