Gestational metabolic syndrome (GMS) not only causes maternal morbidities during pregnancy and after childbirth, but also predisposes offspring to long-term adverse health issues. Bile acids have now emerged as key regulatory factors in metabolism, but their role in mediating metabolic adaptations during pregnancy has not been understood completely. In this study, we investigated bile acids mediated lipid metabolism in a new rat GMS model which recapitulates major symptoms of human GMS, including elevated blood pressure, glucose intolerance, obesity and disrupted lipid metabolism, in response to a high fat diet in combination of drinking water with fructose (HFC feed) for only five weeks. We hypothesized that bile acids profile and downstream signaling in the placenta were altered by GMS, disrupting lipid metabolism in trophoblast cells. In Study One, we performed untargeted and targeted metabolomics analyses on the placentas for male and female fetuses at gestational day 20.5 in the control diet and HCF groups. In Study Two, we investigated placental expression of genes related to bile acids production (CYP7A1, CYP27A1, and CYP8B1) by Western blotting analysis. In Study Three, we explored the regulatory role of chenodeoxycholic acid (CDCA) and its receptor Takeda G protein-coupled receptor 5 (TGR5) on expression and/or activities of LPL (major lipase in the placenta ) and angiopoietin-like proteins (ANGTPL) 3, 4, 8 (inhibiting LPL activity), and formation of lipid droplets in human trophoblast cell line JEG-3, after administration of BAR501(TGR5 agonist), SBI-115 (TGR5 antagonist) and gene knockdown. All quantitative data were analyzed by ANOVA or Student’s t-test, to show the effects of feed, sex or cell treatments (n=3-5). The main findings include: 1) The concentrations of total cholesterol, triglyceride, and free fatty acids in the placentas of both male and female fetuses were highly elevated in the HCF group, with more remarkable increase in the placentas of male fetuses compared to those in female fetuses in the HCF group; 2) Several bile acids in placental tissues in the HCF group demonstrated sex difference, and CDCA levels were increased only in the placentas of male fetuses in the HCF group; 3) Unlike the placentas for female fetuses, the protein abundance of CYP7A1 was significantly elevated in the placentas of male fetuses, that of CYP8B1 was decreased and that of CYP27A1 was unaltered; 4) CDCA increased the protein abundance of ANGPTL4, reduced LPL activity and increased lipid droplets in JEG-3 cells, in a dose-dependent manner; 5) BAR501 significantly increased the protein abundance of ANGPTL4 and lipid droplets, while SBI-115 did not alter the protein abundance of ANGPTL4 but blocked CDCA induced lipid accumulation. These results indicate that GMS alters placental lipid metabolism in a sex-specific manner and CDCA/TGR5/ANGPTL4/LPL axis may play an important role in regulating the placental lipid metabolism in human trophoblast cells. Objective:To study the trend of menarche age in Han and Mongolian women born from 1951 to 2005 in Mongolian region.Methods:A cross-sectional cluster sampling survey method was adopted, From 2003 to 2019, a retrospective survey was carried out in three banners/counties in Tongliao region on the female population of Han and Mongols nationalities aged 16 to 46 and conducted under standardized survey procedures and quality control standards. The basic data of menarche age of women born between 1951 and 2005 were obtained. The changes and rules were analyzed by taking 1 year, 5 years and 10 years as nodes.Results:Totally 46 and conducted under standardized survey procedures and quality control standards 928 pepole (24 450 Han and 22 478 Mongolian) were recruited, the survey response rate was 96.09% (46 928/48 836). In one-year-period analysis, the menarche age gradually decreased from 1951 to 2005. The mean menarche age of Han and