目的 探讨INSM1、CD200、突触素在胰腺神经内分泌肿瘤诊断中的作用.方法 以免疫组织化学法为基础对2019年10月—2020年1月山东省聊城市第二人民医院参与研究的66例胰腺神经内分泌肿瘤患者进行病理诊断,选取60例胰腺非神经内分泌肿瘤患者,检测INSM1、CD200、突触素的表达,通过病理结果为金标准对INSM1、CD200、突触素及其组合在胰腺神经内分泌肿瘤中的诊断意义进行分析.结果 胰腺神经内分泌肿瘤中,INSM1、CD200、突触素阳性率高于胰腺非神经内分泌肿瘤,差异有统计学意义(χ2=58.570、67.120、64.156,P<0.05);INSM1、CD200、突触素对胰腺神经内分泌肿瘤的诊断效能差异无统计学意义(P>0.05),三者联合检测时,诊断的敏感度、特异度及准确度为98.48%、100.00%、99.21%,明显高于单项指标的诊断效能,差异有统计学意义(P<0.05).结论 INSM1与CD200、突触素是胰腺神经内分泌肿瘤临床诊断中的新型标志物,其对胰腺神经内分泌肿瘤得到有效诊断有积极意义,将三者联合应用,能进一步提升诊断效能.
目的探讨肝动脉化疗栓塞(TACE)联合经皮射频消融(RFA)治疗原发性肝癌(PHC)的临床疗效。方法选择确诊的中、晚期PHC患者64例,随机分为联合治疗组35例和单纯TACE组29例。两组均先行TACE,间隔3~4周重复1次,治疗2~3次;联合治疗组行TACE治疗2周后再行RFA治疗1~2次。治疗后比较两组近期疗效及生存率。结果联合治疗组和单纯TACE组近期有效率分别为77.1%、51.7%,两组比较差异有统计学意义(P<0.05);联合治疗组1、2、3年生存率分别是79.7%、52.3%、23.0%,单纯TACE组1、2、3年生存率分别为59.4%、20.3%、6.8%,两组生存率比较差异有统计学意义(P均<0.05)。结论 TACE+RFA联合治疗PHC能有效控制肿瘤局部生长,延长患者生存期。
目的 探讨高脂血症对胰腺腺泡细胞三磷酸肌醇(IP3)介导的信号通路的影响.方法 将24只大鼠随机分为两组各12只,观察组建立高脂血症模型,予高脂饲料饮食,对照组予普通饲料饮食,4周后检测两组胰腺腺泡细胞内IP3水平及缩胆囊素(CCK)刺激的腺泡细胞淀粉酶释放率.结果 观察组胰腺腺泡细胞IP3水平、淀粉酶释放率均明显高于对照组,P均<0.01.结论 高脂血症大鼠胰腺腺泡细胞CCK/IP3信号通路发生变化,其信号分子IP3表达增高,腺泡细胞对CCK刺激的外分泌敏感性增加.
Objective This study was designed to examine the relationship between Fas-670 and FasL-844 gene polymorphism and the susceptibility to gastric cancer in North China.Methods Genomic DNA was extracted from peripheral blood leukocytes of 234 gastric cancer patients and 321 healthy volunteers.Genotypes at Fas-670 and FasL-844 polymorphic sites were identified by polymerase chain reaction-restriction fragment length polymorphism(PCR-PFLP).Results No significant difference was found between the two groups in the distribution of the polymorphism at Fas-670 and FasL-844(P0.05).A logistic regression analysis indicated that there was no correlation of Fas-670 and FasL-844 gene polymorphism with susceptibility to gastric cancer.After stratification by smoking status,it was found,however,that smokers carrying an Fas-670GA genotype had an high risk of gastric cancer compared with those with an Fas-670AA genotype(OR=2.438,95% CI 1.067~5.569).Conclusions In North China,the FasL-844 gene polymorphism has no correlation with the susceptibility to gastric cancer.While smokers with an FAS-670GA genotype are more likely to develop gastric cancer relative to those with an Fas-670AA genotype.
目的 FASL-844基因多态性与多种肿瘤易感性相关.本研究旨在探讨FASL-844基因多态性与食管癌易感性的关联.方法 提取204例食管癌患者及248例健康查体者外周血基因组DNA,用聚合酶链反应-限制性片段长度多态性法(PCR-RFLP)确定FASL-844基因型.结果 FASL-844 TT、TC、CC基因型在病例组和对照组中的分布频率分别为3.9%、45.6%、50.5%和4.4%、39.1%、56.5%,差异无统计学意义(P>0.05).Logistic回归分析表明,FASL-844基因多态性与食管癌发生风险无关.进一步分层分析显示,FASL-844基因多态性与食管癌发生风险仍无关联.结论 FASL-844基因多态性与河南安阳地区食管癌发生风险无相关性.
