BACKGROUND: Vitamin D receptor (VDR) gene shows restriction fragment length polymorphism with incision enzyme Bsm Ⅰ ,Apa Ⅰ ,Taq Ⅰ ,which is related to bone mineral density (BMD).However, it is unclear that the relationship between VDR gene (Bsm Ⅰ ) polymorphisms and BMD,osteoporosis.OBJECTIVE: To analyze the distribution regularity of vitamin D receptor gene polymorphism related to BMD in postmenopausal women of Han,Uygur, Kazak and Mongoloid nationality in China. DESIGN: controlled observation.SETTING: Institute of Gerontology,General Hospital of Chinese PLA.PARTICIPANTS: Totally 179 women of Han,who were taking physical examination in General Hospital of Chinese PLA from January 2002 to December 2003, at the average age of (59±3) years,were selected. A total number of 122 women of Uygur with average age of 56.49 years; 63 women of Kazak with average age of (55±3) years; and 112 women of Mongoloid with average age of (57±3) years,who were all taking physical examination in department of geriatrics, Urumqi General Hospital of Lanzhou Military Area Command of Chinese PLA from January 2001 to December 2003.All of them were informed consent.METHODS: VDR genotypes(Bsm Ⅰ ) were defined with polymerase chain reaction-restriction fragment length polymorphism,so as to analyze distribution of Vitamin-D receptor gene (Bsm Ⅰ )polymorphisms of postmenopausal women in Han,Uygur, Kazak,Mongoloid nationality,and compared with the data of USA,Australia,France,Japan,Korea. Enumeration data were compared with Chi-square test.MAIN OUTCOME MEASURES: VDR (Bsm Ⅰ ) polymorphisms in healthy postmenopausal women from Han, Uygur, Kazak, Mongoloid populations in China, which were compared with the data of USA, Australia,France, Japan, Korea.RESULTS: For women of Han, Uygur, Kazak and Mongoloid nationality,the BB genotypes accounted for 0, 4.1%, 6.35% and 4.46%, the bb genotypes accounted for 90.5%, 69.67%, 38.1% and 50% respectively. There was a significant difference between women of Han, Uygur, Kazak, and Mongoloid nationality(P < 0.01). There was insignificant difference in comparation of distribtuion of VDR genotype between Kazak nationality and the west races, but it was significantly different to that in Japan,Korea races.CONCLUSION:VDR genotype polymorphisms is characterized by obvious racial diversify in postmenopausal women of Han,Uygur, Kazak,Mongoloid populations in China;Distribution of VDR gene frequency of Kazak population is similar with the west race ,but is different to Japanese and Korea's race.
?Objective To determine the distribution of vitamin D receptor gene polymorphism in women of Han,Uygur,Kazak and Mongoloid nationality in China.Methods Polymorphism defined by the Bsm1 restriction enzyme using polymerase chain reaction-restriction fragment length polymorphism in healthy postmenopausal women of 179 Han nationality,122 Uygur nationality,63 Kazak nationality and 112 Mongoloid nationality.The genotype frequencies of the VDR were calculated later using the Hardy-Weinberg equilibrium formula.Results For women of Han,Uygur,Kazak and Mongoloid national,the BB genotypes accounted for 0%,4.1%,6.35% and 4.46%,the bb genotypes accounted for 90.5%,69.67%,38.1% and 50% respectively.We found that was a significant difference between women of Han,Uygur,Kazak,and Mongoloid nationality.Compared to Caucasian population,there was no significant in VDR gene in Kazak nationality.Conclusions There was a significant difference between women of Han and Uygur,Kazak,and Mongoloid nationality in VDR genotype.The present study provide further evidence on the different pathogeny of osteoporosis in various ethnic groups.
目的研究我国维吾尔族妇女维生素D受体(VDR)基因多态性分布和骨密度、骨质疏松的关系.方法采用聚合酶链反应限制性片段长度多态性(PCR-RFLP)技术对30例维吾尔族妇女进行VDR基因分型.并进行双能X线骨密度(BMD)仪的检测.结果 VDR基因检出bb、Bb、BB,基因型分别为60%、33.3%、6.7%.bb、Bb、BB基因型的BMD分别降低为61%(11/18)、50%(5/10)、50%(1/2).结论就调查例数而言:中国维吾尔族妇女VDR基因多态性的分布频率以bb基因型最多,其次为Bb型,BB型仅占6.7%.BMD检测显示BMD降低以bb型较显著、Bb型次之.