Mongolian women changed from (16.22±0.52) years and (15.86±1.24) years in 1951 to (12.37±1.15) years and (12.33±0.98) years in 2005, respectively. The mean menarche age of Han and Mongolian women decreased 3.85 years and 3.54 years. The trend of the mean menarche age's change showed a significant negative correlation with the years (all P<0.000 1). In five-year-period analysis, the mean menarche age of Han and Mongolian women changed from (15.54±1.45) years and (15.53±1.48) years from 1951 to 1955 to (12.41±0.97) years and (12.47±0.96) years from 2001 to 2005, the mean menarche age decreased 3.13 years (3.41 months ahead of schedule every 5 years on average) and 3.06 years (3.34 months ahead of schedule every 5 years on average) in Han and Mongolian women respectively. In ten-year-period analysis, the mean menarche age of Han and Mongolian women changed from (15.79±0.95) years and (15.53±1.33) years from 1951 to 1960 to (12.41±0.97) years and (12.47±0.96) years from 2001 to 2005, the mean menarche age decreased 3.38 years (6.76 months ahead of schedule every 10 years on average) and 3.06 years (6.12 months ahead of schedule every 10 years on average) in Han and Mongolian women respectively. During the 15 years from 1951 to 1965, 1966 to 1970, 1971 to 1990, and 1991 to 2000, they were concentrated at the ages of 15-16, 14-15, 13-14, and 12-13, respectively. The proportion of women at 11 years, 12 years and 13 years menarche age were 26.79% (457/1 706), 73.27% (1 250/1 706), and 92.85% (1 584/1 706) during 2001—2005 in Han women, while the proportion were 23.25% (653/2 809), 62.01% (1 742/2 809), and 90.14% (2 532/2 809) in Mongolian women. Conclusion:The menarche age decreased in Han and Mongolian women from 1951 to 2005, and the ethnic groups tended to be the same. It is recommended to start adolescent education at the age of 8-9 years and pay attention to the changing pattern of early onset of menarche.
目的:分析纸质问卷、当面问询、电子问卷3种方法在月经初潮年龄调查中的使用规范和适用场景.方法:对通辽地区汉族及蒙古族女生采用3种方法同时进行月经初潮年龄调查并进行比较.结果:月经初潮年龄,纸质问卷和电子问卷方法在汉族与蒙古族间均有差异(P<0.05),当面问询方法在汉族与蒙古族间无有差异(P>0.05).纸质问卷和当面体检问询方法调查的汉族、蒙古族月经初潮数据无差异(P>0.05),在总人群中有差异(P<0.01).纸质问卷和当面体检问询方法,均与电子问卷方法数据有差异(均P<0.0001).结论:纸质问卷方法及当面问询方法应用于月经初潮年龄调查准确性更高.建议学校对女生从小学3年级开始每年进行不同侧重的青春期健康教育.
Objective: To retrospectively study the association between male iodine status and fecundity. Design: Population-based retrospective cross-sectional study. Setting: 7 geographic areas in Northeast, North, South, East, West, and Central China from 2013 to 2018.Population: Fertile heterosexual couples who conceived within the past 3 months before recruitment in the study.Methods: Questionnaires for couples, semen and spot urine samples of males were collected, semen parameters were examined, and urinary iodine concentration (UIC) was determined as a biomarker of iodine status. Logistic regression model was employed to calculate fecundability odds ratios (FORs) and 95% confidence intervals (CIs), and to study the association between male iodine status and fecundability. A Cox proportional hazards model was used to analyze couple’s time to pregnancy with response to male iodine status. Main outcome measures: Fecundability. Results: 1132 fertile couples were included in this study. 25.4% of male UICs are iodine deficiency (<99μg/l) and 15.8% of them are excessive (≥300μg/l). Among these fertile couples, the adjusted FORs of couples with optimal UICs in male partners are significantly higher than those whose UIC less than 50μg/l (FOR=0.468; 95% CI, 0.349-0.629; P=0.000); 50-99μg/l (FOR=0.462; 95% CI, 0.381-0.560; P=0.000), greater than 300μg/l (FOR= 0.503; 95% CI, 0.414-0.611; P=0.000), and slightly higher than 200-200 (FOR=0.898; 95% CI, 0.764-1.055; P=0.191) without statistical significance.Conclusion: A U-shaped relationship existed between iodine status of male partners and reduced couple fecundity. An UIC between 100 and 299 ug/L is safe for reproduction of fertile man.
