Persistent infection with high-risk human papillomavirus (HR-HPV) is the primary cause of cervical cancer. Understanding genotype distribution and evaluating screening strategies are essential for effective prevention. We retrospectively analyzed cervical cancer screening data from 97,686 women aged 35–64 years in Nanjing (2021–2023). Among these, 30,147 underwent combined cytology and HPV testing and 67,539 TCT alone. HPV genotyping was further performed in 3,362 histologically confirmed cervical cancer cases (3,014 squamous cell carcinomas [SCC] and 348 adenocarcinomas [ADC]) from multiple regions in China. Background: Hepatitis C virus (HCV) is one of the main causes of liver fibrosis, chronic hepatitis, and liver cirrhosis. The aim of this study is to determine the prevalence of HCV, age-dependent prevalence and genotypes distribution in a large number of clinical samples in Sichuan area of China. Methods: In the past five years from 2014 to 2018, a total number of 4,508 individuals received the serum HCV-RNA analysis in the Sichuan Provincial People's Hospital. Viral nucleic acid was extracted from the serum samples and amplified using COBAS AmpliPre/COBAS TaqMan Detection Platform. Five HCV genotypes (1b, 2a, 3a, 3b, and 6a) of serum samples from 469 HCV positive individuals collected from 2016 to 2018 were analyzed using the PCR-fluorescence probe technique. Results: A total of 1,668 individuals had positive results by high precision HCV-RNA quantitative technique, corresponding to a crude prevalence of 37.0% (95% confidence interval: 33.6 - 40.3%). The majority of HCV positive individuals were aged over 41 years, accounting for 80.7% (1,346/1,668, CI: 72.3 - 87.1%). Among the nine age groups, the 41 - 50-year age group had the highest HCV prevalence of 29.8% (497/1,668, CI: 25.6 - 32.3%). Of the 469 HCV-RNA positive serum samples collected in 2016 - 2018, genotype 1b was the most frequent type found in 357 individuals, corresponding to a prevalence of 76.1% (CI: 72.3 - 80.0%). Conclusions: Positive rates of HCV in the years of 2014 to 2018 showed a downward trend year by year, of which a majority of positive cases were aged over 41 years. HCV was distributed with multi-genotype features while genotype 1b yielded a very high prevalence in the Sichuan area. The results have potential for prevention and treatment of HCV infection, as well as epidemiological research.
BACKGROUNDHuman papillomavirus (HPV) is the cause of nearly all cervical cancers and the primary cause of anal cancers. Prevalence of HPV varies largely among countries and regions, and population-based data are largely insufficient. The aim of this study is to determine the prevalence and genotype distribution of HPV infection among the women received a general health check.METHODSIn the years 2015, 2016, and 2017, a total of 553,654 individuals received a general health check in the Sichuan Provincial People's Hospital. Among them, 9,182 unselected and asymptomatic individuals received the HPV screening test. Samples of exfoliated endocervical cells were collected and DNA isolation was performed with a Cell Lysis Kit. Fragments of HPV DNA were amplified by PCR. Twenty-one different HPV genotypes, including HPV 6, 11, 16, 18, 31, 33, 35, 39, 42, 43, 44, 45, 51, 52, 53, 56, 58, 59, 66, 68, and CP8304, were detected from PCR products using a GenoArray Diagnostic Hybridization Kit. HPV genotype was read on the colored position on the array.RESULTSA total of 1,207 individuals were positive for at least one HPV genotype, giving a crude prevalence of 13.2% (95% CI: 12.5 - 13.9%). The prevalence did not differ much among age groups. HPV-positive individuals were 291, 389, and 527 in 2015, 2016, and 2017, respectively. The majority of the HPV-positive participants (960/1,207 = 80%) had one type of virus. Approximately 15% had two genotypes of HPV. One individual had HPV of 6 different genotypes, including 16, 18, 52, 53, 56, and CP8304. The most frequent genotype was 52, followed by CP8304, 58, and 53. The oncogenic types 16 and 18 were found in 112 and 52 participants, corresponding to a prevalence of 0.9% (CI: 0.8 - 1.1%) and 0.4% (CI: 0.3 - 0.6%), respectively, for the 9,182 individuals included in this study.CONCLUSIONSThe prevalence of 13.2% for HPV among unselected and asymptomatic individuals who received a general health check is high in the Sichuan area. Identification of high-risk HPV types is essential for preventing or early detection of cervical cancers and consequently save life.
