Objective This study aimed to investigate the effects of caffeine on the invasion and migration of human glioblastoma U-373 cells and to elucidate its mechanism of action through the regulation of the PI3K/Akt signaling pathway and ferroptosis.Methods U-373 cells were cultured in Dulbecco's Modified Eagle's Medium (DMEM). The effects of caffeine on cell invasion, migration, apoptosis, and ferroptosis were assessed using Transwell assays, flow cytometry, cell scratch assays, and Western blotting techniques. Western blot analysis was specifically used to evaluate the expression of PI3K/Akt-related proteins and markers of ferroptosis.Results Caffeine significantly inhibited the invasion and migration of U-373 cells in a concentration-dependent manner. It also promoted apoptosis and decreased the expression of p-PI3K/PI3K and p-Akt/Akt proteins, as well as markers associated with ferroptosis.Conclusion Caffeine, through its regulation of the PI3K/Akt signaling pathway and modulation of ferroptosis, effectively suppresses the invasion and migration of U-373 glioblastoma cells. These findings suggest that caffeine could be a promising anti-glioblastoma agent and offer new insights into its potential applications in cancer treatment.
目的 通过观察黄连解毒汤(HLJDT)对淀粉样前体蛋白(APP)/早老素(PS)1双转基因小鼠海马区NOD样受体蛋白(NLRP)3表达的影响,探讨其对阿尔茨海默病(AD)的保护作用.方法 将APP/PS1双转基因小鼠随机分为对照组,模型组,HLJDT组(433 g/kg),各组连续灌胃35 d后,采用Morris水迷宫实验,检测APP/PS1小鼠的学习记忆能力,通过体重和脑、脾脏的质量,检测脏器指数变化,采用苏木素-伊红(HE)染色观察小鼠海马区神经元的结构变化,采用实时荧光定量聚合酶链式反应(RT-qPCR)检测小鼠海马区APP、β-淀粉样蛋白(Aβ)1~42和NLRP3 mRNA表达水平,Western印迹实验检测小鼠海马区APP、Aβ1~42和NLRP3蛋白表达水平.结果 与对照组比较,模型组学习记忆能力明显下降,脑和脾脏的脏器指数明显降低,神经元细胞减少,排列疏松,小鼠海马区APP、Aβ1~42和NLRP3 mRNA及蛋白表达均明显升高(均P<0.05);与模型组比较,HLJDT组能明显改善小鼠学习记忆能力(P<0.05),脑和脾脏的脏器指数明显升高,神经元细胞增多,排列较紧密,小鼠海马区APP、Aβ1~42和NLRP3 mRNA及蛋白表达均明显降低(P<0.05).结论 HLJDT可改善APP/PS1双转基因小鼠的学习记忆能力、海马神经元的结构及脏器指数,HLJDT通过抑制NLRP3的表达,使AD在一定程度上得到保护.
Background: Gliomas are the most frequent and dangerous primary cerebral tumors. Therefore, there is a need to develop molecular targets for the diagnosis and treatment for glioma. Methods: In September 2020, we retrieved the expression matrix of glioblastoma (GBM) sufferers and pertinent clinical data from the TCGA (The Cancer Genome Atlas) database. Prognostic differences between various families with sequence similarity 110 member C (FAM110C) expression groups were assessed by Kaplan-Meier with log-rank test. The R platform get used to assess the accuracy of FAM110C delivery in predicting the prognosis of PDAC using a time-dependent receptor operating characteristic (ROC) curve. The delivery level of FAM110C was determined by qRT-PCR and western blot. Gene set enrichment investigated possible mechanisms between different FAM110C expression groups in GBM (GSEA). The impact of FAM110C on glioma cell movement was discovered using migration test. The drug's gene-targeting impact was validated by the CCK8 test. Results: A total of 173 GBM samples were obtained from the TCGA database, with 148 including information on IDH1 mutations and 151 containing information on overall survival. The mRNA expression level of FAM110C was greater in wild-type GBM, according to qRT-PCR data. The connection between FAM110C expression and Hallmark, GO, and KEGG pathway gene sets was investigated using GSEA software. We used migration test to assess the impact of FAM110C on glioma motility in order to confirm the findings of the GSEA analysis. Conclusion: FAM110C might get used as a possible diagnostic and prognostic biomarker for wild-type GBM, and its inhibition could be used to prevention and treatment wild-type GBM.