Objective:To examine the association between parental characteristics and risk of autism spectrum disorder (ASD) in children.Methods:In this case-control study, the cases were defined as children who were diagnosed with ASD and were recruited from June 2018 to February 2019 in Shanghai Mental Center ( n=104). The controls were defined as children who did not have ASD and were recruited in the two community health centers in Jing-an District of Shanghai during the same period ( n=149). All children recruited in this study were 2-6 years old. A multivariate logistic regression model was used to examine the association between parental characteristics and the risk of ASD in offspring, and further to estimate the interaction coefficient. Results:According to multivariate regression analysis, the association between maternal age, previous pregnancy complication and the risk of ASD in children appeared to be not statistically significant. After adjusted, advanced paternal age (≥35 years old)( OR=3.65, 95% CI=1.19-11.15, P=0.023), parental disease before or during pregnancy ( OR=3.34, 95% CI=1.41-7.94, P=0.006) and gender of child (male) ( OR=5.84, 95% CI=2.98-11.44, P<0.001) were associated with increased risk of ASD. The results also showed that the boys whose father was 35 years old or more had a higher risk of ASD than the boys whose fahter was less 35 years old and the girls whose father was 35 years old or more ( P=0.005, P=0.006). Conclusion:Advanced paternal age was associated with increased risk of ASD in offspring and this effect may be more pronounced in boys.
ObjectiveTo establish a system for evaluation of semen quality in fertile men by factor analysis (FA). MethodsThe FA method was used to analyze five sperm test indicators for fertile men (sperm pH, sperm motility, sperm progressive motility, semen density, and total sperm number) to determine the evaluation standard of semen quality. Pearson analysis was adopted for correlation testing. ResultsThe comprehensive score formula for semen quality of normal fertile men was as follows: comprehensive score of semen quality = (0.38272 F-1 + 0.36359 F-2 + 0.20018 F-3)/94.699. Across the whole fertile population, semen quality was found to be correlated with abstinence period, age of first spermatorrhea, and frequency of intercourse. Smoking, drinking, and place of residence were correlated with semen quality in the high semen quality population. In the population with medium semen quality, only the abstinence period was associated with semen quality. ConclusionIt is feasible to evaluate the semen quality of fertile men using the FA method. The comprehensive indicators of semen volume, sperm motility, and semen pH can be used as evaluative measures. Across the whole fertile population, the abstinence period and age of first spermatorrhea were correlated with semen quality. In the high semen quality population, smoking and drinking were negatively correlated with semen quality, and participants living in rural areas had better semen quality.
Objective The objective of this study is to reduce the dimension of several indicators with a strong correlation when conducting semen quality analysis in a small number of comprehensive variables that could retain most of the information in the original variables. Methods A total of 1132 subjects were recruited from the Maternal and Child Health Institutions of seven provinces in mainland China. They completed the questionnaire and provided semen samples. Visualization of the correlation between variables was realized by using a function chart and correlation in the PerformanceAnalytics package of the R programming language (version 3.6.3 [2020-02-29]). Factor analysis was conducted using the principal function in the psych package of R. Principal component analysis, combined with varimax rotation, was used in the operation of the model, and two common factors were selected and measured to provide values for the common factor. The score coefficient was estimated using the regression method. Results The contribution rates of the two common factors to variable X were 43.7% and 33.98%, respectively. When the two common factors were selected, approximately 78% of the information of the original variables could be explained. The correlation coefficients between the first common factor (the quantitative factor) and sperm density, total sperm count, and semen volume were 0.824, 0.984, and 0.544, respectively. The correlation coefficients between the second common factor (the quality factor) and sperm motility and the percentage of forward-moving (progressive spermatozoa) sperm were 0.978 and 0.976, respectively. Conclusion The correlation between the original variables of a semen quality analysis was strong and suitable for dimensionality reduction by factor analysis. Factor analysis and dimensionality reduction provide a fast and accurate assessment of semen quality. Patients with low fertility or infertility can be identified and provided with corresponding treatments.