Interferon induced with helicase C domain 1 (IFIH1), which is a type of cytological RNA helicase protein and an important initiator of innate immune response to RNA virus infection, may play an important role in hepatitis C virus (HCV) infection outcomes. This study was conducted to investigate the association of IFIH1 gene polymorphisms with HCV clearance in Chinese Han population. A total of 1,527 subjects positive for anti-HCV and 357 chronic hepatitis C (CHC) patients were enrolled in this study. Two single nucleotide polymorphisms in IFIH1 gene (rs2111485 and rs1990760) were selected and genotyped by TaqMan real-time polymerase chain reaction. Haplotypes analysis was further performed by HaploView software and PHASE software. Our results demonstrated that the mutant alleles of rs2111485 (dominant model: odds ratio [OR] = 1.41, 95% confidence interval [CI] = 1.11-1.79) and rs1990760 (dominant model: OR = 1.63, 95% CI = 1.30-2.06) decreased the possibility of spontaneous HCV clearance, but it had no association with HCV clearance induced by interferon-alfa (IFN-alpha). And, CHC risk increased with the increasing number of unfavorable alleles (p(trend) < 0.001). In addition, haplotype analysis also showed that the A-C protective haplotype (rs2111485-rs1997060) promoted spontaneous HCV clearance (p < 0.001). Variants of rs2111485 and rs1990760 at IFIH1 may be associated with spontaneous HCV clearance in Chinese Han population, but have no effect on HCV clearance induced by IFN-alpha.
目的:探讨3种方案治疗急性大面积脑梗死的临床效果.方法:收治急性大面积脑梗死患者60例,随机分为3组,每组各20例.A组给予常规治疗,B组在常规治疗基础上给予针刺治疗,C组在常规治疗基础上给予针刺联合亚低温治疗,比较3组治疗前后NIHSS评分、神经功能缺损(ESS)、日常生活能力(ADL)评分、血清一氧化氮(NO)含量、C反应蛋白(CRP)含量.结果:治疗14 d、30 d,A组、B组NIHSS、ESS、ADL评分高于C组,差异有统计学意义(P<0.05).A组NIHSS、ESS、ADL评分高于B组,差异有统计学意义(P<0.05).治疗14 d,C组血清NO、CRP含量低于A组、B组,差异有统计学意义(P<0.05).B组血清NO、CRP含量低于A组,差异有统计学意义(P<0.05).结论:在常规治疗基础上给予针刺联合亚低温治疗急性大面积脑梗死可减轻梗死部位脑组织水肿,利于脑细胞及神经的功能恢复.
目的 研究兔膝骨性关节炎(OA)早期软骨下骨血管再生变化及二磷酸盐的保护作用.方法 60只新西兰大白兔随机分成模型组(A组,24只)、二磷酸盐组(B组,24只)和对照组(C组,12只).术后4周、8周、12周处死兔子,切取手术侧膝关节,进行大体评分及Mankin评分,免疫组化染色测量骨软骨连接处的血管密度.结果 与C组相比,A组4周时已出现软骨退变(P<0.01),8、12周时退变进一步加重(P<0.01),并且骨软骨连接区血管侵犯的密度增加(P<0.01);而B组的软骨退变轻于A组(P<0.05或P<0.01).结论 OA早期关节软骨开始退变,软骨下骨与关节软骨交界区血管增生.二磷酸盐对关节软骨有明显的保护作用.
