Backgrounds: Excessive pepsin can damage both normal laryngeal epithelial cells and laryngeal cancer (LC) cells. Heat shock protein 70 (HSP70) is closely related to pepsin. In this paper, we will explore the different significance of the regulatory role of HSP70 in endoplasmic reticulum stress (ERS) level in pepsin-treated laryngeal epithelial cells and LC cells. Methods: In cell experiments, laryngeal epithelial cells and LC cells were selected and induced by different concentrations of pepsin. Cell activity was detected by CCK8, cell apoptosis was detected by flow cytometry, and autophagy was detected by autophagy detection kit. The expression of ER)-related proteins was detected by immunofluorescence (IF) and Western blot. Cell transfection was used to inhibit HSP70 expression in both cells, and ERS, apoptosis, and autophagy were measured using related techniques. In animal experiments, a mouse model bearing LC was established. TUNEL assay detected apoptosis, autophagy kit detected autophagy, and ER-related protein expression was detected by Western blot. Results: HSP70 was increased in pepsin-stimulated laryngeal epithelial cells and LC cells, thereby inhibiting ER and ER-induced apoptosis and autophagy. Inhibition of HSP70 reduced the expression of glucose regulated protein 78 (GRP78) in pepsin-stimulated laryngeal epithelial cells and LC cells, and only inhibited downstream apoptosis-related pathways in laryngeal epithelial cells rather than in LC cells. Inhibition of HSP70 and ER could significantly promote apoptosis and inhibit tumor growth in the absence of pepsin stimulation in vivo. Conclusion: ER level regulated by HSP70 had different significance in laryngeal epithelial cells and LC cells treated with pepsin.
目的 分析免疫球蛋白(Ig)E高亲和力受体α段(FcεR1α)基因rs2427827位点的单核苷酸多态性(SNP)分布情况,并探讨其与过敏性鼻炎(AR)的关系.方法 采取病例对照研究方法,选择2019年1月至2020年7月于该院耳鼻咽喉头颈外科就诊并确诊为AR的151例患者为AR组,选取同期健康体检志愿者108例为对照组.采集所有受试者外周抗凝血3 mL并提取血液DNA,采用Mass ARRAY iPLEX GOLD分型技术检测两组FcεR1α基因rs2427827位点多态性;采用Logistic回归分析影响AR易感性的相关因素.结果 AR组FcεR1α基因rs2427827位点CC基因型频率为66.2%,尽管高于对照组的65.7%,但是差异无统计学意义(P>0.05).Logistic回归分析结果显示,突变杂合基因型TC(P=0.226,OR=2.15,95%CI:0.63~7.85)与突变纯合基因型CC(P=0.262,OR=1.97,95%CI:0.61~6.90)均与AR的发病无关.突变等位基因型C与野生等位基因型T进行比较,发现没有增加AR的患病风险,差异无统计学意义(P=0.603,OR=1.12,95%CI:0.72~1.75).结论 FcεR1α基因多态性与AR无明显相关性,但由于样本条件的限制,有必要扩大样本量和样本选择范围,以便进一步研究.
目的 探讨眶底及上颌窦前壁原始神经外胚层肿瘤/尤文氏肉瘤(ESFTs)这一罕见疾病的临床特点.方法 报道1例儿童ESFTs的临床特征、影像学表现、病理学特征,探讨其诊断、鉴别诊断、治疗及预后,并通过文献复习了解该病早期诊断要点.结果 该例患儿肿瘤病理上以小圆细胞为主要结构,细胞免疫组化为CD99的高表达.治疗方法 为手术切除加放化疗,患者术后随访21个月未出现肿瘤复发及远处转移征象.结论 头颈部肿瘤高度恶性肿瘤ESFTs影像学检查有一定的诊断意义,但是没有特异性,明确诊断主要依靠免疫组化检查.确诊后应尽可能早期手术,以免贻误治疗时机.预后较差,手术切除加放化疗能改善患者预后.
在新型冠状病毒肺炎疫情下,耳鼻咽喉-头颈外科(ORL-HNS)在门诊和急诊患者的诊治,实施ORL-HNS手术,以及术后管理和随访等环节面临新的挑战.文章结合最新的文献报道,针对以上问题深入思考,探讨并总结一部分临床应用经验,为ORL-HNS同道提供一些参考,以达到保护医护人员安全和保障患者健康的目的.
目的 分析鼻内镜手术治疗鼻乳头状瘤的方法及疗效.方法 回顾性分析2011年1月~2018年12月123例住院鼻内镜手术的鼻乳头状瘤临床资料.根据病变程度和范围,选择经典鼻内镜手术、鼻内镜中下鼻道联合开窗术、鼻内镜辅助下Caldwell-Luc手术、鼻内镜下泪前隐窝入路手术、Draf手术以及鼻内镜下辅助鼻侧切开术等术式.分析术后病理分型比例,病变累及部位、范围、术后复发率.结果 随访2~8年,中位随访时间29个月.病理分型比例为外翻性乳头状瘤32.5%(40例)、内翻性乳头状瘤61.8%(76例)、嗜酸性细胞乳头状瘤5.7%(7例).外翻性乳头状瘤未见复发病例,内翻性乳头状瘤复发率为13.2%(10例)、嗜酸性细胞乳头状瘤复发率为14.3%(1例).结论 针对鼻乳头状瘤的不同病理类型、病变范围、病变累及的部位,采用合适的鼻内镜下手术进路和方式,结合术后密切随访处理,能安全有效的切除病灶,降低复发率.