目的 观察孕期添加多种维生素和矿物质营养补充剂对母鼠和仔鼠自主活动与探究行为的影响,以及对仔鼠发育的影响.方法 SPF级昆明小鼠交配见栓后随机分为低剂量组、中剂量组、高剂量组和溶剂对照组,孕期添加营养补充剂(孕早期、孕中期、孕晚期3个阶段的产品)按人体推荐日摄入量2 g/d换算小鼠的等效剂量,以等效剂量的0.5倍、1倍、2倍作为低、中、高3个剂量组的药物剂量,分别为150 mg/(kg·d)、300 mg/(kg·d)、600 mg/(kg·d),连续灌胃6 d,休息1 d后使用下一阶段产品,直至生产;观察仔鼠出生体质量、离乳体质量和离乳一周体质量;同时对离乳后一周母鼠及仔鼠进行旷场实验,观察其行为学差异.结果 仔鼠出生体质量3个剂量组较溶剂对照组均增加,其中高剂量组与溶剂对照组比较差异有统计学意义(P<0.05).离乳当天仔鼠体质量较溶剂对照组显著增加(P<0.05~0.01).离乳后一周仔鼠体质量3个剂量组较溶剂对照组显著增加(P<0.01).产后母鼠在水平运动和垂直运动两项分析指标无明显变化(P>0.05),在中央区停留时间及穿越中央区次数两项分析指标有增加的趋势.离乳后一周雌性仔鼠水平运动和垂直运动指标无明显差异(P>0.05),但水平运动指标显示在中剂量和高剂量组分别增加5.16%和14.79%;在中剂量和高剂量组中央区停留时间分别增加4.59% 和72.95%,高剂量组明显增加(P<0.01);穿越中央区次数分别增加8.58%和56.89%,高剂量组明显增加(P<0.05).在雄性仔鼠中,三组实验组与溶剂对照组比较,在垂直运动次数、水平运动时间和水平运动次数指标上都有所增加,其中高剂量组的垂直运动次数与溶剂对照组比较差异有统计学意义(P<0.05).结论 妊娠期母鼠添加营养补充剂能够增加仔鼠的体质量,利于仔鼠的生长发育,并且能够在不增加实验动物自主活动的情况下,增加其探索行为.
Background: Iodine is an essential element for hormones synthesized by the human thyroid gland. Thyroid hormone deficiency affects all body tissues, including multiple endocrine changes that alter growth hormones, corticotrophin, glucocorticoids, and gonadal function. In many countries, subfertility is an important problem affecting about 10–15% of all couples trying to conceive. Male subfertile factors have accounted for 25–30% of all cases in their studies. The aim of this study was to study the correlation between urinary iodine concentration (UIC) and semen quality of fertile male in China. Methods: semen and urine samples were collected from males who attended a survey for the reproductive physiological constants of male and female in China. All participants were fertile and they should be free from diseases of male reproductive system and thyroid related diseases. We studied the correlation between UIC and semen quality parameters in fertile males and the effect of iodine concentration on male reproduction. Participants were divided into six groups based on UIC. We measured semen parameters and UIC. Semen parameters or time-to-pregnancy (TTP) were analyzed by Spearman correlation, linear regression and multivariable logistic regression. Results: 1089 men were recruited in the study. The mean and median age of participants were 27.46±0.12 and 27±3.88 years, respectively. UIC of 274 (25.16%) men was lower than the UIC of WHO recommended. UIC of 405 men located in the region of WHO recommended. 62.35% of them were not located in the optimal region. Semen concentration and total sperm count were found to decrease when UIC increases when the UIC≥100 ug/L. Conclusions: A male with deficient UIC or excessive UIC has more risk of lower semen quality such as sperm concentration and total sperm count. In addition, if a male with deficient UIC or excessive UIC, his wife has longer TTP than the males who had normal UIC. Deficient UIC associated with lower semen quality and TTP than excessive UIC.