Objective To observe the biomechanical changes of subchondral bone in the early stage of knee joint instability and effect of diphosphonate therapy so as to investigate the role of the early biomechanical changes of subchondral bone in the onset and development of osteoarthritis (OA).Methods Sixty healthy male New Zealand white rabbits were assigned to model group (n =24),diphosphonate group (n =24) and control group (n =12) according to random number table.Joint destabilization by anterior cruciate ligament transection of the right knee of the rabbits was performed to induce OA models.Rabbits in diphosphonate group received subcutaneous injection of 0.01 mg/kg diphosphonate (risedronate) per day,and isotonic saline solution of the same volume was subcutaneously given to rabbits in model and control groups.Half the animals in each group were killed by aeroembolism at postoperative 4 weeks and 12 weeks respectively.Surgical knee joint with preservation of each 2 cm bone above and below joint surface was dissected to perform gross scoring.Thereafter,two-dimensional image profile was achieved by Micro-CT examination and converted into Ansys for limit element analysis after fitting in Mimics software.Results At four weeks,bone volume fraction (BVF),elastic modulus (EM),reaction force (RF),and mean Von Mises stress were all declined in three groups,the lowest level in the model group (P < 0.01).The diphosphonate group also had lower levels than the control group,with insignificant difference.Bone mineral density (BMD) in the model group was obviously declined in contrast with the diphosphonate and control groups (P < 0.01),but there were no significant difference between the diphosphonate group and the control group.At 12 weeks,the model group showed higher level of BVF and BMD,but lower level of EM,RF and Von Mises stress in comparison with the control and diphosphonate groups (P < 0.01).EM,RF,and Von Mises stress were lower in the diphosphonate group than those in the control group as well,but the difference was statistically insignificant.The model group showed that BVF,BMD,EM,RF,and Von Mises stress at 12 weeks were improved from those at 4 weeks (P < 0.01).Conclusions Biomechanical properties of subchondral bone are affected in the early stage of knee joint instability and a notable decrease of EM is observed in the early stage,followed by an enhancement in late stage.It means that the biomechanical changes of subchondra,l bone in the early stage of knee joint instability may be connected with the bone resorption resulting from abnormal stress.On the contrary,diphosphonate may markedly improve EM of subchondral bone through inhibiting bone resorption.
Objective To study the expression of microtubule-associated protein-2 (MAP-2) in the CA3 region of the hippocampus and any effect of acupuncture combined with exercise training on the recovery of learning and memory after experimental cerebral infarction.Methods Eighty Wistar rats were randomly divided into a sham-operation group (n = 8 ) and an operation group (n = 72 ) subdivided randomly and equally into a model group, an exercisetraining group and an acupuncture plus exercise training group, with 24 rats in each group. After right middle cerebral artery occlusion or a mock operation, the expression of MAP-2 in the CA3 region of the hippocampus was detected with immunohistochemistry 1, 3 and 5 weeks after the operation. Learning and memory function was assessed at the 5th week post operation.Results MAP-2- positive fibers were arranged in an orderly way and distributed densely in the shamoperation group. After cerebral infarction, number of MAP-2 positive neurons and dendrite fibers decreased in the CA3 region of the affected side. A week after the operation, MAP-2 positive fibers had increased significantly in the acupuncture plus exercise group compared with the model group and the exercise group. At the 3rd and 5th week post operation, expression of MAP-2 and optical density had increased significantly compared with the model group and the exercise training group. At the 5th week post operation, Y maze discrimination and learning tests showed that learning and memory had improved significantly more in the acupuncture plus exercise training group compared with the model group and the exercise training group.Conclusions Exercise training combined with acupuncture can promote changes in dendritic plasticity and increase MAP-2 expression in the CA3 region after cerebral infarction. This may be correlated positively with the recovery of learning and memory after cerebral infarction, at least in rats.
