目的 探讨芳香烃受体AHR在溃疡性结肠炎(UC)组织中的表达及其对IL-17信号效应调控的作用.方法 10例UC患者和10例正常的结肠组织切片进行免疫组化检测,分析AHR在UC中的变化体外细胞实验研究AHR对IL-17诱导的炎症因子产生的调控作用.结果 AHR蛋白在UC肠上皮细胞中的表达量显著下降;体外细胞实验发现活化AHR或者过表达AHR均能抑制IL-17介导的趋化因子CCL20、CXCL2和细胞因子IL-6的产生;相反,在Hela细胞中干扰AHR可以促进炎症相关因子CCL20、CXCL2和IL-6的产生.结论 AHR能负向调控IL-17介导的炎症因子的产生,同时其表达量与UC的发生呈负相关,提示AHR有可能通过负相调控IL-17介导的炎症效应,抑制UC的发生,发展.
目的 探讨雷公藤红素逆转乳腺癌细胞对多柔比星的耐药性及作用机制.方法 将多柔比星耐药MDA-MB-231细胞(MDA-MB-231/R细胞)分为对照组、雷公藤红素组、多柔比星组、雷公藤红素+多柔比星组和雷公藤红素+多柔比星+Bim siRNA组,MTT法检测MDA-MB-231/R细胞活力,流式细胞术检测各组MDA-MB-231/R细胞的凋亡,免疫共沉淀法检测MDA-MB-231/R细胞Bim蛋白与Bak及Bax蛋白的相互作用,Western blot检测MDA-MB-231/R细胞色素C释放和Caspase-3活化.结果 MDA-MB-231/R细胞对多柔比星的半数有效浓度(IC50)显著高于常规MDA-MB-231细胞[(18.4±1.1)g/mL比(1.5±0.1)g/mL,P<0.05];雷公藤红素处理后,多柔比星对MDA-MB-231/R细胞活力抑制率和凋亡诱导率均显著提高,与对照组和多柔比星组比较,差异有统计学意义[(57.3±4.1)%比0、(13.0±1.0)%,(36.5±2.3)%比(1.6±0.2)%、(8.8±0.6)%,P均<0.051;雷公藤红素处理能显著上调MDA-MB-231/R细胞Bim蛋白表达水平[(0.51±0.04)比(0.11±0.01)、(0.13±0.01),P均<0.05];用Bim siRNA阻断Bim蛋白上调后,雷公藤红素对多柔比星的协同抗肿瘤效应受到明显抑制[(20.5±2.1)%比(57.3±4.1)%,(12.4±1.1)%比(36.5±2.3)%,P<0.05];雷公藤红素处理后,与MDA-MB-231/R细胞Bim蛋白结合的Bak、Bax蛋白表达水平显著提高[(0.29±0.02)比(0.04±0.01)、(0.05-±0.01),(0.35±0.02)比(0.05±0.01)、(0.04±0.01),P均<0.05)];雷公藤红素处理后,多柔比星对MDA-MB-231/R细胞色素C的释放率和Caspase-3的活化率显著提高[(0.57±0.05)比(0.11±0.02)、(0.06±0.01),P均<0.05;(0.43±0.04)比(0.09±0.02)、(0.03±0.01),P均<0.05].结论 雷公藤红素可通过上调Bim蛋白表达,提高多柔比星耐药乳腺癌细胞对多柔比星的敏感性.
AIM:To detect the expression of matrix metalloproteinases-9(MMP-9)and tissue inhibitor of metalloproteinase-1(TIMP-1)in endometrium after caesarian section,and the change of blood leukocyte count,neutrophil percentage,serum C-reactive protein(CRP)level,and to explore the mechanism of Chonglou Shenghua Tang for preventing lochiorrhagia after caesarian section(CS).METHODS:The prospective study was designed as randomized and single blind research from Jan 2011to Jan 2013.60women underwent caesarian section(CS)indicated by obstetric factors were enrolled from the2ED clinical medical college of Zhejiang Chinese Medical University and assigned into three groups:group of Chonglou Shenghua Tang:20cases were administered by Chonglou Shenghua Tang 100mL,per 12h14dafter CS;group of Classical Shenghua Tang:20cases were administered by Classical Shenghua Tang 100mL,per12h14dafter CS and group of placebo:20cases were administered by placebo 100 mL,per12h14dafter CS.The expressions of MMP-9and TIMP-1in endometrium were determined using immunohistochemistry at 16th days after CS,and blood leukocyte and CRP were detected at 5th days and 16th days after CS.RESULTS:The levels of MMP-9expression decreased gradually in Chonglou Shenghua Tang group,Classical Shenghua Tang group and placebo group at16th days after CS,respectively,with statistical significance(P0.05).The levels of TIMP-9expression and the ratio of MMP-9/TIMP-1increased gradually in Chonglou Shenghua Tang group,Classical Shenghua Tang group and placebo group at 16 th days after CS,respectively,with statistical significance(P0.05).Blood leukocyte count,neutrophil percentage and CRP increased gradually in Chonglou Shenghua Tang group,Classical Shenghua Tang group and placebo group at 5th days and 16 th days after CS,respectively,with statistical significance(P0.05).CONCLUSION:These results suggestthat reduced expressions of MMP-9,and increased TIMP-1,and reduced the ratio MMP-9/TIMP-1in endometrium and reduced inflammatory indexes in the blood may play important roles in Chonglou Shenghua Tang preventing lochia after cesarean section.