目的 探索经鼻内镜泪前隐窝进路上颌窦病变切除的手术方法、疗效及注意事项.方法 回顾性分析自2014年l月到2018年2月东部战区总医院耳鼻咽喉头颈外科住院行经鼻内镜泪前隐窝进路上颌窦病变切除手术的23例患者临床资料,包括一般情况、术中所见、术后症状、内镜检查结果及术后并发症情况.结果 23例经鼻内镜泪前隐窝进路上颌窦病变切除手术中,上颌窦内翻性乳头状瘤19例,Krouse分期均为T3期;上颌窦出血坏死性息肉4例.术后随访8~36个月,中位数随访时间l6个月,无严重并发症发生,无溢泪,无复发病例.术后症状均明显改善或消失.结论 经鼻内镜泪前隐窝进路上颌窦手术暴露良好、无明显死角,病变清除易彻底,鼻泪管及鼻腔结构和功能保留良好.
Histone deacetylase inhibitors (HDACis) -based therapeutic drug tolerance is one of the principal factors of poor prognosis of patients with nasopharyngeal cancer (NPC). Mechanisms of tolerance to HDACis are not well understood. Nowadays, dysregulation of long non-coding RNAs (LncRNAs) and microRNAs (miRNAs) has been reported to provide beneficial or inhibitory effects in drug-tolerance in various cancers. Herein, we established the HDAC inhibitor (SAHA)-tolerant NPC cell sublines, which had decreased apoptosis in response to SAHA treatment. We observed that the expression of miR-129 was significantly reduced in SAHA-tolerant NPC cells. Manipulating the expression of miR-129 overcame SAHA tolerance, and enhanced the SAHA-induced apoptosis. In terms of miR-129 downregulation, we identified that NEAT1 suppresses miR-129 expression. NEAT1 was found to be upregulated in SAHA tolerance cells. The depletion of NEAT1 phenocopied the effect of miR-129 overexpression, which also enhanced SAHA-induced apoptosis. Bcl-2 was the downstream target of miR-129 and contributed to SAHA tolerance in NPC. Our in vivo xenograft experiment confirmed that the administration of miR-129 or inhibition of Bcl-2 overcame the SAHA tolerance in NPC. In conclusion, NEAT1 increases in NPC tissues and manages to facilitate SAHA tolerance by modulating the miR-129/Bcl-2 axis, providing novel therapeutic targets for NPC treatment.
Objective It is to observe the curative effect of Banxia Houpu Decoction combined with Huangqi Jianzhong Decoction plus Omeprazole Magnesium Enteric-Coated Tablets on of paraesthesia pharyngis(spleen deficiency and liver de-pression syndrome),and its influence on hemorheology. Methods Fifty-six cases of patients with paraesthesia pharyngis (spleen deficiency and liver depression syndrome) were randomly divided into observation group and control group. The patients were treated by oral administration of Omeprazole Magnesium Enteric-Coated Tablets in the control group,and with Banxia Houpu Decoction and Huangqi Jianzhong Decoction on the basis of control group in the observation group. There were 4 weeks for 1 courses in the 2 groups. After 2 courses of treatment,the improvement of pharyngeal foreign body sensa-tion score,TCM symptom score,anxiety and depression scores, and hemorheology indexes were analyzed and compared, and the clinical efficacy of the 2 groups was evaluated. Results After treatment, the pharyngeal foreign body sensation score,TCM syndrome score,HADd and HADa score were significantly decreased(all P<0.05),and the improvement of each indexes in the observation group was significantly better than that of the control group (P<0.05). After treatment, the total blood viscosity,plasma viscosity,hematocrit and fibrinogen were significantly decreased in the 2 groups(all P<0.05),and the improvement of the observation group was significantly higher than that of the control group (P<0.05). After treatment,the total effective rate of the observation group was significantly higher than that of the control group(P<0.05). Conclusion Banxia Houpu Decoction and Huangqi Jianzhong Decoction plus Omeprazole Magnesium Enteric-Coa-ted Tablets are effective in the treatment of paraesthesia pharyngis (spleen deficiency and liver depression syndrome). It can effectively promote local microcirculation,improve the local pathophysiological state,it is worthy of clinical promotion to relieve the clinical symptoms of the patients.
Objective To investigate the expression change of inhibitory member of the ASPP family(iASPP) and its relationship with clinicopathological features and prognosis in the patients with nasopharyngeal carcinoma(NPC).Methods One hundred and thirty-three cases of nasopharyngeal carcinoma confirmed by pathology in the hospital were selected,at the same time 30 cases of nasopharyngeal chronic mucositis biopsy paraffin blocks previously collected in the pathology department served as the control group.The immunohistochemical staining was used to compare the expression of iASPP protein in the two groups and the relationship between the expression of iASPP with the clinicopathologic characteristics and prognosis in the patients with NPC was analyzed.Results The positive expression of iASPP protein in the nasopharyngeal carcinoma tissue was 72.18%,which was higher than 20.00% in the control group(P<0.05);the positive expression of iASPP protein in the nasopharyngeal carcinoma tissue had a certain relationship with the primary tumor range and lymph node metastasis(P<0.05);92 cases of nasopharyngeal carcinoma were followed up,including 63 cases of iASPP protein positive expression and 29 cases of iASPP protein negative expression,the 5-year survival rate in the patients with iASPP protein positive expression was 71.43%,which in the patients with iASPP protein negative expression was 86.21%,the difference was not statistically significant(P>0.05).Conclusion The iASPP expression in the nasopharyngeal carcinoma tissue is up-regulated,which is related to the lymph node metastasis and the primary tumor range,but its influence on the patient's prognosis is not significant.