【Objective】To screen serum biomarkers in patients with in situ and invasive carcinoma of the cervix by using surface-enhanced laser desorption and ionization time-of-fight mass spectroscopy(SELDI-TOF-MS) and WCX2(weak cation exchange array) protein chips.【Methods】The study included 44 cases with invasive cervical cancer(ICC),30 cases with in situ cervical cancer(carcinoma in situ,CIS) and 51 controls.The peaks were detected and filtrated by Biomarker Wizard 3.1.The diagnostic model was developed with Biomarker pattern software 5.0.2.【Results】Compared the cervical cancers specimens and controls,125 serum samples randomly divided into training test(n=60,30 cervical cancer patients and 30 controls) for developing an artificial neural networks classifier,and test set(n=65) for checking the classification system.The diagnostic model defined by six specific protein peaks with M/Z at 5642,5912,5905,8700,2288,5660,which could discriminate cervical cancers(ICC and CIS) from controls,it yielded a sensitivity of 95.45%,a specificity of 95.23%.Compared the CIS with ICC specimens,74 serum samples randomly divided into training test(n=30,15 ICC and 15 ICS specimens) and test set(n=44,29 ICC and 15 ICS specimens).Six protein peaks at M/Z at 13776,2796,2697,2070,2768 and 9222 were able to classify ICC samples from the all CIS samples,with 93.10% sensitivity and 93.33% specificity.Compared the CIS with controls,81 serum samples randomly divided into training test(n=30,15 CIS and 15 controls) and test set(n=51,15 CIS and 36 con-trols).Six proteins were able to discriminate CIS from controls,with 60% sensitivity and 98.08% specificity.【Con-clusion】 This study represented that it may be potential to detect and classify in situ and invasive cervical carcino-ma through plasma proteomic profiling.Identification of the proteins distinct in invasive and in situ cancer may be of great value in the studying of cervical cancer invasion and the development of novel therapeutic methods.
Objective To identify serum protein markers for diagnosis of schizophrenia using surface-enhanced laser desorption-ionization time of flight mass spectrometry(SELDI-TOF-MS)and CM10 chip.Methods 66 patients with schizophrenia and 62 normal controls were included.Sample were randomly assigned into two subsets,the training set(n=60,30 in each),and the testing set(n=68,36 patients with schizophrenia and 32 normal controls).The training set was used for identifying the statistically significant peaks as well as for developing artificial neural network(ANN)model.And the testing set was used for blind test to validate the diagnostic efficiency of ANN model.The SELDI-TOF-MS and CM10 protein chips were performed to screen for differentially expressed proteins in serum from patients and mormal controls.Results Total 15 differentially expressed proteins between patients and controls were identified.Among them,five proteins(P10-4,m/z at 2820,3219,3317,4284 and 4347)were chosen to develop ANA based diagnostic model.The model was blindly tested with the testing set for diagnosing schizophrenia.The sensitivity and specificity was 91.7% and 93.8%,respectively.Conclusions The preliminary results suggested that patients with schizophrenia may have specific serum protein expression different from normal controls.These biomarkers may have the potential in diagnosis of schizophrenia.
目的:观察综合康复疗法佐治糖尿病足的临床疗效。方法:将98例糖尿病足患者分为康复组(48例)及常规组(50例)。康复组在给予治疗糖尿病足常规药物及足部创面处理的基础上再予以综合康复疗法,包括电针刺穴、超短波治疗、体外反搏治疗、红外线照射治疗;常规组则予以治疗糖尿病足常规药物及足部创面处理。治疗1个月后比较两组0级糖尿病足患者的血液流变学变化,治疗2个月后比较2组1级及以上糖尿病足患者的疗效及痊愈时间。结果:治疗1个月后,康复组0级糖尿病足患者的血液流变学参数改善较常规组更明显;治疗2个月后康复组1级及以上糖尿病足患者的治疗总有效率(100%)明显高于常规组(79%),且其痊愈患者的痊愈时间亦较常规组短。结论:综合康复疗法辅助治疗糖尿病足疗效较好,并可缩短治疗时间,值得推广。