Background This study aimed to investigate FAM84B expression in glioma tissues and explore the role of FAM84B in promoting the proliferation of glioma cells and the mechanism of regulating the cell cycle pathways. Methods The TCGA database was adopted to analyze FAM84B expression in glioma tissues. The FAM84B expression was detected by qRT-PCR in patients with glioma, especially that in glioma cells, U251, LN-229, U98, and U87. Two glioma cell lines U87 and T98 were selected for siRNA transfection, which were divided into si-NC si-FAM84B-1 and si-FAM84B-2 groups. The effect of FAM84B on the proliferation of glioma cells was detected with the MTT experiment and that on the glioma cell cycle was detected with the flow cytometry. The signaling pathways potentially regulated by FAM84B in glioma were analyzed through the bioinformatics analysis. The expression of proteins, Cyclin D1, CDK4, Cdk6, and p21, in the cell cycle-related pathways in cells of each group was detected by the Western blot. Results TCGA database results showed a significantly higher FAM84B expression in glioma tissues than that in paracancerous tissues. According to the detection of qRT-PCR, FAM84B expressed the highest in the glioma cell line U87 ( P < 0.05). Compared with the serum of healthy controls, FAM84B mRNA expression significantly increased in patients with gliomas. And compared with the si-NC group, the proliferation ability of U87 and T98 cells decreased and the cell cycle was blocked in the G0/G1 phase in both si-FAM84B transfection groups ( P < 0.05). According to the bioinformatics analysis, FAM84B regulated the cell cycle pathways in glioma. FAM84B siRNA inhibited the expression of key proteins, Cyclin D1, CDK2, CDK4, and Cdk6, of the cell cycle pathways in glioma cells and promoted the expression of P53 and P21 proteins. Conclusions In conclusion, FAM84B may inhibit the proliferation of glioma cells by regulating the cell cycle pathways.
目的 探讨咪喹莫特(IMQ)对脑胶质细胞瘤U87细胞增殖的影响.方法 U87细胞分为对照组、1 mmol/L IMQ 组、5 mmol/L IMQ 组、1 mmol/L IMQ+STAT3 抑制剂(inhibitor)(STAT3-IN)组和 5 mmol/L IMQ+STAT3-IN组.5-乙炔基-2'-脱氧尿苷(EdU)和MTT实验检测各组U87细胞EdU标记的细胞数目或增殖吸光度值;Real-time PCR、ELISA检测各组U87细胞白细胞介素6(IL-6)mRNA、肿瘤坏死因子-α(TNF-α)mRNA及其蛋白含量;Western blotting 检测各组 U87 细胞 STAT3、磷酸化 STAT3(p-STAT3)、核因子(NF)-κB、磷酸化 NF-κB(p-NF-κB)蛋白表达.结果 相对于对照组,1mmol/LIMQ组和5 mmol/LIMQ组U87细胞的EdU标记细胞数目和吸光度值依次降低,并呈现剂量依赖性(P<0.01,n=10),IMQ+STAT3-IN组和5 mmol/L IMQ+STAT3-IN组EdU标记的U87细胞数目、吸光度值均降低.1 mmol/L IMQ 组和 5 mmol/L IMQ 组 U87 细胞 STAT3、p-STAT3、NF-κB、p-NF-κB 或IL-6、TNF-α 蛋白均比对照组低表达(P<0.01,n=10),1 mmol/L IMQ+STAT3-IN 组和 5 mmol/L IMQ+STAT3-IN 组上述蛋白持续低表达(P<0.01,n=10).结论 咪喹莫特通过下调STAT3/NF-κB通路降低IL-6、TNF-α含量,进而抑制U87细胞增殖.