目的探讨Fas-1377基因多态性与胃癌易感性的关系。方法提取234例胃癌患者(观察组)及321例健康献血者(对照组)的外周血基因组DNA,用聚合酶链反应—限制性片段长度多态性法确定Fas-1377基因型。结果 Fas-1377AA、GA、GG基因型在观察组和对照组中的分布频率分别为13.25%、44.44%、42.31%和12.15%、47.04%、40.81%,组间比较无统计学差异(P>0.05)。Logistic回归分析表明,Fas-1377基因多态性与胃癌发生风险无关。结论 Fas-1377基因多态性与华北地区胃癌发生风险无明显相关性。
Objective: To investigate the effect of Bozhi injection on the cell-mediated immunity in patients.Methods: Eighty-four patients with condyloma acuminatum(CA) were confirmed by clinical characteristics,pathological examination and PCR.They were randomly divided into treatment group and Bozhi group.Twelve health persons were selected as controls.ELISA was carried out to detect the levels of serum IL-2 and IFN-γ.Results: After a month treatment,the serum IL-2 and IFN-γ in Bozhi group increased to normal level.Conclusion: Bozhi injection can increase serum level of IL-2 and IFN-γ and may have a potential role in reducing the recurrence of clinical CA.
OBJECTIVE:To evaluate the effects of antiprogestins mifepristone and lilopristone on proliferation and expressions of nuclear factor-kappa B (NF-kappaB) of ectopic stromal cells in vitro.METHODS:The ectopic stromal cells of ovary were cultured in vitro. Methyl thiazolyl tetrazolium method was used to evaluate proliferative activity of ectopic stromal cells. Cells were divided into five groups according to drug concentration: control group, group I and group II of mifepristone and of lilopristone. The expressions of NF-kappaB P65 and NF-kappaB P65 mRNA of ectopic stromal cells were determined by immunocytochemistry and in situ hybridization of cell slice.RESULTS:Antiprogestins mifepristone and lilopristone were able to significantly suppress the proliferation of ectopic stromal cells in a time- and dose-dependent manner in vitro. The expressions of NF-kappaB P65 and NF-kappaB P65 mRNA of ectopic stromal cells in the control group were higher than that of group I and group II. Their expressions in mifepristone groups were higher than that of lilopristone groups.CONCLUSIONS:Antiprogestin mifepristone and lilopristone could significantly suppress the proliferation of ectopic stromal cells in a time- and dose-dependent manner in vitro. The action mechanisms may be associated with the suppression of expression of NF-kappaB P65 mRNA and NF-kappaB P65.
Objective: To investigate the effects of Mifepristone and Tamoxifene on the expression of NF-κ B p65 mRNA, NF-κ B, PDGF-A and TGF-β of the adenomyosis. Methods: 46 cases of adenomyosis were randomly divided into 3 groups: Mifepristone group, Tamoxifene group, and the control group. Cytokines NF-κ B p65 mRNA was measured by ISH. The expression of NF-κ B, PDGF-A, and TGF-β was assayed by immunohistochemistry. Results: The expression intensity of NF-κ B p65 mRNA ,NF-κ B,PDGF-A and TGF-β was higher in control group, orderly decreased to Tamoxifene group, and then Mifepristone group, there was significant difference among them. The expression of NF-κ B was positive correlation with the expression of PDGF-A and TGF-β, respectively. Conclusions: Mifepristone and Tamoxifene could inhibit the production of NF-κ B p65 mRNA and NF-κ B in adenomyosis, and induced the down regulation of the expression of PDGF-A and TGF-β also.

探讨抗孕激素治疗子宫内膜异位症的作用机理.方法:分离、传代培养卵巢异位子宫内膜,培养液中分别加入不同浓度的米非司酮和利洛司酮.ELISA检测间质细胞培养上清液中表皮生长因子(EGF)和巨噬细胞集落生长因子(GM-CSF)浓度;RT-PCR检测异位间质细胞GM-CSF的表达.结果:培养72 h后,米非司酮组培养上清中EGF和GM-CSF的浓度显著高于利洛司酮组(P<0.05);异位间质细胞中GM-CSF mRNA电泳带光密度比值,米非司酮组也显著高于利洛司酮组(P<0.05);而两种抗孕激素的空白对照组EGF和GM-CSF及GM-CSF mRNA的表达均显著高于实验组的1 0-6 mol/L组和1 0-5 mol/L组(P<0.05).结论:米非司酮和利洛司酮均可降调节异位子宫内膜间质细胞生长因子EGF和GM-CSF的表达,抗孕激素可能通过此环节抑制异位子宫内膜间质细胞的生长.