目的 探讨中小学生校园欺凌现状及其与不良心理健康结局的关联.方法 2019年4月,采取分层整群抽样的方法,选取甘肃省民乐县的3所中小学校1575名在校学生,调查其遭受校园欺凌行为情况及心理健康状况.结果 有83名学生自报在过去的2个月内遭受过欺凌(每月2~3次及以上),校园欺凌发生率为5.27%,不同性别发生率差异无统计学意义(x2=1.75,P=0.19);欺凌发生率小学生(9.77%)>初中生(4.63%)>高中生(0.56%),差异有统计学意义(x2=36.15,P<0.01).被欺凌的青少年中不良心理的发生率高,抑郁症状、焦虑症状、自杀意念的发生率分别是26.51%、22.89%、14.46%,而一般青少年的发生率仅为7.77%、6.57%、7.98%,差异均有统计学意义(x2=34.51、30.47、4.33,P值均<0.05).按性别分层进行多因素logistic回归分析发现,在女生中被欺凌是抑郁(OR=8.61)、焦虑(OR=4.71)和自杀(OR=3.30)的危险因素,而在男生中并未发现该关联.结论 校园欺凌发生率小学>初中>高中,且被欺凌青少年不良心理发生率高,应尽早开展干预,并重点关注校园欺凌行为对女生心理健康的影响.
目的:采用偏度系数-中位数-变异系数法(LMS法)研究藏族儿童肌肉脂肪生长发育特点.方法:2018年调查拉萨市曲水县6个乡镇6~15岁在校中小学生3288名.按照《2010年全国学生体质健康调研工作手册》规定的方法进行身体测量;采用生物电阻抗方法进行身体成分测定.以LMS法拟合男生、女生身高、体重、体脂肪含量、体质指数(BMI)和体脂率随年龄变化的百分位数曲线图.结果:LMS曲线显示,不同指标中,男生女生各百分位曲线的变化趋势、曲线间的距离、曲线光滑程度有差别,11~13岁时,男生体重和BMI的增长低于女生,男生的体脂率和体脂肪含量增长速度小于女性.从12岁开始,男生骨骼肌含量增加速度快于女生.结论:藏族儿童青少年的身高、体重、BMI、骨骼肌含量,体脂肪含量和体脂率总体随年龄增加而增加.11~13岁时,体重、BMI、体脂总量、体脂率和骨骼肌含量在性别间差异大.
目的:分析孕期营养补充剂对妊娠期妇女贫血发生的影响。方法:对358例进行产前检查孕妇随机分为观察组(188例)和对照组(170例),分别给予孕期营养补充剂和叶酸,并在孕早、中、晚期测定血红蛋白(Hb)、血清铁离子、铁蛋白、维生素B12和叶酸的浓度,对产妇新生儿进行阿氏评分。结果:观察组孕晚期和临产前孕妇体质指数高于对照组,血红蛋白水平孕中、晚期高于对照组,晚期血清铁离子浓度高于对照组(均P<0.05);孕期血清铁蛋白、维生素B12和叶酸与对照组比较无差异(P>0.05);新生儿阿氏评分高于对照组(P=0.001)。结论:孕期给予营养补充剂可显著提高孕妇孕晚期和血清铁离子水平,从而有效提高孕妇血红蛋白水平,预防孕期贫血的发生。
目的:研究桃仁在小鼠生理性月经模型中的作用.方法:假孕雌鼠蜕膜化诱导后切除双侧卵巢,引发孕酮撤退,造成蜕膜化子宫内膜崩解坏死,模拟月经样出血,建立小鼠生理性月经样模型.在孕酮撤退前至孕酮撤退后24h给予模型小鼠桃仁30mg/(kg·次),观察小鼠子宫大体形态与组织形态变化.结果:孕酮撤退24h后,与对照组相比,给予桃仁的小鼠子宫颜色较深,充血明显;崩解的蜕膜化组织以及崩解组织外围的基底层,有大量红细胞渗出.而空白对照组未见此现象.桃仁处理小鼠脱膜相对崩解面积与对照组未见显著差异,而其相对充血面积高于对照组(P=0.009).结论:桃仁能够促进小鼠月经样模型崩解期的红细胞渗出,其药理作用与作用机制有待进一步研究.