目的 通过观察黄连解毒汤(Huanglian Jiedutang,HLJDT)对APP/PS1双转基因小鼠行为学以及白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)表达的影响,探讨HLJDT对阿尔茨海默病(AD)的作用机制.方法 以5月龄的APP/PS1双转基因小鼠为研究对象,设为模型组、HLJDT组(433g·kg-1)处理,对照组为5月龄同背景小鼠,每组各8只.采用Morris水迷宫和新物体识别实验检测HLJDT对AD小鼠学习记忆能力的改善;采用实时荧光定量聚合酶链式反应(RT-qPCR)检测IL-1β和TNF-αmRNA表达水平.结果 HLJDT可以改善APP/PS1双转基因小鼠空间及非空间学习记忆能力,并降低IL-1β和TNF-αmRNA表达水平(P<0.05).结论 HLJDT的抗炎功效,可能是通过调控IL-1β和TNF-α的表达实现的.

Accumulating evidence has indicated the important roles of circular RNAs (circRNAs) in different tumors. However, their detailed regulatory mechanisms in glioma are not fully understood. In this study, the functional role of a novel circRNA, circ-EZH2, was investigated by cell counting kit-8 (CCK-8), colony formation, flow cytometry, and transwell experiments. The regulatory mechanism of circ-EZH2 was explored by bioinformatics analysis, quantitative real-time PCR (qRT-PCR), Western blot and dual-luciferase reporter assay. We identified that circ-EZH2 was overexpressed in glioma tissues and cell lines. Further studies revealed that ectopic expression of circ-EZH2 significantly promoted cell growth, migration and invasion but inhibited cell apoptosis. By contrast, silencing of circ-EZH2 induced the opposite effects. Additionally, we found circ-EZH2 served as a miRNA sponge for miR-1265 to release its suppression on DDAH1 and CBX3. Rescue assays further revealed that the oncogenic function of circ-EZH2 was partly dependent on its modulation of DDAH1 and CBX3. Our study unraveled a novel molecular pathway in glioma and may provide a new perspective for the treatment of glioma.
在神经外科临床教学中采用模拟教学结合问题教学(PBL)方法,有利于开展实习生教学,提高临床教学质量和学生综合素质,培养出与社会发展相适应的医学人才.
目的 总结对模拟教学结合问题教学教学模式在医学教学中的应用效果调查与分析,得出此方法 在教学中的应用价值.方法对2016级临床医学专业学生采用模拟教学结合问题教学模式,同时与2015级临床医学专业采用传统教学模式的学生成绩进行对比,比较两组学生的教学效果.结果 观察组实习生的理论成绩(35.17±0.82)分和实践操作考核成绩(56.85±0.96)分均显著高于对照组学生的理论成绩(27.26±0.27)分和实践成绩(42.38±0.92)分,(t=10.022、11.126,P=0.000).结论 模拟教学结合问题教学的教学模式能够有效提高学生的学习效果,在提高学生主动学习同时,更养成了学生积极正确的临床思维,该教学法可以在临床教学中广泛推广.
目的:分析内镜下经鼻蝶手术对于复杂性垂体腺瘤的治疗效果.方法:在2016年1月1日至2017年9月1日间,选择齐齐哈尔医学院附属第三医院神经外科内镜下经鼻蝶窦行手术切除的45例垂体瘤患者资料进行回顾性总结分析,仅选择垂体泌乳素(PRL)腺瘤与垂体生长激素(GH)腺瘤患者,评价患者的治疗效果、生活质量评分、并发症情况以及激素改善情况等.结果:45例患者治疗后,经过MRI复查后,镜下全切率为84.44%(38/45),大部分切除率11.11%(5/45),部分切除率4.44%(2/45);垂体泌乳素(PRL)水平比较,男性患者(t=7.928,P=0.000),女性患者(t=6.983,P=0.000),垂体生长激素(GH)治疗前后比较(t=8.291,P=0.000),生活质量评分比较(t=-10.265,P=0.000),均具有统计学意义(P<0.05).结论:内镜下经鼻蝶手术治疗复杂性垂体腺瘤,疗效稳定,效果可靠.
