Исследовано регуляторное действие двух оксипроизводных ненасыщенных жирных кислот (оксилипинов) 18-гидрокси-(9Z,12Z)-октадекадиеновой кислоты (18-HODE) и 18-гидрокси-(9Z,12Z,15Z)-октадекатриеновой кислоты (18-HOTrE), на половое и бесполое размножение штаммов Neurospora crassa дикого типа и мутантов wc-1 и wc-2. Показано, что у штамма дикого типа 18-НОDE, в отличие от 18-HOTrE, является стимулятором образования протоперитециев в темноте и на свету. У этого же штамма исследованные оксилипины оказывали влияние на конидиогенез только в условиях освещения: 18-HODE стимулировала, а 18-HOTrE ингибировала образование конидий. Оксилипины не влияли на образование протоперитециев у мутантов по фоторецепторному комплексу, что, по-видимому, свидетельствовало о его участии в передаче сигнала у N. crassa. Стимуляция образования конидий у wc-1 под влиянием исследованных оксилипинов и отсутствие их действия на wc-2 может указывать на наличие альтернативных путей передачи сигнала оксилипинов при конидиогенезе.
Исследовали влияние оксилипинов природной структуры, 3(R)-гидрокси-(5Z,8Z,11Z,14Z)-эйкозатетраеновой кислоты (3-HETE) и 18-гидрокси-(9Z,12Z)-октадекадиеновой кислоты (18-HODE), на рост и агрегацию гиф, а также на ряд светозависимых процессов биосинтез каротиноидов и образование протоперитециев (половой цикл) и конидий (бесполый цикл) у аскомицета Neurospora crassa. 3-HETE, 18-HODE и линолевая кислота вызывали агрегацию гиф и замедление их роста. При концентрациях от 5 до 50 мкМ данные соединения не оказывали существенного влияния на индуцируемое светом образование каротиноидов. Вместе с тем, в этом же диапазоне концентраций 3-HETE и 18-HODE, в отличие от линолевой кислоты, вызывали увеличение образования протоперитециев в темноте. При концентрации 5 мкМ было выявлено аддитивное действие оксилипинов и света. Оксилипины по-разному влияли на бесполое размножение гриба 3-HETE стимулировала образование конидий в темноте, а 18-HODE на свету. Обсуждается участие оксилипинов в регуляции процессов полового и бесполого размножения N. crassa.
Предложена простая модификация спектрофотометрического метода быстрого выявления микроорганизмов по выделению или поглощению ими летучих компонентов. Метод обеспечивает возможность бесконтактного контроля роста бактерий при концентрации выше 107 кл/мл. Он позволяет также различать мутанты гриба Neurospora crassa, дефектные по метаболизму азота, от штаммов дикого типа. По-видимому, ферменты нитритредуктаза и нитратредуктаза, активность которых регулируется генами nit-2 и nit-6, участвуют в образовании водорастворимых летучих компонентов у этого организма.
The specific activity of lipoxygenase from several strains of the zygomycete Mortierella varied from 1.02 to 2.02 microMol diene per min per mg protein). The enzyme equally used linoleic or arachidonic acid as a substrate. An increase in lipoxygenase activity was observed after adding corn oil to the culture medium. Tests with inhibitors having different chemical structures revealed that the lipoxygenase activity from Mortierella cells was inhibited only by esculetin, ethanol and nordihydroguaiaretic acid (NDGA). NDGA inhibited the enzyme in vitro (IC50 = 142 microM), but its addition in the exponential phase of growth activated the enzyme.
The specific activity and molecular forms of NAD-kinase during ontogenesis of Neurospora crassa were investigated. The specific activity of the enzyme increased drastically at critical stages of fungal development, i.e. during conidia germination and during transition from the logarithmic to stationary growth stage, reaching 85 nmole NADP/hr/mg protein. By polyacrylamide gel electrophoresis four forms of NAD-kinase were revealed that had the following molecular masses: I-338,000, II-306,000, III-229,000, and IV-203,000. The vegetative mycelium contained predominantly form III, and conidia showed a high content of high-molecular-weight forms I and II.
Illumination with a visible light is known to induce accumulation of colored carotenoid pigments in mycelial cells on Neurospora crassa and sporulation of starved macroplasmodia of Physarum polycephalum. In both microorganisms, the temperature, at which they are cultivated in the dark, controls manifestation of the photoregulation processes in the cells subsequently exposed to light. The maximal rate of photoinduced accumulation of carotenoids in N. crassa is observed, when the cells are grown in the dark at 25-28 degrees C; the highest photoinduction of P. polycephalum sporulation takes place, if microplasmodia are grown in the dark at 21-23 degrees C and macroplasmodia are starved at no more than 23 degrees C. Both a 5 degree C rise and fall of the temperature of dark cultivation from the optimum dramatically inhibits the cell ability for photoresponse. This inhibition is not associated with suppression of cell growth processes: temperature rise at the dark period up to 32 degrees C for N. crassa and 25-27 degrees C for P. polycephalum brings the subsequent photoresponse to a minimum but does not produce any negative effect on the growth rate. It is concluded that temperature affects specifically processes of formation of functionally active photoregulatory systems.