维生素D受体(VDR)基因与骨密度(BMD)的关系各家报道不一.本研究对乌鲁木齐地区维吾尔、哈萨克、蒙古三个民族绝经后妇女进行VDR基因多态性研究,同时测定BMD,探讨骨质疏松与遗传的关系.
Polymorphyism of the vitamin D receptor (VDR) gene is reported to play a major role in variations of the genetic regulation of bone mass. The role of VDR polymorphisms within various ethnic populations is also undetermined. The purpose of the present study was to determine the frequencies of VDR genotypes in the Han and Kazak nationalities in China. We analyzed the polymorphism defined by the Bsm1 restriction enzyme using polymerase chain reaction–restriction fragment length polymorphism in 179 healthy postmenopausal Han women and 63 healthy postmenopausal Kazak women. The genotype frequencies of the VDR were calculated later using the Hardy–Weinberg equilibrium formula. The results showed that for women of Han nationality, the bb, Bb, and BB genotypes accounted for 90.5%, 9.5%, and 0%, respectively. In Kazak women, the respective frequencies were 38.1%, 55.56%, and 6.35%. We found that there was a significant difference between women of Han and Kazak nationality in terms of the frequency of distribution of VDR genotype (P < 0.001). There was a similar distribution of VDR genotypes in Kazak women to that seen for the Caucasian population of the USA. The results of the present study provide further evidence on the different pathogeny of osteoporosis in various ethnic groups.
AIM To explore the difference between Kazakh and the Han in the distribution of vitamin D receptor(VDR) gene polymorphism. METHODS VDR genotypes in 179 health postmenopausal women of the Han nationality (age:59±3 years),and in 93 those of the Kazakh (age: 50±2 years),and also in 17cases of osteoporosis patients were analyzes by polymerase chain reaction restriction fragment length polymorphism. RESULTS Frequencies of bb,Bb and BB genotype are 90%,10% and 0% in the the Han,and 47%,48% and 4% in the Kazakh respectively. A significant difference is found in the frequency distribution of VDR genotype between the Han and the Kazakh (P<0.005). CONCLUSION Han and Kazakh differ in the frequency distribution of VDR genotype.
目的研究我国乌鲁木齐维吾尔族妇女维生素D受体(VDR)基因多态性分布,探讨不同地区、不同民族之间基因型频率分布.为从VDR基因水平研究与骨质疏松的关系提供依据.方法采用聚合酶链反应(PCR)限制性片段长度多态性技术,对55例维吾尔族、179例汉族健康妇女进行VDR基因检测.结果维吾尔族55例中bb型40例(72.72%),Bb型12例(21.81%),BB型3例(5.45%),汉族179例,bb型162例(90.50%),Bb型17例(9.50%),未见BB型.结论中国维吾尔族VDR基因多态性分布与中国汉族比较,差异有显著性意义(P<0.05).
本文对20年中2805例住院死亡病例进行回顾分析,并进行前后10年死亡病因对照.重点显示死因顺位与年代变化规律、年龄、性别特征,旨在为疾病防治提供参考资料.
目的研究甲状腺素随增龄变化规律,探讨甲状腺素与衰老的关系.方法用TT3、TT4放免药盒,对286例45岁以上健康人群,进行TT3、TT4测定,人群按年龄分组,并进行组间对照,男女性别对照分析.结果女性TT3、TT4随增龄渐下降,男性TT3、TT4变化不明显.无论男性与女性均在60~69岁组出现明显下降.该年龄组与其它各年龄组比较(P<0.05).结论内分泌功能障碍是促进机体衰老的重要因素之一.60岁年龄组衰老加速,各种疾病发生率、死亡率增加可能与甲状腺素下降有关.此期适当调节内分泌平衡,可能对延缓衰老减少各种疾病的发生有一定作用.