目的 探讨TBL教学法在规培生神经外科临床操作教学中的应用效果.方法 将20例住院医师规范化培训的学生随机分为实验组和对照组,实验组采用TBL教学法(课前预习准备→个人测试及小组测试→教师示范→分组操作练习,组内、组间互相提问、考核重点→组内、组间总结操作错误及遗漏,教师给予点评→考试),对照组应用常规讲授教学法(教师示范→学生操作练习→考试),培训完成后进行成绩比照及问卷调查.结果 实验组临床操作技能考核成绩优于对照组,实验组对教学效果评价的满意度高于对照组.结论 采用TBL教学法进行规培生神经外科临床操作教学有利于调动学生主动性,强化了学生将基础理论与实际操作的联系,使学生更好的理解了临床技能操作重点,为临床操作考核的顺利通过提供确切保障.
目的 探讨脑胶质瘤与DNMT2表达之间的关系.方法 采用免疫组织化学方法检测不同级别脑胶质瘤DNMT2表达水平变化.结果 与分化程度为Ⅰ级脑胶质瘤样本比较,Ⅱ级、Ⅲ级、Ⅳ级的脑胶质瘤DNMT2的表达上调(P<0.05);与分化程度为Ⅱ级脑胶质瘤样本比较,Ⅲ级、Ⅳ级的脑胶质瘤DNMT2的表达上调(P<0.05);但是Ⅲ级和Ⅳ级之间的脑胶质瘤DNMT2的表达无统计学差异(P>0.05).结论 DNMT2与脑胶质瘤的恶性程度相关,可能成为脑胶质瘤的客观诊断标准.
OBJECTIVE To explore cerebral angiography after vascular spasm associated factors and corresponding preventive measures. METHODS From January 2010 - December 2011, patients with cerebral angiography examination were selected as the research objects, and a total of 80 cases were collected. Comparative analysis was taken to analyze the vasospasm incidence , multivariate regression analysis analyzed the incidence of vasospasm and related factors. RESULTS The incidence of skilled technical operations were significantly lower than the technical operation is not skilled (P﹤0.05), material selection for inci- dence was significantly lower than the material selection is not suitable for persons (P﹤0.05), and the operation time was sig- nificantly higher than that of elderly incidence of short operation time (P﹤0.05). There is a special disease incidence was sig- nificantly higher than that of there is no special disease (P﹤0.05), urokinase incidence was significantly lower than that with- out application of urokinase person (P﹤0.05), and application of nimodipine incidence was significantly lower than that with- out application of nimodipine (P ﹤ 0.05). The incidences of vasospasm and technical operation, material selection, urokinase and nimodipine were negative correlation, and the operation time and special disease were significantly positively correlated (P ﹤0.05). CONCLUSION The related factors of cerebral angiography after occurrence of vasospasm were technology, material selection, operation time, special disease, urokinase and nimodipine application. We should strengthen the corresponding mea- sures for environment, so as to reduce the incidence of postoperative cerebral vasospasm.
目的观察钻孔引流引流管注入尿激酶对高血压脑出血患者的疗效。方法高血压脑出血患者行钻孔引流术,术后引流管注入尿激酶,观察治疗效果。结果 64例患者好转出院,5例因术后加重行血肿清除去骨瓣减压术,7例死亡。结论钻孔引流术引流管注入尿激酶治疗脑出血疗效确切。
目的探讨齐齐哈尔市颅脑交通伤的流行病学特点。方法收集齐齐哈尔市2006~2008年交通事故资料,并收集相应年份的人口资料,建立数据库,采用流行病学描述性研究方法,并应用SPSS13.0统计软件包进行统计学分析,P<0.05为差异有统计学意义。结果3年共发生事故6496起,呈逐年下降趋势,伤者多分布在20~50岁,以30~40岁为高峰,男性多于女性,第三季度为高发期,占33.82%,周六、周日高于平时,多发时段在10:00~12:00,14:00~16:00,19:00~21:00,凌晨0:00~1:00。驾驶摩托车者、行人和乘非机动车使用者占绝大多数(72.41%)。结论齐齐哈尔市第三季度为颅脑交通伤高发期,应加强交通意外伤害的防控,制定有力的干预措施。