Objective To study the effect of lycium barbarum polysaccharide (LBP) on the function of spermatogenesis and spermatogonial stem cells.Methods Totally 92 of 120 male Kunming mice were randomly selected and treated with 35 mg/kg busulfan to build the animal model with spermatogenesis impairment.And 14 mice were selected as solvent control,16 selected as normal control.After 40 d,the model animals were randomly divided into spontaneous recovery group (n=23),LBP low-dose group (n=16,75 mg/kg),LBP middle-dose group (n=19,150 mg/kg),LBP high-dose group (n=16,300 mg/kg).After treatment for 70 d (two spermatogenic cycles),the changes of physiological status and reproductive function were observed.Results In terms of the appearance,behaviors,body weight,reproductive organs,sperm count in epididymis,sperm count in testis,histology and morphology of testis and expression of c-kit,three kinds dosages of LBP group were better than the spontaneous recovery group with dose-effect relationship,but still not achieve the level of normal control.Conclusion LBP can promote the spermatogenic function on the busulfan-treated infertile mouse,and increase the spermatogonial stem cells,quality of sperm in epididymis and sperm count in testis.
Paeoniflorn (PF), the principal bioactive component of Paeoniae radix prescribed in traditional Chinese medicine, possesses a wide range of biological effects and exhibits neuroprotective effects in numerous diseases. Previous studies have demonstrated that PF significantly attenuates memory impairment in rats with vascular dementia (VD). In the present study, a bilateral common carotid artery occlusion (BCCAO) rat model was used to explore the underlying mechanisms of PF. The expression levels of neuron-specific enolase (NSE), S100β, B-cell lymphoma 2 (Bcl-2), Bcl-2 associated X protein, cytochrome c and brain-derived neurotrophic factor (BDNF) in the hippocampus were measured by western blot analysis. The results showed that administration of PF for 28 days significantly decreased the expression levels of NSE and S100β, both sensitive markers for brain damage, in vascular dementia (VD) model rats. In addition, PF inhibited the initiation of apoptotic cell death and attenuated the decreased expression levels of BDNF induced by bilateral common carotid artery occlusion. These data confirm the neuroprotective effects of PF on VD and provide a novel insight into the long-term use of PF as a potential treatment in the stages of early cognitive impairment in VD.
目的:通过调查通辽市女性月经初潮年龄,建立蒙古族地区汉族和蒙古族女性发育基础数据.方法:采用横断面分层整群抽样方法,在规范调查流程和质量控制标准下于2015年9~12月对本地区3所高中1728名在校女生进行回顾性问卷调查,采集母女月经初潮年龄相关数据,以农村蒙古族、农村汉族、城镇蒙古族、城镇汉族4组进行分析.结果:问卷回收率97.97%,合格率96.16%;女学生月经初潮发生在调查前2~5年者占92.39%,母女记忆明确率均>94.01%.女儿和母亲的月经初潮年龄分别为12.6±1.2(9~16)岁和14.8±1.7(11~ 18)岁,女儿比母亲平均提前了2.14岁(P<0.0001),且城镇早于农村(P<0.05、P<0.001).蒙古族与汉族间未见差异.女儿月经初潮发生发生在11、12、13、14岁的比例分别为6.45%、46.11%、81.57%、94.60%,母亲分别为0.42%、18.50%、38.98%、61.46%.月经初潮发生率按季节从高到低在母女均表现为夏季、冬季、秋季、春季的顺序.女生月经初潮发生在小学阶段者占50.28%,在初中1、2年级时分别达到了80.44%、96.22%.结论:通辽地区属于蒙古族聚集地,其女性月经初潮年龄有逐渐提前趋势,建议对该地区青少年的青春期教育应从小学4年级开始.
