目的 考察大孔树脂纯化淫羊藿中朝藿定与淫羊藿苷的最佳工艺条件,并且对最佳工艺富集部位进行初步表征.方法 以朝藿定A1、A、B、C及淫羊藿苷为检测指标,采用静态吸附实验对5种大孔树脂进行筛选,通过动态吸附实验考察上柱液质量浓度、最大上样量、上样体积流量、水洗体积、除杂乙醇及洗脱乙醇体积分数、除杂乙醇及洗脱乙醇体积、径高比来优选最佳工艺条件,最后以UPLC-Q-TOF/MS、HPLC、紫外分光光度法对最佳工艺富集部位进行表征.结果 确定最佳大孔树脂型号为AB-8,柱径高比为1:7,以生药0.5 g/mL的淫羊藿上柱液上样6 BV,以6 BV/h的体积流量进行动态吸附,以5 BV水和5 BV 20%乙醇除杂,以6 BV 50%乙醇洗脱,除杂及洗脱体积流量均为6 BV/h.纯化后总黄酮质量分数为63.29%,朝藿定A1、A、B、C及淫羊藿苷总量为40.48%,朝藿定A1、A、B、C及淫羊藿苷质量分数分别为1.63%、2.52%、16.36%、5.51%、14.46%.结论 经验证实验,AB-8型大孔树脂纯化淫羊藿中朝藿定与淫羊藿苷的工艺稳定、合理、可行.经化学表征,富集部位主要为黄酮类成分,且黄酮类成分中以朝藿定与淫羊藿苷为主,表明最佳纯化工艺可以用于此类成分的纯化富集.
Objective To evaluate the protection effect of Hope I decoction on ozone inhalationinduced injury in rats. Methods 48 male SD rats were randomly divided into 6 groups,i. e.,control,model,VE( 0. 0135 g / kg),Hope I decoction at three dosages( 0. 045,0. 090,0. 180 g / kg respectively).Modeling rats were exposed to ozone in an exposure cabinet( 10 h / d × 45d),then some of them were intragastrically administrated with vitamin E or Hope I decoction. Then,the level of SOD,LPO,CAT and GSHPx in rat serum were detected and pathologic change of lung tissue were observed,Immunohistochemistry for surfactant protein A( SP-A) content were observed. Results The activity of SOD,CAT and GSH-Px in serum were increased and LPO level were reduced in all Hope I groups. Histopathological slides of these three groups also showed an attenuated extent of alveoli dilatation and collapse induced by ozone exposure,surfactant protein A of these three groups were significantly enhanced. Conclusion Hope I decoction hasgreat protection effect on ozone inhalation-induced injury in rats.
Objective: To study on the influence of gansui with different extracting manners in Gansui Banxia decoction on aldosterone and functions of kidneys on rat models of malignant ascites. Methods: 80 Wistar rats were divided into four groups: blank group, model group, Gansui Banxia Decoction(gansui decocted together with other herb), Gansui Banxia Decoction(gansui powder into the ready decoction). The walker-256 carcinosarcoma cells were injected intraperitoneally into the rats except blank group. Examine changes of aldosterone, renal function and morphology of rats. Results: The levels of ALD in each medication groups were significantly lower than in model group. Gansui Banxia decoction(gansui decocted together with other herbs) was with less nephrotoxicity. Conclusion: Gansui Banxia Decoction(gansui decocted together with other herbs) with less nephrotoxicity at the same time reduces the level of ALD significantly. The experiment also proves the theoretical basis for clinic.
目的:比较海藻玉壶汤(生甘草)组、海藻玉壶汤(炙甘草)组、去海藻(生甘草)组、去海藻(炙甘草)组、去甘草组、去海藻甘草组的小鼠急性毒性,评价海藻-甘草反药组合在复方中应用的毒性.方法:灌胃给与不同配伍组合不同剂量的海藻玉壶汤,计算出LD50,不符合LD50评价条件的组别,采用连续给药28 d,即累计半数致死量的方法来进行毒性评价;眼球采血,通过检测小鼠血清肝、肾功能相关指标评价其对小鼠肝、肾系统的影响.结果:各给药组均未得出LD50,海藻玉壶汤(生甘草)组LD50(16)为71.08 g·kg-1,药物剂量21 g·kg-1;海藻玉壶汤(炙甘草)组LD50(3)为68.28 g·kg-1,剂量21 g·kg-1;去甘草组LD5002)为55.78 g·kg-1,剂量20 g·kg-1;去海藻(生甘草)组LD50(22)为72.27 g·kg-,剂量21 g·kg-1;去海藻甘草组LD50(21)为79.62 g·kg-,剂量18.1 g·kg-1;去海藻(炙甘草)组小鼠连续给药28 d仍未达到半数致死量,最大耐受量为生药80 g·kg-,药物剂量20 g·kg-1;给药各组对小鼠的肝、肾脏功能均未见明显影响.结论:给药各组LD50(n)均属无毒的范畴,且对小鼠的肝、肾脏功能指标未见明显影响.
On the basis of single factor experiment,five influencing factors,namely extracting time,ethanol concentration,extraction temperature,material-liquid ratio and extraction times for extracting flavonoids from mung bean coat were established.Thus a L16(45) orthogonal experiment was applied to optimize the extraction conditions,using spectrophotometry to determine the total flavonoids at 510nm wavelength.Results: The obtained optimal extraction conditions are extracting time 150 min,ethanol concentration 50%,extraction temperature 80 ℃,material-liquid ratio 1∶10 and extraction times 2 times.Under the conditions the extraction rate of flavonoids is 3.879 mg/g,the average recovery is 100.84% and RSD is 0.18%.
[OBJECTIVE]To find out the effects of different trace element on the photosynthesis and secondary metabolites of Leonurus japonicus Houtt.;[METHOD]Pot experiments were carried out,different trace element were sprayed on the leaf during the developmental stage of medicines,and the leaf photosynthesis characteristids and contents of stachydrine of Leonurus japonicus Houtt.in different fertilization treatment were calculated and compared;[RESULTS]The result indicate that trace element of different concentration could facilitate the photosynthesis and stachydrine contents of Leonurus japonicus Houtt.in various degree.Among them,the effect of normal concentration of Mn,Zn and Fe were more distinguished,they elevate the stachydrine contents 96%,62%and 57%more than the control group.And stachydrine content were significant correlated with the max-photosynthesis;[CONCLUSION]It illustrate that trace element can impact the content of the end product by regulating the metabolism of physiology and the precursor and kinase of the secondary metabolite.