目的:研究亚硝酸钠短期低剂量暴露对雄性小鼠生殖的影响.方法:将小鼠随机分为正常对照组,亚硝酸钠本底对照组,亚硝酸钠低、中、高剂量组.称量雄性小鼠体重及肝、脾、睾丸、附睾并计算各脏器系数.取出附睾计数精子、观察其活动度.结果:与正常对照组比较,亚硝酸钠各组小鼠肝脏、脾脏的脏器系数明显增加;附睾脏器系数明显减小,精子数目明显下降,精子活率下降,但a、b级精子所占比例变化不明显,c级精子所占比例下降;组织形态学观察发现,与正常对照相比,使用亚硝酸钠的各组小鼠睾丸发生不同程度的损伤.结论:亚硝酸钠短期暴露可导致小鼠肝、脾、睾丸和附睾组织受损,精子数目减少、活率下降,组织和精子损伤程度随剂量增加而加重,亚硝酸钠的短期暴露可以损伤雄性小鼠的生殖功能.
目的 通过对ICR、NIH、Balb/c三个品系雄性小鼠腹腔单次注射白消安,寻求成功建立小鼠无精子症模型的最佳剂量. 方法 三个品系小鼠分批次进行实验共1 096只,分别给予白消安45、40、35和30 mg/kg四个剂量,40 d后存活小鼠为模型动物,以正常动物为对照(无处理).造模后连续观察2个生精周期共70 d,观察记录小鼠体重变化和死亡情况.分别于造模后0.5、5、7.5、10周取材,以附睾精子质量检测和睾丸组织形态学观察评价造模效果和生精能力恢复情况. 结果 无精子症模型动物外观体征明显改变,以NIH小鼠变化更为明显;NIH小鼠30 mg/kg白消安无精子症模型造模失败,而35、40和45 mg/kg白消安在所研究的三个品系小鼠均可成功建立无精子症模型;动物死亡率在NIH小鼠呈显著剂量效应关系(P<0.01),45 mg/kg白消安剂量组最高,为69.2%;在NIH和ICR小鼠,35 mg/kg剂量组存活率分别为71.3%(194/272)和94.4%(51/54),均显著高于40mg/kg剂量组(P<0.01). 结论 使用临界剂量35 mg/kg白消安在3个品系小鼠均可成功建立无精子症模型,并较40 mg/kg和45 mg/kg白消安组造模成功动物的成活率明显提高,提示可以不采用通常建议的40~45 mg/kg剂量.白消安无精子症小鼠模型在2个生精周期(70 d)内维持基本稳定,处于无精子症或低生精能力状态.
目的:研究慢性不可预知性温和刺激对雄性大鼠生殖系统的影响.方法:将SD雄性大鼠随机分为正常对照组和模型组.模型组大鼠每天给予多种低强度刺激,连续28 d,构建模型.称量大鼠体重、睾丸和附睾重量.取出附睾计数精子、观察精子活率.HE染色观察大鼠睾丸组织结构.结果:与正常对照组相比,模型大鼠体重、睾丸重量、附睾重量显著减少,精子活率显著下降,a、c级精子所占比例明显下降;组织形态学与对照组相比,模型组大鼠睾丸组织受到明显损伤.结论:慢性不可预知性温和刺激可损伤雄性大鼠的生殖功能.
